Modification of Acetobacter xylinum Bacterial Cellulose Using Dextransucrase and Alternansucrase

  • In addition to catalyzing the synthesis of glucan from sucrose as a primary reaction, glucansucrase also catalyzes the transfer of glucose from sucrose to other carbohydrates that are present or are added to the reaction digest. Using dextransucrase and altemansucrase, prepared from Leuconostoc mesenteroides B-742CBM and B-1355C, respectively, we modified the bacterial cellulose in Acetobacter xylinum ATCC10821 culture, and then produced a characteristic cellulose that is soluble and has a new structure. There were also some partially modified insoluble cellulose and oligosaccharides in the modification culture. After methylation and following acid hydrolysis of both the soluble and insoluble glucans, there were ($1{\rightarrow}4$) as well as ($1{\rightarrow}6$) and ($1{\rightarrow}3$) glycosidic linkages in the soluble glucan.

Purification and Characterization of Branching Specificity of a Novel Extracellular Amylolytic Enzyme from Marine Hyperthermophilic Rhodothermus marinus

  • An extracellular enzyme (RMEBE) possessing ${\alpha}-(1{\rightarrow}4)-(1{\rightarrow}6)$-transferring activity was purified to homogeneity from Rhodothermus marin us by combination of ammonium sulfate precipitation, Q-Sepharose ion-exchange, and Superdex-200 gel filtration chromatographies, and preparative native polyacrylamide gel electrophoresis. The purified enzyme had an optimum pH of 6.0 and was highly thermostable with a maximal activity at $80^{\circ}C$. Its half-life was determined to be 73.7 and 16.7 min at 80 and $85^{\circ}C$, respectively. The enzyme was also halophilic and highly halotolerant up to about 2M NaCl, with a maximal activity at 0.5M. The substrate specificity of RMEBE suggested that it possesses partial characteristics of both glucan branching enzyme and neopullulanase. RMEBE clearly produced branched glucans from amylose, with partial ${\alpha}-(1{\rightarrow}4)$-hydrolysis of amylose and starch. At the same time, it hydrolyzed pullulan partly to panose, and exhibited ${\alpha}-(1{\rightarrow}4)-(1{\rightarrow}6)$-transferase activity for small maltooligosaccharides, producing disproportionated ${\alpha}-(1{\rightarrow}6)$-branched maltooligosaccharides. The enzyme preferred maltopentaose and maltohexaose to smaller maltooligosaccharides for production of longer branched products. Thus, the results suggest that RMEBE might be applied for production of branched oligosaccharides from small maltodextrins at high temperature or even at high salinity.

Effect of Temperature on the Extraction of β-Glucan from Different Jeju Barley Varieties

  • The effect of different temperatures (45, 55, 65, and $75^{\circ}C$) on the extraction of ${\beta}$-glucan and the properties of extracted ${\beta}$-glucan were investigated with four different varieties of barley. Jeju naked barley, blue barley, beer barley, and black barley contained 6.85, 5.13, 3.58, and 4.16% of ${\beta}$-glucan, respectively. ${\beta}$-glucan in barley was extracted in the range of 64.88 to 93.84% depending on the extraction temperature and barley variety. The ${\beta}$-glucans in Jeju naked barley, Jeju blue barley, and black barley were optimally extracted at $65^{\circ}C$ for 3 h and Jeju beer barley at $75^{\circ}C$. The extracted ${\beta}$-glucan resolubilized to 43.48-81.73% and the ratio of ${\beta}(1{\rightarrow}3)$ to ${\beta}(1{\rightarrow}4)$ linkage was in the range of 1:3.8-5.8. These results suggest that purification and properties of ${\beta}$-glucan depend not only on the water extraction temperature, but also on the barley variety.

Cyclosophoraose as a Novel Chiral Stationary Phase for Enantioseparation

  • Cyclosophoraoses (Cys), cyclic ${\beta}-(1{\rightarrow}2)-D-glucans$ produced by Rhizobium meliloti 2011, were used as a novel chiral stationary phase for the enantiomeric separation. A novel Cys stationary phase, chemically immobilized onto porous silica via aminopropyltrimethoxysilane as a molecular linker, showed good separation for each racemate of bupivacain (separation factor, $\alpha$=1.3), propranolol ($\alpha$=1.3), and fenoprofen ($\alpha$=2.9), respectively, under the mobile phase of water: methanol (80:20, v/v) at a constant flow rate of 0.9 ml/min at pH7.

