Anti-inflammatory Activity of an Ethanol Extract of Laminaria japonica Root on Lipopolysaccharide-induced Inflammatory Responses in RAW 264.7 Cells (Lipopolysaccharide로 유도된 RAW 264.7 세포에서 다시마 뿌리 에탄올 추출물의 항염증 효과)

  • Laminaria japonica roots have not been used practically in Korea. In this study, in order to promote the use of these by-products, the anti-inflammatory activity of an ethanol extract of Laminaria japonica root (LJREE) was investigated using a lipopolysaccharide (LPS) to induce an inflammatory response in RAW 264.7 cells. To examine the potential anti-inflammatory effects of LJREE, levels of nitric oxide (NO) and pro-inflammatory cytokines, such as interleukin-6 (IL-6), tumor necrosis factor-${\alpha}$ (TNF-${\alpha}$), and interleukin-$1{\beta}$ (IL-$1{\beta}$), and cell proliferation were measured. We found that NO levels decreased in a dose-dependent manner, the production of IL-6, TNF-${\alpha}$, and IL-$1{\beta}$ was suppressed, and the production of IL-$1{\beta}$ was inhibited over 30% after treatment with $100{\mu}g/mL$ LJREE. In conclusion, the proliferation of RAW 264.7 cells, measured by MTT assay, confirmed that LJREE may have significant effects on inflammatory factors without any cytotoxicity, making it a potential anti-inflammatory agent.

Simulation of Local Climate and Crop Productivity in Andong after Multi-Purpose Dam Construction (임하 다목적댐 건설 후 주변지역 기후 및 작물생산력 변화)

  • A simulation study was carried out to delineate potential effects of the lake-induced climate change on crop productivity around Lake Imha which was formed after a multi-purpose dam construction in Andong, Korea. Twenty seven cropping zones were identified within the 30 km by 25 km study area. Five automated weather stations were installed within the study area and operated for five years after the lake formation. A geostatistical method was used to calculate the monthly climatological normals of daily maximum and minimum temperature, solar radiation and precipitation for each cropping zone before and after the dam construction. Daily weather data sets for 30 years were generated for each cropping zone from the monthly normals data representing "No lake" and "After lake" climatic scenarios, respectively. They were fed into crop models (ORYZA1 for rice, SOYGRO for soybean, CERES-maize for corn) to simulate the yield potential of each cropping zone. Calculated daily maximum temperature was higher after the dam construction for the period of October through March and lower for the remaining months except June and July. Decrease in daily minimum temperature was predicted for the period of April through August. Monthly total radiation was predicted to decrease after the lake formation in all the months except February, June, and September and the largest drop was found in winter. But there was no consistent pattern in precipitation change. According to the model calculation, the number of cropping zones which showed a decreased yield potential was 2 for soybean and 6 for corn out of 27 zones with a 10 to 17% yield drop. Little change in yield potential was found at most cropping zones in the case of paddy rice, but interannual variation was predicted to increase after the lake formation. the lake formation.

Effect of Sargassum sagamianum Extract on Shelf-life and Improved Quality of Morning Bread (비틀대 모자반(Sargassum sagamianum) 추출물이 모닝빵의 저장성 및 품질에 미치는 영향)

  • This study was performed to test the effect of Sargassum sagamianum extract (SSE) on shelf-life and improved quality in bread with 0.25, 0.5 and 0.75% added SSE. Bread with added SSE had reduced total microbial counts by 2 log cycles and mold cell counts by 3 log cycles. No changes in moisture content or pH occurred from days 3 to 9. In addition, bread with SSE had a lower yield of malonaldehyde than that of the control as shown by the TBARS assay. Yellowness increased in bread with added SSE, whereas lightness and redness decreased. In the sensory evaluation, taste, total preference, inner shape, and color of the bread containing 0.25 and 0.5% SSE were preferred. These results suggest that the adding 0.25 and 0.5% SSE to bread improved shelf-life and quality.

Geometrical Interpretation on the Development Sequence and the Movement Sense of Fractures in the Cheongsong Granite, Gilan-myeon Area, Uiseong Block of Gyeongsang Basin, Korea (경상분지 의성지괴 길안면지역에서 청송화강암의 단열 발달사 및 운동성에 대한 기하학적 해석)

