In the course of screening for anti-acidosis and anti-diabetes agent from natural products, the inhibitory activities of Nepales plant extracts against microbial $\alpha$-amylase and $\alpha$-glucosidase were evaluated. Among the 100 different kinds of ethanol extracts, Cedrus deodara (Roxb.) G. Don and Myrica nagi Thunb showed strong inhibition activities against $\alpha$-amylase. The $IC_{50}$ values of C. deodara (Roxb.) G. Don, M. nagi Thunb and acarose, a commercial available anti-diabetes agent, were 44.5, 47.5 and $50.5\;{\mu}g/mL$, respectively. Considering the crude extract of C. deodara (Roxb.) G. Don, and M. nagi Thunb, these extracts have strong potentials as anti-acidosis or anti-diabates agent. In a while, Cleistocalyx operculatus (Roxb.) extract showed a good inhibition activity to $\alpha$-amylase and $\alpha$-glucosidase, even it was recently reported. The selected three extracts did not show any hemolysis activity against human red blood cell up to 1 mg/mL, and the inhibition activities were maintained by heat or acid treatment. Moreover, treatment of HCl (0.01N) for 1 h into C. operculatus (Roxb.) and M. nagi Thunb increased the inhibition activity from 50% to 70%. Our results suggest that C. deodara (Roxb.) G. Don, M. nagi Thunb, and C. operculatus (Roxb.) are potential as anti-acidosis and anti-diabetes agents.

In this study, we examined the anti-diabetic activity in vitro by the mixed Korean herb water extracts(JDG) composed of Lycium chinense, Cordyceps militaris, and Acanthopanax senticosus on hepatic glucose-regulating enzyme activities such as glucokinase(GK), acetyl-CoA carboxylase(ACC), and inhibition activity of $\alpha$-glucosidase. The hepatic cytosol fraction of a type II diabetic animal(Goto-Kakizaki rat) was used in GK and ACC activity assays. JDG mixed water extracts significantly increased the hepatic GK and ACC activity. The highest anti-$\alpha$-glucosidase activity was observed in JDG 5 water extract when compared to the acarbose control, inhibitor of $\alpha$-glucosidase. We suggest that Lycium chinense, Cordyceps militaris, and Acanthopanax senticosus mixed water extracts may exert an anti-diabetic activity by enhancing the glucose metabolism and may be used as natural $\alpha$-glucosidase inhibitors in type 2 diabetic conditions.

To develop anti-acidosis and anti-diabetes agentsfrom natural products, the inhibitory activities of Brazilian plant extracts against microbial $\alpha$-amylase and $\alpha$-glucosidase were evaluated. Among 100 different ethanol extracts tested, those of Acacia jurema Mart., Anacardium humile A. St.-Hil., Cedrela odorata L., and Guazuma ulmifolia Lam showed good inhibitoryactivities toward both enzymes. In addition, an extract of Plumeria drastica Mart. showed specific inhibition of $\alpha$-amylase, whereas that of Eugenia uniflora L. demonstrated strong inhibition of the enzyme. IC50 values of $\alpha$-amylase inhibition suggested that the extract of A. humile A. St.-Hil., which has been used as an anti-diabetes medicine in Brazil, had potent inhibitory activity. The IC50 for the A. humile A. St.-Hil. extract ($91.2{\mu}g/mL$) was similar to that of acarbose ($50.5{\mu}g/mL$). This activity of A. humile A. St.-Hil. was not reduced by heat or acid treatment. Moreover, treatment with HCl (0.01 M) for 1 h increased the inhibitory activity from 57.5% to 81.2%. Also, the extract did not cause hemolysis of human red blood cells at levels up to 1 mg/mL. The results indicate that the extract of A. humile A. St.-Hil. is potentially useful as an anti-acidosis and anti-diabetes agent.
