Disease Progression from Chronic Hepatitis C to Cirrhosis and Hepatocellular Carcinoma is Associated with Increasing DNA Promoter Methylation

  • Background: Changes in DNA methylation patterns are believed to be early events in hepatocarcinogenesis. A better understanding of methylation states and how they correlate with disease progression will aid in finding potential strategies for early detection of HCC. The aim of our study was to analyze the methylation frequency of tumor suppressor genes, P14, P15, and P73, and a mismatch repair gene (O6MGMT) in HCV related chronic liver disease and HCC to identify candidate epigenetic biomarkers for HCC prediction. Materials and Methods: 516 Egyptian patients with HCV-related liver disease were recruited from Kasr Alaini multidisciplinary HCC clinic from April 2010 to January 2012. Subjects were divided into 4 different clinically defined groups - HCC group (n=208), liver cirrhosis group (n=108), chronic hepatitis C group (n=100), and control group (n=100) - to analyze the methylation status of the target genes in patient plasma using EpiTect Methyl qPCR Array technology. Methylation was considered to be hypermethylated if >10% and/or intermediately methylated if >60%. Results: In our series, a significant difference in the hypermethylation status of all studied genes was noted within the different stages of chronic liver disease and ultimately HCC. Hypermethylation of the P14 gene was detected in 100/208 (48.1%), 52/108 (48.1%), 16/100 (16%) and 8/100 (8%) among HCC, liver cirrhosis, chronic hepatitis and control groups, respectively, with a statistically significant difference between the studied groups (p-value 0.008). We also detected P15 hypermethylation in 92/208 (44.2%), 36/108 (33.3%), 20/100 (20%) and 4/100 (4%), respectively (p-value 0.006). In addition, hypermethylation of P73 was detected in 136/208 (65.4%), 72/108 (66.7%), 32/100 (32%) and 4/100 (4%) (p-value <0.001). Also, we detected O6MGMT hypermethylation in 84/208 (40.4%), 60/108 (55.3%), 20/100 (20%) and 4/100 (4%), respectively (p value <0.001. Conclusions: The epigenetic changes observed in this study indicate that HCC tumors exhibit specific DNA methylation signatures with potential clinical applications in diagnosis and prognosis. In addition, methylation frequency could be used to monitor whether a patient with chronic hepatitis C is likely to progress to liver cirrhosis or even HCC. We can conclude that methylation processes are not just early events in hepatocarcinogenesis but accumulate with progression to cancer.

True Digestibility of Phosphorus in Different Resources of Feed Ingredients in Growing Pigs

  • To determine the true digestible phosphorus (TDP) requirement of growing pigs, two experiments were designed with the experimental diets containing five true digestible P levels (0.16%, 0.20%, 0.23%, 0.26% and 0.39%) and the ratio of total calcium to true digestible P (TDP) kept at 2:1. In Experiment 1, five barrows (Duroc${\times}$Landrace${\times}$Yorkshire) with an average initial body weight of 27.9 kg were used in a $5{\times}5$ Latin-square design to evaluate the effect of different dietary P levels on the digestibility and output of P and nitrogen. In Experiment 2, sixty healthy growing pigs (Duroc${\times}$Landrace${\times}$Yorkshire) with an average body weight (BW) of 21.4 kg were assigned randomly to one of the five dietary treatments (12 pigs/diet), and were used to determine the true digestible phosphorus (TDP) requirement of growing pigs on the basis of growth performance and serum biochemical indices. The results indicated that the true digestibility of P increased (p<0.05) linearly with increasing dietary TDP level below 0.26%. The true P digestibility was highest (56.6%) when dietary TDP was 0.34%. Expressed as g/kg dry matter intake (DMI), fecal P output increased (p<0.05) linearly with increasing P input. On the basis of g/kg fecal dry matter (DM), fecal P output was lowest for Diet 4 and highest (p<0.05) for Diet 5. The apparent digestibility of crude protein (CP) did not differ (p>0.05) among the five diets, with the average nitrogen output of 12.14 g/d and nitrogen retention of 66% to 74% (p>0.05), which suggested that there was no interaction between dietary P and CP protein levels. During the 28-d experimental period of Experiment 2, the average daily gain (ADG) of pigs was affected by dietary TDP levels as described by Eq. (1): $y=-809,532x^4+788,079x^3-276,250x^2+42,114x-1,759$; ($R^2=0.99$; p<0.01; y = ADG, g/d; x = dietary TDP, %), F/G for pigs by Eq. (2): $y=3,651.1x^4-3,480.4x^3+1,183.8x^2-172.5x+10.9$ ($R^2=0.99$; p<0.01; y = F/G; x = dietary TDP, %), and Total P concentrations in serum by Eq. (3): $y=-3,311.7x^4+3,342.7x^3-1,224.6x^2+195.6x-8.7$ (R2 = 0.99; p<0.01; y = total serum P concentration and x = dietary TDP, %). The highest ADG (782 g/d), the lowest F/G (1.07) and the highest total serum P concentration (3.1 mmol/L) were obtained when dietary TDP level was 0.34%. Collectively, these results indicate that the optimal TDP requirement of growing pigs is 0.34% of the diet at a total Ca to TDP ratio of 2:1.

