Effect of Cryoprotectants on Quality Properties of Chicken Breast Surimi Manufactured by pH Adjustment (냉동변성방지제 조건에 따른 닭가슴살 수리미의 저장특성)

  • This study was conducted to determine the effect of pH adjustment and the addition of cryoprotectants on the storage properties of chicken breast surimi. Surimi as a control was prepared with Alaska pollack by two times of washing treatment and addition of cryoprotectants(4% sugar, 5% sorbitol and 0.3% polyphosphate). Three types of surimi were manufactured by different cryoprotectant conditions(T1: 5% sorbitol+0.3% polyphosphate, T2: 4% sugar+5% sorbitol+0.3% polyphosphate, T3: 2% salt+4% sugar+5% sorbitol+0.3% polyphosphate) and frozen after adjusting the surimi to pH 11.0. The amino acid and saturated fatty acid composition were significantly higher in the control. EAA(essential amino acid), FAA(amino acid in relation to flavor), SAAA(amino acid in relation to saccarinity) and FRAA (fragrant amino acid) were significantly higher in the control. The TBARS(thiobarbituric acid reactive substances) and VBN(volatile basic nitrogen) were increased with storage times. TBARS and VBN values were significantly higher in the control and T3 than the other treatments. The VBN was significantly higher in control and T3 than other surimi samples. Chicken breast surimi adjusted to pH 11.0 had higher in microorganisms than the control, and T1 sample had the highest total plate count. Sensory evaluations were significantly higher in the control and T3 than the other samples. Especially, the overall acceptability of T3 was similar to the control.

The Effect of Conjugated Linoleic Acid Isomers on the Cell Proliferation, Apotosis and Expressions of Uncoupling Protein (Ucp) Genes during Differentiation of 3T3-L1 Preadipocytes (Conjugated Linoleic Acid 이성체가 3T3-L1 지방전구세포 분화중 세포증식, 세포사멸 및 Ucp 유전자 발현에 미치는 영향)

  • It has been reported that CLA decreases fat deposition in vivo and in vitro experiments. Among CLA isomers, c9t11 and t10c12 have been shown to exert active biological activities. For example, t10c12 reduces body weight and increases lean body mass, whereas, c9t11 has little effect on body fattness. However, the underlying molecular mechanism for the anti-obesity action of CLA isomers are not well understood. The purpose of this study was to examine the effects of t10c12 and c9t11 on lipid accumulation, cell proliferation, cell death and the expression levels of Ucp genes which are proposed as targets for anti-obesity in 3T3-L1 preadipocytes. Isomers of CLA at 50$\mu$M were added into preadipocyte differentiation medium for 3, 6 and 9days. Control cells received only the vehicle in the differentiation medium. Cytochemical analyses for lipid accumulation, cell proliferation and apotosis were carried out to compare lipidogenesis and cellular activity. RT-PCR analysis of GAPDH, Ucp 2,3 and 4 were also performed to find any modulatory effects of CLA isomers on the metabolic genes. Lipid accumulation indicated by Oil Red-O staining was inhibited in CLA isomers as compared to the control. T10c12 isomer showed less lipidogenesis than c9t11 did. A decrease occurred in CLA isomers as shown by BrdU incorporation. Apotosis has occured at higher level in t10c12 when compared to that of t9c11. Ucp 2, 3 and 4 genes were also upregulated in CLA isomers. T10c12 showed higher level of Ucp gene expressions than the c9t11 did. The biological activities of CLA isomers were also found to be different during differentiation of 3T3-L1 preadipocytes, suggesting that different isomers may be active in certain stage of lipidogenesis. The results indicate that both c9t11 and t10c12 CLA isomers decrease lipidogenesis, inhibit cell proliferation, increase cell death and upregulate in Ucp gene expressions during 3T3-L1 preadipocyte differentiation. T10c12 isomer was more effective than c9t11 in overall anti-obesity activity.

