Effect of Soil Textures and Fertilizer Application Conditions on the Growth, Yield and Quality of Scutellaria baicalensis G. (토성(土性) 및 시비조건(施肥條件)이 황금(黃芩)의 생육(生育), 수량(收量) 및 품질(品質)에 미치는 영향(影響))

  • Scutellaria baicalensis G. was cultivated in plots of different soil textures which were sand loam(SL), loam(L) and clay loam(CL). Also three fertilizer conditions were used; non application (NA)treatment, rice straw manure(RSM) treatment and N-P-K, chemical fertilization(CF) treatment. The chemical-physical properties, such as organic matter, available phosphate, $K_2O$, CaO, clay contents and porosity ratio in CL plot with RSM application were the most proper in CL plot and RSM application for the culture of S. baicalensis plants. RSM had very high contents of total nitrogen, 2.25% and C/N ratios, 21.4. Thus the growth of shoot and root in loam plot with RSM treatment were greater compared to that of CL plot with NA treatment. Whereas, The highest baicalin, baicalein and wogonin contents in roots were found in CL plot with RSM treatment. There was significantly positive correlation between aerial and underground parts of plant, yield and contents of T-N, $K_2O$ but negatively correlated with the contents of baicalin, baicalein and wogonin in S. baicalensis roots.

Protoplast Formation, Regeneration and Reversion in Pleurotus ostreatus and P. sajor-caju (느타리버섯과 여름느타리버섯의 원형질체(原形質體) 나출(裸出)과 재생(再生))

  • The studies were carried out to obtain the basic data for maximizing the protoplast yields from the mycelia of P. ostreatus and P. sajor-caju. Some factors affecting the regeneration of the protoplast of both species and the productivity of their reversion were also examined. The maximum yields of protoplasts were obtained from four days cultured mycelia of both species on cellophan membrane placed on the surface of PSA or MCM media in a petri dish. The optimal concentration of lytic enzyme Novozym 234 for protoplast releasing was 5 mg per ml of 0.5 M phosphate buffer solution with 0.6 M sucrose or 0.6 M $MgSO_4$ at pH 6.0. The greatest number of protoplasts was released 3 hours after incubation of the mycelia of P. ostreatus and after 4 hours for the P. sajor-caju in the lytic enzyme solution. Among the osmotic stabilizer solutions tested 0.6 M sucrose and 0.6 M KCl showed the best regeneration rates of the protoplasts of both species. When 0.75 % agar solution was over-layed on the regeneration media immediately after inoculation of the protoplast the regeneration rates were greatly enhanced. The ampicillin added to the agar solution prevented bacteria from infection. The reverted isolates produced the sporophores and basidial spores just like their parents without any mutations when they were cultivated in a broad mouth bottle with sawdust substrates.

  • Several antagonistic bacteria, SD-1, 4, 10, 11, 14, 15, and 16, which have strong CMCase and amylase activities, were isolated from the fermented mushroom media. Among them, SD-1, 10, 11, and 15 have strong antibacterial activities against the mushroom pathogenic bacteria Pseudomonas sp., and SD-1, 10, 11, 14, and 16 have strong antifungal activities against the mushroom pathogenic fungi, Trichoderma sp. SD-14, 15, and 16 did not inhibit the growth of mushroom Pleurotus eryngii ASI-2302, and Pleurotus ostreatus ASI-2042 and ASI-2180. When the culture broth mixture of the seven bacterial strains was applied to the mushroom media, the growths of pathogens, Pseudomonas sp. and Trichoderma sp., were inhibited.

