Cell-meditated studies on blooming and growth of potentially ichthyotoxic Cochlodinium polykrikoides(Dinophyceae)

  • The fluctuations of biochemical and molecular activities III the harmful dinoflagellate, Cochlodinium polykrikoides, depending on water temperatures, were studied. In genomic DNA concentration, a similar value of 0.6 was shown at $12^{\circ}C$ and $15^{\circ}C$, but significantly increasing DNA from $18^{\circ}C$ (p<0.05), with a maximum of 1.8 at $24^{\circ}C$. After$24^{\circ}C$, the DNA significantly decreased to 0.6. Likely, the concentrations of RNA and total protein were at their highest values of 1.7 and 0.07 g $mL^1$ at $24^{\circ}C$, respectively. In contrast to ONA, RNA and total protein began to increase at $15^{\circ}C$. Oxygen availability between lower and higher temperatures was significantly different and increased from $18^{\circ}C$ according to light intensity, regardless of wavelengths (p<0.05). At $24^{\circ}C$, the highest value of the maximum electron transport rate (ETRmax), ranging from 537.9 (Ch 1) to 602.5 mol electrons $g^{-1}$ Ch1 a $s^{-1}$ (Ch 4), was also shown. Nitrate reductase (NR) and ATPase activities were at their highest values of 0.11 mol $NO_2^-g^{-1}$ Ch1 a $h^{-1}$ and 0.78 pmol 100 $mg^{-1}$ $at^2$ $4^{\circ}C$, respectively. When the cells cultured at $15^{\circ}C$, NR and ATPase activities significantly increased compared to $12^{\circ}C$ (p<0.05). In an analysis of CHN, the concentration of C and N also significantly increased (p<0.05). However, at $27^{\circ}C$, most of the molecular and biochemical movements were much lower, compared to $24^{\circ}C$. These results suggest that C. polykrikoides is very sensitive biochemical and molecular activities depending on water temperatures. Possibly, it is desirable to estimate at $18^{\circ}C$ the initiation of the massive blooming development of C. polykrikoides. In nature, it will be very difficult to maintain the massive blooms after $24^{\circ}C$ because of the possibility of significantly decreasing the molecular movement and activity of C. polykrikoides.

  • In this investigation, we prepared a 1 and 3-parallel serpentine single PEMFC, which has an active area of $100\;cm^2$ and a flow channel cross section of $1{\times}1mm$. Distribution and transport of water in a non-operating PEMFC were observed by varying flow types and the flow rates (250, 400, and 850 cc/min). This investigation was performed at the neutron imaging facility at the CO1d Neutron RAdiography facility (CONRAD), HMI, Germany of which the collimation ratio and neutron fluence rate are 250, $1{\times}10^{6}n/s/cm^2$, respectively. The neutron image was continuously recorded by a scintillator and lens-CCD coupled detector system every 10 seconds. It has been observed that although the distilled water was supplied into the cathode channel only, the neutron image showed a water movement from the cathode to the anode channel. The water at the cathode channel was completely discharged as soon as the pressurized air was supplied. But the water at the anode channel was not easily removed by the pressurized air except for the 3-parallel serpentine type with 850cc/min of air flow rate. Moreover, the water at the MEA wasn't removed for any of the cases.

