Evaluation Antioxidant and Anti-inflammatory Activity of Ethanolic Extracts of Myriophyllum spicatum L. in Lipopolysaccharide-stimulated RAW 264.7 Cells (이삭물수세미(Myriophyllum spicatum L.) 에탄올 추출물의 항산화와 항염증 효과)

  • Myriophyllum spicatum L. has been used as an ornamental in ponds and aquariums, and as a folk remedy for inflammation and pus. Nevertheless, the biological activity and underlying mechanisms of anti-inflammatory effects are unclear. This study is aimed at investigating the antioxidative and anti-inflammatory activities of ethanol extract of Myriophyllum spicatum L. (EMS) in lipopolysaccharide (LPS)-stimulated RAW 264.7 cells. Antioxidant activity of EMS was assessed by radical-scavenging effects on ferric reducing antioxidant power (FRAP) and 2,2-diphenyl-1-picrylhydrazyl (DPPH) free radicals. As inflammatory response parameters produced by LPS-stimulated RAW 264.7 cells were quantified to assess the anti-inflammatory activity of EMS. Our results showed that EMS increased FRAP and DPPH radical-scavenging activity. In EMS-treated RAW 264.7 cells, the production of NO, PGE2, TNF-α and IL-1β was significantly inhibited at the non-cytotoxic concentration. In addition, EMS significantly attenuated LPS-stimulated the toll-like receptor (TLR) 4/myeloid differentiation protein (MyD) 88 signaling pathway, and inhibited nuclear translocation of nuclear factor-kappa B(NF-κB). Positive correlations were noted between anti-inflammatory activity and antioxidant activity. In conclusion, it was indicated that EMS suppresses the transcription of inflammatory factors by inhibiting the TLR4/MyD88/NF-κB signaling pathway, thereby suppressing LPS-stimulated inflammation in RAW 264.7 cells. This study highlights the potential role of EMS against inflammation and associated diseases.

Hesperidin Ameliorates TNF-${\alpha}$-Mediated Insulin Resistance in Differentiated 3T3-L1 Cells

  • Adipose inflammation is linked to the development of insulin resistance and type 2 diabetes. Hesperidin (HES) is a flavonoid with antioxidant, anti-inflammatory and anti-diabetic properties. However, whether HES improves inflammation-mediated insulin resistance in adipose tissues remains unclear. The purpose of this study was to investigate whether HES attenuates inflammation-mediated insulin resistance in adipose tissue. Herein, RAW 264.7 cells and differentiated 3T3-L1 adipocytes were pretreated with various concentrations of HES in complete media for 1 h and then cultured in the presence or absence of LPS or TNF-${\alpha}$. Our results demonstrated that HES remarkably inhibited LPS-induced production of IL-6, TNF-${\alpha}$, and NO by RAW 264.7 cells in a dose-dependent manner. Also, HES inhibited TNF-${\alpha}$-induced production of IL-6 and $PGE_2$ in differentiated 3T3-L1 cells, while upregulated TNF-${\alpha}$-suppressed expression of adiponectin and PPAR-${\gamma}$ mRNA. These findings suggest that HES may ameliorate inflammation-mediated insulin resistance in adipose tissue.

Protective Effect of Baicalin on the TNF-${\alpha}$-Mediated Development of Insulin Resistance in Differentiated 3T3-L1 Cells

  • Adipose tissue-derived chronic inflammation contributes to development of insulin resistance in obesity, leading to type 2 diabetes and cardiovascular disease. Baicalin, a flavonoid, has antioxidant, anti-inflammatory, antihyperglycemic, anti-adipogenic, and antiobesity effects. However, whether baicalin attenuates adipose tissue-derived development of insulin resistance remains still unclear. This study was to investigate effect of baicalin on the inflammatory changes involved in the development of insulin resistance in adipose tissue. RAW 264.7 cells and differentiated 3T3-L1 adipocytes were pretreated with various concentrations of baicalin in complete media for 1 h and then cultured in the presence or absence of LPS or TNF-${\alpha}$. Our results demonstrated that baicalin remarkably inhibited LPS-induced production of TNF-${\alpha}$, IL-6, and NO by RAW 264.7 cells in a dose-dependent manner. Baicalin also inhibited TNF-${\alpha}$-induced production of IL-6 and $PGE_2$ in differentiated 3T3-L1 cells in a dose-dependent manner, while upregulated TNF-${\alpha}$-suppressed expression of adiponectin and PPAR-${\gamma}$ mRNA and IRS-1 protein. These findings suggest that baicalin may prevent the adipose tissue-derived development of insulin resistance in obesity.