Ethanol extracts from the edible mushroom hinmogi (Tremella fuciformis, TF) were used in the investigation of effects on $\alpha$-glucosidase in vitro and on glucose-uptake in 3T3-L1 mature adipocytes. Addition of the extract significantly inhibited $\alpha$-glucosidase from small intestine of porcine and of rat (about 42 and 35% of control, respectively), and stimulated glucose uptake (about 100% of control), of which activity was higher than that of maitake (Grifola frondosa) X-fraction, a well known anti-diabetic substance. When the ethanol extracts were further partitioned successively by organic solvents and purified by silica gel chromatography, the non-polar layer (F-7) from hexane layer showed highest stimulatory activity of glucose-uptake among layers tested. The major components of the F-7 were 1-monooleoylglycerol and 1-monopalmitoylglycerol. Our report is the first description of TF with stimulatory activity of glucose-uptake. These results suggest that TF extracts may constitute a new source of glucose transport activator and could be employed as a potential anti-diabetic material for treatment and preventing diabetes.
To evaluate the anti-oxidant and anti-tumor potential of the green pepper (Capsicum annuum L. cv. DangZo), total polyphenol content, radical scavenging activities and anti-tumor properties were measured. The total polyphenol content of the 70% ethanol extracts from green pepper (Capsicum annuum L. cv. DangZo) was 30.29 mg gallic acid equivalent/g extract. The DPPH radical and hydroxyl radical scavenging activities of 70% ethanol extracts of green pepper (Capsicum annuum L. cv. DangZo) were documented at 2.87 and 10.55, respectively. For ${\alpha}$-glucosidase and ${\alpha}$-amylase inhibitory activity, 70% ethanol extracts of green pepper (Capsicum annuum L. cv. DangZo) were documented at 35.67% and 58.41% respectively. The green pepper (Capsicum annuum L. cv. DangZo) demonstrated greater capability in terms of anti-neoplastic activity vis-a-vis colon cancer cell lines when compared to other cancer cell lines.s. er (Capsicum annuum L. cv. DangZo) higher activities of anticancer activities on colon cancer cell lines compared to other cancer cell lines.
In order to develop a new anti-diabetic ${\alpha}$-glucosidase inhibitor, we compared the ${\alpha}$-glucosidase inhibitory activity of the cell-free extracts of 48 strains of yeasts isolated from Korean fermented foods, and found that Pichia burtonii Y257-7 exhibited the highest ${\alpha}$-glucosidase inhibitory activity of 55.6%. The ${\alpha}$-glucosidase inhibitor was maximally produced when Pichia burtonii Y257-7 was cultured in LB broth (initial pH of 6.0) at $28^{\circ}C$ for 24 h. The ${\alpha}$-glucosidase inhibitor, partially purified by Sephadex G-50 gel permeation chromatography and systematic solvents extraction, revealed potent hypoglycemic effects in normal rats and streptozotocin-induced diabetic rats after the oral administration of starch.

The purpose of this study was to measure total phenolic compounds as a measure of antioxidant activity as well as ${\alpha}$-amylase inhibitory and ${\alpha}$-glucosidase inhibitory activities as a measure of anti-diabetic efficacy in methanol extracts from 23 kinds of medicinal plants. Extracts of three medicinal plant species showing high total polyphenol contents were selected (Euonymus alatus stem, Taxus cuspidata fruit, and Eucommia ulmoides leaf). Extracts of six medicinal plant species showing over 60% DPPH radical scavenging activity were also selected [Eucommia ulmoides barks (80.10%), Lycium chinense roots (64.25%), Euonymus alatus stem (73.59%), Lespedeza cuneata (78.20%), Taxus cuspidata fruits (70.52%), and Tilia taquetii leaf and stem (67.81%)]. Regarding ${\alpha}$-glucosidase and ${\alpha}$-amylase inhibitory activities acarbose showing approximately 80% inhibitory activity was selected as a control group, and six species (Eucommia ulmoides heartwood, Eucommia ulmoides bark, Euonymus alatus stem, Dioscorea batatas, Coix lachryma-jobi, and Phaseolus radiatus) showed greater than 80% ${\alpha}$-glucosidase inhibitory activity. Extracts of nine medicinal plant. species showing over 80% ${\alpha}$-amylase inhibitory activity (Pueraria thunbergiana root, Eucommia ulmoides bark, Eucommia ulmoides leaf, Lycium chinense fruits, Euonymus alatus leaf and stem, Euonymus alatus stem, Sasa borealis whole, Dioscorea batatas leaf and stem, and Tilia taquetii leaf and stem). Based on these results, medicinal plants showing high antioxidant and antidiabetic activities can be used as fundamental products in developing new medicines, as well as functional foods to prevent adult disease.

