Optimization of Environmental Parameters for Extracellular Chitinase Production by Trichoderma harzianum SJG-99721 in Bioreactor (Trichoderma harzianum SJG-99721의 체외 분비 chitinase 생산에 미치는 생물 반응기에서의 반응 최적화 연구)

  • A self-directing optimization procedure was applied to determine the best environmental factors in operating the bioreactor. The self-directing optimization process was employed to determine the best conditional combination of multi parameters, pH, temperature, aeration rate and mixing rate toy maximal production of chitinase by Trichoderma harzianum SJG-99721 in batch mode fermentation. Among these factors, the parameters of pH and aeration rate were found to be particularly important on mycellial growth and chitinase activity. pH 4.89, an aeration rate of 3.22 ι per minute and an agitation rate of 225 rpm was found to be the best combination. By the optimization, chitinase activity was dramatically increased from an initial value of 4.221 U under basic conditions to n final value of 16.825 U.

Circadian Changes in Pharmacokinetics of Sulfamethoxazole Administered Orally to Rabbits

  • Circadian variations of sulfamethoxazole pharmacokinetics were studied after a single oral administration of sulfamethoxazole, 50 mg/kg, to rabbits at 09:00 (a.m.) and 22 :00 (p.m.). The profiles of plasma sulfamethoxazole concentration showed from 6h to 24 h significant statistical difference (p<0.05) between 09:00 and 22:00. The half-life $t_{1/2} was significantly shorter in the morning (11.2 $\pm$3.2 h) when compared to the nighttime (15.4 $\pm$ 3.5h) (7< 0.05). The AUC was significantly decreased in the morning (1325 $\pm$ 264${u}g/ml$.h) than that in the nighttime (2059 $\pm$ 379${u}g/ml$. h) (p<0.05). Tota1 body clearance ($Cl_t$ was significantly higher when sulfamethoxazole was given in the morning (6.65 $\pm$ 0.23 ml/min) versus in the nighttime (4.28 $\pm$ 0.20 ml/min) (p<0.05).

Studies on Improved Amylases Developed by Protoplast Fusion of Aspergillus species

  • Improved amylases were developed from protoplast fusants of two amylase-producing Aspergillus species. Twenty regenerated fusants were screened for amylase production using Remazol Brilliant Blue agar. Crude enzyme extracts produced by solid state fermentation of rice bran were assayed for activity. Three variable factors (temperature, pH and enzyme type) were optimized to increase the amylase activity of the parents and selected fusants using rice bran medium and solid state fermentation. Analysis of this optimization was completed using the Central Composite Design (CCD) of the Response Surface Methodology (RSM). Amylase activity assays conducted at room temperature and 80℃ demonstrated that Aspergillus designates, T5 (920.21 U/ml, 966.67 U/ml), T13 (430 U/ml, 1011.11 U/ml) and T14 (500.63 U/ml, 1012.00 U/ml) all exhibited improved function making them the preferred fusants. Amylases produced from these fusants were observed to be active over the entire pH range evaluated in this study. Fusants T5 and T14 demonstrated optimal activity under acidic and alkaline conditions, respectively. Fusants T13 and T14 produced the most amylase at 72 h while parents TA, TC and fusant T5 produced the most amylase after 96 h of incubation. Response surface methodology examinations revealed that the enzyme from fusant T5 was the optimal enzyme demonstrating the highest activity (1055.17 U/ml) at pH 4 and a temperature of 40℃. This enzyme lost activity with further increases in temperature. Starch hydrolysis using fusant T5 gave the highest yield of glucose (1.6158 g/100 ml). The significant activities of the selected fusants at 28 ± 2℃ and 80℃ and the higher sugar yields from cassava starch hydrolysis over their parental strains indicate that it is possible to improve amylase activity using the protoplast fusion technique.

Microbiological Studies on the Rice Makkulli (Part 2) Nucleic Acid Degrading Enzymes and Their Related Substances during Rice Makkulli Koji Making (쌀막걸리의 미생물학적 연구 (제2보)쌀막걸리 제국중 핵산분해효소 및 핵산관련물질)

  • Changes of nucleic acid related substances and their enzymes during rice makkulli koji making were observed and enzymological properties of crude enzymes were examined. The results obtained were as follows : (1) The amounst of acid soluble phosphorus were increased, while no remarkable changes were observed in the component of total phosphorus during koji making. (2) AMP and IMP were increased, while ADP and ATP were decreased gradually in the course of process. (3) Activities of nucleic acid degrading enzymes were increased with the lapse of time. (4) In the crude enzyme solution extracted from rice makkulli koji, the optimal pH of RNase was 4.0~5.0 and those of PDase PNase were 5.0. (5) RNase and PMase were stable at the range of pH 4.0~5.0 and PDase was stable at the pH 4.0. (6) The optimal temperature of RNase was 55$^{\circ}C$, and that of PDase was at the range of 50~55$^{\circ}C$, and 5$0^{\circ}C$ for PMase. (7) Among the three enzymes, the heat stability was in order RNase, PDase and PMase, and especially PMase was so heat labile that it was almost inactivated at 7$0^{\circ}C$ for 10 min. (8) Inhibition by metal ions and other inhibitors was disclosed : C $u^{++}$ and Z $n^{++}$ inhibited the activity of RNase, and C $u^{++}$, NaF and N $a_2$HP $O_4$ inhibited that of PDase, while C $u^{++}$ and NaF inhibited the PMase activity.ctivity.

