Molecular Cloning of the Antiapoptotic Gene, p35, from Bombyx mori Nuclear Polyhedrosis Virus K1

  • We have cloned and characterized an antiapoptotic gene, p35, which blocks apoptosis, from Bombyx mori nuclear polyhedrosis virus (BmNPV) K1 strain. The 897 bp p35 has an open reading frame of 299 amino acids. The BmNPV-K1 p35 showed a high identity to Autographa californica nuclear polyhedrosis virus and BmNPV T3 strain. The BmNPV-K1 p35 was different from the amino acid sequences of BmNPV T3 at 6 positions. The p35 gene of BmNPV-K1 was 99.2% identical at the nucleotide level and 98% identical at the amino acid level to BmNPV T3. The location of p35 gene in the BmNPV-K1 genome was confirmed by Southern blot analysis and its expression patterns at the transcriptional level in the infected cells were con- firmed by Northern hybridization analysis.

  • Consider a graph G of order n and maximum degree ${\Delta}$. Let $f:V(G){\rightarrow}\{0,1,{\cdots},{\lceil}{\frac{{\Delta}}{2}}{\rceil}+1\}$ be a function that labels the vertices of G. Let $B_0=\{v{\in}V(G):f(v)=0\}$. The function f is a strong Roman dominating function for G if every $v{\in}B_0$ has a neighbor w such that $f(w){\geq}1+{\lceil}{\frac{1}{2}}{\mid}N(w){\cap}B_0{\mid}{\rceil}$. In this paper, we study the bounds on strong Roman domination numbers of the Cartesian product $P_m{\square}P_k$ of paths $P_m$ and paths $P_k$. We compute the exact values for the strong Roman domination number of the Cartesian product $P_2{\square}P_k$ and $P_3{\square}P_k$. We also show that the strong Roman domination number of the Cartesian product $P_4{\square}P_k$ is between ${\lceil}{\frac{1}{3}}(8k-{\lfloor}{\frac{k}{8}}{\rfloor}+1){\rceil}$ and ${\lceil}{\frac{8k}{3}}{\rceil}$ for $k{\geq}8$, and that both bounds are sharp bounds.

Effects of Tween 80 on In Vitro Fermentation of Silages and Interactive Effects of Tween 80, Monensin and Exogenous Fibrolytic Enzymes on Growth Performance by Feedlot Cattle

  • The effects of monensin, Tween 80 and exogenous fibrolytic enzymes on ruminal fermentation and animal performance were studied in vitro and in vivo. In Expt 1, the effects of the surfactant Tween 80 (0.2% wt/wt, DM basis) on ruminal fermentation of alfalfa, corn and orchardgrass silages were investigated using in vitro gas production techniques. Tween 80 did not affect (p>0.05) cumulative gas production at 24 h, but it reduced (p<0.05) the lag in fermentation of all three silages. With corn silage and orchardgrass silage, gas production rates and concentrations of total volatile fatty acids (VFA) were increased (p<0.05) by Tween 80; with alfalfa silage, they were reduced (p<0.05). Tween 80 increased (p<0.05) the proportion of propionate in total VFA, and reduced (p<0.05) acetate to propionate ratios (A:P) with all three silages. In Expt 2, exogenous fibrolytic enzymes (E; at 0, 37.5 or 75 g/tonne DM), monensin (M; at 0 or 25 ppm and Tween 80 (T; at 0 or 2 L/tonne DM) were added alone or in combination to backgrounding and finishing diets fed to 320 crossbred steers in a feeding trial with a $3{\times}2{\times}$2 factorial arrangement of treatments. The backgrounding and finishing diets contained barley grain and barley silage in ratios of 57.8:42.2 and 93.5:6.5 (DM basis), respectively. Added alone, none of the additives affected DM intake (p>0.1) in the backgrounding or in the finishing period, but interactive $M{\times}T$ effects were observed in the finishing period (p=0.02) and overall (p=0.04). In the finishing period, T without M tended to reduce DM intake (p=0.11), but T with M increased (p=0.05) DM intake. Monensin increased average daily gain (ADG) during backgrounding (p=0.07) and finishing (p=0.01), and this ionophore also improved overall feed efficiency (p=0.02). Warm carcass weight was increased (p<0.001) by M, but dressing percentage was reduced (p=0.07). In the backgrounding period, T increased ADG by 7% (p=0.06). Enzymes increased (p=0.07) ADG by 5 and 6% (low and high application rates, respectively) during backgrounding, but did not affect (p>0.10) ADG during finishing, or overall feed efficiency. Whereas T enhanced the positive effects of M on ADG during backgrounding (p=0.04) and overall (p=0.05), it had no impact (p>0.1) on the effects of E. Interactions between M and T suggest that the surfactant may have potential for enhancing the positive effects of monensin on beef production, but this requires further research.

