Antifungal Activity of Bioactive Fractions on the Dermatophytic Fungus

  • Cytotoxic and antifungal activities were strongly shown by Fr. 4 (218 mg, P388 IC/sub 50/ 652 ng/ml at 75 μg/disc, IC50 834 ng/ml at 30 μg/disc and HM 8). The subfraction 4 - 3 that showed the most activity was cytotoxic to murine leukaemia cells ATCC CCl 46 P388D1, (44.0 mg, P388 IC50 302 ng/ml at 30 μg/disc).

  • A new flavonol derivative, meliglabrin (1) along with three known flavonols, ternatin (2), meliternatin (3), and 5,4'-dihydroxy-3,7,3'-trimethoxyflavon (4) were isolated from the leaves of Melicope glabra (Blume) T.G. Hartley. Their structures were determined using extensive spectroscopic methods, including UV, IR, HRESIMS, 1D and 2D NMR. Compounds 1 - 4 were evaluated for their cytotoxicity against murine leukemia P-388 cells, compound 4 showed moderate activity.

  • The effects of chloroform extract from Lepidolaena tayiorii (L. tayiorii) on antifungal activity were investigated. The crude chloroform extract of L. tayiorii inhibited the growth of the Gram positive bacteria Bacillus subtilis ATCC 19659, (5 mm inhibition zone at $150{\mu}g/disc$) and the dermatophytic fungus Trichophyton mentagrophytes ATCC 28185, (6 mm inhibition zone at $150{\mu}g/disc$), and cytotoxic to P388 murine leukaemia cells ATCC CCL 46 P388D1, ($IC_{50}\;405.0{\mu}g/mL\;at\;150{\mu}g$/disc) and cytotoxic to BSC monkey kidney cells (@ 5 mg/mL, $150{\mu}g/disc;$ +++: 100% activity). We suppose that this crude chloroform extract of L. tayiorii is the strong antimicrobial and cytotoxic activities.

Comparative Antitumor Activity of Different Solvent Fractions from an Auricularia auricula-judae Ethanol Extract in P388D1 and Sarcoma 180 Cells

  • The objective of this study was to evaluate and compare the antitumor activity of different solvent fractions (ethanol, dichloromethane, ethyl acetate, butanol and water) of the Auricularia auricula-judae 70% ethanol extract on the P388D1 macrophage and sarcoma 180 cells. A dose-dependent antitumor activity of each solvent fraction (from 0.01 mg/ml to 0.3 mg/ml) was shown against both cell types. These cytotoxic effects of all the tested fractions were confirmed on the MTT and SRB assays, without statistical differences each other. $IC_{50}$ value of dichloromethane fraction was 94.2 ${\mu}g/ml$ against sarcoma 180 cells lower than any other solvent fractions. The potent antitumor effect of the dichloromethane (DCM) fraction was also found against solid tumor in BALB/c mice. The splenomegaly and higher splenic index were found in tumor-bearing mice, with the DCM fraction returning to the negative control values. Thus, the results indicated the dichloromethane fraction may have potential ingredients as antitumor candidates.

  • MAP kinases are a family of serine/threonine specific protein kinases becoming activated in response to different proliferative stimuli by phosphorylation at both threonine and tyrosine residue. Present study shows that MAP kinase was purified from P388 murine leukema cells by SP sephadex C-50, phenyl superose and Mono Q column chromatography and identified with anti-ERKl antibody by western blotting. Immnublotting analysis to the crude extract of P388 cell lysate shows 44 kD and other minor bands but partial purified fraction eluted from phenyl supherose column have 44kD and 66 kD isoform. Subcloned GST-fusion protein from N-terminal of $p56^{kk}$ was tested as a substrate for MAP kinase phosphorylation. It was showed that the wild type and mutant forms(S42A) were fully phosporylated by purified MAP kinase fraction as com-pare with the other mutant form(S59A). This finding suggest that those GST-fusion proteins may be used as substrate for the in vitro test of MAP kinase.