  • Hydrogen could be produced from any substance containing hydrogen atoms, such as water, hydrocarbon (HC) fuels, acids or bases. Hydrocarbon fuels couold be converted to hydrogen-rich gas through reforming process for hydrogen production. Even though fuel cell have high efficiency with pure hydrogen from gas tank, it is more beneficial to generate hydrogen from city gas (mainly methane) in residential application such as domestic or office environments. Thus hydrogen is generated by reforming process using hydrocarbon. Unfortunately, the reforming process for hydrogen production is accompanied with unavoidable impurities. Impurities such as CO, $CO_2$, $H_2S$, $NH_3$, and $CH_4$ in hydrogen could cause negative effects on fuel cell performance. Those effects are kinetic losses due to poisoning of electrode catalysts, ohmic losses due to proton conductivity reduction including membrane and catalyst ionomer layers, and mass transport losses due to degrading catalyst layer structure and hydrophobic property. Hydrogen produced from reformer eventually contains around 73% of $H_2$, 20% or less of $CO_2$, 5.8% of less of $N_2$, or 2% less of $CH_4$, and 10ppm or less of CO. Most impurities are removed using pressure swing adsorption (PSA) process to get high purity hydrogen. However, high purity hydrogen production requires high operation cost of reforming process. The effect of carbon dioxide on fuel cell performance was investigated in this experiment. The performance of PEM fuel cell was investigated using current vs. potential experiment, long run (10 hr) test, and electrochemical impedance measurement when the concentrations of carbon dioxide were 10%, 20% and 30%. Also, the concentration of impurity supplied to the fuel cell was verified by gas chromatography (GC).

  • As a new technical approach, a hydraulic and magnetic clamp device was developed to realize a magnetic clamp crane system by simultaneously actuating eight individual hydraulic cylinders. In this approach, an Sr-type of ferritic permanent magnet (SrO· 6Fe2O3), rather than the previous electromagnet, was utilized for the purpose of lifting and transporting the large curved steel plates used for manufacturing ships. This study had the goal of developing and manufacturing a hydraulic, magnetic clamp prototype composed of three main parts, including the base frame, cylinder joint, and magnet joint, in order to safely transport curved steel plates. Furthermore, this research included a performance evaluation of the manufactured prototype and acquired the purposed quantity value in the performance test. The most significant item, the magnetic adhesive force (G), was evaluated in a performance test, which utilized a ferritic permanent magnet (Sr type) with 3700~4000 G of residual induction (Br) and 2640/2770 Oe of coercive force (Hc). In particular, relevant items such as the hoist tension (kN), transportation time (s), and applied load (Kgf) on the hydraulic cylinders were also evaluated in order to determine the optimum values.

Cloning and Sequence Analysis of Glyceraldehyde-3-Phosphate Dehydrogenase Gene in Yak

  • In order to study the biological function of gapdh gene in yak, and prove whether the gapdh gene was a useful intra-reference gene that can be given an important role in molecular biology research of yak, the cDNA sequence encoding glyceraldehyde-3-phosphate dehydrogenase from yak was cloned by the RT-PCR method using gene specific PCR primers. The sequence results indicated that the cloned cDNA fragment (1,008 bp) contained a 1,002 bp open reading frame, encoding 333 amino acids (AAs) with a molecular mass of 35.753 kDa. The deduced amino acids sequence showed a high level of sequence identity to Bos Taurus (99.70%), Xenopus laevis (94.29%), Homo sapiens (97.01%), Mus musculus (97.90%) and Sus scrofa (98.20%). The expression of yak's gapdh gene in heart, spleen, kidney and brain tissues was also detected; the results showed that the gapdh gene was expressed in all these tissues. Further analysis of yak GAPDH amino acid sequence implied that it contained a complete glyceraldehyde-3-phosphate dehydrogenase active site (ASCTTNCL) which ranged from 148 to 155 amino acid residues. It also contained two conserved domains, a NAD binding domain in its N-terminal and a complete catalytic domain of sugar transport in its C-terminal. The phylogenetic analysis showed that yak and Bos taurus were the closest species. The prediction of secondary structures indicated that GAPDH of yak had a similar secondary structure to other isolated GAPDH. The results of this study suggested that the gapdh gene of yak was similar to other species and could be used as the intra-reference to analyze the expression of other genes in yak.

