Differences in isolates of Tomato yellow leaf curl virus in tomato fields located in Daejeon and Chungcheongnam-do between 2017 and 2018

  • To follow up on a 2017 survey of tomato virus diseases, samples with virus-like symptoms were collected from the same areas (Buyeo-gun, Chungchungnam-Do and Daejeon, Korea) in 2018. While in 2017 mixed infections of Tomato mosaic virus with either Tomato yellow leaf curl virus (TYLCV) or Tomato chlorosis virus were detected, only TYLCV was detected in symptomatic samples in 2018. TYLCV amplicons of c.777 bp representing the coat protein (CP) coding region were cloned from the TYLCV positive samples, and the sequence data showed a 97.17% to 98.84% nucleotide and 98.45% to 99.22% amino acid identity with the 2017 Buyeo-gun isolate (MG787542), which had the highest amino acid (aa) sequence identity of up to 99.2% with four 2018 Buyeo-gun sequences (MK521830, MK521833, MK521834, and MK521835). The lowest aa sequence identity of 98.45% was found in a 2018 Daejeon isolate (MK521836); the distance between Buyeo-gun and Daejeon is about 45 km. Phylogenetic analysis indicated that the currently reported CP sequences are most closely related to Korean sequences from Masan (HM130912), Goseong (JN680149), Busan (GQ141873), Boseong (GU325634), and the 2017 isolate TYLCV-N (MG787543) in the 'Japan' cluster of TYLCV isolates and distinct from the 'China' cluster isolates from Nonsan (GU325632), Jeonju (HM130913) and Jeju (GU325633, HM130914). Our survey data from 2017 and 2018 suggest that TYLCV has become established in Korea and may be spread by whitefly vectors from weed reservoirs within the farm environment.

Suggestion of Product Planning Process for Small and Medium Sized Design Company : Focused on the Case of Baby Bathtub Design Concept Development (중소 디자인 기업을 위한 제품 기획 프로세스 제안 : 유아욕조 디자인 콘셉트 개발 사례를 중심으로)

  • A number of small and medium design companies in Korea are making efforts to develop distinguished products with an aim to survive and prosper. However, it is quite difficult to succeed due to insufficient experience in product planning and the challenges in applying known methodologies, which are based on a large amount of data presented as best practices in designing process, in the actual small and medium enterprise operations. To this end, this study suggested the usefulness of the user participation process as the methodology for small and medium design companies and chose the user FGD method implemented by Company P which is a small design company as an empirical case study. The following are the processes used in the case study; First, the problems of existing baby bath were derived through user FGD. Second, opinions were collected from various classes of users through in-depth interviews. Third, the ideas derived were analyzed with the KJ method and grouped based on similar elements, through which six design directions and detailed design concepts covering size, material, safety, purchase factors of existing product, direction of improvement, additional elements were derived. Through the case study, this study verified that the FGD method of Company P could improve the practical verification, integration and promptness of the product planning process in small and medium enterprises. This is valuable as a realistic process that small and medium enterprises with limited capital and manpower may adopt.

  • This article considered generally gradational change aspect, character of community for the tombs of the Three kingdoms prepared by some certain homogenous groups. It was completed by tracing the transfering process to grasp character of social structure and organizing System in the community. For this reason, it's analyzed that Wooden chamber tomb was formed in some certain Time-Space of the Yaksadong Buk-dong archaeological site in the Three Kingdom Period. Constructed group was distinguished by the five classes. So perhaps scale of Wooden chamber and sort, material of burial relics, the top layer and the upper layer were established two status those are active the ruling class led the Wooden chamber tomb's society, the general public hold a large majority from the middle class to the lowest class. And it depends on age(nonage-adult) that differed in relation of arrangement, character. It's changed with function or roles, property, purpose etc. of social community in accordance with these individual position, age. Received signification, value of social status were considered with social worth, faith. Members in a community had been conducted under premeditated, organised system for common target, relatedness. Also it seems to attempt organization's own maintenance, persistiveness in norms that given at common law. To do that, relation for combination between members who consisted systems had to be organic. The relation for combination was considered to form relation for the much larger community in based on blood relationship that most vital part. One social structure had established family(house, household) community in based on individual blood relationship, family communities gather up to develop into relative(blood relative, same race) community gradually. Furthermore, relative(blood relative, same race) communities, that focus on the public good gather up to develop into social community in based on regionalism. It's considered to go through the Development Process of family(house, household) community - relative(blood relative, same race) community - social community.