Relationships between Air Pollution by SO2 and Soluble Sulphur Contents in the Leaves and Bark pH in Urban Forest Trees (도심지역(都心地域)의 아황산(亞黃酸) 가스에 의(依)한 대기오염(大氣汚染)과 수목내(樹木內) 엽(燁)의 수용성(水溶性) 유황(硫黃) 함량(含量) 및 수피산도(樹皮酸度)와의 관계(關係))

  • This study was carried out to investigate relationships between atmospheric $SO_2$ concentration and both soluble sulphur contents in the tree leaves and bark pH to test the possibility of using them as bioindicators for air pollution. Mt. Kwanak, Mt. Nam, Mt. Bukhan (located in Seoul) and Mt. Paldal (located in Suwon, Kyonggi-do) as polluted areas and Pyongchang, Kwangwon-do as an unpolluted area were selected for this study. Soluble sulphur contents in the leaves and hark pH of two tree species (Pines densiflora S. et Z. and Quercus mongolica Fisch.) were analyzed in May, August, and October, 1990 by $BaSO_4$ precipitation method and pH measurement, respectively. In both species, concentration of soluble sulphur in the leaves increased with increasing concentration of atmospheric $SO_2$ (correlation coefficient : 0.52). Soluble sulphur contents in the 2-year-old needles of Pines densiflora (0.170%) and current year leaves of Quercus mongolica (0.081%) in Mt. Nam in the center of Seoul were higher than those in unpolluted Pyongchang area (0.023% and 0.034%, respectively). Bark pH decreased with increasing atmospheric $SO_2$ concentration (correlation coefficient : -0.52). Bark pH of P. densiflora (pH 3.42) and Q. mongolica (pH 3.63) in Mt. Nam were lower than those in Pyongchang area (pH 3.94 and pH 4.93, respectively). Both soluble S content in the leaves and bark pH were recognized as suitable bioindicators for air pollution by $SO_2$. Especially, bark pH showed more sensitive response to air pollution by $SO_2$ than soluble S concentration in the leaves. The lowest concentration of soluble sulphur and the highest bark pH in August were considered to be due to heavy rain during the rainy season. Soluble S content in the leaves and bark pH were not significantly different at 5% level between the two species in polluted areas.

A Study on the Effects of Temperature Rise of Irrigation Water Passed Through the Warm Water Pool. (온수지에 의한 관개용수의 수온상승 효과에 관한 연구)