Influence of Moisture, pH, Depth of Burial and Submerged Conditions on Seed Germination and Seedling Emergence of Major Weed Species in Coconut Plantations of Sri Lanka

  • The laboratory and green house studies evaluated the effect of three different environmental factors on the seed germination, seedling emergence and survival of four major weed species in coconut plantations, Mimosa pudica, Ureana lobata, Panicum maximum and Pennisetum polystachyon. Germination percentage of all the weed species was significantly reduced with increasing soil moisture stress, no germination was observed at -0.9 MPa. Germination of both grass seeds ranged from 8% to 25% and 10% to 45% as moisture stress decreased from -0.4 MPa to 0 MPa, respectively. In contrast, seeds of M. pudica, and U. lobata were moderately tolerant to soil moisture stress and best adapted to moist environment. All the weed species seeds germinated over a wide range of soil pH values with the highest germination occurring at pH 6. In all the species, seedling emergence was declined rapidly with increasing depth with the exception of U. lobata. Seedling emergence significantly declined when the duration of flooding was three days or longer in dicotyledonous weed species and two days or longer in monocotyledonous weeds. This study illustrates the adaptability of these weeds to different environmental conditions which would enable the development of management strategies to reduce their populations below economic threshold levels in coconut plantations.

  • Serum levels of human placental lactogen have been measured by hemagglutination-inhibition reaction in 67 normal pregnant state and in 15 postpartum 24 hour state, HAIR is less sensitive and reliable method than radioimmunoassay, but simple, rapid, less expensive and fairly accurate, so it is more helpful in screening of large antenatal population with or without high risk complications. 1) Sensitivity of HPL-HAIR test kit was $0.1{\mu}g$/ml of H.P.L. serum level and had no cross reaction to HCG or male serum or non-pregenant female or newborn infant, 2) H.P.L. value was around $2{\mu}g$/ml until 24th week of pregnancy and rose to $6{\sim}8$ ${\mu}g$/ml continuously until about 36th week of pregnancy and then slightly decreased or stationary. 3) H.P.L. value in postpartum 24 hour state was undetectable. 4) There was poor correlation between maternal serum H.P.L. value at term and baby weight.

Optimization of Submerged Culture Conditions for Protease Production and Its Enzymatic Properties (Protease 생산을 위한 최적 배양조건 및 생산된 Protease의 특성)

  • This study was performed to investigate the optimum condition of protease production from submerged culture of oak mushroom (Lentinula edodes, Sanlim No. 5) and its enzymatic features. Among several combinations of media, the combination of wheat bran, corn flour, water and corn oil (WB+CF+W+ CO) yielded 84.8 U/g of maximum protease activity. This combination of ingredients, in spite of not being particularly protein-rich in comparison to the other media, allowed for good growth of the fungus and maximal protease production. Comparison of different growth medium liquids indicated that demineralized water afforded the best growth of the fungus and the highest protease activity. Acetate buffer and acidified water negatively affected The protease production peaked around 72 hr of incubation, and decreased thereafter. The molecular weights of produced protease were about 45,000 by Sephadex G-75 chromatography. The pH optimum for protease activity was 4, while maximal activity incubated at 37℃ for 1 hr was observed between pH 4~6. The optimum temperature of this protease was 55℃, and the enzyme was active over a broad temperature range (30~60℃), indicating that this protease would be suitable for a wide range of applications where. different pH and temperature are necessary, such as digestive aids, food industry, beer and tannery industries.

Effects of Human or Mouse Leukemia Inhibitory Factors on the Development of Bovine IVM/IVF Embryos (사람 및 생쥐 백혈병 억제인자가 소 체외성숙, 체외수정란의 발육에 미치는 효과)

  • The effects of human or mouse leukemia inhibitory factor(hLIF or mLIF) were examined as a means of increasing the development of in vitro matured(IVM) and in vitro fertilized (IVF) oocytes into morulae or blastocysts. Cell numbers of blastocysts were also counted using Hochest dye staining. Two-to 8-cell embryos derived from bovine IVM/IVF oocytes were cultured 5 to 6 days in CRI aa with or without mLIF or hLIF. All culture media were contained 3mg/ml bovine serum albumin. In experiment 1, the proportion of embryos developed to morulae and blastocysts in CRI aa containing 5,000U/ml mLIF(37.8%) was slightly higher than those of CRIaa containing 1,000U/ml mLIF(34.6%) and 0 U/ml mLIF(27.4%; P>0.05). In experiment 2, 0, 1,000 and 5,000U/ml of hLIF added to CR1aa media yielded 27.6%, 43.0% and 35.5% morulae and blastocysts, respectively(p>0.05). These were no significant increases in cell number among treatments(p>0.05). These results were indicating that mLIF or hLF can increase the proportion of embryos that develop into morulae and blastocysts without and increase in the cell number.

  • The conditions for ${\beta}$-galactosidase production from alkalophilic, thermophilic Bacillus sp. TA-11 were investigated. The maximal enzyme production was obtained when the strain was cultured at $50^{\circ}C$ for 5 days with fed-batch culture in the optimal medium containing 1.5% lactose, 0.6% yeast extract 0.15% $K_2HP0_4$and initial pH 9.5, and then final enzyme activity under the above conditions was 5200 unit/ml of cell free extract.