The physiological activities of Strychnos nux-vomica extracts were investigated through the total phenolic contents (TPC), 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical scavenging activity (RSA), comet assay, and $\alpha$-glucosidase inhibitory activity. S. nux-vomica extracts were prepared with methanol, ethanol, and acetone. The methanol extract showed the highest phenolic content (71 mg/100 g gallic acid equivalents). Pretreatment with S. nux-vomica extracts resulted in significant reductions in oxidative DNA damage at all of the concentrations tested ($1{\sim}50\;{\mu}g/mL$). The $\alpha$-glucosidase inhibitory activity of a methanol extract was 12.8% at the concentration of 1 mg/mL. Therefore, these results indicate that S. nux-vomica might be a noble potential candidate exhibiting antioxidant and $\alpha$-glucosidase inhibitory activity.
After a mixed carbohydrate diet, inhibition of ${\alpha}$-amylase and ${\alpha}$-glucosidase involved in the digestion and absorption of carbohydrates can significantly decrease the postprandial increase of blood glucose level. In the course of screening these useful enzyme inhibitors, we selected five kinds of bean, using an in-vitro enzyme inhibition assay method. To evaluate the effect of germination process on the functionality of the bean, we investigated the inhibitory activities of the water extracts of non-germinated bean and germinated bean against ${\alpha}$-amylase and ${\alpha}$-glucosidase, relevant to postprandial hyperglycemia. We also investigated the oxygen radical absorbance capacity(ORAC), total phenolics content, and postprandial blood glucose lowering effect in rats(Sprague-Dawley rat model). Most germinated beans showed significantly higher ${\alpha}$-glucosidase inhibitory activity, compared with non-germinated beans. Among germinated beans, Glycine max had the highest ${\alpha}$-glucosidase inhibitory activity(53.3%). The water extract of germinated Phaseolus vulgaris L. had the highest ${\alpha}$-amylase inhibitory activity(95.1%), followed by Glycine max(58.7%), and Glycine max L. Merr(54.1%). Furthermore, the five germinated beans also showed high antioxidant activities in ORAC assay. Results suggested that the germination process may improve and enhance the anti-hyperglycemia potential and antioxidant activity of the bean.
We studied the total polyphenol content, DPPH radical scavenging activity, hydroxyl radical scavenging activity and ${\alpha}$-glucosidase inhibition of water extracts from 17 medicinal plants. Total polyphenol contents ranged from 10.0 (Coix lachryma-jobi L, CL) ~ 279.7 (Perilla sikokiana, PS)mg/g. The water extract from medicinal plants were evaluated for its free radical scavenging activities and compared with a commercial antioxidant, ascorbic acid. DPPH radical scavenging activity of Pyrus pyrifolia (PP), Chamaecyparis obtusa L. (COL), Chamaecyparis obtusa F. (COF), and PS were higher than positive control. Higher hydroxyl radical scavenging activity were shown in Acanthopanax senticosus (AS) and Cordyceps militaris (CM) than the other plants. The highest anti-${\alpha}$-glucosidase activity was observed in Cornus officinalis (CO) and Paeonia suffruticosa Andrews (PSA) water extracts. PSA showed not only the higher DPPH radical scavenging activity but also the anti-${\alpha}$-glucosidase activity. The results of our study that PP, COL, COF, PS, AS, CM, CO and PSA could be potential candidates for natural antioxidants.
