Identification of Differentially Expressed Genes in Human Small Cell Lung Carcinoma Using Subtractive Hybridization

  • Lung cancer is a leading cause of cancer death worldwide; however, despite major advances in cancer treatment during the past two decades, the prognostic outcome of lung cancer patients has improved only minimally. This is largely due to the inadequacy of the traditional screening approach of diagnosis in lung cancer, which detects only well­established overt cancers and fails to identify precursor lesions in premalignant conditions of the bronchial tree. In recent years this situation has fundamentally changed with the identification of molecular abnormalities characteristic of premalignant changes; these concern tumour suppressor genes, loss of heterozygosity at crucial sites and activation of oncogenes. Basic knowledge at the molecular level has extremely important clinical implications with regard to early diagnosis, risk assessment and prevention, and therapeutic targets. In this study we used a 'cap-finder' subtractive hybridization method, 'long distance' polymerase chain reaction (PCR), streptavidin magnetic beads mediated subtraction, and spin column chromatography to detect differential expression genes of human small cell lung carcinoma. We have now isolated ninety two genes that expressed differentially in the human small cell lung carcinoma cells and analyzed of 12 clones with sequencing, nine cDNAs include tapasin (NGS-17) mRNA, BC200 alpha scRNA, chromosome 12q24 PAC RPCI3-462E2, protein phosphatase 1 (PPPICA), translocation protein 1 (TLOC1), ribosomal protein S24 (RPS24) mRNA, protein phosphatase (PPEF2), cathepsin Z, MDM2 gene and three novel genes. They may be oncogenesis­related proteins.

Construction of New Transfer Vector of Nuclear Polyhedrosis Virus of the Silkworm, Bombyx mori (누에 핵다각체병 바이러스를 이용한 새로운 전이 벡터의 제작)

  • In order to develope baculovirus expression vector system, we constructed new transfer vector of nuclear polyhedrosis virus of the silkworm, Bombyx mori. The promoter region containing only adenine of translation start codon of polyhedrin gene was cloned by polymerase chain reaction technique. And the 5' and 3' leader regions of polyhedrin gene was sequentially cloned. The polyhedrin coding gene was deleted from the +2 to the +597 position. As the result, we constructed new transfer vector which has EcoRI, SacI and KpnI sites for the cloning sites of foreign gene. New transfer vector was named as pBmKSKl. Escherichia coli $\beta$-galactosidase gene as foreign gene was inserted into pBmKSKl, under the control of the polyhedrin promoter and expressed in B. mori cells. The result showed that the new transfer vector pBmKSK1 is functional.

Characteristics of Cucumber mosaic virus isolated from Zea mays in Korea

  • A virus causing mottle and stunt symptom on Zea mays was observed around Ulleng-do, Korea and identified as Cucumber mosaic virus (CMV-ZM) based upon biological, serological, and molecular characteristics. In host range studies, the CMV-ZM isolate produced local lesions on Datura stramonium, Vigna unguiculata, Cucurbita moschata, Chenopodium amaranticolor, Ch. quinoa, whereas this isolate produced systemic mosaic on Nicotiana tabacum cv. 'Xanthi-nc', Capsicum annuum, Solanum lycopersicum, Solanum melongena, Cucurbita pepo, and Z. mays. In addition, chlorotic local rings on inoculated leaves along with severe mosaic, malformation, and fern leaf symptoms on upper systemic leaves were shown in N. glutinosa plants. Complete nucleotide sequences of each genomic RNA segment was determined and compared to those of the other CMV strains. Comparison of the nucleotide sequence of 1a open reading frame (ORF) revealed approximately 89.2-92.4% sequence identity with each CMV subgroup IA and IB strain, while showing only 78% sequence identity with CMV subgroup II. Nucleotide sequence analysis of RNA2 ORFs revealed 85.3-97.6% sequence identity with subgroup I. In ORFs of RNA3, levels of nucleotide sequence identities were higher than 92-99.2% with CMV subgroup I and lower than 82% with CMV isolates of subgroup II. These results suggest that CMV-ZM isolate is more closely related to subgroup I than subgroup II and therefore, CMV-ZM isolate might be classified into as CMV subgroup I based on biological and molecular analysis.