  • The study was to estimate the effect of the rise of water temperature in the warm water pool and to make contribution to the establishment of reducing to a damage of cool water as well as to the planning for warm water pool. This observation was performed in Wudu warm water pool located at Wudu-Dong of Chuncheon for two years from 1975 to 1976. The results were showed as follows; 1. The daily variation of water temperature was the least for inset (No.1; 0.6$^{\circ}C$) the second for middle overflow (No2: 3$^{\circ}C$, No.3; 2.3$^{\circ}C$) and another for outflet (No.4; 3.6$^{\circ}C$, No.5; 3.8$^{\circ}C$) And the highest reaching time of water temperature in each block was later about 1 hour than the time at which air temperature happend in the daytime. So, the variation of water temperature was sensitive to the variation of air temperature 2. The monthly variation of water temperature at each measuring point was plotted to be increased with increase in air temperature till August (Mean monthly rising degree; No.1; 1.15$^{\circ}C$, No.2; 1.7$^{\circ}C$, No.3; 1.73$^{\circ}C$, No.4; 2.08$^{\circ}C$, No.5; 2.0$^{\circ}C$), and expressed gradually descended influence upon water temperature after August. 3. The mean temperature of inflow folwed in warm Water pool was 7.5∼12.5$^{\circ}C$, and outflow temperature was described as 13.4∼22.5$^{\circ}C$ to be climbed. And So, the rising interval of water temperature was shown as 6.7∼10.4$^{\circ}C$. 4. The correlation between the rising of water temperature and the weather condition was found out highly significant. As the result, their correlation coefficents of water temperature depending on mean air temperature, ground temperature, wind velocity and relative humidity were to be 0.93, 0.90, - 0.83 and 0.71 respectively. But there was no confrimation of the correlation on the clouds, sunlight time, volume of evaporation, and heat capacity of horizontal place. 5. The water temperature of balance during the period of rice growing in Chuncheon district was shown as table 10, and the mean of whole period was calculated as about 23.7$^{\circ}C$. 6. The observed value of the outflow temperature passed through the warm water pool was higher than that of computed, the mean difference between two value was marked as 1.15$^{\circ}C$ for blockl, 1.18$^{\circ}C$ for block2, and 0.47$^{\circ}C$ for block3, respectivly. Therefore, the ratio on the rising degree between the observed and computed were shown as 53%, 44%, and 18%, mean 38% through each block warm water pool (referring item $\circled9$ of table 11,12, and 13). As the result, we can apply the coefficient in order to plan and to construct a new warm water pool.

Applicability of plate tectonics to the post-late Cretaceous igneous activities and mineralization in the southern part of South Korea( I ) (한국남부(韓國南部)의 백악기말(白堊紀末) 이후(以後)의 화성활동(火成活動)과 광화작용(鑛化作用)에 대(對)한 판구조론(板構造論)의 적용성(適用性) 연구(硏究)( I ))

  • Petrochemical, K-Ar dating, Sand Rb/Sr isotopes, metallogenic zoning, paleomagnetic and geotectonic studies of the Gyongsang basin were carried out to examine applicability of plate tectonics to the post-late Cretaceous igneous activity and metallogeny in the southeastern part of Korean Peninsula. The results obtained are as follows: 1. Bulgugsa granitic rocks range from granite to adamellite, whose Q-Ab-Or triangular diagram indicates that the depth and pressure at which the magma consolidated increase from coast to inland varying from 6 km, 0.5-3.3 kb in the coastal area to 17 km, 0.5-10 kb in the inland area. 2. The volcanic rocks in Gyongsang basin range from andesitic to basaltic rocks, and the basaltic rocks are generally tholeiitic in the coastal area and alkali basalt in the inland area. 3. The volcanic rocks of the area have the initial ratio of Sr^{87}/Sr^{86} varying from 0.706 to 0.707 which suggests a continental origin; the ratio of Rb/Sr changing from 0.079-0.157 in the coastal area to 0.021-0.034 in the inland area suggests that the volcanism is getting younger toward coastal side, which may indicate a retreat in stage of differentiation if they were derived from a same magma. The K_2O/SiO_2 (60%) increases from about 1.0 in the coastal area to about 3.0 in the inland area, which may suggest an increase indepth of the Benioff zone, if existed, toward inland side. 4. The K-Ar ages of volcanic rocks were measured to be 79.4 m.y. near Daegu, and 61.7 m.y. near Busan indicating a southeastward decrease in age. The ages of plutonic rocks also decrease toward the same direction with 73 m.y. near Daegu, and 58 m.y. near Busan, so that the volcanism predated the plutonism by 6 m.y. in the continental interior and 4 m.y. along the coast. Such igneous activities provide a positive evidence for an applicability of plate tectonics to this area. 5. Sulfur isotope analyses of sulfide minerals from 8 mines revealed that these deposits were genetically connected with the spacially associated ingeous rocks showing relatively narrow range of ${\delta}^{34}S$ values (-0.9‰ to +7.5‰ except for +13.3 from Mulgum Mine). A sequence of metallogenic zones from the coast to the inland is delineated to be in the order of Fe-Cu zone, Cu-Pb-Zn zone, and W-Mo zone. A few porphyry type copper deposits are found in the Fe-Cu zone. These two facts enable the sequence to be comparable with that of Andean type in South America. 6. The VGP's of Cretaceous and post Cretaceous rocks from Korea are located near the ones($71^{\circ}N$, $180^{\circ}E$ and $90^{\circ}N$, $110^{\circ}E$) obtained from continents of northern hemisphere. This suggests that the Korean peninsula has been stable tectonically since Cretaceous, belonging to the Eurasian continent. 7. Different polar wandering path between Korean peninsula and Japanese islands delineates that there has been some relative movement between them. 8. The variational feature of declination of NRM toward northwestern inland side from southeastern extremity of Korean peninsula suggests that the age of rocks becomes older toward inland side. 9. The geological structure(mainly faults) and trends of lineaments interpreted from the Landsat imagery reveal that NNE-, NWW- and NEE-trends are predominant in the decreasing order of intensity. 10. The NNE-trending structures were originated by tensional and/or compressional forces, the directions of which were parallel and perpendicular respectively to the subduction boundary of the Kula plate during about 90 m.y. B.P. The NWW-trending structures were originated as shear fractures by the same compressional forces. The NEE-trending structures are considered to be priginated as tension fractures parallel to the subduction boundary of the Kula plate during about 70 m.y. B.P. when Japanese islands had drifted toward southeast leaving the Sea of Japan behind. It was clearly demonstrated by many authors that the drifting of Japanese islands was accompanied with a rotational movement of a clock-wise direction, so that it is inferred that subduction boundary had changed from NNE- to NEE-direction. A number of facts and features mentioned above provide a suite of positive evidences enabling application of plate tectonics to the late Cretaceous-early Tertiary igneous activity and metallogeny in the area. Synthesizing these facts, an arc-trench system of continental margin-type is adopted by reconstructing paleogeographic models for the evolution of Korean peninsula and Japan islands. The models involve an extention mechanism behind the are(proto-Japan), by which proto-Japan as of northeastern continuation of Gyongsang zone has been drifted rotationally toward southeast. The zone of igneous activity has also been migrated from the inland in late-Cretaceous to the peninsula margin and southwestern Japan in Tertiary.

