광합성세균의 생리 및 이용에 관한 최근의 동향 (General characters and applications of photosynthetic bacteria)

  • 배양조건에 따른 aflattoxin 생성능을 알기위하여 증대두배지에 Aspergillus flavus를 PH5.0 PH7.0 PH9.0 수분30%,45%,63%, 온도 2$0^{\circ}C$ 3$0^{\circ}C$ 4$0^{\circ}C$등의 조건으로 각각 배양한 27종의 시료와 Bacillus와 혼합배양한 3종의 시료를 AOAC 공정법에 의하여 aflatoxin의 생성량을 실험한 결과 다음과 같은 겨로가를 얻었다. 1. 각 PH에 의한 aflatoxin의 생성량은 PH5.0의 경우가 높았으며 PH7.0의 경우도 비교적 높은 편이었으나 PH9.0에서는 aflatoxin생성량이 현저히 감소되었다. 2. 수분합량에 따른 aflatoxin의 생성량은 수분이 적은 30%의 경우, 비교적 많은 양의 aflatoxin을 얻을 수 있었으며, 그 다음이 45%, 63% 으로서 수분함량이 많아질수록 aflatoxin의 생성이 감소되었다. 3.온도조건이 의한 결과는 Aspergillus flavus를 2$0^{\circ}C$로 배양하였을 때 가장 높은 aflatoxin의 생성량을 볼 수 있었으며, 3$0^{\circ}C$의 경우도 비교적 높은 양의 aflatoxin 생성을 볼 수 있으나 4$0^{\circ}C$의 경우는 aflatoxin의 생성은 낮았다. 4.여러가지 배양조건에 따른 Aspergillus flavus의 aflatoxin생성량은 수분 30% PH5.0 온도2$0^{\circ}C$일때 crude aflatoxin은 5.093ppm로 가장 높았으며 수분함량 63% PH 9.0 온도 4$0^{\circ}C$일때 crude aflatoxin의 량은 2.197ppm으로 aflatoxin 생성이 가장 낮았다. 5. Aspergillus flavus를 여러 조건하에서 배양한 결과 가장 많이 생성되는 배양조건인 온도 20 $^{\circ}C$ 수분 30%의 조건하에서 Bacillus subtilis와 혼합배양한 결과 Aspergillus flavus 단독배양의 경우보다 Bacillus subtilis균과 혼합배양하였을때 aflatoxin은 약 27%나 감소되었다.

  • Poly(HEMA)는 소프트 콘택트렌즈의 재료로써 널리 사용되어 왔다. $phSiH_3$, $phMeSiH_2$, $ph2SiH_2$와 같은 다양한 hydrosilane들과 hydroxyethyl methacrylate(HEMA)와의 열중합은 Phenylsilyl 말단기를 포함하는 poly(HEMA)를 생성시켰다. 열중합 반응 동안에는 Phenylsilane의 농도가 증가함에 따라 고분자의 분자량과 고분자 수득률이 감소됨을 보였으며 광중합 반응에서 보다 중합 수득률, 고분자의 분자량, TGA 잔여 수율 등이 더 높았다.

Reciprocal effect of ethyl formate and phosphine gas on two quarantine pests, Tetranychus urticae(Acari: Tetranychidae) and Myzus persicae(Hemiptera: Aphididae)

  • Fumigation of fruits and vegetables during quarantine and pre-shipment(QPS) treatment should be effective with a shorter fumigation time to minimize phytotoxicity. In this research study, a shorter fumigation time, 2 hours exposure which is shorter than that of the current commercial fumigation procedures using a lower dose of ethyl formate (EF) mixed with phosphine (PH3) on strawberry was investigated. The reciprocal effect between EF and PH3 against nymphs and adult Myzus persicae (Sulzer) and Tetranychus urticae (Koch) was evaluated. In addition, L(Ct)50 and L(Ct)99 of EF only and EF mixed with PH3 were analyzed at 5℃ and 20℃. The synergistic ratio (SR) of L(Ct)50 and L(Ct)99 for the nymph and adult stages of M. persicae were >1.0, which indicated a synergistic effect between EF and PH3. However, the SR values of L(Ct)50 and L(Ct)99 of the nymph and adult stages of T. urticae were ≤1.0 indicating that there was no synergistic effect between the two fumigants against T. urticae. Our results showed that the reciprocal effect between EF and PH3 has different effects on M. persicae and T. urticae. This could be attributed to the biological and physical differences between the class Arachnida and Insecta. The synergistic effect between EF and PH3 against M. persicae within a shorter exposure period and without phytotoxicity on fruits and vegetables will significantly benefit the horticultural industry.

