Association between PPARGC1A Genetic Polymorphisms and Type 2 Diabetes Mellitus in the Korean Population (한국인 대상의 PPARGC1A 유전적 다형성과 제2형 당뇨병과의 상관성)

  • The prevalence of type 2 diabetes mellitus (T2DM) is increasing worldwide. T2DM is one of the most common types of diabetes and is caused by increased insulin resistance and reduced insulin secretion. Peroxisome proliferator-activated receptor γ coactivator 1 alpha (PPARGC1A) is a master modulator of mitochondrial biogenesis and of gluconeogenesis in liver. In this study, we analyzed genetic polymorphisms of PPARGC1A gene in a middle-aged Korean population with T2DM. Using the genotype data of 736 T2DM cases and 4544 healthy controls obtained from the Korean Association Resource (KARE), we analyzed genetic correlations between single nucleotide polymorphisms (SNPs) of PPARGC1A and T2DM. Fifteen SNPs of PPARGC1A demonstrated a statistically significant association with T2DM. Of these, rs10212638 exhibited the strongest correlation with T2DM (P-value=0.015, OR=1.29, CI=1.05~1.59), and the minor G allele of PPARGC1A increased the risk of T2DM. This is the first study to report a significant association between genetic polymorphisms in PPARGC1A and T2DM and suggests that SNPs of PPARGC1A display genetic correlations to the etiology of T2DM.

A Study on Optimum Insemination Interval in Aged Egg-Type Fowl (산란노계의 인공수정에 있어서 적정주정간격에 관한 연구)

  • Present study was carried out to find out an optimum interval for artificial insemination in aged and poor laying hens, Brown colored, hundred and twenty commercial laying hens at about 530 days old and 20 S. C. W. Leghorn males at 580 days old were adopted Egg production rate during the experiment was 61.6% and number of eggs examined was 2,283. The intervals for the insemination were 2, 4 and 6 days for experimental groups T-1, T-2 and T-3, respectively. Eggs were collected on every 6th day and examined for fertility by candling after 5 days of incubation. Average fertility rates for T-1, T-2 and T-3 groups appeared 87.5, 81.1, and 65.1%, respectively. The fertility of T-3 group was significantly lower than those of T-1 and T-2 groups(P<0.05). The highest fertility rate was obtained on the second day after the insemination for all groups, i. e. 90.7, 84.0 and 71.7% for T-1, T-2 and T-3, respectively. Thereafter, a tendency of gradual decline in fertility was observed. This study suggests that the optimum interval for artificial insemination in aged hens is 2 days. The fertility tended to be improved by repeated injection. For the 2-day interval group, the highest rate (98.2%) was obtained on the 6th insemination.

  • 5주령시 병아리에 대하여 볶은 콩을 이용한 고열량 사료 1030Cal구 ($T_2$ 구)를 만들고 이를 830Cal의 관용사료구(C구)와 5주간 비교시험 한 결과 다음과 같은 성적을 얻었다. 1) 증체량에 있어서 $T_1$, $T_2$구는 C구에 비하여 증체량이 높으며 5%수준의 유의차가 있었다. 한편 L.S.D.-Test에 의하면 $T_1$과 C구 사이에만 5%의 유의차를 보였다. 2) 사료섭취량에 있어서는 $T_2$, $T_1$, C구의 순으로 사료를 절약할 수 있는데 L.S.D.-Test에 의하면 $T_2$구와 C구간에 1%의 유의차가 있으며 약 9%의 사료가 절약되었고 $T_1$구와 C구간에는 5%의 유의성이 있고 약 3%의 사료가 절약되었다. 3) 경제성에 있어서 추일수당 사료비는 $T_1$, $T_2$구 모두 C구보다 고가의 경향이 있어 볶은 콩의 실용가치를 감소시킨다. 그러나 육계 판매가격에 있어서 $T_1$구는 24.85원의 이익으로 C구 26.43원보다 하회하는 결과를 보인다. The Effects of Live Yeast(Saaccharomyces cerevisiae) Supplementation on the Performance of Laying Hens (활성효모 첨가가 산란계의 생산성에 미치는 영향)

