New Species and Eight Undescribed Species Belonging to the Families Aspergillaceae and Trichocomaceae in Korea

  • During a survey of fungal diversity associated with insects, mud, soil, and freshwater niches in different areas in Korea, nine interesting fungal strains were isolated. Based on their morphological characteristics and molecular phylogeny analyses, using a combined data set of b-tubulin (BenA), calmodulin (CaM), and second largest subunit of RNA polymerase (RPB2) sequences, the strains CNUFC AM-44, CNUFC JCW3-4, CNUFC S708, CNUFC WT202, CNUFC AS1-29, CNUFC JCW3-5, CNUFC JDP37, and CNUFC JDP62 were identified as Aspergillus alabamensis, A. floridensis, A. subversicolor, Penicillium flavigenum, P. laevigatum, P. lenticrescens, Talaromyces adpressus, and T. beijingensis, respectively. The strain CNUFC JT1301 belongs to series Westlingiorum in section Citrina and is phylogenetically related to P. manginii. However, slow growth when cultivated on CYA, MEA, CREA is observed and the property can be used to easily distinguish the new species from these species. Additionally, P. manginii is known to produce sclerotia, while CNUFC JT1301 strain does not. Herein, the new fungal species is proposed as P. aquadulcis sp. nov. Eight species, A. alabamensis, A. floridensis, A. subversicolor, P. flavigenum, P. laevigatum, P. lenticrescens, T. adpressus, and T. beijingensis, have not been previously reported in Korea. The present study expands the known distribution of fungal species belonging to the families Aspergillaceae and Trichocomaceae in Korea.

Identification of 12 radiation-resistant bacterial species in the phylum Proteobacteria new to Korea

  • In 2019, after a comprehensive investigation of indigenous prokaryotic species in Korea, a total of 12 bacterial strains assigned to the phylum Proteobacteria were isolated from soil. With the high 16S rRNA gene sequence similarity (>98.8%) and formation of a robust phylogenetic clade with the closest species, it was determined that each strain belonged to independent, predefined bacterial species. This study identified two species in the family Burkholderiaceae, one species in the family Comamonadaceae, two species in the family Oxalobacteraceae, one species in the family Micrococcaceae, one species in the family Bradyrhizobiaceae, one species in the family Methylobacteriaceae, one species in the family Rhizobiaceae, one species in the family Rhodocyclaceae, and one species in the family Sphingomonadaceae. There is no official report about these 12 species in Korea, so are described as unreported bacterial species in Korea in this study. Gram reaction, basic biochemical characteristics, colony, and cell morphology are also described in the species description section.

A report of nine unrecorded bacterial species in the phylum Bacteroidetes collected from freshwater environments in Korea

  • During a comprehensive study of indigenous prokaryotic species in South Korea, nine bacterial species in the phylum Bacteroidetes were isolated from freshwater environmental samples that were collected from three major rivers in the Republic of Korea. High 16S rRNA gene sequence similarity (${\geq}98.7%$) and robust phylogenetic clades with the closely related species suggest that each strain was correctly assigned to an independent and predefined bacterial species. There were no previous reports of these nine species in Korea. Within the phylum Bacteroidetes, four species were assigned to the genus Flavobacterium, order Flavobacteriales, and five species to three genera of two families in the order Cytophagales. Gram reaction, colony and cell morphology, basic biochemical characteristics, isolation source, and strain IDs are described in the species description section.

Analysis of Seasonal Water Pollution According to the Reuse and Treatment of Wastewater from Anyang Stream (안양천 하수처리 재이용에 따른 계절별 수질오염 분석)

  • Anyang Stream Restoration Movement was started from 2001 for the construction of ecological city. The facilities for reuse of treated sewerage have been used since 2003 for improvement of water quality, maintenance of water quantity, river ecological restoration, and hydrophilic space. Thus, the Anyang city has been gradually transformed to eco-friendly city after the construction of Anyang stream and Hakui stream as natural rivers. In this study, biological and chemical methods as well as ecological indicators for Anyang mainstream and major tributaries were monitored for four years in between 2008 and 2012. The water quality and the diversity of species in most of the streams were found to be good except Anyang main stream. It appears that the influence of seasonal drying stream is almost disappeared except Sammak stream. Thus, the values for BOD (Biochemical Oxygen Demand) and BIP (Biological Index of Pollution) for Anyang main stream were found to be 5.27~3.42mg/l and 4.51~5.50, respectively. This is considered to be caused by the reused water quality of treated wastewater being exceeded the design criteria or by the non-point source of contaminants around the stream. However, entire section of tributaries shows I, II grade as a good water quality.