Agrobacterium을 이용한 Saccharomyces cerevisiae Acid Phosphatse 유전자 (PHO5) 의 식물체로의 도입 (Agrobacterium-Mediated Transformation on a Plant with Saccharomyces cerevisiae Acid Phosphatse Gene(PHO5))

  • Agronacterium tumefaciens(strain LBA4404)를 매개로 한 외래 유전자의 식물체로의 도입에 관한 실험을 하여 아래와 가은 결과를 얻었다. 모델 식물로는 담배(Nivotuana tabacum c.v. samsun)를 사용하였으며, 외래 유전자는 효모 유래의 Acid phosphatase 유전자인 PH05를 사용하였다. pVC727G에서 PH05를 잘라내어 전기영동 및 graphical estimation법으로 확인한 결과, PH05의 크기는 1.5kb였다. pVC727G와 광역plasmid인 qBI121을 이용하여 식물체 형질전환을 위한 vector인 pBKJ I을 만들었다. pBKJ I을 Agronacterium LBA4404에 subcloninggksgn 담배의 leaf dise를 Agronacterium LBA4404과 공배양하여 형질전환을 유도하였다. kanamycindmf 첨가한 MS-n/B음지에서 형질전환된 shoots를 얻었으며, 이들의 재분화를 실시하여 형질전환된 식물체를 얻었다. 형질전환된 식물체의 genomic DNA를 PH05로서 southern hybridization하여 형질전환을 확인하였으며, 식물체의 잎, 줄기 및 뿌리의 Apase(PH05)활성을 측정하여 도입한 PH05가 발현됨을 확인하였다.

정확성 평가결과에 따른 태권도 평원 품새 동작의 운동학적 분석 (Kinematic Analysis of Accuracy Evaluation of Pyeongwon Poomsae)

  • Objective : The purpose of this study was to investigate the relations between accuracy score and the motions which are performed in PyeongWon Poomsae, thereby developing objectivity in accuracy evaluations for Poomsae competitions. Method : The subjects were six male Poomsae players (age: $21.2{\pm}1.17yrs$, height: $173.4{\pm}3.95cm$, weight: $67.7{\pm}7.65kg$). A three-dimensional motion analysis was performed on the subjects using six high-speed cameras (60 frames/sec) and subjects' performed motions were evaluated by 5 evaluators. The entire Pyeong Poomsae was divided into 13 events and 9 phases; there were three pairs of symmetric phases among them: front kick & turning side kick phases (3PH, 3-1PH), arm motion & stance phases (4PH, 4-1PH), side kick with Hakdari-stance phases (5PH, 5-1PH). Performance time, change and range of COM, height of vertex, and foot of side kicks were analysed. The Data was analysed utilizing correlation analysis. Results : There was a positive correlation between accuracy score and the difference between right and left range of COM (X direction) at 4PH (r=0.921, p=0.009). Conclusion : The results of our study indicate that it is necessary to consider some of objective criterion such as performance time, COM range, and symmetrical movements in accuracy evaluations of Poomsae competitions.

  • [[Pt(dppe)$Cl_2]$]와 1 당량 $AgNO_3$ 및 1 당량 AgOTf (OTf=$CF_3SO_3$)의 순차적 반응으로 표제 착물 [Pt(dppe)($NO_3$)($CF_3SO_3$)] (dppe=1,2-bis(diphenylphosphino)ethane, $Ph_2PCH_2CH_2PH_2$)이 합성되었다. Pt 금속은 dppe 리간드의 두 P 원자, nitrato ($NO^-_3$) 리간드의 O 원자, 그리고 triflato (trifluoromethylsulfonato, $OTf^-$) 리간드의 O 원자로 배위되어 있다. Pt 금속의 배위권은 일그러진 평면 사각형으로 기술될 수 있다.

한의약건강증진사업 발전을 위한 공중보건한의사 역할 탐색 (Exploring the roles of Korean public health doctors in the development of Korean medicine health promotion project)

