Purpose: Frostbite can affect still soldiers. Initial clinical manifestations are similar for superficial and deep frostbite, so early treatment is identical. It is under-estimated by physicians. We try to identify the challenges of managing these complex tissue injuries. Methods: A retrospective analysis of 84 patients hospitalized at AFCH from 2009 to 2015 was conducted. We investigated differences of epidemiological characteristics, identification of soft tissue injury, treatment and complications between superficial (SF: 43; 51.2%) and deep (DF: 41; 48.8%) frostbite. Results: The major (94.0%) developed frostbite in dry circumstances (89.3%). Wet circumstances (66.7%) were more susceptible to DF rather than dry (46.7%). The 38 (45.2%) arrived to specialist within 7days. Most prone sites were feet, followed by hands. Toes had more deep injuries. DF presented more increased levels of ALT, CPK, CKMB, CRP. The bone scan of W+S+ was 48.3%, 87.1% and W+S- was 20.7%, 12.9%, respectively. The treatment resulted in improved or normalized perfusion scan with matching clinical improvement. It was a good tool to assess treatment response. Eighteen normal and 8 stenotic type of PCR resulted in normal with matching clinical improvement. One continuous obstructive waveform led to minor amputation. Twelve underwent both PCR and MRA. Among 6 normal PCR, 5 showed normal and one stenosis in MRA. All 5 stenosis and one obstruction showed the same findings in MRA. It was a good tool to evaluate vascular compromise. They were treated with rapid rewarming (11.6%, 22.0%), hydrotherapy (16.3%, 29.3%), respectively. Six (14.6%) underwent STSG, 2 (4.9%) had digital amputation in DF. Berasil, Ibuprofen, Trental were commonly administered. PGE1 was administered selectively for 6.8, 10.8 days, respectively. Raynaud's syndrome (16.3%), CRPS (4.7%), LOM (14.6%) and toe deformity (4.9%) were specific sequelae. Conclusion: We should recommend intensive foot care education, early rewarming and evacuation to specialized units. The bone scanning and PCR should allow for a more aggressive and active approach to the management of tissue viability.
Objectives : Taraxaci Herba et Radix (THR) is widely used as a food and medicinal herb in Korea. It has been used for treatment of virus inflammatory disease, liver diseases and gastritis. So far, anti-inflammatory effects and constituents of various species in THR has not been studied for comparison. The aim of this study is to compare the anti-inflammatory effects of the aerial part and root from various THR. Also, we have compared the contents of its known constituents with each. Methods : In this study, we estimated anti-inflammatory effect and compared their constituent by HPLC. For the determination of anti-inflammatory effects, we investigated NO and $PGE_2$ production by ELISA. The expressions of iNOS was determined by western blotting in LPS-induced RAW 264.7 macrophage cells. And, standard compounds which are methyl gallate, gallic acid, syringic acid and esculetin of THR were analyzed by HPLC using a $C_{18}$ column. Results : Methanol extracts of THR decreased NO and $PGE_2$ production. The expressions of iNOS protein were also decreased in methanol extracts of THR. As a result, HPLC analysis showed that they showed similar patterns. Methyl gallate and esculetin showed the highest content. Methyl gallate was included over 10% content in each aerial part and root of THR. Conclusions : These results indicate that most of THR distributed in Korea might represent therapeutic agent for treatment of inflammatory diseases.
Objectives GyejigaChulBuTang (GCBT) is a prescription used to treat acute and chronic arthritis in Korea, China, and Japan. This study assessed the anti-inflammatory and anti-oxidant activities of GCBT on lipopolysaccharide (LPS)-stimulated RAW264.7 macrophage cells. Methods Raw264.7 cells were pretreated with or without GCBT for 1 hour prior to incubation with LPS. Anti-inflammatory activity of GCBT was evaluated with reference to gene expression and production levels of proinflammatory cytokines ($TNF{\alpha}$, IL-$1{\beta}$, IL-6, GM-CSF and $INF{\gamma}$) and inflammatory mediators (iNOS, COX-2, NO and $PGE_2$). In addition, intracellular ROS generation and signal transduction of MAPK family, PI3K/Akt and $I{\kappa}B{\alpha}/NF{\kappa}B$ was investigated. Results Prior treatment with GCBT inhibited elevation of $TNF{\alpha}$, IL-$1{\beta}$, IL-6, GM-CSF, $INF{\gamma}$, NO and $PGE_2$, together with their cognate mRNAs in a dose-dependent manner. Intracellular ROS contents were similarly reduced. These effects were due to inhibition of LPS-induced phosphorylation of MAPK family, PI3K/Akt and $I{\kappa}B{\alpha}$ as well as nuclear translocation of $NF{\kappa}B$. Conclusions GCBT suppresses pro-inflammatory mediators. GCBT has potential in the treatment of juvenile rheumatoid arthritis associated with inflammation.
