Biological Activities of Magnolia denudata Desr. Flower Extracts (목련(Magnolia denudata Desr.) 꽃 추출물의 생리활성)

  • The antioxidant, antiproliferation, and nitrate synthesis inhibitory effects of Magnolia denudata extracts (ME) were evaluated. The ME was extracted with 70% (v/v) ethanol and fractionated with solvents of hexane, chloroform, ethyl acetate, n-buthanol and aqueous. The ethyl acetate fraction contained the highest phenolic and flavonoid contents of 427.10 mg garlic acid eq/g and 356.05 mg catechin eq/g, respectively. The ethyl acetate fraction showed strong 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical scavenging activity with a 50% inhibition concentration ($IC_{50}$) of 0.20 mg/mL and total antioxidant activity was 0.90 mg AA eq/100 mg. From the results of cytotoxic effects of HCT116, NCL-H460, and HepG2 human cancer cells by MTT assay on the ME and its solvent fraction, chloroform fraction showed the highest cytotoxic effect ($IC_{50}$ value: 0.14, 0.37, and 0.41 mg/mL, respectively). Nitrate synthesis inhibitory effect of ME and its solvent fractions on nitric oxide synthase activity in LPS stimulated RAW 264.7 cells were decreased in dose-dependent manners, and $IC_{50}$ value of hexane and chloroform fractions were 0.39 and 0.49 mg/mL, respectively.

Antioxidant Compounds and Activities as well as Caffeine Content of Aronia melanocarpa Leaf Tea according to Pan-Roasting Conditions (아로니아잎차의 덖음조건에 따른 항산화 성분과 항산화 활성 및 카페인 함량)

  • Differences in bioactive compounds and antioxidant activities of aronia leaf (AL) extracts according to manufacturing conditions such as different number of pan-roasting and different temperatures were investigated. Both total polyphenolic compounds and total flavonoids contents were the highest in six time-pan-roasted AL tea extract ($37.96{\pm}0.48mg$ catechin equivalent/g and $19.96{\pm}0.44mg$ quercetin equivalent/g, respectively) among four tea samples. Antioxidative activities were also the highest in six time-pan-roasted AL tea extract with $IC_{50}$ 0.43 mg/mL and $IC_{50}$ 0.27 mg/mL based on DPPH and ABTS radical scavenging activities, respectively. HPLC analysis revealed that AL tea infusion did not have caffeine regardless of manufacturing conditions, whereas green tea infusion had 3.8 mg/g caffeine. Results demonstrated that AL tea can be expected as caffeine free leaf tea containing antioxidant benefits. Moreover, specific pan-roasting conditions of AL tea would be very important for its functional and sensory attributes.

Comparison of Nutritional Components and Physicochemical Properties of Small Colored Potatoes and Small Regular Potatoes (칼라꼬마감자와 일반꼬마감자의 영양성분 및 이화학적 특성)

  • This study examined the nutritional components and physicochemical properties of small colored potatoes and small regular potatoes as a natural health food source. To accomplish this, the general and antioxidative contents of small colored potatoes and small regular potatoes were measured. Total contents of carbohydrates, crude protein, crude lipid, and ash were 88.1%, 4.9%, 0.9%, and 6.4%, respectively. Small colored potatoes contained 76.5 kcal, while their total dietary fiber was 4.0%. Total proteins consisted of 17 different kinds of amino acids. Regarding their mineral contents, K was the most abundant mineral, followed by P, Mg, and Ca. Total phenol contents of the 70% ethanolic extracts of small colored potatoes were $48.2{\pm}1.2$ mg GAE/g. Total flavonoid contents of the 70% ethanolic extracts were $13.1{\pm}0.3$ mg RE/g. Overall, small colored potatoes had higher amounts of nutrients and physicochemical properties than small regular potatoes. The general nutrients and other antioxidant bioactive materials in small colored potatoes were also potential materials for good health food. It is expected that follow up studies of small colored potatoes through developing processed food and evaluation of their functional properties would provide useful information as a source of functional foods.

