Molecular Characterization of FprB (Ferredoxin-$NADP^+$ Reductase) in Pseudomonas putida KT2440

  • The fpr gene, which encodes a ferredoxin-$NADP^+$ reductase, is known to participate in the reversible redox reactions between $NADP^+$/NADPH and electron carriers, such as ferredoxin or flavodoxin. The role of Fpr and its regulatory protein, FinR, in Pseudomonas putida KT2440 on the oxidative and osmotic stress responses has already been characterized [Lee at al. (2006). Biochem. Biophys. Res. Commun. 339, 1246-1254]. In the genome of P. putida KT2440, another Fpr homolog (FprB) has a 35.3% amino acid identity with Fpr. The fprB gene was cloned and expressed in Escherichia coli. The diaphorase activity assay was conducted using purified FprB to identify the function of FprB. In contrast to the fpr gene, the induction of fprB was not affected by oxidative stress agents, such as paraquat, menadione, $H_2O_2$, and t-butyl hydroperoxide. However, a higher level of fprB induction was observed under osmotic stress. Targeted disruption of fprB by homologous recombination resulted in a growth defect under high osmotic conditions. Recovery of oxidatively damaged aconitase activity was faster for the fprB mutant than for the fpr mutant, yet still slower than that for the wild type. Therefore, these data suggest that the catalytic function of FprB may have evolved to augment the function of Fpr in P. putida KT2440.

Relationship between roar sound and regional groups of Steller sea lion, Eumetopias jubatus (큰바다사자의 명음과 지역적 그룹과의 관계)

  • Hundreds of Steller sea lions, Eumetopias jubatus, migrate from Sakhalin and the northern Kuril Islands to Hokkaido in Japan every winter. During this migration, they may use their roaring sounds to navigate and to maintain their groups. We recorded the roars of wild Steller sea lions that had landed on reefs on the west coast of Hokkaido, and those of captive sea lions, while making video recordings. The fundamental frequency ($F_0$), Formant frequency ($F_1$), pulse repetition rate (PRR), and duration of syllables (T) were analyzed using a sonagraph. $F_0$, $F_1$, and PRR of the roars emitted by captive sea lions increased in the order male, female. By contrast, the $F_1$ of wild males was lower than that of females, while the $F_0$ and PRR of wild males and females did not differ statistically. These results suggest regional differences between the five groups showed that.

Genetic Relationship Between Korean and Mongolian Populations Based on the Y Chromosome DNA Variation

  • We analyzed seven Y chromosome binary markers (YAP, RPS4Y_711,\;M9,\;M175,\;LINE1,\;SRY_+465$ and 47z) in samples from a total of 254 males from Koreans and tow Mongolian ethnic groups (Buryat and Khalkh) to study the genetic relationship among these populations. We found eight distinct Y haplogroups constructed from the seven binary markers. Haplogroup DE-YAP was present at extremely low frequencies (∼2%) in the Korean and Mongolian populations. This result is consistent with earlier reports that showed the YAP+ chromosomes to be highly polymorphic only in populations from Japan and Tibet in east Asia. The observed high frequency of haplogroup $C-RPS4Y_711$ in the Mongolian populations (∼40%) is concordant with recent findings, showing that the $RPS4Y_711$-T chromosomes were distributed at high frequencies in Siberian and Mongolian populations compared with most other populations from east Asia. Thus, the relatively moderate frequency of haplogroup $C-RPS4Y_711$ in Korean (∼15%) can be seen as genetic evidence for probable interaction with Mongolian and/or Siberian populations. In contrast, the majority (∼75%) of modern Koreans studied here had high frequencies of Y chromosome lineages of haplogroup O-M175 and additional haplogroupts that define sublineage of O-M175, which are most likely related with modern populations in China. In conclusion, our data on the Y chromosome haplogroup distribution may provide evidence for interaction between Korean and Mongolian populations, but Korean tend to be much more related with those from southern-to-northern populations of China than to Mongolians in east Asia.