  • The Gilan area in the central-northern part of Uiseong Block of Cretaceous Gyeongsang Basin is composed of Precambrian metamorphic rocks, Triassic Cheongsong granite, Early Cretaceous Hayans Group, and Late Cretaceous-Paleocene igneous rocks. In this area, the faults of various directions are developed: Oksan fault of $NS{\sim}NNW$ trend, Gilan fault of NW trend, Hwanghaksan fault of WNW trend, and Imbongsan fault of EW trend. Several fracture sets with various geometric indicators, which determine their relative timing (sequence and coexistence relationships) and shear sense, we well observed in the Cheongsong granite, the basement of Gyeongsang Basin. The aim of this study is to determine the development sequence of extension fractures and the movement sense of shear fractures in the Gitan area on the basis of detailed analysis of their geometric indicators (connection, termination, intersection patterns, and cross-cutting relations). This study suggests that the fracture system of the Gilan area was formed at least through seven different fracturing events, named as Pre-Dn to Dn +5 phases. The orientations of fracture sets show (W) NW, NNW, NNE, EW, NE in descending order of frequency. The orientation and frequency patterns are concordant with those of faults around and in the Gilan area on a geological map scale. The development sequence and movement sense of fracture sets are summarized as follows. (1) Pre-Dn phase: extension fracturing event of $NS{\sim}NNW$ and/or $WNW{\sim}ENE$ trend. The joint sets of $NS{\sim}NNW$ trend and of $WNW{\sim}ENE$ trend underwent the reactivation histories of sinistral ${\rightarrow}$dextral${\rightarrow}$sinistral shearing and of (dextral${\rightarrow}$) sinistral shearing with the change of stress field afterward, respectively. (2) Dn phase: that of NW trend. The joint set experienced the reactivations of sinistral${\rightarrow}$dextral shearing. (3) Dn + 1 phase: that of $NNE{\sim}NE$ trend. The joint set was reactivated as a sinistral shear fracture afterward. (4) Dn +2 phase: that of $ENE{\sim}EW$ trend. (5) Dn +3 phase: that of $WNW{\sim}NW$ trend. (6) Dn+4 phase: that of NNW trend. The joint set underwent a dextral shearing after this. (7) The last Dn +5 phase: that of NNE trend.

Soil Salinity and Salt Spray Drift Tolerance of Native Trees on the Coastal Windbreaks in the South-Sea, Korea (한국 남해안방풍림 자생수종의 내염성 및 내조성 수종 선발)

  • This study was carried out to investigate the soil salinity and salt spray drift of the indigenous windbreak trees, and its main purpose was to provide basic data for the selection of salt-tolerant trees in the saline coastal region in the South Sea of Korea. The soil salinity($EC_{1:5}$)was $0.18dSm^{-1}$, which was an average degree of the whole areas of investigation whose salinity degree ranged from $0.05dSm^{-1}$ to $0.58dSm^{-1}$. The level of soil salinity gradually decreased as it moved farther inland, except the belt I. The level of decreasing soil salinity was found to be in the following order: belt II, belt III, belt I, belt IV. The degree of soil salinity was $EC_{1:5}$ $0.22dSm_{1:5}$, $0.22dSm_{1:5}$ $0.19dSm^{-1}$ and $0.13dSm^{-1}$ respectively. The total 110 taxa, which consisted of 45 families, 74 genus, 101 species, and 9 varieties, were found to be tolerant to both soil salinity and salt spray drift. The trees that grow in the highest degree of salinity($EC_{1:5}$ $0.50dSm^{-1}$)were Parthenocissus tricuspidata(Siebold & Zucc.), Planch and Lonicera japonica Thunb. The next group of trees that grow in the high degree of salinity ranging from $EC_{1:5}$ 0.41 to $0.50dSm^{-1}$ was Cudrania tricuspidata(Carr.) Bureau ex Lavall$\acute{e}$e, Rubus parvifolius L., Zanthoxylum schinifolium(Siebold & Zucc.), Hedera rhombea(Miq.) Bean., Robinia pseudoacacia L., Quercus serrata Thunb., Callicarpa dichotoma(Lour.) K. Koch, and so on. The woody species which grew in the entire belts were Pueraria lobata(Willd.) Ohwi and Vitis flexuosa Thunb., and Vitex rotundifolia L. f. which was known to be highly tolerant to salt spray drift was found only in belt I. The woody species with high important value(IV) were Zelkova serrata(Thunb.) Makino., Celtis sinensis Pers., Koelreuteria paniculata Laxmann, Mallotusjaponicus(Thunb.) Muell. Arg., Trachelospermum asiaticum(Siebold & Zucc.) NAKAI, and Pueraria lobata(Willd.) Ohwi. These species were classified as native windbreak trees that are comparatively more tolerant to salt spray drift than other kinds.

