Plasma Levels of Hormones and Metabolites as Affected by the Forages Type in Two Different Types of Crossbred Holstein Cattle

  • An experiment was carried out to study plasma levels of hormones and metabolites of crossbred Holstein cattle during late pregnancy (28 days pre partum), early lactation (30 days post partum), mid-lactation (120 days post partum) and late lactation (210 days post partum). Two breed types of Holstein $Friesian{\times}Red$ Sindhi (50:50 = 50%HF) and Holstein $Friesian{\times}Red$ Sindhi (87.5:12.5 = 87.5%HF) were divided into four groups of four animals each. Two groups of each breed were fed with either rice straw treated with 5% urea or pangola hay (Digitaria decumbens) as the source of roughage throughout the experiments. There were a substantial increases in the mean levels of total triiodothyronine ($T_3$), insulin and glucagon at the onset of lactation, and maintained in a high levels during lactation advance for all groups of experiments. The mean levels of prolactin and thyroxine ($T_4$) were not significantly different among groups of animals, but the plasma cortisol concentration was slightly higher in both groups of 50%HF in comparison with those of 87.5%HF animals. The mean levels of plasma growth hormone (GH) of both groups of 87.5%HF animals feeding on either hay or urea treated rice straw markedly rose in the early period of lactation and markedly reduced in mid- and late lactation. These changes were accompanied with changes of milk yield. In contrast to 50%HF animals, plasma GH levels were considerably higher in the late pregnant period than in the early period of lactation and it remained constant as its value at the early lactation throughout the experimental period. The high levels of both plasma progesterone and estradiol concentration significantly declined after parturition and remained low through lactating period. The plasma glucose level in the 50%HF animals feeding on either hay or urea treated rice straw was higher than the 87.5%HF animals in all periods of experiments. Changes in plasma FFA levels of both types of crossbred animals were depended on the endocrine status during late pregnancy and lactation. The levels of plasma FFA of 50%HF animals were significantly higher (p<0.05) than those of 87.5%HF animals during late pregnancy. Both plasma ${\beta}$-hydroxybutyrate and lactate concentrations were not affected by feeding on either hay or urea treated rice straw during late pregnancy and lactation. These data demonstrate that there were no differences in the physiological performances in the same crossbred animals fed either hay or urea treated rice straw. The 87.5%HF animal has the genetic potential for a high milk yield and homeorhetic adaptation for mammary function differed from 50%HF animals during periods of lactation. Altering lactation persistency in 87.5%HF is regulated mainly by chronically acting growth hormones through the period of lactation.

Effects of Emergency Department Nurses' Emotional Labor on Professional Quality of Life -Focusing on Mediating Effects of Emotional Dissonance- (응급실 간호사의 감정노동이 전문직 삶의 질에 미치는 영향 -감정부조화의 매개효과를 중심으로-)

  • The purpose of this descriptive survey study was to explore the effects of emotional labor on professional quality of life of emergency department nurses and to elucidate any mediating effects of emotional dissonance between emotional labor and professional quality of life. A total of 227 nurses, including general, charge, and head nurses, who have worked in an emergency department at university hospitals, general hospitals, and junior general hospitals located in Gyeongnam, Daegu and Gyeongbuk areas over a period of 6 months. Data were collected between December 12 and December 29 of 2015. We analyzed the data with descriptive statistics, t-test, ANOVA, Pearson's correlation coefficient, and three-step mediated regression analysis using SPSS/WIN 21.0 program. The average scores of emotional labor, compassion satisfaction, compassion fatigue, burnout, and emotional dissonance were 4.50 out of 7.00, 31.62 out of 50.00, 29.48 out of 50.00, 29.27 out of 50.00, and 5.02 out of 7.00, respectively. Emotional labor had significant effects on compassion fatigue and burnout, showing explanatory power of 12% and 29%, respectively. Emotional dissonance had partial mediating effects in the relationship between emotional labor and compassion fatigue and complete mediating effects in the relationship between emotional labor and burnout. In summary, to improve the professional quality of life of nurses, it is necessary to develop effective strategies to minimize their emotional labor and emotional dissonance.