  • The calculation of the resulting fluid motion is an important problem of ship hydrodynamics. For a partially immersed body the condition of constant pressure at the free surface can be linearized. The resulting linear boundary-value problem for the velocity potential is the Neumann-Kelvin problem. The two-dimensional Neumann-Kelvin problem is studied for the half-immersed circular cylinder by Ursell. Maruo introduced a slender body approach to simplify the Neumann-Kelvin problem in such a way that the integral equation which determines the singularity distribution over the hull surface can be solved by a marching procedure of step by step integration starting at bow. In the present pater for the two-dimensional Neumann-Kelvin problem, it has been suggested that any solution of the problem must have singularities in the corners between the body surface and free surface. There can be infinitely many solutions depending on the singularities in the coroners.

Technical Development of Korean Type Hot sauce (한국식 핫소스의 제조기술 개발)

  • To develope a manufacturing process of the Korean type hot sauce, ingredients and chemical components of the imported hot sauces were investigated. The major ingredients of the imported hot sauce were chili, vinegar and salt. Chemical analysis of the imported hot sauce showed: moisture; 44.73-95.66%, total nitrogen; 0.11-1.06%, reducing sugar; 0.03-3.18%, crude fiber; 0.42-2.51%, salt; 0.87-10.44%, pH; 3.22-4.05, titratable acidity; 1.18-3.62%, capsanthin; 0.44-1.06% and capsaicin; 2.40-4.28 mg%. With the red pepper powder and/or kochujang, 20 Korean type hot sauces were prepared. Chemical analysis of the Korean type hot sauce showed: moisture; 53.07-78.30%, total nitrogen; 0.34-0.68%, reducing sugar; 1.60-4.34%, curde fiber; 1.31-2.54%, salt; 4.07-5.56%, pH; 3.37-2.54, titratable acidity; 1.15-3.06%, capsanthin; 0.11-1.36% and capsaicin; 0.55-1.42 mg%. Chemical components except capsaicin of the Korean type hot sauce were similar to those of the imported one. As the results of sensory evaluation on the 20 Korean type hot sauces with red pepper powder and/or kochujang developed, three Korean type hot sauces were finally selected. As compared with Sriracha hot sauce, an imported hot sauce, the Korean type hot sauces were evaluated to be superior to the imported one.