Antioxidant and Anticancer Activities of Artemisia annua L. and Determination of Functional Compounds (개똥쑥의 항산화 및 항암활성과 기능성 물질의 탐색)

  • The antioxidant and anticancer activities of leaves and stems from Gaeddongssuk (Artemisia annua L.) were investigated. The leaves and stems were extracted with water and 80% ethanol, respectively. The antioxidant and growth inhibition activities toward cancer cells by the 4 kinds of extracts were tested. In addition, phenolic compounds from A. annua were identified through quantitative analysis using HPLC. Antioxidant activities significantly increased in a does-dependent manner, and those of water and ethanol extracts of the leaves were stronger than those of the stems. Growth inhibition activities of the leaf extracts on HeLa and AGS cells were higher than those of the stem extracts. In particular, the ethanol extract of the leaves had growth inhibition activities of 61.07% and 57.24% against HeLa and AGS cells, at the concentration of $500\;{\mu}g$/mL, respectively, which were the highest among all the extracts. Phenolic acid and catechin contents of the A. annua extracts as determined by HPLC were higher in the leaves than in the stems. Flavonols were identified only in the leaves. The data suggest that the antioxidant and anticancer activities of A. annua extracts were due to phenolic compounds as well as unknown biological compounds in A. annua.

Effect of Heat-epimerized-catechin-mixture Rich in Gallocatechin-3-gallate on Skin Barrier Recovery (갈로카테킨-3-갈레이트가 풍부한 열전환 카테킨의 피부 장벽 회복에 대한 개선 효과)

  • Until now, (-)-epigallocatechin-3-gallate(EGCG) is known as the most powerful antioxidant among green tea catechins having many beneficial effects on human skin. Considering that the content of catechins is variable according to many conditions such as solvent, temperature and pressure, we prepared the heat-epimerized-EGCG-mixture (HE-EGCG-mix) containing high content of gallocatechin-3-gallate(GCG) by epimerization during autoclaving process and found out its optimal condition for maximizing conversion from EGCG to GCG. To investigate the effects of EGCG and HE-EGCG-mix on skin barrier function, we performed in vivo experiments with hairless mice. We found that HE-EGCG-mix has more potent stimulating activity than EGCG for the production of involucrin 7(INV7) and for recovery of barrier function in SKH-1 mice. Also, we found that GCG stimulates $PPAR-{\alpha}$ transactivation more effectively than EGCG in vitro by transient transfection assay for $PPAR-{\alpha}$ activation activity. These imply that HE-EGCG-mix consisting of high content of GCG should stimulate more efficiently recovery of skin barrier through PPAR-mediated-kerationocyte differentiation than EGCG. In conclusion, our study may provide a possibility that GCG, the C-2 epimer of EGCG, could be a potentially effective agent for development of new cosmetics or health foods for recovery of skin barrier.

Physical and Chemical Ingredients Components and Physiological Activity of Chungtaejeon and Green Tea Extracts (청태전과 녹차 추출물의 이화학적 성분과 생리 활성)

  • This study was carried out to help restore and spread broaden the consumption of Chungtaejeon, a our traditional tea, Chungtaejeon in Korea, and as well as to gather the basic data on Chutaejeon tea. We have also compared the various ingredients components and physiological activities of Chungtaejeon tea extracts with that of green tea extracts. According to the results, the total phenol contents in of the Chungtaejeon and green tea extracts were 90.56 and 98.70 mg/100 mL, respectively, and their tannin contents levels in Chungtaejeon and green tea extracts were 214.14 mg/100 mL and 259.60 mg/100 mL, respectively, showing a difference in content. The catechin contents of the Chetaejeon and green tea extracts were in the order of EGC($15.80{\sim}16.50\;mg/100mL$), EGCG($10.40{\sim}10.60\;mg/100 mL$), and ECG($9.55{\sim}9.88\;mg/100 mL$), however, with the exception of EC(7.86 mg/100 mL) in the green tea extracts, there were not significant differences between that in them Chetaejeon and green tea extracts. Quercetin was the main flavonoid ingredient component in those both the Chungtaejeon and green tea extracts was quercetin, at levels of 0.08 mg/100 mL and 0.06 mg/100 mL and its contents in Chungtaejeon and green tea extracts were 0.08 mg/100 mL and 0.06 mg/100 mL, respectively. The primary amino acid in both tea extracts was theanine main amino acid at levels of, theanine contents were most increased in 8.02 mg/100mL and 10.80 mg/100 mL, respectively, Chungtaejeon tea extracts by 8.02 mg/100 mL and green tea extracts by 10.80 mg/100 mL. And the arginine contents of the Chungtaejeon and green tea extracts in Chungtaejeon and green tea extracts were 1.78 mg/100 mL and 1.86 mg/100 mL, respectively. In terms of mineral composition, potassium content was highest contents in the Chungtaejeon and green tea extracts by 19.80 and 18.04 mg/100 mL, respectively. Finally, the Chungtaejeon tea extracts offered the greatest anti-oxidation and ACE inhibition activities were most increased in the Chungtaejeon tea extracts, and the green tea extracts showed the highest level of nitrite radical scavenging activity at 90% was extremely much increased in the green tea extracts by 90%.