감자와 무를 재배하는 사질양토 고랭지 밭의 시비량에 따른 비점오염 발생량 비교 (Comparison of Non-Point Pollution Occurrence by Amount of Fertilizer Applicetion from Sandy Loam Alpine Fields which Cultivetes Poteto and Radish in Korea)

  • This study was performed to monitor the runoff of sandy soils on alpine uplands between March 2008 and December 2009, and assess non-point source pollution load. The fields were used to cultivete poteto in 2008 and radish in 2009. The fertilizers used in 200S, compared to those used in 2009, contained 2.1 times of nitrogen, 1.9 times of phosphorous, and 2.3 times of potassium. In 2008, the annual pollution load indiceted SS 2,908.47kg/ha/yr, COD 67.95kg/ha/yr, BOD 50.72kg/ha/yr, TN l3.29kg/ha/yr, and TP 9.97kg/ha/yr. In 2009, the annual pollution load indiceted SS 3,908.34kg/ha/yr, COD 225.04kg/ha/yr, BOD 156.96kg/ha/yr, TN 18.88kg/ha/yr, and TP 36.41kg/ha/yr. The amount of fertilizers used was about twice greeter in 2008, but the amounts of TN in pollution load per unit of rainfall were similar by 0.031kg/ha/mm to 0.029kg/ha/mm, whereas the amounts of COD (0.16kg/ha/mm to 0.35kg/ha/mm), BOD (0.12kg/ha/mm to 0.24kg/ha/mm), and TP (0.023kg/ha/mm to 0.057kg/ha/mm) doubled in 2009. We can infer thet the surface covering by the growth of crop mainly affected the transport of T-N through the subsurface flow to reduce non-point source pollution.

세포내인자로서의 정교한 기능을 하는 molecular chaperone (Molecular chaperone as a sophisticated intracellular membership)

  • Molecular chaperone의 발견은 생명과학자들에게 살아있는 세포 내에서 어떻게 생체활성단백질이 만들어지고 유지되는지에 대한 자극과 함께 그것을 증명하기 위한 실험동기를 부여하였다. 초기에는 Molecular chaperone이 nucleosomes의 assembly에 관여하는 단백질을 설명하기 위하여 사용되었으나, 지금은 기본적인 세포생리기능의 하나인 단백질의 folding과 assembly를 돕는 assistant protein으로 주로 사용된다. 단백질합성 뿐만 아니라 단백질수송, oligomeric structure의 assembly와 disassembly, heat shock을 포함한 각종 내, 외부스트래스에 의해서 변성된 단백질의 세로내분화와 회복에도 Molecular chaperone이 관여하고 있다. 그러나 아직까지는 Molecular chaperone들의 3차구조와 그들간의 상호작용에 관한 정보가 부족하여 크게 진전되지 못하고 있지만, 많은 연구자에 의한 정보축적으로 인하여 빠른 시일 내에 Molecular chaperone에 세포내역할이 분명하게밝혀질 것이다.

분배성 추적자 시험법을 이용한 불균질 지반의 유류 오염도 평가 (The Evaluation of Petroleum Contamination in Heterogeneous Media Using Partitioning Tracer Method)

  • For the remediation of the subsurface contaminated by nonaqueous phase liquids(NAPLs), it is important to characterize the NAPL zone properly. Conventional characterization methods provide data at discrete points. To overcome the weak points of conventional characterization methods, the partitioning tracer method has been developed and studied. The average saturation of NAPL($S_n$), which is the representative and continuous saturation value within contaminated site, can be calculated by comparing the transport of the partitioning tracers to that of the conservative tracer in the partitioning tracer method. In this study, the application of the partitioning tracer method in heterogeneous media was investigated. To represent the heterogeneous condition of subsurface, a two-dimensional soil box was divided into four layers and each layer contained different sized soils. Soils in the soil box were contaminated by the mixture of kerosene and diesel, and partitioning tracer tests were conducted before and after the contamination using methanol as conservative tracer and 4-methyl-2-pentanol, 2-ethyl-1-butanol, and hexanol as partitioning tracers. The response curves of partitioning tracers from contaminated soils were separated and retarded in comparison with those from non-contaminated soils. The contamination of soils by NAPLs, therefore, can be detected by partitioning tracer method considering these retardations of tracers. From our experiment condition, the average saturation of NAPLs calculated by partitioning tracer method using the methanol as conservative tracer and hexanol as partitioning tracer showed the highest accuracy, though all results were underestimated. Further studies, therefore, were needed for improving the accuracy using the partitioning tracer test in heterogeneous media.

