유기수은의 신경독성에 대한 셀레늄의 보상작용 (Interaction of Sodium Selenite on Neurotoxicity Induced by Methylmercuric Chloride)

  • This study was conducted to investigate the mechanism of protective effect by sodium selenite in methylmercuric chloride neurotoxicity, increasing intracellular $Ca^{2+}$concentration of the neuron. Methylmercuric chloride of 3mg/kg of body weight was administered simultaneously with sodium selenite of 5mg/kg and pretreatment of sodium selenite via intraperitoneal injection to rats. Also, effect of methylmercuric chloride($25{\mu}M,\;50{\mu}M,\;100{\mu}M$) and sodium selenite($200{\mu}M$) on free intrasynaptosomal $Ca^{2+}$ concentration were studied using the fluorescent $Ca^{2+}$ indicator fura -2 in vitro. After the treatment, at 6, 24, and 48 hours later, mercury in the cerebral cortex, liver and kidney tissues, succlnic dehydrogenase activities, adenosin-5'-triphosphate concentration, acetylcholinesterase activities, and intracellular $Ca^{2+}$ concentration in the cerebral cortex were determined in vivo. Cerebral synaptosomes of rats were incubated with methylmercuric chloride and sodium selenite in Hepes buffer for 10 minutes and free intrasynaptosomal $Ca^{2+}$ concentration were measured with fura-2 in vitro. The results were summarized as follows ; 1. The combined administration of $CH_3HgCl$ and $Na_2SeO_3$ and pretreatment of $Na_2SeO_3$ according to time significantly more increased in the cerebral cortex and decreased in the liver, kidney mercury concentrations compared to the administration of $CH_3HgCl$ only. 2. The combined administration of $CH_3HgCl$ and $Na_2SeO_3$ and pretreatment of $Na_2SeO_3$ increased more succinic dehydrogenase and acetylcholinesterase activities compared to the administration of $CH_3HgCl$ only. Particularly pretreatment of $Na_2SeO_3$ significantly more compared to the administration of $CH_3HgCl$ only. The concentration of adenosine-5'-triphosphate in $Na_2SeO_3$ treatment groups revealed a favourable effect compared to the administration of $CH_3HgCl$ only. 3. Intracellular $Ca^{2+}$ concentration in administration of $CH_3HgCl$ only was increased significantly more than control group in all test hours but was increased significantly more at 48 hours only after treatment in combined administration of $CH_3HgCl$ and $Na_2SeO_3$ and pretreatment of $Na_2SeO_3$ according to time interval more decreased significantly intracellular $Ca^{2+}$ concentration compared to the administration of $CH_3HgCl$ only. 4. Free intrasynaptosomal $Ca^{2+}$ concentration in the combined administration of $CH_3HgCl$ and $Na_2SeO_3$ was decreased ($24%{\sim}40%$) significantly more than the administration of $CH_3HgCl$ only. From the above results, the specific dosage of $Na_2SeO_3$ decreased increment of intracellular $Ca^{2+}$ concentration induced by administration of $CH_3HgCl$. These findings suggest the protective mechanism of $Na_2SeO_3$ on the neurotoxicity of $CH_3HgCl$.

스테아르산과 인지질 혼합물의 농도변화에 대한 유기초박막의 안정성에 관한 연구 (A Study on the Stability of Ultra-Thin Film Mixed with Stearic Acid and Phospholipid)

  • 스테아르산과 인지질혼합물의 농도변화에 띠르는 유기초박막에 대한 안정성을 조사하였다. 스테아르산과 인지질 혼합물 유기초박막은 ITO glass에 LB법을 사용하여 제막하였다. 전기화학적 특성은 $NaClO_4$ 용액에서 3 전극 시스템으로 순환전압전류법을 사용하여 초기 1650 mV에서 최종 퍼텐셜 -1350 mV 까지 측정하였다. 그 결과 스테아르산과 인지질의 혼합물 유기초막은 순환전압전류도표로부터 산화전류로 인한 비가역공정으로 나타났다. 스테아르산과 인지질혼합물 LB막(몰비 1:1, 1:2, 1:3)에서 확산계수(D)는 $0.01N\;NaClO_4$에서 각각 $1.4{\times}10^{-3}$, $1.7{\times}10^{-3}$$1.6{\times}10^{-3}(cm^2/s)$로 산출되었다.

膠質土酸, Alkali 處理에 의한 Dawsonite의 合成에 관한 硏究 (Synthesis of Dawsonite (Basic Sodium Aluminum Carbonate) from Colloidal Earth (Aoolphane) by Treatment with Acid and Alkali)

  • Aoolphane was treated with 30% Hydrochloric acid at $18^{\circ}C$ for two hows with stirring in order to obtain the insoluble form of SiO2 gel and to extract quantitatively both $Al_2O_3$as and $Fe_2O_3AlCl_3{\cdot}6H_2O Fe$ and $Cl_3{\cdot}6H_2O$ forms, respectively, at the same time. $SiO_2$ gel was filtered and to the filterate Ammonia was added to precipitate $Al(OH)_3[Fe(OH)_3 Contaminated ]$ The precipitate was separated by filteration and the filterate was recovered as the form of $NH_4Cl$. The precipitate was treated with 200g (NaOH)/l Concentration of NaOH a little excessively to the equivalent at $65~70^{\circ}C$ as $Fe(OH)_3$ formed was insoluble, it was filtered of and to the filterate containing $NaAl(OH)_4(OH_2)_2$Carbon dioxide gas was bubbled at $50^{\circ}C~90^{\circ}C$ to obtain the precipitate with excellent filterability and crystallinity. The product was certified to be Dawsonite $(NaAl(OH)_2CO_3)$ by X-Ray diffraction analysis at below $40^{\circ}C$, when $CO_2$ gas was bubbled into the relatively lower concentration of $NaAl(OH)_4(OH_2)_2$ solution, the precipitate of very fine particles was formed, which was hard to filter and with the Composition of $\alpha-Al_2O_3-H_2O$ (Boehmite).

