Isolation of a Pseudomonas aeruginosa Strain Producing an Extracellular Alkaline Lipase Catabolitely Regulated by Glucose, and Purification of the Lipase (포도당에 의해 합성억제되는 알카리성 Lipase를 생성분비하는 Pseudomonas aeruginosa 균주의 분리 및 이 효소의 정제)

  • Producing an extracellular alkaline lipase, this isolate JM123 was identified as a Pseudomonas aeruginosa strain from the results of the analyses of its morphological, biochemical and physiological properties. This strain showed the highest productivity of alkaline lipase when grown at pH 9.0 and 30C for 13-20 hours in the medium of 2% starch, 1% soytone, 0.5% peptone and 1% MgSO4.7H2O. However, this enzyme was greatly repressed when grown in the glucose containing medium. The culture broth was fractionated by the order of the ammonium sulfate precipitation, Sephadex G-200 gel filtration, DEAE-cellulose column chromatography, and Sephadex G-150 gel filtration.

  • In order to obtain a good microorganism capable of degrading microcrystaline cellulose (avicel), the screening test was carried out from soil and brown-rot wood. 8 strains which had good avicel-hydrolyzing activity were isolated. Among them, HK 47 which exhibited the highest avicel hydrolyzing activity was identified as Trichoderma sp. HK 47. Maximum avicel-hydrolyzing enzyme production from Trichoderma sp. HK 47 was obtained with the optimum medium contained carboxymethylcellulose 1.5% as carbon source, NaN030.75% as nitrogen source, KH2P040.5%, MgSO4.7H2O 0.1%, Tween 800.005% (V/V) during stationary cultivation at pH 6.0, 3$0^{\circ}C$ In this case, the production of avicel-hydrolyzing enzyme was 0.028 U/ml.

  • The growth characteristics of Candida tropicalis KIST 359, isolated from soil samples collected at an oil depot in Korea, have been studied by cultivating batches under varying conditions. The conclusions of the study were: 1. The yeast easily assimilates hydrocarbons in a range of $C_{14}-C_{17}$, and the optimum cultivation temperature and pH are $30^{\circ}C$ and 5.5, respectively. 2. Using this strain of micro-organism, gas oil gives a higher cell yield than kerosine and with gas oil except urea all other nitrogen sources $(NH_4NO_3,\;NH_4Cl,\;(NH_4)_2SO_4\;and\;(NH_4)_2HPO_4)$ similarly support a satisfactory growth of the yeast. 3. The highest yield is obtained with a gas oil level of 10%(v/v), and concentrations of nitrogen source and $MgSO_4{\cdot}7H_2O$ of 0.5 and 0.05%(w/v), respectively. 4. The protein content of dried yeast cells is 59.8%. Its amino acid composition can be compared well with that of FAO provisional patterns, but with a low methionine and a high lysine content.

  • Serratia marcescens CK-3, decomposing chitin which is a mar component of cell wall in phyitopathogenic fungi, was isolated from the continuous cropping rhizosphere of pepper and cucumber and its enzymatic property was examined. S. marcescens CK-3 was found tn have an tagonistic effects against, Fusarium axysporum and Rhizoctonia solani and to have complex enzyme system such as chitinase, laminarinase, and proteinase. The preferable composition of the medium for production of chitinase was fond and was as follows : colloidal chitin 1.5%, tryptone 0.5%, glucose 1.0%, peptone 0.2%, $MgSO_4{\cdot}7H_2O\;0.1%,\;K_2HPO_4\;0.1%,\;and\;NaCl\;0.1%$(w/v), pH 6.8. The maximum enzyme production was observed after culture of 72 hours at $30^{\circ}C$ using a medium containing the above chemical composition. The optimal pH and temperature for in vitro activity of chitinase from S. marcescens CK-3 were pH 7.5 and $50^{\circ}C$, respectively. The enzyme activity in-creased by metal ions such as$Ag^+$ and $Mn^{++}$.

