Antisense gibberellin 3β-hydroxylase발현 형질전환벼 (Antisense GA 3β-Hydroxylase Gene Transferred to Rice Plants.)

  • GA 생합성에 결정적 역할을 하는 GA 3$\beta$-hydroxylase를 pIG121-Hm 벡터에 GUS유전자를 빼고 antisense로 클로닝하여 이를 동진벼에 도입한 결과 17개체의 절간신장이 억제된 형질전환한 식물체를 얻을 수 있었다. 일반 재배 동진벼를 대조군으로 하여 비교하였을때 antisense GA 3$\beta$-hydroxylase 유전자가 형질 도입된 식물체의 획득형질은 평균적으로 대조군에 비해 절간 신장의 억제가 확인되었다. 절간신장의 억제가 보인 개체의 엽육조직을 co취하여 Southen blot hybridization분석 결과 3개의 line에서 모두 single copy로 도입된 것으로 나타났다. 이로써$T_o$ 식물체 내에 antisense GA 3$\beta$-hydroxylase 유전자를 내포하고 있는 것으로 확인되었다. 이것은 antisense GA 3$\beta$-hydroxylase 유전자가 생체내에서 직접 또는 간접적으로 GA 3$\beta$-hydroxylase유전자의 발현에 관여한 것이라 사료되어진다.

Generation of Hydrogen Peroxide by Single-Atom Clusters Pd Anchored on t-BaTiO3 for Piezoelectric Degradation of Tetracycline

  • Single-atom Pd clusters anchored on t-BaTiO3 material was synthesized using hydrothermal and ultrasonic methods for the effective piezoelectric catalytic degradation of pollutants using vibration energy. XRD patterns of BaTiO3 loaded with monoatomic Pd were obtained before and after calcining, and showed typical cubic-phase BTO. TEM and HAADF-STEM images indicated single-atom Pd clusters were successfully introduced into the BaTiO3. The piezoelectric current density of the prepared Pd-BaTiO3 binary composite was significantly higher than that of the pristine BaTiO3. Under mechanical vibration, the nanomaterial exhibited a tetracycline decomposition rate of ~95 % within 7 h, which is much higher than the degradation rate of 56.7 % observed with pure BaTiO3. Many of the piezo-induced electrons escaped to the Pd-doped BaTiO3 interface because of Pd's excellent conductivity. Single-atom Pd clusters help promote the separation of the piezo-induced electrons, thereby achieving synergistic catalysis. This work demonstrates the feasibility of combining ultrasonic technology with the piezoelectric effect and provides a promising strategy for the development of ultrasonic and piezoelectric materials.

아염소산나트륨의 무격막 전기분해에 의한 이산화염소 생성: 양전극 재질에 따른 영향 (Electrochemical Generation of Chlorine Dioxide from Sodium Chlorite Using Un-Divided Electrochemical Cell: Effect of Anode Materials)

  • 아염소산나트륨($NaClO_2$)의 무격막 전기분해(un-divided electrolysis)에 의한 이산화염소(chlorine dioxide; $ClO_2$) 제조에서 양전극(anode) 재질에 따른 이산화염소수 발생특성을 조사하였다. 양전극으로는 $IrO_2$-coated Ti, $RuO_2$-coated Ti, DSA(dimensionally stable anode) 전극을 사용하였으며, 음전극으로는 Pt-coated Ti 전극을 사용하였다. 다양한 양전극을 사용한 무격막 전해셀(un-divided electrochemical cell) 시스템에서 이산화염소의 전구체인 아염소산나트륨 ($NaClO_2$) 농도, 전해질로 사용된 염화나트륨(NaCl) 농도 그리고 전구체 용액의 전해셀 체류시간(cell residence time;$t_R$), 전구체 용액의 초기 pH 그리고 무격막 전해셀에 공급된 전류(current; A)와 같은 운전 파라미터가 이산화염소수 발생에 미치는 영향을 조사하고 최적 운전조건을 도출하였다. $IrO_2$-coated Ti, $RuO_2$-coated Ti 그리고 DSA 양전극 시스템에서 최적 전해셀 체류시간은 각각 약 2.27, 1.52, 1.52 s, 전구체 용액의 초기 pH는 약 2.3, 최적 아염소산나트륨 농도는 $IrO_2$-coated Ti와 $RuO_2$-coated Ti 양전극 시스템이 약 0.43 g/L, DSA 양전극 시스템이 약 0.32 g/L 그리고 최적전해질 농도는 약 5.85 g/L로 나타났으며 무격막 전해셀에 공급된 최적 전류는 약 0.6 A로 나타났다. 산출된 최적 무격막 전해셀 조건에서 이산화염소수 발생을 위한 $IrO_2$-coated Ti, $RuO_2$-coated Ti 그리고 DSA 양전극 시스템의 전류효율(current efficiency; C.E.%)과 에너지 소모율(energy consumption; E.C. $W{\cdot}hr/g-ClO_2$)은 각각 약 79.80, 114.70, 70.99% 그리고 1.38, 1.03, $1.61W{\cdot}hr/g-ClO_2$로 나타났다.