Background: The objective of this study was to determine the effects of Daegangwhal-Tang (DGHT) hot aqueous extract on production of inflammatory mediators and antioxidants in RAW 264.7 macrophage. Methods: DGHT was extracted with water, filtered, concentrated and freeze-dried to perform. Cytotoxicity of DGHT extract was performed by MTT assay. Activated macrophages were treated with varying concentrations of DGHT extract (10, 50, 100 and $200{\mu}g/mL$), and nitric oxide (NO) and prostaglandin E2 ($PGE_2$) concentrations were measured to detect anti-oxidative effects. Interleukin-6 (IL-6), interleukin-1 beta ($IL-1{\beta}$) and tumor necrosis factor-alpha($TNF-{\alpha}$) concentrations were also measured to detect inflammatory responses to DGHT Results: Cytotoxicity of DGHT extract at concentrations of 10, 50, 100 and $200{\mu}g/mL$ were not observed. NO production was significantly decreased in the DGHT hot aqueous extract $200{\mu}g/mL$ concentration group. $PGE_2$, IL-6, $IL-1{\beta}$ and $TNF-{\alpha}$ production was significantly decreased in the DGHT hot aqueous extract 100 and $200{\mu}g/mL$ concentration groups. DGHT hot aqueous extract appeared to have DPPH free radical scavenging capability at all of concentrations, but did not exceed 50%. Conclusion: These results suggest that DGHT hot aqueous extract has concentration-dependent anti-inflammatory and anti-oxidative effect.
Background: The purpose of this study was to investigate whether Sibseonsan (SSS) is an effective antiinflammatory, anti-wrinkling, and whitening agent. Methods: To determine whether SSS had an anti-inflammatory effect, a murine macrophage cell line was used (RAW 264.7) and production of DPPH, NO, TNF-α, and PGE2 were measured. To ascertain potential anti-wrinkle effects of SSS in these cells, collagenase and elastase production were measured. To verify whether SSS had a whitening effect, tyrosinase activity and DOPA staining were performed using a melanoma cell line (B16/F10). Results: There was no significant reduction in survival of SSS-treated RAW 264.7 cells, up to 400 ㎍/mL. Free radical scavenging (23.96 ± 1.85%) was observed in RAW 264.7 cells treated with SSS at a concentration of 400 ㎍/mL. The SSS treatment group (400 ㎍/mL) significantly inhibited NO production compared with the LPS stimulated treatment group. The SSS treatment of macrophage cells appeared to reduce production of TNF-α in a concentration dependent manner. There was a significant reduction in the concentration of PGE2 by about 25% in the SSS treatment (400 ㎍/mL) group (p = 0.05). Compared with the control, the production of collagenase and elastase in B16/F10 cells treated with SSS (400 ㎍/mL) was greater by 26.37% and 45.71%, respectively. The SSS treatment (400 ㎍/mL) group showed a significant reduction by about 17% in tyrosinase production in B16/F10 cells. The SSS treatment group showed little change in DOPA staining. Conclusion: SSS extract may be useful for the treatment and prevention of inflammatory diseases and may have anti-wrinkle and whitening effects. These results may support the use of SSS in clinical practice.
This study was conducted to evaluate the effects of Padina arborescens and 1-O-myristoyl-2-O-oleoyl-3-O-(α-D-glucopyranosyl)-glycerol(MOGG), its active component, on the prevention of hair loss. The P. arborescens extract and MOGG inhibited the activity of 5α-reductase, which converts testosterone to dihydrotestosterone(DHT), a main cause of androgenetic alopecia. When immortalized rat vibrissa dermal papilla cells were treated with MOGG, the proliferation of dermal papilla cells significantly increased. In addition, we found that the P. arborescens extract and MOGG could open the KATP channel, which may contribute to increase hair growth. Furthermore, MOGG promoted PGE2 production in HaCaT cells. The results suggest that MOGG from the P. arborescens extract has the potential to treat alopecia via 5α-reductase inhibition, the proliferation of dermal papilla, the opening of the KATP channel and/or increase of PGE2 production.