$LLC-PK_1$ 세포에서의 퓨모너신 $B_1$에 의해 유도된 스핑고리피드 대사 (Fumonisin $B_1$-induced Alteration of Sphingolipid Metabolism in $LLC-PK_1$ Cells)

  • The purpose of this study was to determine the effect of sulfinpyrazone on fumonisin $B_1$-induced elevation of free sphingoid bases in LLC-$PK_1$ cells. Fumonis ins are a family of mycotoxins produced by the fungi Fusarium moniliforme which is common contaminant in corn. Fumonisins are also potent inhibiors of sphingosine and sphinganine N-acyltransferases (ceramide synthases), key enzymes in sphingolipid metabolism resulting in the elevation of free sphinganine. The cytosolic concentration of fumonisin B1 was known to be closely proportional to the elevation of free sphinganine in LLC-PK1 cells [Yoo, H.-S., Norred, W.P., Wang, E., Merrill, A.H., Jr., and Riley, R.T. (1992) Toxicol. Appl.Pharmacol. 114. 9-15]. Sulfinpyrazone, an anion transport inhibitor, reduced the elevated level of free sphinganine resulting from fumonisin B1 inhibition of de novo sphingolipid biosynthesis. Fumonisin B1 at a concentration of 20${\mu}$M showed approximately 120pmol/$10^6$ cells relative to 3-10pmol/$10^6$ cells in control cultures, and sulfinpyrazone at a concentration of 200${\mu}$M partially reversed the increased level of free sphinganine induced by fumonisin $B_1$ down to normal level after exposure to fumonisin $B_1$ for 8 to 24hr. However, the reduced effect of sulfinpyrazone on the fumonisin $B_1$-induced elevation of intracellular sphinganine was not shown after 24hr. Fumonisin $B_1$ exposure to LLC-PK1 cells for 36 and 48hr showed approximately 74 and 80pmol per $10^6$ cells relative to 82 and 76pmol,respectively, in fumonisin $B_1$ plus sulfinpyrazone-treated cultures. Sulfinpyrazone-induced less elevation of free sphinganine in confluent cells after exposure to fumonisin $B_1$ suggested that either sulfinpyrazone may block the availability of fumonisin $B_1$ to cells or act on the fumonisin $B_1$ interaction with ceramide synthase.

Effects of STS and 1-MCP on Flower Opening and Lifespan of Potted Kalanchoe blossfeldiana Exported to Japan

  • This study was conducted to determine the effects of silver thiosulfate (STS) and 1-methylcyclopropene (1-MCP) on flower opening and lifespan of potted Kalanchoe blossfeldiana 'Oriba' for exportation. Ethylene inhibitors, STS and 1-MCP were applied to the kalanchoe plants prior to their export to Japan. STS 0.5 mM with 1% Tween 20 surfactant was directly sprayed (20 mL per plant) to leaves, buds, and flowers and 1-MCP 100 $nL{\cdot}L^{-1}$ was injected into sealed glass chambers containing kalanchoe plants, which were placed on the chambers for 6 hours. After transport to Japan, the plants were immediately transferred to a simulated retail condition room (80 ${\mu}mol{\cdot}m^{-2}{\cdot}s^{-1}$ for 12 hours of photoperiod at $22^{\circ}C$ and 64% RH) at Toyko University. The numbers of buds, open florets, and wilted florets in the middle inflorescence for each plant were counted right after export, 1 week after export, and 6 weeks after export. The percentages of open florets and wilted florets were calculated from the numbers. STS treatment resulted in 35% more open florets than the control and only 11% of wilted florets at 6 weeks after export to Japan which indicate the extension of lifespan of potted kalanchoe plants. Meanwhile, the plants exposed to 1-MCP before export did not show any significant differences in the numbers of buds and open florets and the percentages of open and wilted florets compared to control plants. In conclusion, STS 0.5 mM treatment strikingly induced better opening florets and lifespan of kalanchoe plants from 1 week to 6 weeks after export than control.