12Cr 강의 이동 화염경화 공정 특성 (Characteristics of Flame Hardening Process for 12Cr Steels)

  • In this study, the movable flame hardening process of 12Cr steel for a uniform hardness and desirable residual stress have been investigated. For this, the temperature cycles have been controlled accurately as a function of the three processing variables, the flame intensity $I_f$, the scanning velocity $V_s$, and the initial flame holding time $t_h$, where the standard surface temperature $T_{s,\;max}$, was maintained at $960^{\circ}C$. The optimized conditions were $V_s=0.68mn/s\;and\;t_h=67sec$ for the $C_3H_8:O_2\;=\;5:20l/min,\;V_s=0.80mm/s$ and $t_h=56sec$ for the $C_3H_8:O_2=6:24l/min,\;V_s=1.01mm/s\;and\;t_h=48sec$ for the $C_3H_8:O_2=7:28l/min,\;and\;V_s=1.15mm/s$ and $t_h=39sec$ for the $C_3H_8:O_2$=8:32 l/min. The optimally flame-hardened surface exhibited uniform distributions of the hardness and residual compressive stress over the treated area with moderate levels of $470{\sim}490HV_{0.2}$in hardness and $-300{\sim}-450MPa$ in residual stress, which were acceptable on the basis of the acceptance criteria of Siemens AG-KWU and GE Power Generation Engineering.

화력발전소 효율개선 측정에 대한 디비지아분해기법의 적용 (An Application of Divisia Decomposition Analysis to the Measurement of Thermal Efficiency Improvement of Power Generation)

  • Since improved thermal efficiency reduces capacity requirements and energy costs, electricity producers often treat thermal efficiency as a measure of management or economic performance. The conventional measure of the thermal efficiency of a fossil-fuel generation system is the ratio of total electricity generation to the simple sum of energy inputs. As a refined approach, we present a novel thermal efficiency measure using the concept of the Divisia index number. Application of this approach to the Korean power sector shows improvement of thermal efficiency of 1.1% per year during 1970-1998. This is higher than the 0.9% improvement per year given by the conventional method. The difference is attributable to the effect of fuel substitution. In the Divisia decomposition context, we also show the limitations of the popular $T{\ddot{o}}rnqvist$ index formula and the superiority of the Sato-Vartia formula.

펄스 전류 하에서 AZ31 마그네슘 합금의 플라즈마전해산화 피막의 형성 거동 (PEO Film Formation Behavior of AZ31 Mg Alloy under Pulse Current)

  • In this study, PEO (plasma electrolytic oxidation) film formation behavior of AZ31 Mg alloy under application of 300 Hz pulse current was studied by the analyses of V-t curve, arc generation behavior, PEO film thickness and morphology of PEO films with treatment time in 0.05 M NaOH + 0.05 M Na2SiO3 + 0.1 M NaF solution. PEO films was observed to grow after 10 s of application of pulse current together with generation of micro-arcs. PEO film grew linearly with treatment time at a growth rate of about 5.58 ㎛/min at 200 mA/cm2 of pulse current but increasing rate of film formation voltage became lowered largely with increasing treatment time after passing about 250 V, suggesting that resistivity of PEO films during micro-arc generation decreases with increasing film formation voltage at more than 250 V.

  • As the maximum power operating point (MPOP) of photovoltaic (PV) power generation systems changes with varying atmospheric conditions such as solar radiation and temperature, an important consideration in the design of efficient PV system is to track the MPOP correctly. Although the efficiency of these Maximum Power Point Tracking algorithms is usually high, it drops noticeably in case of rapidly changing atmospheric conditions. This paper describes common MPPT control algorithm: Constant Voltage Control, Perturbation and Observation(P&O), Incremental Conductance (IncCnd) and proposes a new MPPT algorithm based on P&O algorithm. The conception and control principles of the proposed MPPT method are explained in detail and its validity of the proposed method is verified through several simulated results. As it doesn't use digital signal processor, this MPPT method has the merits of both a cost efficiency and a simple control circuit design. Therefore, it is considered that the proposed MPPT method is proper to low power, low cost PV applications.