아가리쿠스로부터 분리한 $\beta$-glucan과 그 올리고당류의 HT-29 인체 대장암 세포에 대한 항암 활성에 관한 연구 (Study on the Anti-HT-29 Human Colon Cancer Activity of $\beta$-Glucans and Their Enzymatically Hydrolyzed Oligosaccharides from Agalicus blazei Murill)

  • 칡을 첨가하여 배양한 아가리쿠스버섯(Agaricus blaxei Murill)으로 부터 열수추출, gel filtration chromatography, DEAE ion exchange chromatography를 통하여 아가리쿠스 $\beta$-glucan (AG)을 추출하였다. 추출한 아가리쿠스 $\beta$-glucan에 Bacillus megaterium 유래의 endo-$\beta$-(1$\rightarrow$6)-glucanase를 처리하여 올리고당류(AO)를 얻었다. 이렇게 얻은 AG와 AO를 이용하여 HT-29 인체 대장암 세포에 대한 항암 활성을 조사한 결과, 암세포의 성장 억제 효과는 시료의 처리 농도에 의존적으로 증가하였으며, apoptosis assay에서 암세포의 apoptosis 유발이 농도에 의존적으로 증가되었다. 또한, 암세포의 세포 주기를 분석한 결과, apoptosis 발생을 뜻하는 G0 (sub-G1)기와 G1기의 비율이 증가한 반면 S기와 G2/M기는 대조군에 비해 감소되었다. 이러한 결과를 바탕으로 암세포의 apoptosis 증가에 대한 AO의 작용이 어떤 유전자와 연관이 있는지를 알아보기 위하여 cDNA microarray를 통해 유전자의 발현율을 검색한 결과, apoptosis의 내부 외부 경로에 영향을 주는 유전자(TNESE9, TNFRSF9, FADD, CASP8, BAD, CRADD, CASP9 등)의 발현이 증가되었고 세포 분열 주기의 진행과 관련된 유전자(CCND2와 CDK2)의 발현이 감소되었으며, 세포 분열 주기를 지체시키는 유전자 (CDKN2A)의 발현은 증가되었다. 또한, 사이토카인을 암호하는 유전자 (IL6, IL18, IL6R 등)와 tumor suppressor와 관련된 유전자 (CEACAM1, TP53BP2, IRF1및 PHB)의 발현이 2배 이상 증가된 것을 확인할 수 있었다. 따라서 HT-29 인체 대장암세포에 대한 AO의 성장 억제 작용은 G0/Gl기를 지체시켜 암세포 증식을 억제하고 apoptosis에 의해 암세포를 사멸시키는 항암 활성을 나타내는 것으로 확인되었으며, 특허 AO가 AG보다 현저한 활성을 보였다. 더 나아가 아가리쿠스 $\beta$-glucan (AG)과 올리고당류 (AO)는 항암 활성을 가진 대체 의약 소재로 개발될 수 있을 것으로 기대된다.

영지(Ganoderma lucidum) 균사체의 액체배양에 의한 세포외 항암활성 다당류의 구조분석 (Structural Analysis of the Antitumor Active Exo-polysaccharide Produced by Submerged Cultivation of Ganoderma lucidum Mycelium)

  • 영지 균사체의 액체 배양에 의하여 생산된 세포의 다당시료 분획 중 우수한 항암활성을 나타내었던 산성분획인 BWS-DA-GI의 구조를 균사체 유래의 산성 다당 분획(MWS-DA-GI)과 비교하면서 조사하였다. BWS-DA-GI의 구성성분을 검토한 결과, 다량의 glucose, mannose 및 galactose와 미량의 xylose 및 fucose를 함유하였으며, glucose, galactose 및 mannose의 몰비는 2.5:2.1:2.5이었다. 반면, 균사체 유래의 산성 다당 분획인 MWS-DA-GI는 다량의 galactose, fucose, mannose 및 glucose와 미량의 xylose를 함유하였다. 또 BWS-DA-GI의 구조를 균사체 유래의 다당 분획인 MWS-DA-GI와 비교하면서 과요오드 산화, Smith분해, affinity chromatography 및 메칠화 분석으로 조사한 결과, 두 시료 모두 ${\beta}-1,3-glucan$이었고, BWS-DA-GI는 1,6분지를 보유하고 있는 반면, MWS-DA-GI는 1,4 및 1,6분지를 함께 보유하는 것으로 추정되었으며, 분자량은 각각 1,200,000 및 1,000,000 dalton이었다.