  • In order to study the effects of supplementation of live yeast(Saccharomyces cerevisiae) on the performance of laying hens, five experiments were conducted. Two experiment were conducted during summer period, one with 37 wk old Dekalb-Delta strain laying birds(Exp. 1) and the other one with 100 wk old molted Nick Chick Brown laying birds(Esp.2) . In each experiment, 240 birds were divided into 12 groups of 20 birds each and randomly distributed. Each of the two experimental diets(Control 71 and 0.05% live yeast supplemented 72) was fed to 6 groups for 4 wks in Exp.1 and 3 wks in Exp.2. Three experiments were conducted during winter period, Exp.2 with 54 wk old Hy-Line strain laying birds, Exp.4 with 52 wk old Hy-Line strain laying birds, and Exp.5 with 36 wk old broiler breeder(Indian River strain). In each experiment, 540 birds were divided into 18 groups of 30 birds each and randomly distributed. Each of the 3 experimental diets(Control:$T_1$0.05% live yeast supplemented:$T_2$ and 0.1% live yeast supplemented : T$_3$) was fed to 9 groups for 6 wks in Exp.3, 9 wks in Exp.4 and 4 wks in Exp.5. In Exp.4, Latin Square design was employed to determine the effects of switching feeds at 3 wk intervals. All hens were housed in cages of commercial farm and experimental diets were made with commercial layer feeds. In Experiment 1, egg production was significantly(P<0.05) higher in $T_2$. Feed intake was significantly (P<higher in 72 at 1st wk but 4 wk average was not significantly different. Feed efficiency was significantly(P<0.01) better in 72 at End wk but 4 wk average was not significantly different. Other parameters, such as weight, soft egg production, cracked egg production and mortality were not significantly different. In Experiment 2, egg production was significantly(P<0.05) higher in $T_2$. Feed efficiency was significantly (P<0.05 and P<0.01) better at End wk and 3rd wk but 3 wk average was not significantly different. Soft egg production was significantly(P<0.05) higher in 72. Other parameters were not significantly different. In Experiment 3, egg productions were significantly(P<0.05) different among treatments : $T_3$ was higher than $T_1$ and $T_2$ was higher than $T_1$. Egg weight of $T_1$ and $T_2$was significantly(P<0.05) heavier than $T_3$. Feed intake of $T_2$ and $T_3$ was significantly(P<0.05) higher than $T_1$ at 6th wk but overall average was not significantly different. Soft egg production were significantly(P<0.01) different among treatments:$T_1$ was higher than $T_3$ was higher than $T_2$. Feed efficiency cracked e99 Production and mortality were not signifcantly different. In Experiment 4, egg production tended to increase as the level of live yeast supplementation increased but they were not statistically different. In Experiment 5, egg production of broiler breeders of $T_3$ was significantly(P<0.01) higher than $T_1$. Feed intake of $T_3$ was significantly(P<0.05) greater than $T_1$ and $T_2$ at 3rd wk but overall average was not significantly different. Fertility and hatchability tended to be higher in the supplemented groups than in the control.

Utilization of Plant Phytase to Improve Phosphorous Availability for Broiler (육계의 인 이용율 향상을 위한 식물성 Phytase의 이용)

  • This study was conducted to evaluate the efficacy of wheat and wheat bran  as the source of phytase in a 5 week broiler feeding trial. One thousand day-old broiler chickens(Ross$^{(R)}$) were divided into 20 pens of 50 broilers(25 male and 25 female) each. Four pens were randomly arranged to one of the five dietary treatments: T1, control diet containing normal nonphytate P(NPP) ;  T2, T1 - 0.1% NPP; T3, T2 + 600IU microbial phytase(NOVO$^{(R)}$) per kg diet; T4, T2 + 600IU plant phytase from wheat and wheat bran; T5, T2 + 600IU plant phytase from wheat and hydrothermally treated wheat bran. Reduction of NPP level by 0.1%(T2) reduced weight gain and feed intake but plant phytase treatments(T4 and T5) recovered the lost performance. Plant phytase treatments showed better (p<0.05) weight gain and intake than the microbial phytase treatment(T3). There was no difference between regular wheat bran treatment(T4) and hydrothermally treated wheat bran treatment(T5). Mortality was the highest by low NPP diet(T2). Availability of ether extract and crude ash of grower diet was the highest(p<0.05) in normal wheat bran diet(T4). Availability of Ca and P of grower diet was the highest(p<0.05) in T4 followed by T3 and T5. Availability of Mg, Fe and Zn was drastically improved by phytase treatments(T3, T4 and T5). Excretion of Ca, P, Mg, Fe and Zn was the lowest(p<0.05) with microbial phytase treatment(T3). Serum level of Ca and Mg was the highest(p<0.05) with the low NPP treatment(T2). Tibial ash content of T2 and T3 was lower(p<0.05) than that of T1, T4 and T5. However, tibial Ca content was higher(p<0.05) in T1 and T2 than other treatments. Tibial P and Mg contents were the highest(p<0.05) in T1. It was concluded that plant phytase from wheat bran can be effectively used to improve P utilization. Hydrothermal treatment of wheat bran prior to inclusion in the diet had no beneficial effects.

