c-Jun N-terminal Kinase Contributes to Norepinephrine-Induced Contraction Through Phosphorylation of Caldesmon in Rat Aortic Smooth Muscle

  • Vascular smooth muscle contraction is mediated by activation of extracellular signal-regulated kinase (ERK) 1/2, an isoform of mitogen-activated protein kinase (MAPK). However, the role of stress-activated protein kinase/c-Jun N-terminal kinase (JNK) in vascular smooth muscle contraction has not been defined. We investigated the role of JNK in the contractile response to norepinephrine (NE) in rat aortic smooth muscle. NE evoked contraction in a dose-dependent manner, and this effect was inhibited by the JNK inhibitor SP600125. NE increased the phosphorylation of JNK, which was greater in aortic smooth muscle from hypertensive rats than from normotensive rats. NE-induced JNK phosphorylation was significantly inhibited by SP600125 and the conventional-type PKC (cPKC) inhibitor Go6976, but not by the Rho kinase inhibitor Y27632 or the phosphatidylinositol 3-kinase inhibitor LY294002. Thymeleatoxin, a selective activator of cPKC, increased JNK phosphorylation, which was inhibited by $G{\ddot{o}}6976$. SP600125 attenuated the phosphorylation of caldesmon, an actin-binding protein whose phosphorylation is increased by NE. These results show that JNK contributes to NE-mediated contraction through phosphorylation of caldesmon in rat aortic smooth muscle, and that this effect is regulated by the PKC pathway, especially cPKC.

A Study on the Area of Reinforcement in the Cross Tunnel by Laboratory Model Test (실내모형실험을 통한 교차터널의 보강영역에 관한 연구)

  • With the increase in infrastructure needs and tunnel construction, more complicated geometries have come to light, especially in cross tunnel design and construction. The major factors to influence existing tunnels are spacing between tunnels, relative position, size of the new tunnel, construction techniques, topographic and geologic conditions, structure, and alternative construction. In this study, settlement from an excavation for a new tunnel near an existing tunnel, settlement from a change in position of the new tunnel relative to an existing tunnel, and the distribution of deformations of the existing tunnel as a result of excavating the cross-location are analyzed through laboratory model tests. As the results, in condition of the new tunnels go through below the existing tunnel, not only analysed through the standard of the diameter of the tunnel, so it would need to set up to strengthen the field within each side of the 1D, but also determined the part of the cross in the existing and the new tunnel, should implement the reinforcement from the part of new tunnel to the existing tunneling influence of excavation.

  • Mechanical properties of carbon fiber reinforced thermoplastic composites (CFRTPs) are affected by various factors. One of the them are poor compatibility of the epoxy sizing layer on the carbon fiber surface with thermoplastic matrix, which causes the inferior interfacial strength between fibers and matrix. In addition, the high molten-viscosity of thermoplastics attributes to the poor impregnation state. Consequently, many voids in the composite materials were generated, which leads to poor mechanical properties of the thermoplastic composites. In this study, the epoxy sizing on the carbon fiber surface was removed and the polyamide 6,6 solution was coated on the de-sized carbon fiber surface to improve the impregnation state and mechanical properties. Interlaminar shear strength (ILSS) of CFRPTs was estimated by implementing short beam shear tests. In addition, flexural strength was measured and the impregnation state of the composites was evaluated by calculating void content.

Optimum Stocking Density of Juvenile Abalone (Haliotis discus hannai) Fed the formulated Diet or Macroalgae (Undaria) (실험 배합사료 및 미역 공급시 참전복 치패의 적정 사육밀도)

  • A feeding trial was carried out to investigate the optimum density of juvenile abalone (Haliotis discus hannai) fed the formulated diet or macroalgae ( Undaria). Abalone with initial average shell length of 8.41 mm were stocked into three replicate tanks at four different densities of 1430, 2860, 4290, and 5720 individual/$m^2$ for 15 weeks. The formulated diet produced higher body weight and shell growth, soft body weight and survival rate than Undaria in abalone. Abalone fed the formulated diet have significantly (P<0,05) higher lipid and ash, and lower moisture and protein in soft body. Higher density resulted in lower growth and survival (P<0.05). Whereas shell growth, survival rate and weight gain were not significantly (P>0.05) different within the abalone stocked at stocking densities of 1430 and 2860 individual/$m^2$ in each diet group. Body composition of abalone was not affected by increasing density. These study indicate that the number of abalone (shell length ranges of between 8 mm and 17 mm) below 2860 individual/$m^2$ were predicted to effective density in the indoor tank culture system.