  • For the purpose to verify the grain and evaluate milling properties of Korean waxy wheat, com­parison analysis between waxy wheat lines and their respective maternal parents were performed. The waxy lines showed various grain yields of 4.76${\~}$5.79 t/ha depending on parentages, which were corresponding to $80{\~}96\%$ levels of their respective maternal parents. One thousand grain weights of waxy lines were also lighter than its respective part in its parentage by exhibiting 32.8${\~}$34.6 g compared to 32.9${\~}$45.2 g of their parents. Test weights of waxy lines and their parents were 720${\~}$798 g/l and 786${\~}$797 g/l, respectively. The proportions of the grains above 2.5 mm in width were higher in order of Keumgang, Olgeuru, Geuru, SW97134, Suwon 292, Woori, SW97105, and SW97110. Waxy lines exhibited low milling properties by showing the straight flour yields ranging from $61.8\%$ to $67.1\%$ compared with the yields of their parents ranging from $66.1\%$ to $72.5\%$; the waxy lines were significantly lower in first break flour (Bl) and first reduction flour (Rl) yields in the Buller test mill, while significantly higher in the yields of second and third reduction flour (R2 and R3) than the respective ones of their parent wheat.

Dynamic Behavior of Model Set Net in the Flow (모형 정치망의 흐름에 대한 거동)

  • This experiment was carried out to measure the sinking depth of each buoy, the change in the net shape of the net, and the tension of sand bag line according to the R (from bag net to the fish court) and L (from fish court to the bag net) current directions and their velocity by the model experiment. The model net was one-fiftieth of the real net, and its size was determined after considering the Tauti’s Similarity Law and the dimension of the experimental tank. 1. The changes of the net shape were as follows : In the current R, the end net of fish court moved 20mm down the lowerward tide and 10mm upper part. So the whole model net moved up at 0.2m/sec. The shape of the net showed an almost linear state from bag net to the fish court at 0.6m/sec. In the current L, the door net moved 242mm down the lowerward tide and 18mm upper part. So the whole model net moved up at 0.2m/sec. The net shape showed an almost linear state from the fish court to the bag net at 0.5m/sec. 2. The sinking depths of each buoy were as follows: In the current R, the head buoy started sinking at 0.2m/sec and sank 20mm, 99mm at 0.3m/sec and 0.6m/sec, respectively. The end buoy didn't sink from 0m/sec to 0.6m/sec but showed a slight quake. In the current L, the end buoy started sinking at 0.1m/sec, and sank 5mm and 108mm at 0.2m/sec and 0.6m/sec, respectively. The whole model net sank at 0.5m/sec except the head buoy. 3. The changes of the sand bag line tension were as follows: In the current R, the tension affected by the sand bag line of the head buoy showed 273.51g at 0.1m/sec increased to 1298.40g at 0.6m/sec. In the current L, the tension affected by the sand bag line of the end buoy on one side showed 137.08g at 0.1m/sec increased to 646.00g at 0.6m/sec. The changes in the sand bag line tension were concentrated on the sand bag line of the upperward tide with increasing velocity at the R and L current directions. However, no significant increase in tension was observed in the other sand bag lines.

Clinical Findings and Gene Analysis of 3-Methylcrotonyl-CoA Carboxylase Deficiency (3-methylcrotonyl-CoA carboxylase 결핍증의 임상 양상과 유전자 분석)

  • Purpose: 3-methylcrotonyl CoA carboxylase deficiency (3MCCD) is leucine metabolic disorder caused by mutation in MCCC1 or MCCC2 gene. Clinical manifestations are variable, ranging from fatal neonatal onset to asymptomatic individuals. There is no retrospective study of Korean patients undergoing long-term treatment for 3MCCD. We reported this study to find out clinical symptoms and gene analysis of 3MCCD patients. Methods: This study was based on data of patients diagnosed with 3MCCD in Soonchunhyang university hospital between April 2009 and September 2013. We report clinical, enzymatic and mutation data of 3MCCD patients found by newborn screening. Results: In tandem mass spectrometry, 3-OH-isovalerylcarnitine (C5OH) of all patients increased. And all 7 patients were elevated 3-methylcrotonylglycine (3MCG) and 3-hydroxyisovaleric acid (3HIVA) in urine. MCCC mutation was identified in 2 patients and MCCC2 was mutated in 5 patients. We found mutation occurred in 8 different parts of nucleotide and such mutation caused 7 different types of changes in amino acid. All patients are on medication of L-carnitine and L-glycine. 4 patients are taking biotin. And 4 patients are eating leucine free formula. After starting treatment, there were no significant changes of urine 3MCG and 3HIVA levels. Conclusions: According to our data, MCCC2 gene mutation was more common than MCCC1 gene mutation. But the level of 3HIVA or 3MCG in urine has no correlation with phenotype. All patients has no symptoms and are shown normal development.