Diagnostic Significance of the Serum Thyroid Hormone Indicies in Various Thyroid Diseases (각종 갑상선질환에서 혈청 갑상선홀몬치의 진단적 의의)

  • In an attempt to evaluate the diagnostic significance of the serum thyroid hormones in various thyroid function states, the author measured serum $T_3$ uptake, serum $T_3$, serum $T_4$, serum free $T_4$ and free $T_3$ index in 27 cases of normal subjects, 11 cases of hypothyroidism, 152 cases of euthyroidism and 81 cases of hyperthyroidism by the radioimmunoassay method. The results were as follows: 1. The ranges of serum thyroid hormones in normal subjects were, serum $T_3$ uptake; $27.4{\sim}42.1%$, serum $T_3;\;93{\sim}245ng/dl$, serum $T_4;\;4.08{\sim}12.9ug/dl$ and serum free $T_4;\;0.57{\sim}1.53ng/dl(M{\pm}2S.D.)$. 2. Free $T_4$ index and serum $T_4$ show relatively high diagnostic value in euthyroidism group, and serum $T_3\;and\;T_4$ in hypothyroidism group, while serum $T_3$, free $T_4\;and\;T_4$ show relatively high diagnostic value in hyperthyroidism group. 3. There were significant correlation between free $T_4$ index and serum $T_4$(r=0.68), and between free $T_4$ index and serum free $T_4$(r=0.67) in hyperthyroidism group.

Effect of Red Ginseng Marc, Mineral and Black Sugar® Supplementation on the Production of Holstein Steers (Bos taurus) (홍삼박, 무기물® 및 흑설탕® 첨가 급여가 Holstein 거세 비육우 생산성에 미치는 영향)

  • This study was carried out to investigate the effects of a dietary addition of red ginseng marc, mineral® and black sugar® on the dry matter intake, daily gain, yield grade and quality grade of Holstein steers(Bos taurus). A total of 32 fattening Holstein steers(avg. BW 590kg) were assigned to four treatments which included a control diet(mixed concentrate and forage; namely, C) and three treatments diet(T1; control diet+red ginseng marc 200g, T2; control diet+red ginseng marc 100g+mineral 50g, T3; control diet+mineral 50g+ black sugar 20g). The results are summarized as follows; the total feed intake and daily feed intake were significantly(p<0.05) higher in T2 and T3 than in the other two treatments(C and T1). The daily gain was higher in T2 than in the other treatments, but there was no significant difference. The back fat thickness, longissimus muscle area and meat yield grade were no significant difference. The marbling score showed significantly(p<0.05) higher in order of T3(4.5) > T1(4.0) > T2(2.7) > C(2.5). The meat color, fat color, texture and maturity were no significant difference. The quality grade was higher in T3 than in the other treatments(C, T1 and T2), but there was no significant difference. T3, which numerically had a higher daily gain and carcass quality grade, showed higher economic efficiency, whereas C showed low economic efficiency. Based on the above results, T3 treatment compared to the other treatments improved the daily gain, carcass quality grade and economic efficiency.

Effects of Fermented Feedstuff with Wet Brewer’s Grain and Soybean on Fattening Performance and Carcass Grade in hanwoo Steers (맥주박 발효사료 및 대두의 급여가 한우 거세우의 육성성적 및 도체등급에 미치는 영향)

  • This study was conducted to examine the feeding effects of FFWBG (fermented feedstuff with wet brewer’s grain) and GS(grinding soybean) in Hanwoo steers. For this research, total twenty steers(average body weight : 455.7$\pm$25.9kg) were grouped into control(formula feed), T1(formula feed:FFWBG=60:40), T2(formula feed:GS=80:20), T3(formula feed:FFWBG:GS=50:40:10), and T4(formula feed: FFWBG:GS=40:40:20), each treatment was allocated to four steers. There was a tendency to be increased average daily gain in T1, T2, T3 and T4 compared with the control even though no statistical significance was found. The feed conversion of T1, T2, T3 and T4 were beneficially better than that of control(P<0.05; 8.25, 9.44, 6.69 and 7.71 vs 11.90, respectively). The blood urea nitrogen concentration of T4 was higher than that of control(P<0.05), but the creatinine concentration of T4 was lower than that of control(P<0.05). The glucose concentration of T1, T2, T3 and T4 were higher than that of control(P<0.05; 59.39, 62.44, 62.17 and 54.00 vs 51.00mg/$d\ell$, respectively). The back-fat thickness of T3 was thicker than that of control, T1, T2 and T4. The rib-eye area of T1, T3 and T4 was wider than that of control. Appearances percentage of ‘A’ ranked meat quantity were 66, 33, 75 and 33% in control, T1, T2 and T4, respectively.