Biomimetics of Nano-pillar (나노섬모의 자연모사 기술)

  • The cochlea of the inner ear has two core components, basilar membrane and hair cells. The basilar membrane disperses incoming sound waves by their frequencies. The hair cells are on the basilar membrane, and they are the sensory receptors generating bioelectric signals. In this paper, a biomimetic technology using ZnO piezoelectric nano-pillar was studied as the part of developing process for artificial cochlea and novel artificial mechanosensory system mimicking human auditory senses. In particular, ZnO piezoelectric nano-pillar was fabricated by both low and high temperature growth methods. ZnO piezoelectric nano-pillars were grown on solid (high temperature growth) and flexible (low temperature growth) substrates. The substrates were patterned prior to ZnO nano-pillar growth so that we can selectively grow ZnO nano-pillar on the substrates. A multi-physical simulation was also conducted to understand the behavior of ZnO nano-pillar. The simulation results show electric potential, von Mises stress, and deformation in the ZnO nano-pillar. Both the experimental and computational works help characterize and optimize ZnO nano-pillar.

식초산 발효에 관한 연구

  • 식초산 발효 실험을 통하여 다음과 같은 결과를 얻었다. 1. 자연에서 초산균 52균주를 분리하고 그중 식초산 농도 5.5% 이상되는 초산균 CAU-4, CAU-15, CAU-17, CAU-46, 4균주를 선별하였다. 2. 정치배양과 진탕배양을 실시한 결과 산생성랑은 진탕배양에서 월등한 효과를 보였다. 3. 유기태 질소원의 첨가시 peptone을 0.4%까지 증가를 보였고, yeast extract는 0.1~0.2%가 최적량이고 그 이상은 산량이 감소되었다. 4. $NH_4NO_3의$ 자화성을 조사한 결과 CAU-17만이 자화하지 못하였고, 무기태 질소원으로서는 $(NH_2)_2CO가$ 가장 좋았다. 5. 무기염류는 $KH/_2PO_4,$ $MgSO_4.7H_2$ O 0~0.1% 첨가하여 약간의 효과를 보았다. 6. Glucose의 첨가량별 산량의 증가는 0.25% 첨가시 0% 첨가시 보다 증가를 보인 반면 1% 첨가시는 오히려 감소하였다. 7. 초발 alcohol농도가 낮을수록 유도기와 대수기가 단축되며 10%이상에서는 4균주 모두 산을 생성하지 못하였다. 8. 초발산도가 높을 경우 유도기가 길어지고 산생성량이 줄어 들었다. 9. 초산균의 배양온도는 $30^{\circ}C,$ $35^{\circ}C에서$ 산생성 속도가 가장 빨랐다. 10. 발효시 산생성량에 따른 세균수의 증가는 접종후 2~3일 사이에 가장 많았다. 11. 초산의 증가와 더불어 균체량은 증가하였고 pH는 약간색 감소하나 PH는 2.5이하로는 내려가지 않았다. 12. alcohol 잔량은 발효 개시 후 5 ~6일의 정지기에 가장 낮았다.

Biocidal Effects of Chlorine Dioxide on Isolated and Identified Pathogens from Nosocomial Environment - Biochemical and Technical Covergence (병원내 환경으로부터 분리 및 확인된 병원균에 대한 이산화염소의 살균 효과 - 생화학 및 기술 융합)