Carboxylic Acids as Biomarkers of Biomphalaria alexandrina Snails Infected with Schistosoma mansoni

  • Biomphalaria alexandrina snails play an indispensable role in transmission of schistosomiasis. Infection rates in field populations of snails are routinely determined by cercarial shedding neglecting prepatent snail infections, because of lack of a suitable method for diagnosis. The present study aimed at separation and quantification of oxalic, malic, acetic, pyruvic, and fumaric acids using ion-suppression reversed-phase high performance liquid chromatography (HPLC) to test the potentiality of these acids to be used as diagnostic and therapeutic biomarkers. The assay was done in both hemolymph and digestive gland-gonad complex (DGG) samples in a total of 300 B. alexandrina snails. All of the studied acids in both the hemolymph and tissue samples except for the fumaric acid in hemolymph appeared to be good diagnostic biomarkers as they provide not only a good discrimination between the infected snails from the control but also between the studied stages of infection from each other. The most sensitive discriminating acid was malic acid in hemolymph samples as it showed the highest F-ratio. Using the Z-score, malic acid was found to be a good potential therapeutic biomarker in the prepatency stage, oxalic acid and acetic acid in the stage of patency, and malic acid and acetic acid at 2 weeks after patency. Quantification of carboxylic acids, using HPLC strategy, was fast, easy, and accurate in prediction of infected and uninfected snails and possibly to detect the stage of infection. It seems also useful for detection of the most suitable acids to be used as drug targets.

Infections with Digenean Trematode Metacercariae in Two Invasive Alien Fish, Micropterus salmoides and Lepomis macrochirus, in Two Rivers in Chungcheongbuk-do, Republic of Korea

  • Present study was performed to survey infection status of digenetic trematode metacercariae in 2 alien fish species, Micropterus salmoides (largemouth bass) and Lepomis macrochirus (bluegill), in 2 rivers draining Chungcheongbuk-do, Republic of Korea. A total of 107 largemouth bass and 244 bluegills were caught in Daecheong-ho (ho=lake) and Musim-cheon (a branch of Geum-gang), in Chungcheongbuk-do April-July 2015. Additionally, 68 native fish of 5 species, i.e., Zacco platypus, Hemibarbus longirostris, Carassius auratus, Pseudogobio esocinus and Puntungia herzi, were caught from the same water bodies. All of the fish collected were examined by artificial digestion method. The metacercariae of Centrocestus armatus, Clinostomum complanatum, Metagonimus sp. and Diplostomum spp. were detected from 4 out of 5 native fish species in Daecheong-ho. However, any metacercariae were not found from 87 M. salmoides and 177 L. macrochirus in Daecheong-ho. In Musim-cheon, metacercariae of Exorchis oviformis and Metacercaria hasegawai were detected from 78% Z. platypus and 34% L. macrochirus, but any metacercariae not found in M. salmoides. We report here that the 2 alien fish species were less infected with the metacercariae than the native ones. Surveys on the metacercariae in the alien fish species in geographically various rivers should be undertaken for better understanding on the role of alien fish species in the trematode infections in Republic of Korea.

The Growth and Nutrient Removal Efficiency of Hydrophytes at an Artificial Vegetation Island, Lake Paldang (팔당호 인공 수초재배섬에서 수생식물의 생장 및 영양염류 제거 효율)

  • We investigated temporal changes of composition, habitat area, growth rate and elements content of hydrophytes at the artificial vegetation island (AVI) in Kyungan Stream within Lake Paldang. We also assessed nutrient removal rate through cutting off the emergent part of hydrophytes. The kinds of hydrophytes have increased from four species (P. australis, P. japonica, T. angustifolia and the Z. latifolia) at initial stage of installation to more than 29 species for 6-year operation. P. japonica was most dominant species at the AVI in 2006. The habitat area of have increased about 2.5 times compared to the initial planting area, occupying 63% of AVI's vegetation area. The incoming species of S. fluviatilis and B. frondosa have adapted successively and expanded habitat area in the AVI. The relative growth rate of P. japonica and P. australis was highest in spring sprouting period. Their hights and weights have increased until summer, by the time they were cutting off the emergent part. They started regrowth immediately after cutting and continued to grow until September and withered away in November. The carbon contents of P. japonica and P. australis have increased during growth phase, on the contrary, the nitrogen and phosphorus contents have decreased. By cutting off and removing the emergent part (leaves and branches) of hydrophytes twice from AVI, $17.6gN/m^2/y$ of nitrogen and $1.3gP/m^2/y$ of phosphorus was removed from AVI in 2006.

Fish Fauna and Community Structure in Lake Paldang and its Inflows (팔당호의 어류상과 군집동태)