Analysis of microstructure for glass-ceramics made of silicate glasses containing EAF dust (제강분진이 첨가된 규산염계 결정화유리의 미세구조 분석)

  • Microstructures of free surface and interior of glass-ceramics obtained by heat treating silicate glass specimen containing electric arc furnace dust(EAF dust) were observed. The crystallization temperature, $T_c$ of glassy specimen was measured around $850^{\circ}C$ from the result of different thermal analysis so heat treatment temperature to obtain glass-ceramic specimen was selected as $950^{\circ}C$ for 1 hr. Glass specimens containing 50 wt% dust were amorphous, while glass specimens containing 70 wt% dust showed spinel crystal peaks in XRD results. In case of glass-ceramic specimens, spinel crystalline phase was appeared with willemite, and willemite crystal peak intensity increased with increasing dust contents. The fractured surface of glass specimens containing 50 wt% dust was smooth like mirror surface, but that containing 70 wt% dust showed spinel crystals of 10 ${\mu}m$ size in glass matrix. In case of glass-ceramic specimens, ZnO crystal particles of $2{\sim}5{\mu}m$ size were produced in free surface and glassy phase, spinel and willemite crystal phases existed in interior. There were no crystals in glasses containing 50 wt% dust, while glass containing 70 wt% dust had 14 vol% crystals. Crystallinity of glass-ceramic specimens containing 50 and 70 wt% dust were 19 and 43%, respectively. When microstructures of glass and glass-ceramic specimens were observed through SEM after TCLP experiment, glass specimens showed flaking phenomenon while glass-ceramic specimens showed a slight corrosion evidence without any cracks.

Development of Kiln Drying Schedule of Lesser-Known Species Imported from Solomon (수입 솔로몬산(産) 미이용(미利用) 수종(樹種)의 인공건조(人工乾燥)스케쥴 개발(開發))

  • A study was conducted to determine the physical properties related to drying characteristics, the seasonal air drying curves and the kiln drying schedule for taun lumber imported and utilized. This kiln drying schedule was found by oven drying and developed by pilot testing of green lumber and partially air dried lumber. The results of this study were as follows; 1. Average green specific gravity and standard deviation of heartwood lumber were 0.60${\pm}$0.03 and those of sapwood lumber were 0.64${\pm}$0.02. 2. Radial shrinkage from green to air dry and from green to oven dry were 3.05 percent and 5.96 percent respectively, and tangential shrinkage from green to air dry and to oven dry were 5.49 percent and 8.74 percent respectively. 3. Drying time for 25mm thick green lumber (50 percent moisture content) air dried to 30 percent moisture content were 14 days in springtime. 6 days in summertime, and 12 days in autumntime, whereas for 50mm thick lumber in 36 days in springtime, 18 days in summertime, 38 days in autumntime. 4. Kiln drying schedules developed by oven drying were T8-B3 for 25mm thick lumber and T5-B2 for 50mm thick lumber. 5. Kiln drying curves of green 25mm and 50mm thick lumber were similar to those of partially air dried lumber from the level of 30 percent average moisture content. Green 25mm thick lumber (55.7 percent moisture content) was dried to 9.3 percent moisture content in 101.5 hours and green 50mm thick lumber (65.6 percent moisture content) was dried to 11.5 percent moisture content in 526 hours. 6. End checking for green 25mm thick lumber occured in 49.6 percent moisture content and reached maximum amount in 27.6 percent moisture content and closed in 15.8 percent moisture content. 7. End checking for green 50mm thick lumber and partially air dried lumber developed and reached maximum amount earlier then for 25mm thick lumber. 8. Final moisture content of surface layer for 50mm thick lumber was one half of that of core, and moisture content equalized in the lumber after nine days of room conditioning. 9. Casehardening for 50mm thick lumber was slight and was conditioned after nine days of room stroage. 10. Drying defects, such as end checking and surface checking, were not observed and the quality of dry lumber was first.

A Study on the Women's Bodysuit Sleeve Block Construction Using Stretch fabrics (Stretch 소재를 사용한 여성용 Bodysuit Sleeve 원형 설계에 관한 연구)

  • The study aimed firstly, to develop the women's bodysuit sleeve block construction method adopting the appropriate pattern reduction rates according to the fabric stretch property. Secondly, the details applied to the bodysuit sleeve block drafting (Dr the educational and industrial usage were proposed. For these, several distinguishing bodysuit sleeve pattern making methods(i.e. Joseph-Armstrong: T1, Shoben & Ward: T2, Esmod: T3 and Mixed Joseph-Armstrong: T4) were analyzed and divided into two categories that adopt 1) the equally (i.e., T1) and 2) the differently(i.e., T2, T3 & T4) distributed front and back armhole length measurements. Women's sleeve samples were made for the research using the same stretch fabric($50\%\;and\; 70\%$ in wale and course each) to the previous research. A group comprising 5 relevant experts evaluated the fit and comfort features of the samples. Experiments analyzed the appearance of sleeve samples focused on total 13 evaluation parts(including the front/side/back fit tolerance, sleeve centre line, sleeve length, appropriateness of the sleeve appearance balanced with the bodysuit and etc.): and performed the comfort test evaluating three kinds(vertical-front/vertical-side/ horizontal) of arm movements. The most appropriate bodysuit sleeve to fulfil the original aims of the study was suggested. The findings and suggestions throughout the study were: 1) the measurements and required reduction rates for the bodysuit sleeve block developed: outer sleeve length (with 1.0 RR), crown height(with 0.7 RR), front and back armhole lengths measured on the bodysuit blocks ($0\%$ ease amount), elbow width(0.9/0.95 RR), wrist girth measurements(from $12\%\;to\;18\%$ tolerances can be given to): and 2) the differently distributed front and back armhole length measurements resulted in the better fit and comfort through the research.

