Dynamic Model Considering the Biases in SP Panel data (SP 패널데이터의 Bias를 고려한 동적모델)

  • Stated Preference (SP) data has been regarded as more useful than Revealed Preference (RP) data, because researchers can investigate the respondents\` Preference and attitude for a traffic condition or a new traffic system by using the SP data. However, the SP data has two bias: the first one is the bias inherent in SP data and the latter one is the attrition bias in SP panel data. If the biases do not corrected, the choice model using SP data may predict a erroneous future demand. In this Paper, six route choice models are constructed to deal with the SP biases, and. these six models are classified into cross-sectional models (model I∼IH) and dynamic models (model IV∼VI) From the six models. some remarkable results are obtained. The cross-sectional model that incorporate RP choice results of responders with SP cross-sectional model can correct the biases inherent in SP data, and also the dynamic models can consider the temporal variations of the effectiveness of state dependence in SP responses by assuming a simple exponential function of the state dependence. WESML method that use the estimated attrition probability is also adopted to correct the attrition bias in SP Panel data. The results can be contributed to the dynamic modeling of SP Panel data and also useful to predict more exact demand.

Production of 6-hydroxynicotinic acid by nicotinic acid-assimilating Pseudomonas sp. (Pseudomonas sp.에 의한 nicotinic acid로부터 6-hydroxynicotinic acid의 생산)

  • For the production of 6-hydroxynicotinic acid from nicotinic acid, bacteria capable of assimilating nicotinic acid as a sole carbon, nitrogen and energy source were isolated from soils. Among them, SH-007, newly isolated strain having the best acitivity of nicotinic acid hydroxylase was selected and identified as Pseudomonas sp. The specific acitivity of nicotinic acid hydroxylase of Pseudomonas sp. was highest when the strain was cultured at $30^{\circ}C$ for 24 hrs in the medium (pH 7.5) containing 2 g nicotinic acid, 1 g $(NH_4)_2SO_4$, and 0.5 g peptone per liter. The addition of 1.5 g/l of nicotinic acid into the 24 hrs incubated culture medium resulted in the 12% higher specific acitivity of nicotinic acid hydroxylase than that of the non-added control after further 18 hrs incubation. In the 6-hydroxynicotinic acid production through the reaction with resting cells, 2.22 g/l of 6-hydroxynicotinic acid was produced from 2 g/l of nicotinic acid after incubation of 3 hrs under optimum conditions, which correpsonds to 98.2% of theoretical 6-hydroxynicotinic acid yield.

Effects of Fermentation to Improve Hygienic Quality of Powdered Raw Grains and Vegetables Raw Grains and Vegetables Using Lactobacillus sp. Isolated from Kimchi (김치분리균주 Lactobacillus sp.를 Starter로 한 발효생식 제조에서의 위생미생물 살균효과)

  • Improvement of hygienic quality of powdered raw grains and vegetables by fermentation was investigated. Luc-tobacillus sp. isolated from kimchi was used as a starter. The cell counts of coliform group and SS enteric bacteria on the SS agar plate in raw grains and vegetables were 2.3$\times$103 cfu/mL and 8.6$\times$10$^3$ cfu/mL, respectively. The starlet, Lactobacillu sp., reached 10$^{7}$ cfu/mL after 48 hr in fermentation. At that time, the coliform group and enteric bacteria on the SS agar plate were gradually inactivated and eliminated after 60 hr of fermentation. During the fermentation process, pH of the suspension was lowered and acidity increased. Antimicrobial activity of the acidic supernatant of fermented raw grains and vegetal]les against the E. coli sp. and Salmonella sp. was higher than that of lactic acid solution or neutralized supernatant. Therefore, it was considered that antimicrobial effect of the fermented raw grains and vegetal]les might be accelerated by tile synergic effect of acid and bacteriocin, and liquid fermentation of powdered raw grains and vegetables will be effective for inactivation of hygienic microorganisms.

The Anti-inflammatory Effects of Probiotic-produced Exopolysaccharide (프로바이오틱스 생산 exopolysaccharide에 의한 항염증 활성)

  • The present study isolated seven different kinds of probiotics from various food sources and identified them with Bacillus sp. and Lactobacillus sp. by 16S rDNA sequencing. Their supernatants were prepared after a 24 hr culture, and their effects on nitric oxide (NO) production in mouse RAW 264.7 cells were investigated. Among the treated samples, the culture supernatants of two strains (Bacillus sp. FG-1 and Lactobacillus sp. FG-6) significantly decreased NO production in LPS-activated RAW 264.7 cells. Moreover, they dramatically reduced the expression of pro-inflammatory genes such as COX-2, iNOS, and TNF-α. To examine whether exopolysaccharide (EPS) is responsible for the anti-inflammatory effects of probiotics, EPS was purified from the culture supernatants of Bacillus sp. FG-1 and Lactobacillus sp. FG-6 strains. The EPS treatment produced by FG-1 and FG-6 strains decreased NO production in a dose-dependent manner in LPS-stimulated RAW 264.7 cells without affecting cell viability, while also reducing pro-inflammatory gene expression. Overall, these results suggest that EPS might be one of the key molecules responsible for the anti-inflammatory effects of probiotics.