Nucleotide Sequence Analyses of p10 Gene and its Promoter of Hyphantria cunea Nuclear Polyhedrosis Virus (Hyphantria cunea Nuclear Polyhedrosis Virus p10유전자와 프로모터의 염기서열 결정)

  • The sequences of p10 gene its promoter of Hyphantria cunea NPV were determined. According to the sequence analysis, the putative p10 gene ORF has 285 bp. The 5'-non-coding leader sequence of the p10 gene promoter contained the TATA box and the putative transcription initiation site TAAG motif. Poly (A) tail signals, AATAAA sequence was at site 65 base upstream from the 3' terminus. The deduced amino acid sequence of p10 protein was 95 with a predicted molecular weight of 10.26 kDa. In the p10 protein sequence, a hydrophobic region was present at the N-terminus of the protein, whereas the C-terminus was highly hydrophilic. The p10 protein of H. cunea NPV did not contain cysteine, histidine, trytophan, tryptophane, tyrosine, glutamine and asparagine residues.

Hsa-miR-181a-5p Expression and Effects on Cell Proliferation in Gastric Cancer

  • Purpose: MicroRNAs (miRNAs) are small endogenous, non-coding, single-stranded RNAs (approximately 22 nt). Accumulating evidence has shown that aberrant miRNA expression is pronounced and correlated with gastric cancer genesis and progression. Materials and Methods: Expression levels of miR-181a-5p in GC tissues and cell lines were assessed by qRT-PCR and tested for correlation with clinical features. In addition, effects of miR-181a-5p on GC cell growth were investigated. Results: Our findings indicate that miR-181a-5p is upregulated in GC, in correlation with lymph node invasion, nerve invasion and vascular invasion (P<0.05). Enforced expression of miR-181a -5p promoted cell proliferation ability. Conclusions: This study suggested that increased miR-181a-5p is related to GC progression. MiR-181a-5p may represent a potential therapeutic target for GC.

Determination of antigenic domain in GST fused major surface protein (Nc-p43) of Neospora caninum

  • The antigenic domain of the major surface protein (Nc-p43) of Neospora caninum was examined by polymerase chain reaction of its gene fragments and recombinant expression as GST fusion proteins. The fragments of Nc-p43 were as follow: a total open reading frame (OFR), T: OFR without signal sequence and C-terminal hydrophobic sequence, S: N-terminal 2/3 parts of S, A: C-terminal 2/3 parts, P; N-terminal 1/3 part, X: middle 1/3 part Y; and C-terminal 1/3 part, Z, respectively. The DNA fragments were cloned into pGEX-47 vector. Recombinant plasmids transformed into Escherichia coli of BL21 pLysS (DE3) strain were induced to express GST or GST fused fragments of Nc-p43 such as 69 kDa protein for T,66 kDa for S, 52 kDa for A,53 kDa for P, and 40 kDa proteins for X, Y, and Z, respectively in SDS-PAGE. The Nc-p43 fragments of T, S, and P reacted with a bovine serum of neosporosis while those of A, X, Y, and Z together with GST did not in the western blot. These findings suggest that the antigenic domain of Nc-p43 of N. caninum may be localized in the C-terminal 2/3 parts. Together with Al9 clone in SAGI of Toxoplasma gondii (Nam et at., 1996), the P fragment of Nc-p43 could be used as efficient antigens to diagnose and differentiate those infections with both species .