  • Objectives : In an effort to suggest a developmental plan of Traditional Korean Medicine health promotion project(TKM-HPP), this study was conducted to identify the problems experienced by public health Traditional Korean Medicine(PH-TKM) doctors in duty and to analyze successful cases of the health promotion project. Methods : Two PH-TKM doctors in duty at the Public Health Centre branch and Public Health Centre respectively, and one TKM doctor in charge of Public Health Centre participated in the in-depth interview. The research procedure was as follows; 1) inform study objectives and interview questions to participants, 2) in-depth interview with semi-structured questions, and 3) post-confirmation and verification. The interview was recorded and transcripted. In-depth interview was composed of 6 subsections: 1) Introducing participants and share purpose of interview, 2) Confirmation of the job status and difficulty of public health centre, 3) Reflecting and sharing experiences of public health clinic job performance cases, 4) Current status and improvement needs of TKM-HPP, 5) Discussions on improvement of TKM-HPP in public health center, 6) Sharing and reconfirmation of interview results. Thematic analysis was conducted from the narratives. Results : The study showed that; 1) the support from the local government was critical for the successful TKM-HPP and participation of the PH-TKMs; 2) the major problems experienced by PH-TKMs were lack of training for TKM-HPP in the field and lack of knowledge at local government level; 3) the suggestions to provide PH-TKMs with manual to educate residents effectively and redefine roles of PH-TKMs in the project. It also suggests that TKM-HPP model should be developed by the district in a way of reflecting regional characteristics. Conclusions : Firstly, more efforts should be made on the projects rather than medical treatment in public health centers, except for areas where medical institutions are scarce. Secondly, in order to facilitate participation of PH-TKM doctors, each local office should reduce medical care and increase efforts related with projects. Thirdly, the institution has to develop feasible TKM-HPP plans that can be carried out by PH-TKMs by the manual. Lastly, pre-training should be provided to PH-TKMs to understand contents of TKM-HPP related to public health centre work before the placement.

  • Hydrocarbon solution of $Ir(PPh_3)_2(CO)OClO_3$ reacts with $Ph_2PC_6H_4$-o-CHO and 3-methyl-2-aminopyridinyl aldimine to yield ${\bar{Ir(Ph_2PC_6H_4-o-CO)}}\;(PPh_3)_2(CO)(H)ClO_4$(1) and ${\bar{Ir(NC_6H_6NC}}C_6H_5)(PPh_3)_2(CO)(H)ClO_4$(2), respectively. The compound $Ir(PPh_3)_2N_2Cl$ also reacts with $Ph_2PC_6H_4$-o-CHO and 3-methyl-2-aminopyridinyl aldimine to give ${\bar{Ir(Ph_2PC_6H_4-o-C}}O)(PPh_3)_2(H)Cl(3)$ and $Ir(NC_6H_5NCC_6H_5(PPh_3)_2(H)Cl(4)$, respectively. Compounds 1, 2, 3, and 4 were characterized by infrared, $^1H$ NMR, $^{31}p$ NMR, UV spectra, and conductivity measurements.

A Kinetic Study on Solvolysis of Diphenyl Thiophosphorochloridate

  • Rates of solvolyses of diphenyl thiophosphorochloridate ($(PhO)_2$PSCl, 1) in ethanol, methanol, and aqueous binary mixtures incorporating ethanol, methanol, acetone and 2,2,2-trifluoroethanol (TFE) are reported. For four representative solvents, studies were made at several temperatures and activation parameters were determined. The 30 solvents gave a reasonably precise extended Grunwald-Winstein plot, correlation coefficient (R) of 0.989. The sensitivity values (l = 1.29 and m = 0.64) of diphenyl thiophosphorochloridate ($(PhO)_2$PSCl, 1) were similar to those obtained for diphenyl phosphorochloridate ($(PhO)_2$POCl, 2), diphenylphosphinyl chloride ($Ph_2$POCl, 3) and diphenylthiophosphinyl chloride ($Ph_2$PSCl, 4). As with the previously studied of 3~4 solvolyses, an $S_N$ pathway is proposed for the solvolyses of diphenyl thiophosphorochloridate (1). The activation parameters, ${\Delta}H^{\neq}\;(=11.6{\sim}13.9\;kcal{\cdot}mol^{-1})\;and\;{\Delta}S^{\neq}\; (=\;-32.1\;{\sim}\;-42.7\;cal{\cdot}mol^{-1}{\cdot}K^{-1})$, were determined, and they were in line with values expected for an $S_N$2 reaction. The large kinetic solvent isotope effects (KSIE, 2.44 in MeOH/MeOD and 3.46 in $H_2O/D_2$O) are also well explained by the proposed $S_N$2 mechanism.

액체배지에서의 Fusarium moniliforme에 의한 Fusarin C생성에 관한 연구 (Fusarin C Production by Fusarium moniliforme in Liquid Media)

  • 세 가지 액체배지를 사용하여 Berkeley대학 Dr. Bjeldanes Lab에서 isolate된 F. moniliforme을 생육시킨 결과 Czapek-Dox medium에서 Fusarin C생성량이 가장 높았으며 옥수수에 비하면 약 1/10정도의 양인 것으로 나타났다. 그리고 Czapek-Dox medium의 경우 $28^{\circ}C$에서 2주간 배양한 때에 Fusarin C의 생성량이 가장 많았으며 옥수수의 경우는 28$^{\circ}C$에서 1주간 배양한 때에 가장 높았다. 또한 Czapek-Dox medium의 initial PH가 6.5인 때에 Fusarin C의 생성량이 최대이었으며 6.3, 5.9인 때도 다른 PH보다 월등히 높은 양이 생성됨을 확인할 수 있었다.