A report of unrecorded bacterial species of Korea isolated in 2016, belonging to the family Deinococcaceae and Planctomycetaceae

  • In 2016, as part of a larger effort to discover indigenous prokaryotic species in Korea, we isolated the family Deinococcaceae and Planctomycetaceae as unrecorded bacterial species. From the high 16S rRNA gene sequence similarity (>98.5%) and formation of a robust phylogenetic clade with known species, it was determined that each strain was a distinct bacterial species. There are no official reports that these two species have been described in Korea; therefore, the bacterial strains of Deinococcus and Blastopirellula are described for the first time in Korea. Gram reaction, colony and cell morphology, basic biochemical characteristics, and isolation sources are also described in the species description section.

Development of a Novel Bioreactor System for the Treatment of Gaseous Benzene

  • A novel, continuous bioreactor system combining a bubble column (absorption section) and a two-phase bioreactor (degradation section) has been designed to treat a gas stream containing benzene. The bubble column contained hexadecane as an absorbent for benzene, and was systemically chosen considering physical, biological, environmental, operational and economic factors. This solvent has infinite solubility for benzene and very low volatility. After absorbing benzene in the bubble column, the hexadecane served as the organic phase of the two-phase partitioning bioreactor, transferring benzene into the aqueous phase where it was degraded by Alcaligenes xylosoxidans Y234. The hexadecane was then continuously recirculated back to the absorber section for the removal of additional benzene. All mass transfer and biodegradation characteristics in this system were investigated prior to operation of the integrated unit, and these included: the mass transfer rate of benzene in the absorption column, the mass transfer rate of benzene from the organic phase into the aqueous phase in the two-phase bioreactor, the stripping rate of benzene out of the two-phase bioreactor, etc. All of these parameters were incorporated into model equations, which were used to investigate the effects of operating conditions on the performance of the system. Several experiments were conducted to show the feasibility of this system. This process is believed to be very practical for the treatment of high concentrations of gaseous pollutants.

Analysis on Urinary N-methylformamide of Korean Workers according to Industrial Classification and Countermeasures for Exposure Control of N,N-dimethylformamide (우리나라 근로자들의 업종별 뇨중 N-methylformamide 분석 및 N,N-dimethylformamide 노출관리 대책)

  • Objectives: This study is aimed to describe the current situation about urinary biomarker N-methylformamide(NMF) for workers exposed to N,N-dimethylformamide(DMF) according to industrial classification. Materials: Special health examination records of the workers who had undergone urinary biological monitoring in 2013 were collected. The numbers and percentage of workers, whose urinary NMF values were above the limit of detection(LOD) and above the biological exposure index(BEI) were calculated. Health relatedness with DMF as judged by their doctors was also described. All description was classified according to the $9^{th}$ Korean Standard Industrial Classification(KSIC). Results: It appeared that most workers exposed to DMF belong to manufacturing section(80.7%). The geometric mean(GM) values of urinary NMF were 6.25 mg/L, 3.54, and 3.86 for the manufacturing section, professional, scientific and technical activities section, and for the construction section respectively. In detail, it seemed that division of textiles(except apparel) (GM 7.51 mg/L), division of leather, luggage and footwear(11.59 mg/L), and division of rubber and plastic products(6.89 mg/L) were highly exposed to DMF with a high percentage of workers with urinary NMF values above BEI. This was probably due to the effect of skin absorption that the division of leather, luggage and footwear showed the highest urine NMF GM. Conclusions: It seemed that workers in manufacture industries such as textile, leather, luggage, footwear, rubber and plastic products were highly exposed to DMF. So, efforts should be focused on those industries in order to effectively diminish worker's exposure. Further studies to compare DMF air-monitoring with bio-monitoring according to industrial classification should be considered.