The Effectiveness of Jeju Island Mixed Reality Tourism Application: Focusing on the Moderating Role of Users' K-Culture Involvement

  • In this study, we introduce a multi-modal mixed reality (MR) application for advertising the main touristic landmarks on Jeju Island, and evaluate its effectiveness based on the Technology Acceptance Model (TAM) with a focus on the user's involvement in Korean culture (K-Culture). Specifically, we deployed several stimuli to enable users to retrieve information, visualize a variety of heritage content such as text, images, videos, virtual artifacts, and immerse in a VR environment to experience the heritage locations. The results of the online survey with 436 participants showed that for consumers with low K-Culture involvement, Perceived Usefulness and Perceived Ease-of-Use did not affect their Intention to Visit the island. Still, it had a significant impact on their Intention to Use the application. On the other hand, for the group with high K-Culture involvement, Perceived Usefulness had no effect on the Intention to Use and Intention to Visit. However, Perceived Ease-of-Use had a significant impact on their Intention to Use the application and Intention to Visit Jeju Island. Therefore, when user involvement in K-Culture is high, convenience of use has played a significant role in sensory media usage and the intention to visit, regardless of the usefulness of the mixed reality content. In contrast, users with low K-Culture involvement primarily focused on the functional aspects of the application. These findings open the path for follow-up studies on K-Culture involvement and immersive media tourism marketing, which will benefit digital tourism marketing and Korea's tourism industry.

  • The purpose of this study is to analyze the change and to compare to the difference of 'selective elements' and 'space-memory' focusing on the theory, 'selective attention' through the survey results. In this study, In this study, the lighting is considered a important factor in the change of 'selective elements'. this survey is to find the selective elements of participants and to measure the spatial sensitivity of respondents through 'self- test'. The analysis in this study is conducted by descriptive statistics, t-test and one way ANOVA by SPSS program 22. The results of this study are as following; Firstly, 'attention-element' could be classified with 4 types, 'shape', 'material', 'contrast' and 'combination'. 'shape' could divide into 'structure' and' furniture and object'. In case of 'material', it could section with 'pattern' and 'color'. Secondly, through the results of study, 'attention-element' is different each space during the day in detail. But we could know that 'shape' is the important element of the 'attention-elements' during the day through comparison of this result. That means users consider this as a important factor when they evaluate the space. Therefore, it is effective way designers to consider 'shape' as the first element when they want to conduct the special sensitivity of users in the space through planning. On the other hand, what selective elements of users are different by the lighting situation should be acknowledged by designers. And they should think the kinds of selective elements are more various when lighting turns on than turns off.. Thirdly, through the results such as the meaningful difference of space-memory of users according to the change of 'attention-elements', designers should judge about which kind of feeling of users to the space do you want lead in the design process. For the effective feedback between spaces and users to induce the same emotion of users, designers need to consider the unified design and the individual design both. Also, we will regard the differences in the users' emotion to the space according to the lighting situation when we design the space.

Imaging Patterns of Bacillus Calmette-Guérin-Related Granulomatous Prostatitis Based on Multiparametric MRI

  • Objective: To categorize multiparametric MRI features of Bacillus Calmette-Guérin (BCG)-related granulomatous prostatitis (GP) and discover potential manifestations for its differential diagnosis from prostate cancer. Materials and Methods: The cases of BCG-related GP in 24 male (mean age ± standard deviation, 66.0 ± 9.4 years; range, 50-88 years) pathologically confirmed between January 2011 and April 2019 were retrospectively reviewed. All patients underwent intravesical BCG therapy followed by a MRI scan. Additional follow-up MRI scans, including diffusion-weighted imaging (DWI), were performed in 19 patients. The BCG-related GP cases were categorized into three: A, B, or C. The lesions with diffusion restriction and homogeneous enhancement were classified as type A. The lesions with diffusion restriction and a poorly enhancing component were classified as type B. A low signal intensity on high b-value DWI (b = 1000 s/mm2) was considered characteristic of type C. Two radiologists independently interpreted the MRI scans before making a consensus about the types. Results: The median lesion size was 22 mm with the interquartile range (IQR) of 18-26 mm as measured using the initial MRI scans. The lesion types were A, B, and C in 7, 15, and 2 patients, respectively. Cohen's kappa value for the inter-reader agreement for the interpretation of the lesion types was 0.837. On the last follow-up MRI scans of 19 patients, the size decreased (median, 5.8 mm; IQR, 3.4-8.5 mm), and the type changed from A or B to C in 11 patients. The lesions resolved in four patients. In five patients who underwent prostatectomy, caseous necrosis on histopathology matched with the non-enhancing components of type B lesions and the entire type C lesions. Conclusion: BCG-related GP demonstrated three imaging patterns on multiparametric MRI. Contrast-enhanced T1-weighted imaging and DWI may play a role in its differential diagnosis from prostate cancer.