  • In this study, microorganisms were isolated from nosocomial environment and are identified by biochemical analysis as the part of biochemical and technical convergence. Microorganisms were collected at intense care unit of general hospital located in Pyeongtak (2014.11.28. - 2014. 11. 30). Using a VITEK2 equipment of biochemical approaches, eleven microorganisms e.g., Micrococcus luteus (or M. lylae), Granulicatella adiacens (M. luteus or M. lylae), Staphylococcus caprae, Sphingomonas paucimobilis, Kocuria kristinae, G elegans, Aerococcus viridans (or Staphylococcus arlettae), Methylobacterium spp., Dermacoccus nishinomiyaensis (or Kytococcus sedentarius), Kocuria kristinae (or M. luteus, M. lylae), Pseudomonas oryzihabitans were identified. And then identified bacteria plates were applied with a plastic stick, so called with "FarmeTok (medistick/Puristic) to produce ClO2. ClO2-releasing plastic stick showed the very strong inhibition of bacterial growth with about 99.9%. There were no bacterial colonies on the ClO2-incubated plate. Taken together, it is suggested that chlorine dioxide should be very strong inhibitor to microorganisms of nosocomial infections.

  • The pilot scale experiments can handle the flue gas up to 1,000 $Nm^3/hr$ for separation of carbon dioxide included in real flue gas at coal-fired power plant. The operational characteristics was analyzed with the main experimental variables such as flue gas flow rate, absorbent circulation rate using chemical absorbents mono-ethanolamine( MEA) and 2-amino-2-methyl-1-propanol(AMP). The more flue gas flow rate decreased in 100 $m^3/hr$ in the MEA 20 wt% experiments, the more carbon dioxide removal efficiency was increased 6.7% on average. Carbon dioxide removal efficiency was increased approximately 2.8% according to raise of the 1,000 kg/hr absorbent circulation rate. It also was more than 90% at $110^{\circ}C$ of re-boiler temperature. Carbon dioxide removal efficiency of the MEA was higher than that of the AMP. In the MEA(20 wt%) experiment, carbon dioxide removal efficiency(85.5%) was 10% higher than result(75.5%) of ASPEN plus simulation.

Studies on Conditions of the Gluconic Acid Production by a Mold isolated from the Soil of Seoul Area (Aspergillus SP. 균주에 의한 Gluconic Acid 생산조건에 관한 연구)

  • Fungi which were capable of producing gluconic acid were isolated from soil and tree leave samples, which had been collected in Seoul ana its vicinity. Among the 19 strains isolated, a strain named arbitrarily KUF-O4 was selected as a test strain chiefty because of its efficiency in gluconic acid production. The strain was identified as an Aspergillus sp. through its morphological properties. Optimum conditions for the gluconic acid production of KUF-O4 were investigated. The results obtained are as follows. 1. An incubation period of at least 30 hours was required for a good yield of gluconic acid. 2. A medium containing 10% glucose needed at least 3 % CaCo$_3$to maintain the optimum pH for the production of gluconic acid during fermentation. 3. As a carbon source, glucose was the most effective one among the carbon sources tested. 4. As a nitrogen source, an ammonium salt was more effective than any other form of nitrogen compounds. 5. As mineral source, a small amount of both KH$_2$PO$_4$and MgSO$_4$was found to be necessary to increase the efficiency of the gluconic acid production.

Medium Optimization for Fibrinolytic Enzyme Production by Bacillus subtilis MG410 Isolated (Bacillus subtilis MG410에 의한 Fibrin 분해효소 생산배지의 최적화)

  • Using the bacteria isolated from Chungkookjang, Bacillus sublilis MG410 which is excellent in fibrinolytic enzyme activity was isolated. In increase the high production of fibrinolytic enzyme from Bacillus sublilis MG410, the effect of various carbon sources, nitrogen sources, inorganic sources, the initial pH of medium were investigated. The most effective carbon and nitrogen sources were founded cellobiose 0.5%(w/v) and soybean meal 2%(w/v) respectively. None of inorganic sources examined had any detectable stimulating effect on fibrinolytic enzyme production except Na₂HPO₄·12H₂O. The initial optimum pH for fibrinolytic enzyme production ranged from 5∼6 and agitation speed was effect at 150rpm. In jar fermentor experiments under optimal culture conditions, the activity of fibrinolytic enzyme reached about 5.050 unit after 48hours.