Exosomal miR-181b-5p Downregulation in Ascites Serves as a Potential Diagnostic Biomarker for Gastric Cancer-associated Malignant Ascites

  • Purpose: Peritoneal carcinomatosis in gastric cancer (GC) patients results in extremely poor prognosis. Malignant ascites samples are the most appropriate biological material to use to evaluate biomarkers for peritoneal carcinomatosis. This study identified exosomal MicroRNAs (miRNAs) differently expressed between benign liver cirrhosis-associated ascites (LC-ascites) and malignant gastric cancer-associated ascites (GC-ascites), and validated their role as diagnostic biomarkers for GC-ascites. Materials and Methods: Total RNA was extracted from exosomes isolated from 165 ascites samples (73 LC-ascites and 92 GC-ascites). Initially, microarrays were used to screen the expression levels of 2,006 miRNAs in the discovery cohort (n=22). Subsequently, quantitative reverse transcriptase-polymerase chain reaction (qRT-PCR) analyses were performed to validate the expression levels of selected exosomal miRNAs in the training (n=70) and validation (n=73) cohorts. Furthermore, carcinoembryonic antigen (CEA) levels were determined in ascites samples. Results: The miR-574-3p, miR-181b-5p, miR-4481, and miR-181d were significantly downregulated in the GC-ascites samples compared to the LC-ascites samples, and miR-181b-5p showed the best diagnostic performance for GC-ascites (area under the curve [AUC]=0.798 and 0.846 for the training and validation cohorts, respectively). The diagnostic performance of CEA for GC-ascites was improved by the combined analysis of miR-181b-5p and CEA (AUC=0.981 and 0.946 for the training and validation cohorts, respectively). Conclusions: We identified exosomal miRNAs capable of distinguishing between non-malignant and GC-ascites, showing that the combined use of miR-181b-5p and CEA could improve diagnosis.

Expression and secretion of CXCL12 are enhanced in autosomal dominant polycystic kidney disease

  • Autosomal dominant polycystic kidney disease (ADPKD), one of the most common human monogenic diseases (frequency of 1/1000-1/400), is characterized by numerous fluid-filled renal cysts (RCs). Inactivation of the PKD1 or PKD2 gene by germline and somatic mutations is necessary for cyst formation in ADPKD. To mechanistically understand cyst formation and growth, we isolated RCs from Korean patients with ADPKD and immortalized them with human telomerase reverse transcriptase (hTERT). Three hTERT-immortalized RC cell lines were characterized as proximal epithelial cells with germline and somatic PKD1 mutations. Thus, we first established hTERT-immortalized proximal cyst cells with somatic PKD1 mutations. Through transcriptome sequencing and Gene Ontology (GO) analysis, we found that upregulated genes were related to cell division and that downregulated genes were related to cell differentiation. We wondered whether the upregulated gene for the chemokine CXCL12 is related to the mTOR signaling pathway in cyst growth in ADPKD. CXCL12 mRNA expression and secretion were increased in RC cell lines. We then examined CXCL12 levels in RC fluids from patients with ADPKD and found increased CXCL12 levels. The CXCL12 receptor CXC chemokine receptor 4 (CXCR4) was upregulated, and the mTOR signaling pathway, which is downstream of the CXCL12/CXCR4 axis, was activated in ADPKD kidney tissue. To confirm activation of the mTOR signaling pathway by CXCL12 via CXCR4, we treated the RC cell lines with recombinant CXCL12 and the CXCR4 antagonist AMD3100; CXCL12 induced the mTOR signaling pathway, but the CXCR4 antagonist AMD3100 blocked the mTOR signaling pathway. Taken together, these results suggest that enhanced CXCL12 in RC fluids activates the mTOR signaling pathway via CXCR4 in ADPKD cyst growth.