Anti-inflammatory and Anti-cancer Effect of Stachys affinis Tubers

  • Stachys affinis tubers are known for its high content of stachyose and eaten as an edible vegetable. In this work, we assessed on the anti-inflammatory and anti-proliferation activity of a various type of extracts derived from S. affinis tubers. The n-hexane and dichloromethane fractions were showed the high cytotoxicity on the cell lines including RAW264.7 macrophages, HEK293 human kidney cell, A549 human lung cancer cell, KB human oral cancer cell, and a PC-3 human prostate cancer cell. N-butanol and water fractions were not exhibited cytotoxicity on the tested cancer cells, limited in anti-inflammatory and anti-cancer activities. Nevertheless, the ethyl acetate fraction showed little harm to RAW264.7 cells but inhibited the production of nitric oxide (NO) and prostaglandin E2 (PGE2) significantly. In addition, it arrests the cell growth in A549, KB, and PC-3 cell while little cytotoxicity on HEK293 cells. Consequently, these results supported that the ethyl acetate fraction of S. affinis tubers could be a potential anti-inflammatory and anti-cancer ingredient.

Antioxidant and Anti-inflammatory Activity of Six Halophytes in Korea

  • The aim of this study was to measure and compare polyphenol content, antioxidant, and anti-inflammatory activity of six halophytes (Limonium tetragonum, Suaeda glauca, Suaeda japonica, Salicornia europaea, Triglochin maritimum, and Sonchus brachyotus). Depending on the total polyphenol content, the plants were categorized into two groups: (1) a high total polyphenol content group that included L. tetragonum, S. brachyotus, and S. europaea, and, (2) a low total polyphenol content group consisting of S. glauca, T. maritima, and S. japonica. Antioxidant activity was evaluated using DPPH and hydroxyl radical scavenging assays, and by measuring ROS. Anti-inflammatory activity was evaluated by measuring NO and $PGE_2$. L. tetragonum and S. brachyotus, that have high polyphenol content, also showed strong antioxidant activity. In addition, L. tetragonum, S. brachyotus, and S. europaea showed good anti-inflammatory activity. Consequently, the total polyphenol content was thought to be related to antioxidant and anti-inflammatory activity. Therefore, S. brachyotus and L. tetragonum are good candidates for use in pharmaceuticals and functional foods.

Effects of Dioscorea daemona Roxb. Stem Extract on the Inflammatory Responses, Antioxidant System and Lipid Levels in Vivo and the Production of Inflammatory Mediators in RAW264.7 Cells (Dioscorea daemona Roxb. (Stem) 추출물이 Rat의 염증반응 및 항산화 체계에 미치는 영향과 RAW264.7 세포주의 염증성 매개물 생성에 미치는 영향)

  • 본 연구에서는 다양한 동물 모델을 사용하여 Dioscorea daemona Roxb. 줄기 메탄을 추출물(DD)의 항염증 활성을 측정하였으며 DD가 생체내에서 항산화 체계의 변화를 유도할 수 있는지도 살펴보았다. DD를 200mg/kg용량으로 3주간 경구투여하였을 때 동물실험모델에서 항염증 및 type IV 알레르기 억제 효과를 나타내었으며 혈청의 Catalase 활성, 지질 과산화, TG 및 HDL cholesterol 수치가 영향을 받았다. DD와 이를 클로로포름과 부탄올로 순차적으로 분획하여 얻은 fraction이 lipopolysaccharide(LPS)로 유도한 RAW264.7 대식세포주의 nitric oxide(NO), prostaglandin $E_2(PGE_2)$, tumor necrosis $factor-\alpha(TNF-\alpha)$, interleukin 6(IL6)의 생성을 억제하는지도 연구하였다. DD와 그 분획물들은 $4\~100{\mu}g/mL$ 농도에서 세포 독성을 나타내지 않고 LPS가 유도한 RAW264.7 세포주의 NO, $TNF-\alpha$, IL-6 생성을 억제하였다. LPS가 유도한 $PGE_2$ 생성은 DD의 클로로포름 분획에서 유의적으로 감소하였다(p<0.05). 따라서 Dioscorea daemona 추출물은 대식세포의 염증성 매개물의 억제를 통하여 항염증 활성을 나타내는 것으로 사료된다.