Effects of Feeding Dehulled Soybean Meal on Growth Performance, Carcass Characteristics and Meat Quality Parameters in Berkshire Pigs (탈피대두박 급여가 흑돼지의 생산성, 도체 및 육질 특성에 미치는 영향)

  • A total of 360 Berkshire(hog) weighing 25kg were fed experimental diets containing either dehulled soybean meal(DSBM) or non-dehulled soybean meal(NDSBM) as a major protein source until they reach 80kg. The pigs were then fed the finisher diets either high(3,265 kcal/kg) or low(2,940 kcal/kg) in ME to evaluate the effects of dietary energy level for finisher phase on the growth performances, carcass and meat quality parameter. In growth performance, daily weight gain(kg/day) was higher(P<0.05) in T1 than in other groups. Feed conversion(feed/gain) was lower(P<0.05) in other groups. In carcass parameter, carcass percentage was higher(P<0.05) in T2 than in T1 and T4. Backfat thickness was lower(P<0.05) in T2 than in T1 and T3. The ratio of up B grade in carcass grade tender to be higher in T2 than in other groups. In meat quality, chemical composition was similar between groups. WHC was slighter higher in T1 than in other groups. Shear force was tender to be higher in T1 than in other groups. Hunter L and a in meat color were tender to be higher in T2 than in other groups. Hunter L in backfat color was thender to be higher in T1 than in other groups. The results indicate that growth performance and carcass parameter of Berkshire were improved by the dehulled soybean meal as comparison with non-dehulled soybean meal during growing and finishing periods.

Activation of Pro-Apoptotic Multidomain Bcl-2 Family Member Bak and Mitochondria-Dependent Caspase Cascade are Involved in p-Coumaric Acid-Induced Apoptosis in Human Jurkat T Cells (p-Coumaric acid에 의해 유도되는 인체 Jurkat T 세포의 에폽토시스 기전)

  • The apoptogenic effect of p-coumaric acid, a phenolic acid found in various edible plants, on human acute leukemia Jurkat T cells was investigated. Exposure of Jurkat T cells to p-coumaric acid (50-$150{\mu}M$) caused cytotoxicity and TdT-mediated dUTP nick-end labeling (TUNEL)-positive apoptotic DNA fragmentation along with Bak activation, ${\Delta}{\psi}m$ loss, activation of caspase-9, -3, -7, and -8, and PARP degradation in a dose-dependent manner. However,these apoptotic events were completely abrogated in Jurkat T cells overexpressing Bcl-2.Under these conditions, necrosis was not accompanied. Pretreatment of the cells with the pan-caspase inhibitor (z-VAD-fmk) could prevent p-coumaric acid-induced sub-$G_1$ peak representing apoptotic cells, whereas it failed to block ${\Delta}{\psi}m$ loss, indicating that the activation of caspase cascade was prerequisite for p-coumaric acid-induced apoptosis as a downstream event of ${\Delta}{\psi}m$ loss. FADD- and caspase-8-positive wild-type Jurkat T cell clone A3, FADD-deficient Jurkat T cell clone I2.1, and caspase-8-deficient Jurkat T cell clone I9.2 exhibited similar susceptibilities to the cytotoxicity of p-coumaric acid, excluding an involvement of Fas/FasL system in triggering the apoptosis. The apoptogenic activity of p-coumaric acid is more potent in malignant Jurkat T cells than in normal human peripheral T cells. Together, these results demonstrated that p-coumaric acid-induced apoptogenic activity in Jurkat T cellswas mediated by Bak activation, ${\Delta}{\psi}m$ loss, and subsequent activation of multiple caspases such as caspase-9, -3, -7, and-8, and PARP degradation, which could be regulated by anti-apoptotic protein Bcl-2.