Distribution of Inorganic Phosphorus Fractions in Sediments of the South Han River over a Rainy Season

  • Rain events are extremely important for phosphorus (P) dynamics in rivers since large portions of annual river P loads can be transported in particulate forms during only a few major events. Despite their importance, a precise estimation of P contribution in river sediments after rainy seasons has rarely been reported. This study estimated the longitudinal variation in the concentrations of different inorganic P fractions in bed sediments of the South Han River over a rainy season, through using the sequential extraction method. Non-apatite P was the dominant form, representing more than 60% of total inorganic P (TIP) content in sediments. Although no significant variation of TIP contents was observed, the proportion of bioavailable P in TIP pools decreased after the rainy season. The concentrations of individual inorganic P fractions ($NH_4Cl-P$, $NH_4F-P$, NaOH-P, and $H_2SO_4-P$) were significantly different across sites and after the rainy season (p < 0.05, two-way ANOVA). $NH_4F-P$ and NaOH-P concentrations in sediments increased in a downstream direction. After the rainy season, $NH_4Cl-P$ concentrations in sediments decreased whereas $NH_4F-P$ and $H_2SO_4-P$ concentrations increased. The redistribution of individual P fractions in sediments observed after rainy seasons were possibly due to the changing contribution of various sources of runoff and the variation in flow related particle size. Current estimation of P in bed sediments of the South Han River suggests a lower potential of internal P loading from sediments after the rainy season.

A study on hyphomycetous fungi found on Maejus, a raw material of Korean traditional soysources (메주에서 분리한 불완전균(Hyphomycetes)에 관한 연구)

  • Forty-eight fungal isolates were collected from the Korean traditional maejus and twenty-eight isolates of them were identified to be a hyphomycetous fungus(three genera and 20 species) Aspergillus flavus, A. flavus var columnaris, A. oryzae, A, oryzae var effusus, A. terreus, Scopula­riosis brevicaulis, Penicillium botryosum, P. gorlenkoanum, P. griseo-purpureum, P. citrinum, P. miczynskii, P. gaditanum, P. turolense, P. funiculosum, P. rubicundum, P. godlewskii, P. jensenii, P. roqueforti, P. volguense, P. verrucosum. Various 14 species of Penicillium were isolated from maejus, but all of them were not considered to be involved in the maeju fermentation. Otherwise, S. brevicaulis was especially collected from the maejus of various located areas, but were also related to the good quality of maejus under labratory conditions. A. flavus, A. oryzae P. tulolense, and P. funiculosum were also observed to be involved in the process of maeju.

In this paper, the hypotheses on A and B can be relaxed by usin a Hilbert-Schmidt operator X: Let A be p-quasihyponormal and let $B^*$ be invertible p-quasihyponormal such that AX = XB for a Hilbert-Schmidt operator X and $|||A^*|^{1-p}||{\cdot}|||B^{-1}|^{1-p}||\;{\leq}\;1$.Then $A^*X\;=\;XB^*$.

repABC- Type Replicator Region of Megaplasmid pAtC58 in Agrobacterium tumefaciens C58

  • The region responsible for replication of the megaplasmid pAtC58 in the nopaline-type Agrobacterium tumefaciens strain C58 was determined. A derivative ofa Co1E1 vector, pBluscript SK-, incapable of autonomous replication in Agrobacterium spp, was cloned with a 7.6-kb Bg1II-HindIII fragment from a cosmid clone of pAtC58, which contains a region adjacent to the operon for the utilization of deoxyfructosyl glutamine (DFG). The resulting plasmid conferred resistance to carbenicillin on the A. tumefaciens strain UIA5 that is a plasmidfree derivative of C58. The plasmid was stably maintained in the strain even after consecutive cultures for generations. Analysis of nested deletions of the 7.6-kb fragment showed that a 4.3-kb BglII-XhoI region sufficiently confers replication of the derivative of the ColE1 vector on UIA5. The region comprises three ORFs, which have high homologies with repA, repB, and repC of plasm ids in virulent Agrobacterium spp. including pTiC58, pTiB6S3, pTi-SAKURA, and pRiA4b as well as those of symbiotic plasmids from Rhizobium spp. Phylogenie analysis showed that rep genes in pAtC58 are more closely related to those in pRiA4 than to pTi plasmids including pTiC58, suggesting that the two inborn plasmids, pTiC58 and pAtC58, harbored in C58 evolved from distinct origins.