A Study on the Optimal Design in the Indirect Aerated Fluidized Bed (간접 포기식 유동상의 이상적 설계에 관한 연구)

  • Process intensification without any increase in bed requires the exploitation of fluid mechanical phenomena as the basis for elegant solutions to the process engineering problems which result from the need to retain and control the immobilized biomass, and for biomass recovery. The fluidized bed biological reactor provides a solution to these needs. The wastewater treatment characteristics of the fluidized bed was filled with sand media. Indirect aeration were studied experimentally. The researcher was filled with sand particle size(0.60~0.42mm) in three reactors with different section area(A)/height(H), in the state BOD loading 4.5kg-$BOD_5/m^3$ㆍd, and under the fixed state of hydraulic retention time for around 32 minutes.

Fish Compositions and Distribution Characteristics in the Upstream and Downstream Region of Seungchon Weir (승촌보의 상류구간 및 하류구간에서 어류 종 조성 및 분포특성)

  • Seungchon Weir was constructed in 2010 on the downstream region of Yeongsan River watershed for four major river projects. The purpose of our study was to determine whether fish compositions and distributions differ between the upper ($U_w$) and lower regions ($L_w$) of Seungchon Weir during May - December 2011 along with hydroacoustic fish surveys using Eco-Sounder monitoring ($E_SM$) technique. Total number of fish species sampled by conventional fish sampling at all sites was 29, and 21 and 24 species occurred in the $U_w$ and $L_w$, respectively. Most dominant species was Hemiculter eigenmanni (43%) in the $U_w$ and Opsaiichthys uncirostris amurensis (21%) in the $L_w$, indicating a distinct differences of the species composition. According to hydroacoustic approach of ESM, fish density was significantly higher (p < 0.05) in the $L_w$ section of 250 m than any other sections, This outcome indicates that fishes moving toward the upstream were blocked by weir construction. According to analysis of tolerance guilds, tolerant species (TS) were made of 88% and 79% in the $U_w$ and $L_w$ region, respectively, while the proportions of sensitive species were < 0.1% of the total in the both regions. Trophic guild analysis showed that the proportion of omnivore species were 58% and 40% in the $U_w$ and $L_w$ region, respectively, while insectivores were made of 16% and 21% in the $U_w$ and $L_w$ region, respectively. Both analyses of tolerance guilds and trophic guilds indicate the biological degradation of the waterbody. Fish community structure analysis, based on species diversity, dominance, and evenness indices, showed higher community stability in the $L_w$ region than $U_w$ region. Exotic species of largemouth bass were largely increased in this survey (14%), compared to the past (< 1%), indicating a modification of fish trophic structure.

  • The objectives of this research were to analyse the microhabitat of Cobitis choii which is designated as an endangered fish species (I) and national monument species in Korea (No. 454), and provide valuable information of suitable replacement habitat in the future for a conservation of the population with rapid decreases by environmental pollution. Sampling and microhabitat analysis in three streams such as Baekgok, Yugu and Gap Stream, known as one of the least habitats in Korea showed that the mean number of Cobitis choii observed was 2.6. This result indicated that the richness was too low, so the species conservation was very urgent. Optimal physical microhabitat of the population was determined as environmental conditions with > 60% sand with 1 mm particle size, optimal water depth of 20 - 60 cm in the habitats, and the optimal current velocity of < 0.4 m/s. Under the circumstances of the microhabitat, optimal water volume (discharge) was 0 - 2 m3/s in the each sectional analysis and this reach was mainly composed of the stream section with intermittant slow runs and pools. These microhabitats were largely disturbed by physical modifications of habitat and chemical pollutions due to direct influences of nutrient-rich water inputs from the urban area and intensive agricultural pollutants. For these reasons, optimal habitat replacement are required in the future for the conservation of the species.