The Effects of Oryungsan-gagampang on Leptin Levels, Leptin Receptor Levels and Differentiation of 3T3-L1 Adipocyte (오차산감방이 3T3-L1 adipocyte의 leptin 및 leptin receptor 함량과 differentiation에 미치는 영향)

  • Objectives: This experimental study was designed to investig:ue the effects of Oryungsan-gagampang on leptin and leptin receptor levels and differentiation of 3T3-L1 adipocyte. Methods: After 3T3-L1 adipocytes were incubated with various concentrations of Oryungsan-gagampang and Reductil(r) for 7 days, leptin and leptin receptor levels in 3T3-Ll adipocytes were measured by ELISA. To elucidate the mechanism of inhibitory effects of Oryungsan-gagampang on obesity, the 3T3-L1 adipocytes after oil red 0 staining were taken by digital photo system. Results: 1. Oryungsan-gagampang $1,000{\mu}g/ml$ significantly increased leptin levels in 3T3-L1 adipocytes in comparison with the control group (p<0.05), and Oryungsan-gagampang 0.1 10, $1,000{\mu}g/ml$ significantly increased leptin receptor levels in 3T3-L1 adipocytes in comparison with the control group (p<0.05). 2. Oryungsan-gagampang inhibited of differentiation of 3T3-L1 adipocytes. Conclusions: Oryungsan-gagampang showed significant effects on inhibiting differentiation of 3T3-Ll adipocytes, and increasing leptin levels and leptin receptor levels in 3T3-L1 adipocytes. Therefore, Oryungsan-gagampang could be used to treat obesity, but further studies are required.

Decreased Interaction of Raf-1 with Its Negative Regulator Spry2 as a Mechanism for Acquired Drug Resistance

  • Experiments were carried out to determine the role of Raf-1 kinase in the development of drug resistance to paclitaxel in v-H-ras transformed NIH 3T3 fibroblasts (Ras-NIH 3T3). We established a multidrug-resistant cell line (Ras-NIH 3T3/Mdr) from Ras-NIH 3T3 cells by stepwise increases in paclitaxel. Drug sensitivity assays indicated that the $IC_{50}$ value for drug-resistant Ras-NIH 3T3/Mdr cells was more than 1 ${\mu}M$ paclitaxel, 10- or more-fold higher than for the parental Ras-NIH 3T3 cells. Western blot and RT-PCR analysis showed that the drug efflux pump a P-glycoprotein were highly expressed in Ras-NIH 3T3/Mdr cells, while not being detectable in Ras-NIH 3T3 cells. Additionally, verapamil, which appears to inhibit drug efflux by acting as a substrate for P-glycoprotein, completely reversed resistance to paclitaxel in Ras-NIH 3T3/Mdr cell line, indicating that resistance to paclitaxel is associated with overexpression of the multidrug resistance gene. Interestingly, Ras-NIH 3T3/Mdr cells have higher basal Raf-1 activity compared to Ras-NIH 3T3 cells. Unexpectedly, however, the colocalization of Raf-1 and its negative regulator Spry2 was less observed in cytoplasm of Ras-NIH 3T3/Mdr cells due to translocation of Spry2 around the nucleus in the perinuclear zone, implying that Raf-1 may be released from negative feedback inhibition by interacting with Spry2. We also showed that shRNA-mediated knockdown of Raf-1 caused a moderate increase in cell susceptibility to paclitaxel. Thus, the results presented here suggest that a Raf-1-dependent pathway plays an important role in the development of acquired drug-resistance.

Effect of Grape Skin on Physicochemical and Sensory Characteristics of Ground Pork Meat (포도과피 첨가가 분쇄돈육의 이화학적·관능적 특성에 미치는 영향)

  • Purpose: This study aimed to investigate the effect of grape skin on the physicochemical properties and sensory score of ground pork meat. Methods: Four types of ground pork were evaluated: T0 without grape skin, T1 with 0.3% grape skin, T2 with 0.7% grape skin, and T3 with 1.0% grape skin. Results: There was no significant group wise difference in VBN content, L-value, b-value, chemical composition of raw and cooked meat, cooking yield, water holding capacity, moisture retention, fat retention, hardness, springiness, cohesiveness, gumminess, chewiness, taste, texture, juiciness, or palatability. Total polyphenol content was highest in T3, and DPPH radical scavenging activity was highest in T2 and T3 (p<0.001). The pH was highest in T0, and was lowest in T3 (p<0.001). The a-value of T2 and T3 were significantly higher than that of T0 (p<0.05). Flavor was highest in T2 among samples (p<0.01). Conclusion: The study results suggest that grape skin may be a useful ingredient in ground pork meat in terms of antioxidant potential, color and flavor.