  • This paper attempts to examine how Maryland's Priority Funding Area (PFA) designation and incentive program has influenced the location of infrastructure investment. Is Maryland's PFA program reducing sprawl? In order to answer this question, data on the water and sewer infrastructure investments between 1997 and 2003 are collected from each county in the state. Empirical works are composed of two parts. The first part of the empirical analysis examines the pattern of water and sewer investment that has gone in and outside the PFAs between 1997 and 2003 at the county level. The second part of the empirical study shows at a county level the conditions that influence decisions to go in and outside the PFA. Regression models with various specifications are used for the analysis. The findings reveal that state fund has worked as designed. The coefficients of state fund in all estimations are significant and have expected signs implying that a county with more state fund tends to invest more in PFA as less outside.

Study on the Change of Wind Field and Temperature According to Location of High-rise Building Using Micrometeorology Numerical Model (미기상 수치 모델을 이용한 고층아파트 입지에 따른 바람장 및 기온 변화 연구)

  • This study was carried out to analyze the change of wind filed and heat island according to the location of the high rise building using micrometeorology numerical model Envi-met3.0. In this study, the real urban planning of Jeonju city was used as input for the location and height of buildings. Modeling was performed for two conditions as input data. Case 1 is that wind direction is SSE and case 2 is W. To analyse the change of wind filed, wind speed results were used. To analyze the change of heat island, temperature results were used. Below the building height, wind speed increased 0.2~2.5 m/s at the inflow area and decreased 0.5~2.0 m/s at the area between the buildings. Above the building height, wind speed decreased 0.1~0.8 m/s near the building complex. On the other hand, wind speed increased 0.2~0.4 m/s in the outside area of the building complex. In the case of temperature, below the building height, temperatures increased $0.01{\sim}0.1^{\circ}C$ in the building complex and leeward area. On the other hand, temperature decreased $0.01{\sim}0.005^{\circ}C$ in the outside area of the building complex. Above the buildings height, temperatures decreased $0.05{\sim}0.2^{\circ}C$ in most of the area.

Comparative Analysis for In Vitro Differentiation Potential of Induced Pluripotent Stem Cells, Embryonic Stem Cells, and Multipotent Spermatogonial Stem Cells into Germ-lineage Cells

  • In the present study, embryoid bodies (EBs) obtained from induced pluripotent stem cells (iPSCs) were induced to differentiate into germ lineage cells by treatment with bone morphogenetic protein 4 (BMP4) and retinoic acid (RA). The results were compared to the results for embryonic stem cells (ESCs) and multipotent spermatogonial stem cells (mSSCs) and quantified using immunocytochemical analysis of germ cell-specific markers (integrin-${\alpha}6$, GFR-${\alpha}1$, CD90/Thy1), fluorescence activating cell sorting (FACS), and real time-RT-PCR. We show that the highest levels of germ cell marker-expressing cells were obtained from groups treated with 10 ng/$m{\ell}$ BMP4 or 0.01 ${\mu}M$ RA. In the BMP4-treated group, GFR-${\alpha}1$ and CD90/Thy-1 were highly expressed in the EBs of iPSCs and ESCs compared to EBs of mSSCs. The expression of Nanog was much lower in iPSCs compared to ESCs and mSSCs. In the RA treated group, the level of GFR-${\alpha}1$ and CD90/Thy-1 expression in the EBs of mSSCs Induced pluripotent stem cells, Mouse embryonic stem cells, Multipotent spermatogonial stem cells, Germ cell lineage, Differentiation potential. was much higher than the levels found in the EBs of iPSCs and similar to the levels found in the EBs of ESCs. FACS analysis using integrin-${\alpha}6$, GFR-${\alpha}1$, CD90/Thy1 and immunocytochemistry using GFR-${\alpha}1$ antibody showed similar gene expression results. Therefore our results show that iPSC has the potential to differentiate into germ cells and suggest that a protocol optimizing germ cell induction from iPSC should be developed because of their potential usefulness in clinical applications requiring patient-specific cells.