Practical Propagation Methods for Production of Prothalli and Sporophytes in Deparia pycnosora (Christ) M. Kato

  • Deparia pycnosora (Christ) M. Kato is a fern used as ornamental plant. In addition, it is called "Teol-go-sa-ri" in Korean name. The aim of this study was to develop a practical propagation method of D. pycnosora using tissue culture technique. Prothallus obtained from spore germination was the used as experiment materials. The prothalli (300 mg) used in all experiments were sub-cultured for 8-week intervals. The most suitable media for prothallus propagation were identified by culturing 300 mg of prothalli in $1/4{\times}$, $1/2{\times}$, $1{\times}$, $2{\times}$ MS medium and in Knop medium for 8 weeks. Also, the prothalli were cultured by chopping with a scalpel. In addition, sucrose, activated charcoal, and total nitrogen source were added in different concentrations based on the culture medium selected. Cultures were maintained at a temperature of $25{\pm}1^{\circ}C$, light intensity of $30{\times}1.0{\mu}mol-m-2{\cdot}s-1$, and a photoperiod of 16/8 h (light/dark) in in vitro. The results showed that optimum was achieved prothallus fresh weight and development in $1{\times}$ MS medium. When other components were added to the basic $1{\times}$ MS medium, prothallus propagation was maximized in $1{\times}$ MS medium supplemented with 2% sucrose, 0.2% activated charcoal, and 60 mM total nitrogen. To select a suitable soil mixture for sporophyte formation, 1.0 g of prothallus was blended with distilled water, spread on five combinations of different soil substrates (decomposed granite, horticultural substrates, peat moss, and perlite), and cultivated for 12 weeks. The sporophyte cultures were maintained at a temperature of $25{\pm}1^{\circ}C$, light intensity of $43{\pm}2.0{\mu}mol-m-2{\cdot}s-1$, humidity of $84{\pm}1.4%$, and a photoperiod of 16/8 h (light/dark). As a results, horticultural substrate alone, 2:1 (v:v) mixtures of horticultural substrate and perlite, and 2:1 mixtures of horticultural substrate and decomposed granite induced 208.0, 201.3 and 248.8 sporophytes per pot, respectively.   • In the era of the Fourth Industrial Revolution, various attempts have been made to incorporate ICT technology into mathematics teaching and learning, and the necessity and efficiency of classroom instruction using flipped learning, virtual reality and augmented reality have attracted attention. This leads to an increase in demand for instructional contents and their use in education. Therefore, there is a growing need for the development of instructional contents that can be applied in the field and the study of teaching methods. In this point of view, this research classifies the types of teaching-learning, presents the flipped learning instruction and mathematics contents by teaching-learning types using constructivist mathematics education principles and augmented reality-based mobile applications. These methods and lesson plans can provide a useful framework for teaching-learning in mathematics education.

Research on the Applicability of Experientially Emotional Measurement Method in Product Design - Centred on Self-report Taking Seaside Seat Users- (경험적 감성 측정 방법의 제품 디자인 적용성에 관한 연구 -해변 벤치 이용자 대상의 자기-보고법을 중심으로-)

  • In the era of the rapid development of a tremendous number of new technologies and products, the emotional communication between products and users has become an important research topic. However, the user's emotional measurement methods have not been widely used in the field of product design. Besides, as an important space that can improve people's life quality and urban entertainment at the emotional level, seaside space also creates an atmosphere of frequent emotional communication among tourists, landscape and environmental facilities. To deduce the applicability and advantages and disadvantages of the experiential emotional measurement method in product design, this paper took the seaside seat users of Haeundae as the object and carried out the Self-report of the experiential emotional measurement method. Specifically, the experiment of Self-report was divided into two stages: prior survey and subsequent main investigation. Then the hypothesis was verified by using Bivariate Regression and Multiple Regression through SPSS. Furthermore, through the experiment of the Self-report, the applicability and advantages and disadvantages of experiential emotional measurement method in product design were derived. In the future, based on the applicability, advantages and disadvantages of the experiential emotional measurement method derived in this study, it is expected to become the fundamental and reliable data for the development of emotional measurement methods that can be suitable for product design.