  • Fish fauna and community structure were investigated at 2003, 2004 and 2006 in Lake Paldang and its inflows. In this survey, we collected 54 species belong to 14 families. Family Cyprinidae take 51.9% (28 species), Cobitidae and Gobiidae occupied 7.4% (each 4 species), respectively. E. erythropterus (11.8%) was dominant species and L. macrochirus (10.2%), designated as an ecosystem-invasive alien fish species by Ministry of Environment, M. yaluensis (9.1%), Z. platypus (7.7%) and R. brunneus (6.7%) were dominated in turn. H. molitrix, C. lutheri, P. altivelis, S. microdorsalis, L. costata. S. gracilis majimae, A. rivularis, P. koreanus. S. scherzeri and O. platycephala were rare species (less than 0.1% in relative abundance). In-lake area, 36 species of fishes belonging to 11 families were collected, and 48 species of 13 families were from inflows. The number of Korean endemic species were 19 species (35.2%), and 4 kinds of exotic species (7.4%), that is, C. cuvieri, H. molitrix, L. macrochirus and M. salmoides were found at this survey area. It was revealed by the analysis of fish community that diversity and richness indices were prominent at tributaries, and dominance index was high at the main body of Lake Paldang. Fish fauna showed rapid decline after dam construction which make the mid-Han river systems to lentic ecosystem, so many of meander-riffle fishes were disappeared dramatically. But after 1990s when installation of small scale wastewater treatment plants started in watershed, the water quality of small tributaries was improved. And also, expansion of the natural macrophytic vegetation of littoral zone should contribute to the development of diverse fish fauna in Lake Paldang.

Nitrogen adsorption on the stepped planes of tungsten: II. W(210) and W(310) plane (계단형 텅스텐 결정면의 질소 흡착에 관한 연구: II. W(210) 및 W(310)면)

  • The heat of desorption and the work function change induced by nitrogen adsorption on the slepped tungstein surface plants, W(210) and W(310), are measured using the Field Electron Emission Microscope(FEM). The adsoption sites are predicted from the Thermal Desortion Spectra(TDS). The wirk function change of both W(210) and W(310) planes increase as increasing the nitrogen dose and saturates at the nitrogen dose about 5 Langmuir to 0.29 eV and 0.20 eV respectively. We find three adsorption site on each plane for the low dose range. The TDS result shows that the intensity of $\alpha_1$ state on W(310) is much stronger than that of $\alpha_1$ state on W(210), and the direction of nitrogen dipole moment adsorbed on the sites correspond to $\alpha_1$ and $\beta_2$ state on W(210) and W(310) planes are in the opposite direction to that of the equivalent states on W(100) plane. From this observation we can predict the relative atomic position in the z-direction (perpendicular direction to the surface) of nitrogen molecules/atoms adsorbed on these sites.

Optical and Electrical Properties of Fluorine-Doped Tin Oxide Prepared by Chemical Vapor Deposition at Low Temperature (저온 증착된 불소도핑 주석 산화 박막의 광학적·전기적 특성)

  • The electrical and optical properties of fluorine-doped tin oxide films grown on polyethylene terephthalate film with a hardness of 3 using electron cyclotron resonance plasma with linear microwave of 2.45 GHz of high ionization energy were investigated. Fluorine-doped tin oxide films with a magnetic field of 875 Gauss and the highest resistance uniformity were obtained. In particular, the magnetic field could be controlled by varying the distribution in electron cyclotron deposition positions. The films were deposited at various gas flow rates of hydrogen and carrier gas of an organometallic source. The surface morphology, electrical resistivity, transmittance, and color in the visible range of the deposited film were examined using SEM, a four-point probe instrument, and a spectrophotometer. The electromagnetic field for electron cyclotron resonance condition was uniformly formed in at a position 16 cm from the center along the Z-axis. The plasma spatial distribution of magnetic current on the roll substrate surface in the film was considerably affected by the electron cyclotron systems. The relative resistance uniformity of electrical properties was obtained in film prepared with a magnetic field in the current range of 180~200A. SEM images showing the surface morphologies of a film deposited on PET with a width of 50 cm revealed that the grains were uniformly distributed with sizes in the range of 2~7 nm. In our experimental range, the electrical resistivity of film was able to observe from $1.0{\times}10^{-2}$ to $1.0{\times}10^{-1}{\Omega}cm$ where optical transmittance at 550 nm was 87~89 %. These properties were depended on the flow rate of the gas, hydrogen and carrier gas of the organometallic source, respectively.

Synthesis and Biological Evaluation of Novel GSK-3β Inhibitors as Anticancer Agents

  • A series of isoxazol-indolin-2-one was designed for GSK-3${\beta}$ inhibitors as novel anticancer agents based on their binding mode analysis in GSK-3${\beta}$ crystal structure. Total 21 compounds were synthesized and evaluated for their inhibitory activity against two tumor cell lines (DU145 and HT29). Most of the synthesized compounds were potent with above 80% inhibitory activity at 100 ${\mu}M$, and several compounds were examined for inhibitory activity against GSK-3${\beta}$. Among them, 15(Z) ($R_1$=H, $R_2$=3-Cl-phenyl) was most active with 78% inhibition of tumor cell line (HT29) at 20 ${\mu}M$ and 72% inhibition of GSK-3${\beta}$ at 20 ${\mu}M$.