Production of High-Level Polyphenol Powders from Young Grape Leaves (어린 포도 잎을 이용한 폴리페놀 고함유 분말 제조)

  • Polyphenols are naturally occurring antioxidants and play many important roles in human health. Grape leaves have not yet been reported to contain the antioxidants. In this study, the physicochemical properties of powders from young grape leaves, obtained after hot-air or vacuum freeze drying, were analyzed. Weight loss rangedfrom 76-80%, but no difference was found between samples prepared by hot-air orvacuum freeze drying. The L-, a-, and b- values of grape powder all fell upon hot-air drying. Quercetin, resveratrol, catechin, and epicatechin were detected in all cultivars. The contents of the four polyphenols were generally higher in powders obtained by vacuum freeze dryingthan in those prepared by hot-air drying, indicating that vacuum freeze drying is better for optimal preservation of polyphenols. None of three polyphenols assayed was found in some cultivars or after certain drying treatments. Based on physiochemical properties, our findings suggest that powders obtained after vacuum freeze drying are optimal for industrial use.

Effect of collection time on the chemical composition and levels of thiobarbituric acid reactive substance of Godulbaegi (Youngia sonchifolia M.) (채취시기에 따른 고들빼기의 성분 조성과 산화방지활성)

  • This study analyzes the chemical composition and thiobarbituric acid reactive substance levels of Godulbaegi (Youngia sonchifolia M.) depending on collection time. The moisture and crude fat content in leaf and root decreased, while crude fiber, crude protein, carbohydrate, and ash increased with increases in collection time. The mineral elements tended to increase in each sample with increases in collection time. The content of vitamin B increased as collection time increased. Vitamin C content was approximately five times higher in the leaves than that in the roots. Total amino acids in leaf and root increased considerably as collection time increased content of phenolic compounds in root were higher than that in the leaf and these contents increased. Antioxidant activity of Godulbaegi was higher in the root than in the leaf and increased as collection time increased.

Study on Biological Activities of Extracts for Cosmeceutical Development from Lagerstroemia indica L. Branch (화장품 개발을 위한 배롱나무(Lagerstroemia indica Linnaeus) 가지 추출물의 생리활성에 관한 연구)

  • The aim of the study was to examine the cosmetic and biological activity of Lagerstroemia indica L. and it is possible that can be used as a cosmetic ingredient for application of cosmetic industries. Lagerstroemia indica L. branch was extracted with 70% acetone in water. In the result of DPPH (1,1-diphenyl-2-picryl-hydrazyl) scavenging radical activity, acetone extract of Lagerstroemia indica L. branch were higher than 73% at the 50 ppm concentration. ABTS radical cation decolorization activity by acetone extract were higher than 78% at the 50 ppm. Both examine of DPPH and ABTS showed high antioxidative activities at the 50 ppm. In the result of nitrite scavenging ability, acetone extract were higher than 63% at the 50 ppm. Collagenase inhibition activity by extract were higher than 85% at the 50 ppm. Extract is showed high collagenase inhibition more than comparison group EGCG at all concentration. These results suggest that Lagerstroemia indica L. has a great potential as a cosmeceutical raw material as well as anti-oxidant and anti-inflammatory and collagenase inhibition activity.

Total Phenolics, Total Flavonoids, and Antioxidant Capacity in the Leaves, Bulbs, and Roots of Allium hookeri (삼채의 잎, 인경, 뿌리의 총페놀 함량, 총플라보노이드 함량 및 항산화능)

  • To quantitatively evaluate the total phenolics, total flavonoids, and antioxidant capacity in the leaves, bulbs, and roots of fresh Allium hookeri, they were extracted using various solvents including water, aqueous methanol (20, 40, 60, and 80%; v/v), and absolute methanol. The leaves had the highest levels of total phenolics (240.4-276.6 mg gallic acid equivalents/100 g) and total flavonoids (9.7-34.1 mg catechin equivalents/100 g). The highest antioxidant capacities of 78.7- 103.4 mg vitamin C equivalents (VCE)/100 g, 24.4-59.0 mg VCE/100 g, and 1,798.8-2,169.7 mg VCE/100 g in the leaves were also observed using 2,2'-azino-bis(3-ethylbenzothiazoline-6-sulfonic acid) (ABTS), 2,2-diphenyl-1-picrylhydrazyl (DPPH), and oxygen radical absorbance capacity (ORAC) assays, respectively. The total phenolics had a higher linear correlation with antioxidant capacity than the total flavonoids. In general, 60% (v/v) aqueous methanol extract had higher levels of total phenolics and flavonoids, and higher antioxidant capacity than any other solvents used. This study suggested that A. hookeri might be a good source of phenolics and antioxidants.