Functional food activities of extracts from Pinus densiflora root (동송근(Pinus densiflora root) 추출물의 기능성식품 활성)

  • In this study, the antioxidative activity and functional food activities of water and ethanol extracts from Pinus densiflora root were examined. It was more effective to use ethanol than water when extracting phenolic compounds. The extracted phenolic compounds from Pinus densiflora root for biological activities were examined. The phenolic compounds extracted with water and 80% EtOH were $1.86{\pm}0.04mg/g$ and $6.85{\pm}0.16mg/g$, respectively. DPPH free radical scavenging activity of water and EtOH were each 86% and 85% at $100{\mu}g/mL$ phenolics, respectively. ABTS radical decolorization activity was 48% in water and 68% in EtOH at $200{\mu}g/mL$. Antioxidant Protection Factor (PF) were 1.74 PF in water and 1.96 PF in EtOH at $50{\mu}g/mL$. TBARs of water and EtOH were 93% and 98%, respectively at $100{\mu}g/mL$. The inhibition activity on xanthine oxidase was 83.7% in water extracts and 79.6% in ethanol extracts. Inhibition on xanthine oxidase of water and ethanol extracts showed a higher inhibition effect than allopurinol. The inhibition activity on ${\alpha}$-glucosidase was 14.8% in water extracts and 91.6% in ethanol extracts. The result suggests that P. densiflora root extracts may be useful as as functional food material.

A Study on the Reproduction Experimental of Breaking of dried stalks technique of Recorded in Oju-Yeonmunjang jeon-Sango in the late Joseon Dynasty (조선 후기 『오주연문장전산고』에 기록된 자연건조쇄경식 제섬 기술 재현 실험 연구)

  • Fiber scutching refers to the process of extracting fibers from plants by separating or extracting fibers from the raw materials. As the definition of the term implies, the "Fiber Scutching" is performed on plants with advanced bast fiber as the primary material processing technique performed on plant materials. Some of the most popular phosphorus plants are ramie, hemp, flax, and the paper mulberry, which have a long history of cultivation and a wide range of distribution, making them very universal as a material supporting human life and culture. This study was described in Oju-yeonmunjangjeon-sango but was designed to re-examine the method of breaking dried stalks, which is currently unused in Korea, to examine the feasibility and characteristics of the technology. As a result of sampling and experimenting with hemp bast using the method recorded in the literature, hemp fiber was actually produced. The criteria for removing the shell from the hemp stem were the degree of discoloration and drying, and only when the stalk was completely discolored to yellow could segregation of the stalk from the shell be performed. The amount of sunlight and temperature were conditions that accelerated drying. However, if exposed for a long time, it is confirmed that hemp bast will be in a suitable condition to process, regardless of the amount of sunlight and temperature. 'Breaking of dried stalks', which utilizes the physical power of 'threshing with a flail' is considered a core process of the fiber scutching technique in 'Yukjin' in Hamgyeong-do. The bark and the core of the hemp were separated by tapping, the bast was thinly split, and the shell was peeled off, making it suitable for collecting with thread. The method of collecting the fibers by applying physical power causes downing on the fibers, which is to be generally avoided in the manufacture of bast fabric woven hemp or ramie. However, Hamgyeong-do's fiber scutching method seems to have applied this principle to the method of making fragile fabrics by using it in reverse. This method is distinct from the steaming or boiling of the stalks' in Andong, Korea, and it is similar to the Western method of spinning fabrics.

Studies on the postnatal development of the Leydig cell in rabbits (토끼에서 출생 후 고환간질세포의 발생에 관한 연구)