Effects of Dietary 3,5,3'-Triiodo-L-Thyronine ($T_3$) on Growth and Survival Rate in Juvenile Black Seabream, Acanthopagrus Sclegeli (감성돔, Acanthopagrus schlegeli 치어의 성장과 생존에 미치는 외인성 3,5,3'-triiodo-L-thyronin ($T_3$) 효과)

  • An experiment was conducted to assess the effects of dietary 3,5,3'-triiodo-L-thyronine ($T_3$) at 0, 10, 20, 50, 100 ppm on growth and survival rate in juvenile black seabream held at $27.9{\pm}1.1^{\circ}C$ for 50 days. Fish were fed the $T_{3}$ experimental diet for 4() days by hand to satiation in $2{\~}4$ times per day. After 50 days period, food intake rate (${\%}$), feed efficiency (${\%}$), survival rate (${\%}$), growth of weight and length (specific growth rates), and condition factor were measured. Food intake rate was inversely related to the dietary $T_{3}$ level. But feed efficiency was not changed by $T_{3}$ level. $T_{3}$ slightly improved survival rate of larvae. Survival rate of larvae from 100 ppm was significantly higher than that of fish from control. Dietary $T_{3}$ influenced growth in length and weight. Growth of black seabream fed a diet containing 10 ppm of $T_{3}$ was significantly higher than that of fish fed control. However 100 ppm of $T_{3}$ induced the inhibition for length and weight growth. The condition factor was inversely related to the dietary $T_{3}$ content.

Effect of Domestic Clay Minerals on Growth Performance and Carcass Characteristics in Growing-Fattening Hanwoo Steers (육성비육 거세한우에 대한 점토광물 급여가 성장 및 도체특성에 미치는 영향)

  • This study was conducted to investigate the effects of domestic clay minerals on feed efficiency, meat quantity, meat quality and economic traits in 24 head of Hanwoo steers(166.1kg in body weight) for 540 days from six to 24 months in age. Feeding trial was conducted with 4 treatment(six heads/treatment) which were T1(Control), T2(Control+Kaolinite), T3(Control+Bentonite), T4(Control+Illite). The results obtained are summarized as follows; The range of average daily gains were 0.682 to 0.713, 0.669 to 0.714, 0.690 to 0.840 and 0.699 to 0.756kg in growing, fattening, finishing and over-all period, respectively, and the gains were high in T1 for growing and fattening period but in clay mineral groups for finishing and over-all period, especially it was high in Illite and Bentonite groups. Concentrates and TDN intakes per unit of kg gains were lower in clay mineral groups than in control and was lower especially in Bentonite groups. In carcass characteristics, dressed carcass and fresh meat and retailed cut percent were not apparently difference by treatments, and yield index was 69.3, 68.9, 68.8 and 68.6 in T3, T2, T4 and T1, respectively. Marbling scores were 5.1, 4.6, 4.4 and 3.3 in T3, T2, T4 and T1, respectively, and the range of shear force by treatment was from 3.51 to 6.02kg/cm2. and were improved with significant difference(P<0.05) in clay mineral groups than in control. Also in palatability traits, panel test scores of juiciness, tenderness and flavor were improved in clay mineral feeding groups, especially the flavor was improved with highly significant difference(P<0.01) in clay mineral groups than in control. In total fatty acid contents, the rate of SFA(saturated fatty acid) in longissimus muscle of beef was higher in the order of T2, T3, T1 and T4 while the rate of MUFA(monounsaturated fatty acid) was high in the order of T4, T3, T1 and T2. The content of oleic acid which is major influential factor at the flavor of beef was higher in Illite groups than in any other groups. In composition of amino acids in longissimus muscles of beef, the rate of essential amino acids was high in the order of T1, T2, T3 and T4. and the rate of amino acids in clay mineral groups was smaller than in control.In chemical component in Gom-Tang(soup of bone) made by Hanwoo steer’s leg-bone, the ranges of crude protein, ether extract, and crude ash was 0.81 to 1.24, 0.17 to 0.35 and 0.07 to 0.09%, respectively. In mineral composition, the ranges of Ca, P, Na and Mg was 14.01 to 15.77, 11.45 to 16.40, 37.92 to 49.99 and 0.26 to 0.46ppm, respectively. Chemical composition were not apparently different but mineral composition was increased in clay mineral groups than in control. Income by treatments was 967,096 to 1,524,055 Won per head for 540 days and income of clay mineral groups in comparison with control’s increased by 23.7 to 57.6 percent, and especially it was higher in bentonite and(or) Illite groups than others. According to the above results it may be concluded that clay mineral to growing-fattening Hanwoo steers can be improved the meat quantity, meat quality and income. Especially the effect of bentonite and illite is large and can be recommended for usage to improve animal performance as feed additives of growing-fattening Hanwoo steers.