  • A new cinnamyl acid derivative, willughbein A (1) along with pinoresinol (2), alyterinate A (3), and scopoletin (4), were isolated from the roots of Willughbeia coriacea Wall. The structure of 1 has been determined based on HRESIMS, 1D, and 2D NMR spectral data. All of the isolates were evaluated for their cytotoxicity against three human cancer cells (HeLa, T47D, MCF-7, and P-388). Compound 3 showed moderate activity against P-388 cells with an IC50 value of 3.04 ㎍/mL.

  • The effects of ethanol extract from Lepidolaena clavigera (L. clavigera) on antifungal activity were investigated. The crude ethanol extract of L. clavigera inhibited the growth of the Gram positive bacterium Bacillus subtilis ATCC 19659, (1 mm inhibition zone at 150 ${\mu}g/disc$) and the dermatophytic fungus Trichophyon mentagrophyes ATCC 28185, (2 mm inhibition zone at 150 ${\mu}g/disc$), and cytotoxic to P388 murine leukaemia cells ATCC CCL 46 P388D1, (IC50 >12,500 ${\mu}g/mL\;at\;150\;{\mu}g/disc$) and cytotoxic to BSC monkey kidney cells (@ 5 mg/mL, 150 ${\mu}g/disc$; ++: 50% activity). We suppose that this crude ethanol extract of L. clavigera is the antifungal activity.

  • This study was carried out to evaluate cytotoxicity of the methanol extract from Sophora flavescens Ait. against L1210 (lymphocytic leukemia) and $P388D_1$ (lymphoid neoplasma) Cells in vitro. We have determined cytotoxicity by the MTT (3- (4,5-dimethylthiazol-2-yl)-2,5-diphenyl-2H- tetrazolium bromide) assay. The order of cytotoxicity of Sophora flavescens Ait. extracts against L1210 and $P388D_1$ cells in vitro is as follows: Fr. 4 > Fr. 3 > Fr. 5 > Fr. 2 > Fr. 1. These results suggest that the fraction 4 of the methanol extracts from Sophora flavescens Ait. may be a valuable choice for the development of antitumor agents. In order to develop an antimicrobial agent, dried Sophora flavescens Ait. was extracted with hot methanol, and then antimicrobial activity (MIC test) was investigated. In this study, the fraction 3 of the methanol extracts from the roots of S. flavescens showed strong growth inhibition activity against gram-positive and gram-negative bacteria (MIC, $3.125\;{\mu}g/ml$) such as S. mutans, S. epidermidis and P. putida. These results indicate that fractions 3 and 4 inhibit tumor cells and bacteria.

Biological Activity of Water Extract from Atractylodes macrocephala

  • The effects of water extract from Atractylodes macrocephala Koidz on biological activity were investigated. The crude water extract of A. macrocephala inhibited the growth of the dermatophytic fungus Trichophyton mentagrophytes ATCC 28185, (3 mm inhibition zone at 300 ㎍/disc). However, it did not show growth inhibition activity against Sreptococcus mutans JC-2 (MIC >1,000 ㎍/mL). This extract was cytotoxic to P388 murine leukaemia cells ATCC CCL 46 P388D1, (IC/sub 50/ 62.24 ㎍/mL at 150 ㎍/disc). These results suggest that water extract of A. macrocephala possesses antitumoral, and antimicrobial activities.

Synthesis and Biological Activity of Geranyloxy Compounds

  • Disk assays on the compounds (10 and 12) showed both to have antifungal activity against the dermatophytic fungus Trichophyton mentagrophytes ATCC 28185, (1 and 3 mm inhibition zones at $60\;{\mu}g/disc$), but not against the Gram-positive bacterium B. subtilis or the Gram-negative bacteria Escherichia coli and Pseudomonas aeruginosa or fungi Cladosporium resinae and Candida albicans. However, the compound (13) did not show against antifungal activity. The geranyloxy compounds (10, 12, and 13) were cytotoxic to P388 murine leukaemia cells ATCC CCL 46 P388D1, ($IC_50$ >6,250 ng/mL at $7.5\;{\mu}g/disc$). These results suggest that The geranyloxy compounds possesses antimicrobial and antitumor activities.