Chemical Composition and Fermentation Characteristics of the Corn Silage During Feedout at Yonchon of Gyeonggi-do (연천지역에 있어서 옥수수 사일리지의 개봉 후 경과기간에 따른 시료성분 및 발효품질)

  • The purpose of this study is to discuss the ways to consistently feed high quality corn silage(CS). This study evaluated the effect of the corn silage, after a certain time has elapsed, on the chemical composition and fermentation characteristics after feedout during the winter feeding period of the CS. Six samples of CS from four dairy farms(E1, E2, E3, and L1 ) were taken in order to feed the milking cows over a winter feeding Period from November of 2002 until February of the following year, 2003(six samples were taken at the fellowing dates in the following order: sample one was taken on the 23rd Nov. 2002, sample two on the 5th of Dec. 2002, 3rd sample on the 23rd of Dec. 2002, 4th sample on the 7th of Jan. 2003, 5th sample on the 22nd of Jan. 2003, and the 6th sampling was carried out on the 6th of Feb. 2003) at the three sampling sites after the opening of the trench silos at intervals of 15 days. In the dry matter contents of CS, there wasn't any specific tendency according to the elapsed time in the range of 21.3~$27.3\%$ at all low dairy farm(E1, E2, E3, and L1). And the average dry matter contents were 24.1, 25.9, 23.6, and $20.4\%$. Considering the Proper amount of the dry matter of CS during the ripen yellow stage, the appropriate moisture content was $33\%$ (NRC, 1989), and these dry matter contents were all low. A consistent tendency was not found in the contents of CS. The average of CP contents were 10.2, 8.0, 8.5, and $9.8\%$ at the E1, E2, E3, and L1 farms, and there were significant differences. The TDN contents of CS were not different among forms according to the time elapsed. The pH, according to the time elapsed after opening of the CS, there were no differences at each of E1, E2, E3, md L1 farms. Average pH were 3.5, 3.9, 3.6, and 4.1, md all of them were in normal range. In the lactic acid contents of CS, a consistent tendency was not found among four farms. But according to the time elapsed. there was a goat difference from 1.13~$7.8\%$ The acetate, propionate, and butyrate contents of CS were very low. In this study, there was no significant difference in the CS's chemical composition and fermentation characteristics according to the elapsed time at all four dairy farms. Considering the proper dry matter contents of CS during the ripen yellow stage, the appropriate dry matter content was $33\%$, and dry matter contents of few farms were all low. To enhance the quality of corn silage should be ensiled com at proper dry matter content range from 28 to $35\%$ Therefore, content of the corn plant should be always be closely monitored prior to beginning harvest.

  • The purpose of this study was to identify factors that might distinguish those who have a high level of Intention to purchase each of domestic, foreign designer and national brand jeans from those who have a low intention in terms of evaluative criteria belief, ideal jeans image and clothing altitude. The sample consisted of 198 male and 197 female students from five universities in Seoul. The questionnnaire consisted of 50 seven-point semantic differential scales dealing with evaluative criteria and ideal jeans image, beliefs about and intention to purchase domestic, foreign designer and foreign national brand jeans and 25 Likert type clothing attitude scales. Based on a series of t-tests the results showed that color and design were the most influencing factor among the evaluative criteria belief, regardless of brand type, while durability, accessory, sewing were the least. Sexy image, brand consciousnees and fashion interest were the important factor that distinguish high intention to purchase group fro)m low intention to purchase group.