One unusual species, Coilia sp. (Engraulidae, Pisces) from the Yellow Sea

  • Four specimens of unknown Coilia sp. were collected for the first time from the Yellow Sea in 2008 and compared with Coilia mystus and Coilia nasus. Coilia sp. showed similar morphology to C. mystus and C. nasus, but differed in that its tail was considerably shorter. We conducted an analysis of the morphological and genetic characteristics in an effort to clarify the taxonomic position of Coilia sp. In counts and measurements, Coilia sp. were well distinguished from C. nasus by the number of scutes (42-44 in Coilia sp. vs. 40-45 in C. mystus vs. 45-55 in C. nasus), ratio of dorsal base length to head length (43.4-47.6 vs. 37.9-47.6 vs. 33.0-41.0), and eye length to head length (19.2-20.8 vs. 17.0-22.4 vs. 13.8-18.2). In caudal skeleton of Coilia sp., urostyle, hypural and epural bones were not observed; instead of them, caudal fin rays were supported by the last vertebra, neural and haemal spines' extension. The molecular phylogenetic relationship was analyzed using 414 base-pair 12S rRNA mitochondrial DNA sequences. The Kimura-2-parameter distance between Coilia sp. and C. mystus was 0.3%, but was 1.3% between Coilia sp. and C. nasus. Both the neighbor-joining tree and maximum-likelihood tree showed that Coilia sp. are closely clustered with C. mystus. Therefore, our results suggest that the Coilia sp. may be a deformed fish of C. mystus.

Evaluation of Fracture Toughness Using Small Punch Test for Aluminum 6061-T6 Type-3 Cylinder Liner (소형펀치시험법을 이용한 알루미늄 6061-T6 Type-3 용기 라이너의 파괴인성 평가)

  • Type-3 cylinder liner has a limitation of machining the standard specimen for fracture toughness test because it has approximately 5 mm in thickness as well as a curvature. Hence, it needs to be employed a miniature specimen test technique to evaluate fracture toughness of the cylinder liner. In this study, small punch (SP) test method was employed to evaluate fracture toughness of the cylinder liner. Load-displacement curve result measured from the SP test showed that the liner material was failed during membrane stretching in the general SP load-displacement curve. Additionally, it was shown that liner material was isotropic although the amount of plastic deformation was different depending on the direction due to manufacturing process characteristics. Fracture toughness, $J_{Ic}$, was evaluated using the SP test data. The value of fracture toughness obtained was $13.0kJ/m^2$. This value was similar to that of the same kind of materials. Therefore, the fracture toughness evaluated using the SP test data was reasonable.

Production of Cyclodextrin Glucanotransferase from Alkalophilic Bacillus sp. C-21 (호알칼리성 Bacillus sp. C-21에 의한 Cyclodextrin Glucanotransferase의 생산)

  • A strain of alkalophilic Bacillus sp. C-21 has been Isolated from sold. The strain was capable of producing large amount of cyclodextrin glycosyltransferase (CGTase) in the high alkaline pH medium. The preferable medium composition was determined to be as follows : 1.0% soluble starch, 1.0% peptone, 0.5% yeast extract, 0.1% K2HP04, 0.02% MgSO4.7H2O and 1.0% Na2CO3(pH 10.0) The highest enzyme production was observed after 30hours of cultivation at 33$^{\circ}C$. The optimum temperature and pH for the activity of crude enzyme were 6$0^{\circ}C$ and 6.0, respectively. The enzyme was stable between pH 6.0 and 9.6, and up to 55$^{\circ}C$.

  • This study was conducted to preserve viability of Aspergillus sp. PS-104, a phosphate-solubilizing fungus, for more than 6 months. The number of Aspergillus sp. PS-104 conidia stored at $25^{\circ}C$ with various concentrations of additives was counted on potato dextrose agar at the intervals of 2 weeks. When PEG 200 was added with concentration of 0.1%, viability of the conidia was increased more than 10 times to compare with control.

  • Objectives : The objectives of this clinical study is to evaluate the efficacy of acupuncture treatment (Sojangjeonggyeok(小腸正格), Sammgyo(SP-6)) and Kami-hwal-hyeol-tang(KHHT), Dangkwi-chungung-San for primary dysmenorrheic patient. Methods : The primary dysmenorrheic patient was treated with acupuncture treatment (Sojangjeonggyeok((小腸正格), Sammgyo(SP-6)) and Kami-hwal-hyeol-tang(KHHT), Dangkwi-chungung-San. Conclusion : The acupuncture treatment (Sojangjeonggyeok((小腸正格), Sammgyo(SP-6)) and Kami-hwal-hyeol-tang(KHHT), Dangkwi-chungung-San will be efficacious against primary dysmenorrheic patient.

Regulation of 3-Deoxy-D-arabinoheptulosonate-7-phosphate (DAHP) Synthase of Bacillus sp. B-6 Producing Phenazine-1-carboxylic acid

  • The 3-Deoxy-D-arabinoheptulosonate 7-phosphate (DAHP) synthase is the first enzyme of aromatic amino acid-, folic acid-, and phenazine-1-carboxylic acid biosynthetic pathways. DAHP synthase of Bacillus sp. B-6 that produces phenazine-1-carboxylic acid was feedback inhibited by two intermediary metabolites of aromatic amino acid biosynthetic pathways, prephenate and chorismate, but not by other metabolites, such as anthranilic acid, shikimic acid, p-aminobenzoic acid, and 3-hydroxyanthranilic acid. DAHP synthase of Bacillus sp. B-6 was not inhibited by end products, such as aromatic amino acids, folic acid, and phenazine-1-carboxylic acid. The inhibition of DAHP synthase by prephenate and chorismate was non-competitive with respect to erythrose 4-phosphate and phosphoenolpyruvate. Prephenate and chorismate inhibited 50% of the DAHP synthase activity at concentrations of $2{\times}10^{-5}\;M$ and $1.2{\times}10^{-4}\;M$, respectively The synthesis of DAHP synthase of Bacillus sp. B-6 was not repressed by exogenous aromatic amino acids, folic acid, and phenazine 1-carboxylic acid, single or in combinations.