A study for middle-aged on oral health knowledge, oral health care and satisfaction with prosthetic treatment (중년층의 구강건강지식, 실천 및 보철치료 만족실태 조사)

  • Objectives : The purpose of this study was to examine the oral health knowledge and actual oral health care of the selected subjects, their decision making about prosthetic treatment, the state and characteristics of their prosthetic treatment and their satisfaction with prosthetic treatment in an attempt to provide some information on the improvement of the quality of life related to oral health and the promotion of oral health. Methods : The subjects in this study were 250 people who received education in two different lifelong education institutions in the city of Busan. After a survey was conducted from May 23 to June 15, 2011, the answer sheets from 217 respondents were analyzed. Results : 1. As for the general characteristics of the respondents, the men(52.1%) outnumbered the women. Those who were in their 60 and up(47.5%) made up the largest age group, and the married people(65.4%) outnumbered the unmarried ones. By occupation, the company employees(20.3%) made up the biggest group. By education, the greatest group was high-school graduates(36.1%). By monthly mean income, the biggest group gained an income of 2.01 million won or more(36.9%). As to medical security, community-based insurance was most prevailing (36.9%). In terms of health promotion, the largest group worked out to stay fit(52.4%). 2. Regarding oral health knowledge, they had a good knowledge about the cause of dental caries(56.7%), but they weren't well aware of the right time for regular dental checkup(47.9%). In relation to oral health care, regular scaling wasn't prevailing(16.9%). 3. Regarding connections between the general characteristics and satisfaction with prosthesis, the less-educated respondents expressed significantly more dissatisfaction(p=0.015). By monthly mean income, those who had a smaller income were dissatisfied in general(p=0.028). Conclusions : The findings of the study illustrated that it's required to spread awareness of the importance of oral health among people in general, and that differentiated incremental oral health care should be provided for different age groups. In order to raise the satisfaction of patients with prosthesis, how to relieve their pain and anxiety and how to adjust prosthetic treatment cost properly should carefully be considered. In addition, the government should take measures to offer assistance for the low-income classes in preparation for an increase in the elderly population.

  • Recently, due to global warming, it is easily exposed to various disasters such as fire, flood, and earthquake. In particular, large-scale disasters have continuously been occurring in crowded areas such as traditional markets, facilities for the elderly and children, and public facilities where various people stay. Purpose: This study aims to detect a fire occurred in crowded facilities early in the event to analyze and provide an optimal evacuation route using big data and advanced technology. Method: The researchers propose a new algorithm through context-aware 3D object model technology and A* algorithm optimization and propose a scenario-based optimal evacuation route selection technique. Result: Using the HPA* E algorithm, the evacuation simulation in the event of a fire was reproduced as a 3D model and the optimal evacuation route and evacuation time were calculated for each scenario. Conclusion: It is expected to reduce fatalities and injuries through the evacuation induction technique that enables evacuation of the building in the shortest path by analyzing in real-time via fire detection sensors that detects the temperature, flame, and smoke.

  • Thanks to the inter-connection of information servers throughout the world based on the internet technology, the new sphere which actual transaction can be made like in the visible market has become conspicuous as the virtual space. The movement to realize the new business through the cyber space has been actively ongoing. In the domestic market, a lot of corporations knowing the needs of internet shopping malls have entered into this e-business but they have not made a big success comparing with internet's potentials. And, it can be attributed to the simple planes and the limitations of information provided by the cyber malls, which means that the needs of better information transfer we apparent Accordingly, in this thesis, the research on the 3-D based products and shopping malls has been made through the inter-complementary composition between the 2-D shopping malls and 3-B ones. This research consists of 3 parts. Firstly, through the research on references and existing data, it presents the analysis on consumer's characteristics and sales limits of the internet shopping mall's products. Secondly, the background of 3-D shopping mall's advent and the virtual reality technology data are put together. Finally, it presents how the development of 3-D based product modeling and shopping malls can increase the consumer's purchase power and furthermore the directions of shopping malls to go.
