In vitro Evidence that Purified Yeast Rad27 and Dna2 are not Stably Associated with Each Other Suggests that an Additional Protein(s) is Required for a Complex Formation

  • The saccharomyces cerevisiae Rad27, a structure-specific endonuclease for the okazaski fragment maturation has been known to interact genetically and biochemically with Dna2, an essential enzyme for DNA replication. In an attempt to define the significance of the interaction between the two enzymes, we expressed and purified both Dna2 and Rad27 proteins. In this report, Rad27 could not form a complex with Dna2 in the three different analyses. The analyses included glycerol gradient sedimentation, protein-column chromatography, and coinfection of baculoviruses followed by affinity purification. This is in striking contrast to the previous results that used crude extracts. These results suggest that the interaction between the two proteins is not sufficiently stable or indirect, and thus requires an additional protein(s) in order for Rad27 and Dna2 to form a stable physical complex. This result is consistent with our genetic findings that Schizosaccharomyces pombe Dna2 is capable of interacting with several proteins that include two subunits of polymerase $\delta$, DNA ligase I, as well as Fen-1. In addition, we found that the N-terminal modification of Rad27 abolished its enzymatic activity. Thus, as suspected, we found that on the basis of the structure determination, N-terminal methionine indeed plays an important role in the nucleolytic cleavage reaction.

THE CANONICAL FORM OF INVOLUTARY FUZZY MATRICES

  • We study special types of matrices. The involutary fuzzy matrices are important in various applications and have many interesting properties. Using the graphical method, we have the zero patterns of involutary fuzzy matrix, that is, involutary Boolean matrices. And we give the construction of all involutary fuzzy matrices for some dimensions and suggest the canonical form of involutary fuzzy matrix.

New Crystal form of Valsartan Dipotassium Salt (발사르탄 이칼륨염의 신규 결정형)

  • A new crystal form of valsartan dipotassium was isolated by recrystallization using the one-pot method. The new crystal form was identified as a monohydrate form ($C_{24}H_{27}N_5O_3K_2.H_2O$) and characterized by diffential scanning calorimetry (DSC), thermogravimetric analysis (TGA) and X-ray powder diffractometry (PXRD). The new crystal data demonstrated to be clearly different from those known for the tetrahydrate form ($C_{24}H_{27}N_5O_3K_2.4H_2O$). It was observed that the monohydrate of vasartan dipotassium salt was completely dissolved in water within 1 hour and its dissolution rate was much faster than anhydrous free form of valsartan.

  • In this paper, we construct a Sasakian manifold by the product of real line and Kählerian manifold with exact Kähler form. This result demonstrates the close relation between Sasakian and Kählerian manifold with exact Kähler form. We present an example and an open problem.

  • In order to study the propagation of singularities for solutions to second order quasilinear strictly hyperbolic equations with boundary, we have to consider the regularity of solutions of first order nonlinear equations satisfied by a characteristic hyper-surface. In this paper, we study the regularity compositions of the form v(${\varphi}$(x), x) with v and ${\varphi}$ assumed to have limited Sobolev regularities and we use it to prove the regularity of solutions of the first order nonlinear equations.

  • I consider the optimal consumption and portfolio selection problem with nonnegative wealth constraints using the dynamic programming approach. I use the constant relative risk aversion (CRRA) utility function and disutility to derive the closed-form solutions.

  • I use the dynamic programming approach to study the optimal consumption and investment problem with regime-switching and constant labor income. I derive the optimal solutions in closed-form with constant absolute risk aversion (CARA) utility and constant disutility.

Current situation of porcine reproductive and respiratory syndrome in Korea (한국의 돼지생식기호흡기증후군(PRRS) 발생경향)

  • The 2,078 blood samples from 148 swine farms were collected and tested by IFA for porcine reproductive and respiratory syndrome(PRRS) virus antibody to know what type of PRRS prevails by the area. Clinically reproductive form of PRRS occurred in swine farms of 3/27, 3/87, and 2/34 in eastern, central and western areas, but the seroprevalence of those areas was different as 6.5%, 23.3%, and 17.6%, respectively. However, respiratory form of PRRS occurred more frequently, and the number of farms manifested with the respiratory form of PRRS in the eastern, central and western areas was 22/27, 71/87, and 30/34, respectively. The seroprevalence of that form of PRRS in the eastern, central and western areas was 52.2%, 67.1%, and 51.6%, respectively. Subsequently mixed form of PRRS occurred more frequently in the central area and the number of farms of eastern, central and western areas was 2/27, 13/87, and 2/34, respectively. The PRRS seroprevalence of the eastern, central and western areas was 58.6%, 54.0%, and 19.2%, respectively. Collectively the PRRS seroprevalence of eastern, central and western areas was 43.8%, 59.3%, and 38.2%, respectively. Overall seroprevalence of PRRS in Korea was 51.8%. In conclusion, the reproductive or the respiratory form of PRRS has been still in trouble in the Korean swine industry and PRRS control measures have to be taken in consideration.