Investigation on the Characteristics Variation According to Air Supply Capacity in Layer Manure by Composting (공기공급량에 따른 산란계분의 퇴비화 특성변화에 관한 연구)

  • The composting of layer manure is economical and efficiently process. In this study, the variation of composting characteristics in layer manure was investigated according to air supply capacity. The fermented compost was added in layer manure and mixed with sawdust inside composting reactors. The level of air supply capacity was varied in the range of $50{\sim}200\;{\ell}/m^3/min$. During composting the temperature variations of composting piles was different the temperatures of composting piles for T-1 ($50\;{\ell}/m^3/min$) and T-2 ($100\;{\ell}/m^3/min$) were reached at $40^{\circ}C$ and $50^{\circ}C$ within 2 days, respectively. For T-3 ($150\;{\ell}/m^3/min$) and T-4 ($200\;{\ell}/m^3/min$), their temperatures was $60^{\circ}C$ within same days and maintained during 8 days. Water contents decreased according to the air supply capacity; 8.9%, 15.4%, 18.0% and 18.6% for T-1, T-2, T-3 and T-4. The weight ratios of T-1, T-2, T-3 and T-4 were reduced to 12.8%, 15.6%, 18.1% and 17.9%, respectively. The decreasing volumetric ratios of T-1, T-2, T-3 and T-4 were 18.0%, 21.0%, 22.3% and 22.0%. The oxygen discharge concentrations during composting were 12 ppm for T-1, T-2 and 9 ppm for T-3 and T-4. After composting, fertilizer components such as total nitrogen (TN) and phosphorous pentoxide ($P_2O_5$) were examined at each air supply capacity. Nitrogen contents of the T-1, T-2, T-3 and T-4 were 0.75%, 0.74%, 0.72% and 0.64%. Also, The contents of $P_2O_5$ were 0.35%, 0.40%, 0.38% and 0.42% for T-1, T-2, T-3 and T-4.

EFFECT OF NITROGEN POSITION ON EXCITED STATE PROPERTIES OF 1-(9- ANTHRYL )-2-(n-QUINOLINYL)ETHENES

  • The fluorescence properties and photoisomerization behavior of 1-(9-anthryl)-2-(n-quinolinyl)ethene (n-AQE, n=2-4) have been investigated in various solvents. t-3-AQE is strongly fluorescent, but does not accomplish photoisomerization, similar to parent hydrocarbon compound, t-1-(9-anthryl)-2-phenylethene (t-9-APE) or t-1-(9-anthryl)-2-(1-naphthyl)ethene (t-1-ANE). Fluorescence and photoisomerization oft-2-AQE and t-4-AQE are strongly affected by solvent polarity. Dependence of fluorescence quantum yield on the solvent polarity is moderate for t-2-AQE and large for t-4-AQE. In nonpolar solvent (in n-hexane), they exhibit relatively strong fluorescence, but do not isomerize to cis isomer on irradiation, even if inefficient isomerization is observed for t-4-AQE. However, as solvent polarity increases, their fluorescences become weak with efficient photoisomerization to corresponding cis isomer. Intramolecular charge-transfer excited state is presumed to contribute to photoisomerization. The S$_1$ decay parameters were found to be solvent-dependent due to the charge-transfer character of lowest S$_1$ state. In polar solvents, the activation barrier to twisting is reduced enhancing the isomerization of r-2-AQE and t-4-AQE in the singlet manifold.