Antiinflammatory Activity of Hyperin from Acanthopanax chiisanensis Roots

  • The chloroform and the ethyl acetate fractions from the roots of Acanthopanax chiisanensis exhibited a significant inhibition of prostaglandin E$_2$ (PGE$_2$)production in rat peritoneal macrophages stimulated by the protein kinase C activator, 12-O-tetradecanoylphorbol 13-acetate (TPA). Hyperin was isolated as an active principle from the ethyl acetate fraction. It sup-pressed not only $PGE_2$ production but also nitric oxide (NO) production in vitro in a concentration dependent manner. their $IC_{50}$, being 24.3 and $32.9{\;}{\mu}M$, respectively. Hyperin also caused a significant inhibition of increase in acetic acid-induced vascular permeability in mice in vivo.

  • Objectives: This study was to investigate the effect of high frequency electro-acupuncture (EA) at $ST_{36}$ acupoint on the Freund's Complete Adjuvant (FCA)-induced arthritis in rats. Methods : Arthritis was induced by intradermal injection of FCA into base of tail. Experimental groups were divided into 4 groups; Normal, Control, $ST_{36}$ and Non-Acupoint (NA). $ST_{36}$ group was treated by 120Hz EA at $ST_{36}$ acupoint. Body weight, paw edema volume and ankle joint thickness were measured after treatment. And we investigated the effects of 120Hz EA via WBC count, segmen neurophil, lymphocyte, PGE2 assay and NADPH-d histochemistry. Results and Conclusions : The mean of body weight and ankle joint thickness of $ST_{36}$ group was increased compared with control group. The mean of paw edema volume, WBC count, segment neutrophil and lymphocyte of were not decreased with control group.   • The present study was designed to evaluate effects of the commonly used NSAIDs(acetaminophen, aspirin, and ibuprofen) on human periodontal ligament cells. Human periodontal ligament cells were grown from a cell line provided by Kyungpook National University. Effects of NSAIDs on the proliferation of human periodontal ligament cells were assessed using MTT assays. And then $PGE_2$ concentrations were determined by ELISA and the changes of cellular configuration were found by electron micrograph. The results were as follows; 1. The MTT assay demonstrated that the commonly used NSAIDs(acetaminophen, aspirin, and ibuprofen) had not significant cytotoxic effect on human periodontal ligament cells. 2. NSAIDs inhibited the $PGE_2$ synthesis of human periodontal ligament cells compared with the control group. These inhibitory effects had no significant differences with NSAID type and concentration. 3. Electron micrographs of human periodontal ligament cells treated with NSAIDs showed more narrow and irregular shape.

Evaluation of Antibacterial, Antioxidant Fractionalities and Hair Loss Prevention Effect of Platycodon grandiflorum (도라지 추출성분의 항균·항산화 및 탈모예방 효과)

  • To investigate the effects of antioxidant activities and hair loss prevention of extracts from Platycodon grandiflorum, we've prepared chloroform (CF) and ethylacetate fractions (EA) extracted from P. grandiflorum. In the results of DPPH radical scavenging assay, the two fractions showed dose-dependent antioxidant activities. Furthermore, in the ABTS assay, the two fractions exhibited the inhibitory effect over 90% at 10, 50, 100, 200 mg/mL. To investigate the inflammation inhibitory effect, we used RAW264.7 cells, these extracts were inhibited inflammatory reaction by suppressing the production of nitric oxide (NO) and prostaglandin $E_2$ ($PGE_2$) in dose-dependent manner. In the assay of HaCaT cells' proliferation, the 24 hr treatment of the extracts significantly accelerarted cell proliferation in the range of concentrations used. The two fractions inhibited the proliferation of Malssezia furfur, the bacteria induce the dandruff. Finally, the CF could significantly inhibit the DHT production at 1, 10, 50, $100{\mu}g/mL$, but EA showed the inhibitory effect at the concentration over $50{\mu}g/mL$. The overall results of this study suggest that the chloroform (CF) and ethylacetate fractions (EA) from P. grandiflorum could be a useful raw material for the hair loss prevention products.