Antimicribial and Antioxidant Activities of Ethanol Extracts of Medicinal Plants

  • The objective of this study was to determine the radical scavenging activity, total phenolic content, antimicrobial activity, minimum inhibitory concentration (MIC) of ethanol extracts of 32 medical plant species that have been commonly used in medicinal plants. Total phenolic index of T. chebula exhibited the highest value (498.01㎎/g), followed by R. coreanus miquel (400.33㎎/g), Sanguisorba officinalis (368.25㎎/g), P. thumbergiana (259.74㎎/g) and Eugenia aromaticum (229.38㎎/g). Radical scavenging activity for the DPPH radical was highest in T. chebula (40.91%, p<0.01), followed by C. sappan (36.50%), S. officinalis (32.92%), R. coreanus miquel (26.54%) and P. thumbergiana (24.50%). The extracts from T. chebula, R. coreanus muquel, C. sappan, E. aromaticum, S. officinalis and C. japonica possessed outstanding antimicrobial activity against Escherichia coli, Salmonella Typhimurium, Pseudomonas aeruginosa, Staphylococcus aureus, Bacillus subtilis and Lactobacillus plantarum. MIC was determined on those extracts that showed high efficacy against the test organisms. The most potent MIC values were seen for T. chebula extract against P. aeruginosa, S. aurusa, E. coli, B. subtilis, L. plantarum and S. Typhimurium at 7.8, 7.8, 15.6, 7.8, 125 and 31.2㎍/mL, respectivley. Furthermore, the total phenolic content and radical scavenging activity were very closely correlated for all samples (r=0.78). The coefficient correlations between total phenolic index and antimicrobial activity were 0.91 (E. coli), 0.91 (B. subtillis), 0.79 (P. aeruginosa), 0.79 (S. Typhimurium) and 0.70 (L. plantarum).

Anti-Apoptotic Effects of SERPIN B3 and B4 via STAT6 Activation in Macrophages after Infection with Toxoplasma gondii

  • $Toxoplasma$ $gondii$ penetrates all kinds of nucleated eukaryotic cells but modulates host cells differently for its intracellular survival. In a previous study, we found out that serine protease inhibitors B3 and B4 (SERPIN B3/B4 because of their very high homology) were significantly induced in THP-1-derived macrophages infected with $T.$ $gondii$ through activation of STAT6. In this study, to evaluate the effects of the induced SERPIN B3/B4 on the apoptosis of $T.$ $gondii$-infected THP-1 cells, we designed and tested various small interfering (si-) RNAs of SERPIN B3 or B4 in staurosporine-induced apoptosis of THP-1 cells. Anti-apoptotic characteristics of THP-1 cells after infection with $T.$ $gondii$ disappeared when SERPIN B3/B4 were knock-downed with gene specific si-RNAs transfected into THP-1 cells as detected by the cleaved caspase 3, poly-ADP ribose polymerase and DNA fragmentation. This anti-apoptotic effect was confirmed in SERPIN B3/B4 overexpressed HeLa cells. We also investigated whether inhibition of STAT6 affects the function of SERPIN B3/B4, and vice versa. Inhibition of SERPIN B3/B4 did not influence STAT6 expression but SERPIN B3/B4 expression was inhibited by STAT6 si-RNA transfection, which confirmed that SERPIN B3/B4 was induced under the control of STAT6 activation. These results suggest that $T.$ $gondii$ induces SERPIN B3/B4 expression via STAT6 activation to inhibit the apoptosis of infected THP-1 cells for longer survival of the intracellular parasites themselves.

$Trichuris$ $vulpis$ (Froelich, 1789) Infection in a Child: A Case Report

  • We present a human infection with the canine whipworm, $Trichuris$ $vulpis$, in a child suffering from rhinitis with a diagnosis of rhinitis. $T.$ $vulpis$ eggs resemble those of $T.$ $trichiura$ but they can be differentiated based on their morphological features and egg size, using micrometry with an ocular micrometer. $T.$ $vulpis$ eggs measured an average of 90 ${\mu}m$ by 44 ${\mu}m$ (range 86-99 ${\mu}m$ by 38-47 ${\mu}m$). Prevalence of hookworms (28.1%), $Toxocara$ $canis$ (11.8%), and $Trichuris$ $vulpis$ (3.5%) was found in 292 fecal samples of dogs collected at the peri-domicile, which showed that the risk of infection was not only fortuitous. The treatment of canine whipworm infections is similar to that of $T.$ $trichiura$ infection. We recommend differentiation of the 2 species for their epidemiological and prevention implications.