  • Chloral hydrate is one of the most widely used sedative agents to control the difficult-to-treat young age group in the dental clinic. We are often frustrated to see the patient still awake and cry with agitation even after far more than the normal onset time of Chloral hydrate. In such a case, the patient has to be rescheduled for another sedation visit with different agents and/or routes which greatly disappoints the guardians. This study was designed to test the efficacy of one sedative regimen that can possibly help the clinician complete scheduled treatment without postponement. We have tried sleep induction with mixed gas of Enflurane(2vol%) and $N_2O(50%)-O_2(50%)$ for $60\sim120$ seconds to 35 patients of those who failed to respond properly to the dose(70mg/kg)of oral Chloral hydrate. The Result of this regimen was compare to those of two oral regimen of Chloral hydrate/Hydroxyzine and Chloral hydrate only Analyses of result on vital signs and behavior pattern were performed. The outcome of the study suggest that sleep induction by a short inhalation of low dose of $Enflurane/N_2O-O_2$ provide dentist with suitable condition for the completion of scheduled treatment in the patient who failed to oral Chloral hydrate. Evidence of adverse effect was not detected or reported during and/or after the procedures.

The Effect of Deodeok Contents on the Quality of Deodeok Wine (더덕 함량이 더덕 침출주의 품질에 미치는 영향)

  • The physicochemical characteristics and sensory properties of Deodeok wine, formed by leaching of Deodeok at room temperature for 180 days, were investigated over the following range of Deodeok levels: 10, 15 and 20% (all w/v). The higher the level of Deodeok, the greater were the final values of total sugars, reducing sugars, total polyphenols, and crude saponins. The Hunter's b-value (yellowness) of Deodeok wine varied markedly with Deodeok levels, and yellowness was highest in Deodeok wine containing 20% (w/v) Deodeok. Non-volatile compounds, that form the basis of the liquor tax law, were 0.64, 1.38 and 2.11% (all w/v), respectively, at day 160. Of these values, that of 2.11% (w/v), the level of non-volatile compounds in Deodeok wine containing 20% (w/v) Deodeok, was in accord with the liquor tax law (that requires this figure to be 2.0%). Sensory evaluation showed that Deodeok wine containing 20% (w/v) Deodeok was superior to the other wines tested.

A Study on Application of Environmental-friendly Program for Using Relict Forest in Golf Course - Focusing on the "S" Golf Course in Incheon - (골프장내 잔존림을 활용한 친환경적 프로그램 적용가능성 연구 - 인천시 S 골프장을 대상으로 -)

  • This study was performed to introduce ecological education program as for reporting the current vegetation state within the remaining trees as ecological golf course and to provide basic data. The survey site was S Golf course, which is located in Woonseodong, Incheon city. Its total area was about $3,298,428m^2$, but the relict forest was about $225,143m^2$. Existing landuse, topological structure, the flora, actual vegetation, and plants community structure survey were performed within the relict forest. As result of comparing and analyzing the existing land use, the relict forest was distributed in the forested areas (89.2%) and around the area (10.8%) which had been bare land and SAMMOK earthen ramparts. There were two courses (Ocean and Hanul) with the relict forests. The ocean course was compared of a natural forest, such as Quercus spp. mixed forest, Quercus acutissima forest, Pinus thunbergii forest within the rock fields, and an artificial forest (Ailanthus altissima-Robinia pseudoacacia forest, Robinia pseudoacacia forest) and Quercus acutissima - Elaeagnus umbellata forest. On the Hanul course, Pinus rigida forest and Robinia pseudoacacia forest were the main vegetation, which were artificial forest. It was the contrast aspect of vegetation species in a natural forest, a restoration forest and an artificial forest, which were Q. spp. mixed forest 26~28 species in a natural forest within the vegetation type per investigation area, 3 Pinus thunbergii forest species, and 5~7 artificial forest species on the Hanul course. Based on these vegetation status, the Ocean course was designed into ecological theme spaces named 'Quercus spp.' indigenous forest, 'Pinus thunbergii' restoration forest and ecological story of 'SAMMOK earthen ramparts'. The Hannul course was designed into an artificial forest observation area of 'Robinia pseudoacacia' and 'Pinus rigida' and a fragrance forest area of 'Robinia pseudoacacia'.