Effects of Different Fertilization Levels and Oversowing on Liveweight Gains of Grazing Cattle in Tall fescue (Festuca arundinacea Schreb.) Dominant Pasture (Tall fescue(Festuca arundinacea Schreb.) 우점초지 시비 및 보파에 의한 방목축의 증체 비교)

  • This study was carried out to determine the effects of the fertilization levels and oversowing treatment on liveweight gain of glazing cattle, changs of botanical composition, and dry matter(DM) yield in tall fescue dominant mixed pasture during the grazing period. The treatments were T$_1$(low fertilizing; 120-100-100 kg/ha), T$_2$(medium fertilizing; 280-200-200 kg/ha) and T$_3$(medium fertilizing+oversowing). The botanical composition of tail festuce was increased in T$_1$ and that of tall fescue, orchardgrass and pernnial ryegrass in T$_3$ was 30.5%, 23.8% and 24.1%, respectively. The total forage DM yield was the highest in T$_3$, and the average stocking rate (animal unit; AU) per day during the grazing period in T$_1$, T$_2$ and T$_3$ was 2.4 AU. 3.0 Au and 3.3 AU, respectively. The total grazing days (animal unit day; AUD) in T$_3$(664 AUD) was higher than that of T$_1$, and T$_2$. There is no significant difference in average daily liveweight gain per head among the treatments but daily liveweight gain per ha in T$_3$ was higher than that of T$_1$, and T$_2$. The total liveweight gain per ha during the grazing period in T$_1$, T$_2$ and T$_3$ was 601kg. 762kg and 877kg, respectively. The herbage consumption per day per 100kg LW was similer among the treatments but crude protein, P, K and Ca contents in herbage were increased with medium fertilization levels(T$_2$) and with oversowing(T$_3$).

The Effects of Amino Acid Levels with Protein the Diet on Broiler Performance (사료의 단백질 수준에 따른 아미노산 첨가수준이 브로일러의 생산성에 미치는 영향)

  • This experiment was conducted to study the sparing effects of animal proteins on weight gain, nutrients utilizability and economic analysis of broiler. Experiment diet contained different ratio of animal and plant protein and were supplemented different levels of methionine and lysine for the respective protein levels. A total of 264 broiler chicks were fed four diets of control, $T_1$, $T_2$ and $T_3$ for 42 days from April 7, 1990 to May 19, 1990. Dietary protein levels of control, $T_1$, $T_2$ and $T_3$ for starter and finisher were 20~18, 18~16, 18~16 and 22~20%, respectively. Methionine and lysine levels of control, $T_1$, $T_2$ and $T_3$ were 0.4~1.1, 0.44~1.21, 0.48~1.32 and 0.48~1.32% for starter diet, respectively, and were 0.32~0.90. 0.35~0.99, 0.38~1.08 and 0.38~1.08% for finisher diet, respectively. The results obtained were summarized as follows. 1. The birds fed control diet gained most for overall period. $T_3$ treatment which was high in protein, methionine and lysine levels gained most for finisher period. 2 The birds fed control diet consumed most feed, and the birds fed T$_3$ diet consumed least feed. for overall period. Feed conversion during 1~4 weeks was better in $T_1$ (1.51) and $T_2$ (1.53) than in control (1.61) and $T_3$ (1.63) . During 4~6weeks, feed conversion was better in $T_3$ (1.37) and control(1.58) than T, (2.05) and T, (2.16) (P<0.01) 3. Dry matter, crude fiber and NFE utilizability were increased for 1~4 weeks and decreased for 4~6 weeks as methionine and lysine levels increased and crude protein utilizability tended to be increased as protein levels increased. 4. Abdominal fat content was lowest in bird fed control diet and was high in birds fed low protein diet Carcass percentage was highest at control and the abdominal fat content was higher in bird fed lower protein diet than bird fed other protein diets(P<0.05). 5. Feed cost per kg weight gain was lowest at $T_3$ which contained more soybean oil meal than other feeds and next was control. According to the results of this experiment, it was revealed that optimum protein, methionine and lysine levels for starter and finisher broiler diet were 20~18, 0.4~0.32 and 1.1~0.9%, respectively.