Lipopolysaccharide로 산화 스트레스를 유도한 Mouse에서 사보환(四補丸)의 Peroxynitrite 억제 효과 (Peroxynitrite Scavenging Activity of Sabohwanin Lipopolysaccharide-Induced Oxidatively-Stressed Mice)

  • Objectives : Peroxynitrite (ONOO-), superoxide anion radical (?O2-) and nitric oxide (NO) are cytotoxic because they can oxidize several cellular components such as proteins, lipids and DNA. They have been implicated in the aging process, and age-related diseases such as Alzheimer's disease, rheumatoid arthritis, cancer and atherosclerosis. The aim of this study was to investigate Sabohwan's activity for scavenging ONOO- and its precursors. NO and ?02-. Methods : For this study, the fluorescent probes, namely 2',7'-dichlorodihydrofluorescein diacetate (DCFDA), 4.5-diaminofluorescein (DAF-2) and dihydrorhodamine 123 (DHR 123) were used. Results : Sabohwanblocked tert-butylhydroperoxide (t-BHP)-induced cell death in a dose-dependent fashion. It scavenged t-BHP-induced ONOO-, NO and ?O2- in YPEN cells. Sabohwan inhibited the generation of ONOO-, NO and ?O2- in the lipopolysaccharide (LPS)-treated mouse kidney postmitochondria both in vitro and in vivo. The lipid peroxide level increased and glutathione level decreased in the LPS-treated mice, whereas the ones in the Sabohwanadministered group among the LPS-treated mice reversed toward their natural levels. Conclusions : These results suggest that Sabohwanis an effective ONOO-, ?O2- and NO scavenger, and thereby it might have a potential role as a therapy against the aging process and age-related diseases.

A/R 코팅 변화에 따른 200MHz AOM의 laser damage threshold 증가 (Laser Damage Threshold Increase of A/R Coating Films for 200MHz AOM)

  • 200MHz 응향광변조기 (AOM: Acousto-Optic Modulator)는 제 2고조파(SHG: Second Harmonic Generation)녹색 레이저와 함께 DVDR(Digital Video Disk Recorder)에 적용되어 고밀도 광기록용으로 사용되었다. 이러한 고밀도 광기록 장치로써 사용되기 위해서는 고출력 레이저의 사용이 필수적이며, 레이저 빔이 통과하는 각 광학 소자들의 코팅막은 고출력 레이저 빔에 대해 높은 레이저 damage threshold를 가져야 한다. AOM의 음향공학재료로 사용되는 $TeO_{2}$단결정에 코팅막의 종류 및 증착조건을 변화시키며 E-beam 증착법으로 A/R코팅 시편을 준비하였다. 0.55W의 입력 power를 갖는Ar레이저를 사용하여 코팅의 광손상 정도를 확인하였다. $AI_{2}O_{3}$막에 비해 $ZrO_{2}$$SiO_{2}$막을 사용한 경우 레이저 damage threshold는 크게 향상되었다. 또한 AOM모듈을 제작 후 구동회로와 연결하여 약 20mW의 SHG power를 입력시키며 출력 power long term안정성을 측정하였다.

Radish phospholipid hydroperoxide glutathione peroxidase provides protection against hydroperoxide-mediated injury in mouse 3T3 fibroblasts

  • Overexpression of phospholipid hydroperoxide glutathione peroxidase (PHGPx) genes has been reported to play an important role in protecting host cells from oxidative injury in several model systems. A radish phospholipid hydroperoxide glutathione peroxidase (RsPHGPx) known to have high catalytic activity was applied to mouse 3T3 fibroblasts to determine the protective effects of PHGPx against oxidative injury triggered by hydroperoxides such as hydrogen peroxide ($H_2O_2$), tert-butyl hydroperoxide (t-BHP) and phosphatidylcholine hydroperoxide (PCOOH). We observed that preincubation of cells with RsPHGPx significantly increased cell viability, reduced levels of malondialdehyde (MDA), inhibited generation of reactive oxygen species (ROS), and maintained natural cell shapes after treatment with $H_2O_2$, t-BHP or PCOOH, indicating that the exogenous RsPHGPx can act as an effective hydroperoxide-scavenger and may also protect target cells from oxidative damage. These results suggest the possibility for use of RsPHGPx as a therapeutic protectant.