  • Changes in the rabbit Leydig cell from birth to adulthood were studied in New Zealand white rabbits of 1, 7, 21, 35, 49, 70, 105, 147, 196, and 252 days (n = 8 rabbits per group) of age. The objectives of this study were to understand the fate of the fetal Leydig cells, to determine the changes in serum testosterone levels, and leutenizing hormone-stimulated testosterone production per testis in vitro, and to quantify adult Leydig cells by number and average volume with age. Testes of rabbits were fixed by whole body perfusion using a fixative containing 2.5% glutaraldehyde in cacodylate buffer, processed and embedded in Epon-araldite. Using $1{\mu}m$ sections stained with methylene blue-azure II, qualitative and quantitative (stereological) morphological studies were performed. Testosterone levels in the incubation medium of luteinizing hormone-stimulated (100 ng/ml) testosterone secretion per testis in vitro, and in serum were determined by radioimmunoassay. The average volume of a testis of 1-day-old rabbits was determined as $0.0073cm^3$ and the parameter increased linearly from birth to 252 days ($3.93cm^3$). The volume density of the seminiferous tubules increased with age from 33.76% at day 1 to 88.2% at day 252. The volume density of the interstitium represents 66.24% of the testicular parenchyma at day 1. This proportion progressively diminished during development to reach a value of 11.8% at day 252. The volume density of Leydig cells increased almost linearly from birth (0.001%) to 252 days (2.62%). Leydig cell mass per testis increases from 0.0012 mg to 0.25 mg between days 1 and 35, from 2.66 mg to 44.3 mg between days 49 and 105 and from 65.42 mg and 102.9 mg between days 147 and 252. The absolute numbers of adult Leydig cells per testis increased linearly from birth to 252 days. The average volume of adult Leydig cell on days 1, 7, 21 and 35 was not significantly different; a gradual and continued increase was observed thereafter, reaching a 3-fold increase at 196 and 252 days. Serum testosterone concentrations were not significantly different at day 1 compared days 7, 21, 35. Significant increases were observed at days 49 and 70. Values at days 70 and 105 and days 147, 196, and 252 were not significantly different. LH-stimulated testosterone production per testis in vitro was significantly different at day 1 compared days 7, 21, 35. Significant increases were observed at days 49 and 70. Hormonal values at days 105, 147, 196, and 252 were not significantly different. These data suggested Leydig cell developmental phase can be classified: a neonatal phase (1-7 days), a prepubertal phase (14-49 days) and an adult phase (70-252 days). Immature and mature adult Leydig cells, initially detected at days 7 and 49, respectively, and mature adult Leydig cells were abundant Leydig cell type according to the number and absolute volume per testis form day 49 onwards.

Effect of Addition of Phosvitin and High Pressure Processing on Microbiological Quality and Lipid and Protein Oxidation of Minced Chicken Leg Meat (닭 다리 분쇄육에 초고압 처리시 Phosvitin의 첨가가 미생물학적 품질과 지방 및 단백질 산화에 미치는 효과)

  • The objective of this study was to investigate the effect of high pressure (HP) processing on shelf life, as well as the addition of phosvitin on lipid and protein oxidation stability of minced chicken leg meat. Minced chicken leg meat was mixed with yolk phosvitin at 500 or 1000 mg/kg meat levels, and divided into raw and cooked groups. Then, the samples were subjected to HP at 0.1, 300, and 600 MPa. The total aerobic bacteria, lipid and protein oxidation, along with instrumental meat color ($L^*$, $a^*$, and $b^*$value) of the samples were measured during storage for 7 d at $4^{\circ}C$. In raw meat, the number of total aerobic bacteria was decreased by HP at 300 MPa (4 Log reductions) and 600 MPa (5 Log reductions) after 7 d of storage (p<0.05). HP at 600 MPa increased lipid oxidation of samples at all storage days and protein oxidation of samples during storage at 3 and 7 d. HP induced the changes of meat color by increase of $L^*$ value and decrease of $a^*$ value (p<0.05). The total aerobic bacteria was not detected in the cooked samples, regardless of HP pressure, and the lipid or protein oxidation of the cooked sample treated by 600 MPa was higher than that of the control (0.1 MPa) on day 7 or control on day 3, respectively (p<0.05). The results suggested that HP can improve the shelf life of minced chicken leg meat. However, phosvitin might be a limited antioxidative agent for the improvement of oxidation stability induced by HP.

A Case Study on the Estimation of the Resource Recovery Potentials by Landfill Mining (매립지 정비에 의한 순환이용 가능량 산정 사례 연구)

  • As many local governments have faced increasing conflicts on landfill use and the time of end use, it is difficult to provide an alternative landfill or conclude a consensus of lifespan extension for the existing landfill site. Therefore, the purpose of this study is to contribute improving of the landfill capacity by calculating the resource recovery potentials of landfilled waste previously and in the future by landfill mining. For this, rate of volume increase, weight ratio, and apparent density were adopted as major parameters and their values were calculated through previous cases. The rate of volume increase was calculated to 1.42 by averaging previous cases of three areas. The average weight ratio of soil matter was 45.6% by calculating for the three areas. For the combustible waste and incombustible waste, statistical data can be used. The apparent densities were divided by combustible waste, incombustible waste, and soil matter using an average of two areas value, i.e., $0.35ton/m^3$, $1.40ton/m^3$ and $1.58ton/m^3$. We analyzed the resource recovery potential of Cheongju landfill by using the estimated parameters. The additional landfill capacity was 45% of the existing landfill capacity by recovering landfilled waste by landfill mining. In addition, it is analyzed that the lifespan is extended to 20 years, if the combustible waste of new inputting waste is sorted and combusted for energy recovery and incineration ash, incombustible waste, and soil matter are only reclaimed into the existing Cheongju landfill. It is expected that the methodology and parameters of this study will be used as basic data when resource recovery potential is analyzed for another case study of landfill mining.