The aim of this study was to explore the role and effect of ginsenosides Rg1 on osteoblasts cultured with Ti particles. Osteoblasts from neonatal rats were cultured with particles and different doses of Rg1, the main active ingredient in ginsenosides Rg1. We found that the COX-2, $PGE_2$, TNF-${\alpha}$, IL-1, and IL -6 concentrations in the medium of cells cultured with Ti particles significantly increased as compared with that of the control cells (p<0.05 or p<0.01). In addition, cells cultured with Ti particles alone exhibited the highest concentrations of these molecules. The $PGE_2$, TNF-${\alpha}$, IL-1, and IL-6 levels in the medium of cells cultured with Rg1 were in between those of the control cells and the cells cultured with Ti particles alone. The IL-1ra level in the group cultured with Ti and medium-dose Rg1 was the highest followed by the cells cultured with Ti and high-dose Rg1 and those cultured with Ti and low-dose Rg1 (p<0.05). In conclusion, ginsenosides can reduce the levels of infl ammatory cytokines produced by osteoblasts on induction with Ti particles and can prevent prosthesis loosening.

This study was carried out to prove the effects of CBTLC on the $5{\alpha}$-reductase inhibition, the sterilizing power for Propionibactrium acnes, the cytotoxicity of human monocyte, the inhibition for prosta-glandins($PGE_2$), Interleukins(IL-$1{\beta}$) and TNF-${\alpha}$ in inflammation, and the size of Hamster ear sebaceous gland concerned with Acnes. The result were obtained as follows : 1. On the $5{\alpha}$-reductase inhibition of CBTLC in vitro, An undiluted solution of CBTLC was $71{\%}$ inhibition on $5{\alpha}$-reductase and $\frac{1}{10}$ diluted solution of CBTLC was $20{\%}$ inhibition on $5{\alpha}$-reductase. 2. On the sterilizing power for Propionibactrium acnes concerned in Acnes, an undiluted solution and $\frac{1}{10}$ diluted solution of CBTLC formed 12mm clear zone diameters. 3. CBTLC was showed the cytotoxicity of human monocyte in $0.08{\%}$ and $0.12{\%}$ of CBTLC. 4. $0.01{\%}$ and $0.04{\%}$ of CBTLC inhibited the production of prostaglandins($PGE_2$) in the human monocyte stimulated with P. acnes LPS. 5. $0.08{\%}$ and downward of GMHBS inhibited the production of interleukins(lL-$1{\beta}$) in the human monocyte stimulated with P. acnes LPS. 6. $0.08{\%}$ and $0.12{\%}$ of GMHBS inhihited the production of TNF-${\alpha}$ in the human monocyte stimulated with P. acnes LPS. 7. As the antiandrogenic compound, CBTLC was used in hamster ears with topical application. CBTLC was diminished slightly on the size of hamster ears sebaceous gland.
Growth characteristics and crude protein content of soybean sprouts cultivated with extracts of some Korean herb medicines for 4 days were investigated. Extracts of Korean herb medicines were utilized and adjusted to 100 ppm, 200 ppm, 300 ppm and 400 ppm as culturing water of soybean sprouts. Of Korean herb medicines tested, extract of Panax ginseng was most effective to increase of weight and length of soybean sprouts, followed by Glycyrrhiza glabra. Extracts of Angelica gigas and Cnidium officinale were similar with that of control group. Extract of Coptidis japonica inhibited the growth of soybean sprout. Optimal concentrations of extracts of Glycyrrhiza glabra and Panax ginseng for growth of soybean sprout were 100 ppm and 200 ppm, respectively. Weight of soybean sprout cultivated with extract of Panax ginseng (PGE) was increased to 7.2%(wet basis) as compared with that of control group. Length of soybean sprouts cultivated with 200 ppm of PGE was accelerated to 32% and increased to 50% in soybean sprout cultivated for 3 days. Crude protein contents of soybean sprout cultivated with extract of Angelica gigas or Panax ginseng was slightly increased according to cultivation period. In Cnidium officinale group, crude protein contents was increased at a 300∼400 ppm concentration and 4 days of cultivation.
Piceatannol is a polyphenol that is found in abundant quantities in grapes and wine. Although recent experimental data revealed the anti-cancer potency of piceatannol, the molecular mechanisms underlying the antileukemic activity have not yet been studied in detail. In the present study, we investigated further possible mechanisms by which piceatannol exerts its anti-proliferative action in cultured human leukemia U937 cells. Exposure of U937 cells to piceatannol resulted in growth inhibition and induction of apoptosis as measured by MTT assay and flow cytometry analysis, which was associated with S phase arrest of the cell cycle. Piceatannol treatment markedly inhibited the activity of telomerase, and the levels of human telomerase reverse transcriptase (hTERT) and telomerase-associated protein-1 (TEP-1), main determinants of the telomerase enzymatic activity, were progressively down-regulated by piceatannol treatment in a dose-dependent fashion. However, the levels of cyclooxygenases (COXs) expression and prostaglandin E2 (PGE2) release were not changed in piceatannol-treated U937 cells. Taken together, these findings provide important new insights into the possible molecular mechanisms of the anti-cancer activity of piceatannol.