Radiographic Study of Cobey Method and Modified Cobey Method (Cobey 검사법과 Modified Cobey 검사법에 대한 방사선학적 연구)

  • The Cobey method and the modified Cobey method are most commonly used in clinical practice. Therefore, the purpose of this study was to investigate the radiological differences between Cobey and modified Cobey and provide radiographic information about changes of hindfoot image with X-ray entrance center and tube angle change in modified Cobey. This study was performed on foot and ankle phantom. First, for image comparison of Cobey and modified Cobey, the images obtained by applying the same X-ray entrance center to the ankle joint were compared and analyzed. Second, in the modified Cobey, the X-ray entrance center is set as ankle joint and lateral malleolus. The X-ray tube angle was varied from $10^{\circ}$ to $40^{\circ}$ at $5^{\circ}$ intervals for each X-ray entrance center. The images obtained by varying the X-ray tube angle from $10^{\circ}$ to $40^{\circ}$ at intervals of $5^{\circ}$ for each X-ray entrance center were compared and analyzed. The irradiation conditions were the same with 110 kVp, 200 mA, 10 ms, and 110 cm of source - image receptor distance (SID). Image evaluation was performed by two radiologists. Measurements were made on the lateral point, middle point, and calcaneus width based on a hypothetical line parallel to the calcaneal tuberosity. Data were analyzed by using descriptive statistics as the mean of the distance to each measurement location. The modified Cobey was longer than the Cobey by an average of 3 to 4 mm lateral and medial points, and the calcaneus width was similar (ICC = 0.939). In modified Cobey method, when the X-ray entrance center is ankle joint, the lateral point is about 3 mm and the medial point is about 4.3 mm longer than lateral malleolus. Also, when the X-ray tube angle is more than $20^{\circ}$, the degree of distortion is large. The ICCs for the lateral, medial point, and calcaneus width were 0.998, 0.961, and 0.997, respectively, as the X-ray entrance center and tube angle were changed. There was no significant difference between Modified Cobey and Cobey. Modified Cobey showed no need to compensate the $20^{\circ}$ detector angle of the Cobey. In addition, we suggest that tube angle should be limited within $20^{\circ}$ when modified Cobey is performed.

A Study on Spirituality Experience of Recovering Drug Addicts (회복기 마약중독자의 영성 체험에 관한 연구 -기독교·가톨릭인 회복자를 중심으로-)

  • In order to find out policy and practical implications to support recovering drug addicts in a holistic way, This study conducted in-depth interviews with five research participants using Giorgi's phenomenological study method to understand the meaning of spiritual experience of recovering drug addicts. According to data analysis, these spirituality experiences appeared as two thematic themes: 'experience of relationship with a divine being' and 'experience of inherent transcendence'. Three sub-themes of 'fear and scary existence', 'hanging object' and 'embracing existence' were derived from the essence theme of 'experience of relationship with a divine being' and 'immanent transcendental experience' was conceptualized into four sub-themes: 'facing existence', 'psychological reconstruction', 'mental maturity', and 'a way to go alone'. Based on this, spiritual experiences of recovery drug addicts showed that religion itself exerted a considerable influence on their spiritual experience and their spiritual experience was closely related to the recovery of humanity. Also, 'intrinsic transcendental experience' suggested that two essential themes of spiritual experience were closely interrelated in a mutual cyclical relationship. Based on the results, We discusses the major intervention plan for the recovery drug addicts that the social welfare system can consider.