Transient Splenial Lesions in the Splenium of Corpus Callosum in Seven Patients: MR Findings and Clinical Correlations (뇌량 팽대의 일과성 병변: 7명의 환자에서의 자기공명 영상소견과 임상 양상의 연관성)

  • Purpose : The purpose of this study is to correlate the imaging findings and the clinical findings in patients with transient splenial lesions (TSL). Materials and Methods: Total of 7 patients (M: F = 4: 3; age range 11 - 38 years, mean age 25.5 years) were studied between November 2006 and April 2011. The MRI findings and clinical findings in all patients were retrospectively reviewed. The location, MR signal intensity, restricted diffusion, enhancement pattern and reversibility from the follow up images were reviewed. Clinical features were reviewed with respect to the presented symptoms, signs, treatment and outcome. Results: The lesions were located in the splenium of corpus callosum in all patients. All lesions showed high signal intensity on diffusion weighted imaging (DWI), and six patients showed restricted diffusion on the apparent diffusion coefficient (ADC) map. ADC map was not available in one patient. All lesions (n = 7) showed high signal intensity on the T2 weighted image (T2WI). Five of the patients (71.4%) with follow up images (range 7 - 34 days) showed complete resolution of focal high signal intensity on DWI, with recovery of ADC values as well as T2WI. After contrast media administration, none of the lesions showed any enhancements. All lesions with various etiologies including TB medication were developed in younger age patients and showed reversibility after the acceptable period of minimum 7 days with conservative treatment. Conclusion: All TSL showed nonspecific imaging findings, including restricted diffusion on DWI and ADC map on the initial images. However, reversibility of the lesions and the young age preference can be a characteristic finding of TSL with acceptable period of minimum 7 days. In addition, to keep it in mind that various etiologies including TB medication may cause TSL, is important for radiologists as well as clinicians.

Increase in Neurogenesis of Neural Stem Cells Cultured from Postnatal Mouse Subventricular Zone by Nifedipine (L-type 칼슘 채널을 저해하는 저해제, nifedipine에 의한 쥐 뇌실하 영역 신경줄기세포의 신경세포로의 분화 촉진)

  • The subventricular zone (SVZ) in the brain contains neural stem cells (NSCs) that generate new neurons throughout one's lifetime. Many extracellular and intracellular factors that affect cell proliferation and neuronal differentiation of NSCs are already well-known. Recently, L-type calcium channels have been reported to regulate neural development and are present in NSCs, differentiating neuroblasts, and mature neurons in the SVZ. Nifedipine, a blocker of L-type calcium channels, has been long used as a therapeutic drug for hypertension. However, studies on the use of nifedipine to inhibit L-type calcium channels of NSCs are lacking. Herein, we treated NSCs cultured from mouse postnatal SVZ with nifedipine during neuronal differentiation. Nifedipine increased the number of Tuj1-positive neurons but did not significantly change the number of Olig2-positive oligodendrocytes. Nifedipine increased cell division during early differentiation, which was detected using the 5-ethynyl-2'-deoxyuridine incorporation assay and immunocytochemistry assessment by staining the cells with phosphorylated histone H3, a mitosis marker. Nifedipine increased the transcription of Dlx2, a neurogenic transcription factor, and the level of Mash1, a marker for early neurogenesis. In addition to nifedipine, verapamil, which is also an L-type calcium channel blocker, showed a slight increase in neurogenesis, but its statistical significance was very low. In contrast, pimozide, a T-type calcium channel blocker, did not affect neurogenesis, although T-type calcium channel genes Cav3.1, Cav3.2, and Cav3.3 were expressed. In summary, nifedipine might promote the neuronal fate of NSCs during early differentiation and calcium signaling through L-type calcium channels might be involved in neuronal differentiation, especially during the early stages of differentiation.