Study on the Establishment of Barley Cultivation System using the Power Tiller (동력경운기를 중심으로한 맥류재배의 기계화 -관작업체계 확립에 관한 연구)

  • To maintain the self-sufficiency of food production , it is required to improved the productivity and increase the effective utilization of land in the double cropping areas using improved cultivation technology. The following reasons encourage this cours of action because the overlapping rice harvest and barely seeding seasons complete for labour, the lower price of barley makes it financially less attractive to grow and these two facts together are responsible for a decrease in barley being planted in certain more seasonably marginal double cropping areas. Fro these reasons, it is desirable to carry out tests to improve the current cultivation techniques . In this interest , the following studies were carried out in the experimental field located at Banweol-myeon, Whasung-jkun, Gyeonggi-do from October 1977 to July 1978 in order to establish the mechanical method of barley cultivation using the power tiller. The summarized results are as follows. 1. The work performance in the seeding operation using the rotary barley seeder was 68 minutes per 10 ares which compares favourably with 408 min/10a using the conventional method. This is only one sixth of the time required by th conventional method. The operating costs using the rotary barley seeder was 1, 463 won per 10 areas as against 3, 486 Won per 10 ares for the conventional method, showing a saving of about 50% in comparison with the conventional method. 2. In the manure spreading operation, the work performance was 25 min/10a for the manure spreading machine , compared to 1089 min/10a for the manual operations. This is about one fourth of the time required by the conventional method, The operating costs were 810 Won/10a for the machine and 857 Won/10a for the manual labour. This cost shows little advantage by the machine over the manual labor costs. The conventional method of manure spreading will continue pending decision which will develope and improve the machine.3. Work performance in the rolling operation using the barley rolIer was 30 min/lOa which compares favourably with 135 min/10a using conventional method. This was one fourth of the time required by the conventional method. The operating costs were 514 Won/103. for the machine and 1, 003 Won/lOa for manual labour. In the weed control operation, the work performance and operating costs were 45min/lOa, 1, 399won/10a for the herbcide application using the power sprayer, 1, 149min/10a 8, 541won/lOa for the conventional method respectively. This is 26 times higher efficiency in comparison to the conventional method. 4. In the harvesting operation using the reaper binder and tiller attachment, the work performance was 60min/lOa and represents a cost of 2, 039won/10a. The con\ulcornerventional method took 640min/10a at a cost of 4, 757won/lOa. The reaper binder showns a saving of one tenth over the conventional method. The automatic thresher is already recommended for the current situations, and is now being used. 5. From a comparison of the results of the above trials, the serial cultivation system using the attachment for the power tiller such as the rotary barley seeder is now to be recommended for the current barley cultivation system. It is also recommend from these results that the mechanized technology now available must be used to improve and maintain the increase in barley production. It is seen th1t this is the only course now avaihble to solve the peak seasonal requirements of labour needed for l11rvesting and seeding between rice and barley cultivation.

Effects of Selenium Supplying Methods on the Growth and Se Uptake of Hydroponically Grown Tomato Plants (Selenium공급방법이 수경재배 토마토의 생장과 Se 흡수에 미치는 영향)

  • This study was conducted to clarify the effects of supplying methods of selenium on the growth and Se uptake of hydroponically grown tomato plants. Tomato seeds (Lycopersicum esculentum Mill. cv. Momotaro T-93, Daki Seed Co.) were sown in plug tray with fifty holes, and raised for sixty days. Tomato seedlings transplanted to coco fiber slabs were supplied with the nutrient solutions adjusted to EC $2.3dS{\cdot}m^{-1}$ and pH $5.8\~6.2$ recommended by the Japanese Horticultural Experiment Station. Selenium forms used were inorganic $SeO_2$ (here in after referred to Se) and organic selenium chlenium with sugar fatty acid ester (here in after referred to chelated-Se). 10 ppm selenium solutions were treated to tomato plants with foliar applications, drenching, and foliar application plus drenching. Growth characteristics in terms of plant height, number of leaves, leaf area and chlorophyll content were significantly increased in the plot of foliar application ot Se, and in the plot of foliar application plus drenching of chelated-Se than other plots, respectively. Transported contents of selenium into the tomato fruits were highest as 0.302 ppm in the plot of foliar application plus drenching of chelated-Se. Also, it had tended to be higher in the plot of foliar application plus drenching than in the plots of foliar application or drenching in both of Se and chelated-Se. Foliar application and drenching of organic chelated-Se were effective to produce the functional tomato fruits.