The Effects of Various Extracellular Matrices on Motility of Cultured MC3T3-E1 Cell (다양한 세포외기질이 배양 골아세포의 이동에 미치는 영향)

  • Chemotactic migration of bone forming cell, osteoblast, is an important event during bone formation, bone remodeling, and fracture healing. Migration of cells is mediated by adhesion receptors, such as integrins, that link the cell to extracellular matrix ligands, type I collagen, fibronectin, laminin and depend on interaction between integrin and extracellular ligand. Our study was designed to investigate the effect of extracellular matrix like fibronectin, laminin, type I collagen on migration of osteoblast. Migration distance and speed of MC3T3-E1 cell on extracellular matrix-coated glass were measured for 24 hours using 0.01% type I collagen, 0.01% fibronectin, 100 microliter/ml laminin. The migration distance and speed of MC3T3-E1 cell was compared using a video-microscopy system. To determine migration speed, cells were viewed with a 4 phase- contrast lens and video recorded. Images were captured using a color CCD camera and saved in 8-bit full-color mode. The migration distance on 0.01% type I collagen or 0.01% fibronectin was longer than that on $100{\mu}l/ml$ laminin-coated glass. The migration speed on fibronectin-coated glass was 68 micrometer/hour which was fastest. The migration speed on type I collagen-coated glass was similar with that on fibronectin-coated glass. The latter two migration speeds were faster than that on no-coated glass. On the other hand, the average migration speed on laminin-coated glass was 37micrometer/hour and not different from that of control group. In conclusion, the extracelluar matrix ligands such as type I collagen and fibronectin seem to play an important role in cell migration. The type I collagen or fibronectin coated scaffold is more effective for migration of osteoblast in tissue engineering process.

A Standardization Study of the Korean Version of Eating Attitudes Test-26 I : Reliability and Factor Analysis (한국판 식사태도검사-26(The Korean Version of Eating Attitudes Test-26 : KEAT-26) 표준화 연구 I : 신뢰도 및 요인분석)

  • The purpose of this study was to test a reliability and validity of the Korean version of Eating Attitudes Test-26(KEAT-26). Using multi-stage sampling, we finally got 3,496 subjects(1422 males and 2074 females) who were available for analysis from target 4,400 Korean adults over 18 in the nationwide areas of9 kus, 10 middle or small cities, and 17 kuns. We tried to make T score norm of the KEAT-26 as a cutoff score and STEN score norm as a index of severity for disordered eating behaviors. For the male group, Cronhach's internal consistency was .83 and Spearman-Brown split half correlation coefficiency was .75. For the female group, each of them was .81 and .75, and .81, .75 for the grand total group respectively. Validity test was performed by construct validation analysis. By a iterated principal axis factoring, 4 factors were extracted. There were some differences in the factors of the KEAT-26 by sex. In the male group, factor I was 'self-control of eating and bulimic symptom', factor II was 'food preoccupation and dieting', factor III was 'preoccupation with being thinner', factor N was 'avoidance of sweet foods'. In contrast with the male group, factor I was 'self-control of eating and bulimic symptom', factor II was 'preoccupation with being thinner', factor III was 'food preoccupation' and factor N was 'dieting' in the female group. We used T score 65 as a cutoff score. T score 65 corresponded to raw score 19 in the male group, 22 in the female group and 21 in the grand total group. Severity of disordered eating behaviors was measured by a STEN score. In the male group, each of the score range of 0-10, 11-14, 15-18, 19-22 and over 23 represented the degree of none, subclinical, manifest, moderate and severe severity respectively. Each of the score range of 0-13, 14-17, 18-21, 22-26 and over 27 in the female group, and the score range of 0-12, 13-16, 17-20, 21-25 and over 26 in the grand total group also represented the same degree of severity as like in the male group. These results support that KEAT-26 is a reliable and valid scale for evaluating disordered eating behaviors and eating problems.