  • Ionic strength 및 공존(共存) 음(陰)Ion($Cl^-$, $SO^{2-}\;_4$, $ClO^-\;_4$)이 Zeolite에 의(依)한 중금속(重金屬)의 흡착(吸着)에 미치는 영향(影響)을 조사(調査)한 결과(結果)는 다음과 같다. Ionic strength별(別) Zeolite에 의(依)한 Cd, Cu 및 Zn의 흡착등온선(吸着等溫線)은 Freundlich 식(式)에 잘 부합(符合)되었다. Ionic strength가 증가(增加)할수록 중금속(重金屬)의 흡착량(吸着量)은 3종(種)의 서로 다른 음(陰)Ion이 공존(共存)하여도 모두 감소(減少)하는 경향(傾向)이었다. Background salt의 음(陰)Ion 종류별(種類別) 중금속(重金屬) 흡착량(吸着量)은 $ClO^-\;_4>SO^{2-}\;_4>Cl^-$의 순(順)이었으며 흡착평형용액내(吸着平衡溶液內)에서 음(陰)Ion은 Ligand로 작용(作用)하여 중금속(重金屬) Ion들과 Complex를 형성(形成)하는 능력(能力)에 차이(差異)가 있음이 인정(認定)되었다.

Experimental Study for the Identification of the Nascent Product of OH Heterogeneous Reaction with NaCl using Chemical Ionization Mass Spectrometry

  • An experimental study on the nascent product of the OH heterogeneous reaction with NaCl was performed under dry and wet conditions using a bead-filled flow tube system coupled to a high-pressure chemical ionization mass spectrometer. The ozone concentration in the flow tube for the atomic hydrogen removal was varied in order to control the conversion reaction of molecular chlorine into HCl for the identification of the nascent product. The mass spectrometric observation was that the $O_3$ introduction reduced the concentration HCl, while it increased the concentration of $Cl_2$ and ClO. Based on the experimental results, we suggest that the nascent product of the titled reaction is gaseous $Cl_2$, which is followed by fast conversion into HCl in presence of H. No significant difference in the concentration profile between under dry and wet (RH = 2%) conditions was observed.

  • In this study poly (1-methyl-4-vinylpyridinium iodide-co-styrene) membrane with pyridinium cation as a fixed carrier was synthesized and the transport characteristics of the membrane was examined over various factors. As the concentration of the fixed carrier in the membrane was increased, the water content was increased. Meanwhile, the counter current of the organic anion and the chloride ion, the following results were obtained. Initial flux of Cl-, organic anion and Na+ decreased with the increasing thickness of membrane, and as the concentration of the fixed carrier increases, the initial flux of Cl- and organic anion increase but the initial flux of Na+ decreased. The flux equation of the organic anion, CCl3COO- was obtained from saturation kinetics as follows;$V_{o}=\frac{(8.67{\times}10^{-5}){\cdot}[NaCl]}{9.63{\times}10^{-2}+[NaCl]} mol/cm^2h$

Statistical Optimization of Medium Components for the Production of Biosurfactant by Bacillus licheniformis K51

  • The nutritional medium requirement for biosurfactant production by Bacillus licheniformis K51 was optimized. The important medium components, identified by the initial screening method of Plackett-Burman, were $H_3PO_4,\;CaCl_2,H_3BO_3$, and Na-EDTA. Box-Behnken response surface methodology was applied to further optimize biosurfactant production. The optimal concentrations for higher production of biosurfactants were (g/l): glucose, $1.1;NaNO_3,\;4.4;MgSO_4{\cdot}7H_2O,\;0.8;KCl,\;0.4;CaCl_2,\;0.27;H_3PO_4,\;1.0ml/l;\;and\;trace elements\;(mg/l):H_3BO_3,\;0.25;CuSO_4,\;0.6;MnSO_4,\;2.2;Na_{2}MoO_4,\;0.5;ZnSO_4,\;6.0;FeSO_4,\;8.0;CoCL_2,\;1.0;$ and Na-EDTA, 30.0. Using this statistical optimization method, the relative biosurfactant yield as critical micelle dilution (CMD) was increased from $10{\times}\;to\;105{\times}$, which is ten times higher than the non-optimized rich medium.

E. coli 불활성화와 산화제 생성에 미치는 소독 공정 결합의 영향 (Effect of Disinfection Process Combination on E. coli Deactivation and Oxidants Generation)

  • The aim of this research was to evaluate the effect of combination of disinfection process (electrolysis, UV process) on Escherichia coli (E. coli) disinfection and oxidants (OH radical, $ClO_2$, HOCl, $H_2O_2$ and $O_3$) generation. The effect of electrolyte type (NaCl, KCl and $Na_2SO_4$) on the E. coli disinfection and oxidants generation were evaluated. The experimental results showed that performance of E. coli disinfection of electrolysis and UV single process was similar. Combination of electrolysis and UV process enhanced the E. coli disinfection and 4-carboxybenzaldehyde (4-CBA, indicator of the generation of OH radical) degradation. It is clearly showed synergy effect on disinfection and OH radical formation. However chlorine ($ClO_2$, HOCl) and oxygen type ($H_2O_2$, $O_3$) oxidants were decreased with the combination of two process. In electrolysis + UV complex process, electro-generated $H_2O_2$ and $O_3$ were reacted with UV light of UV-C lamp and increased 4-CBA degradation(increase OH radical). Disinfection of electrolyte of chlorine type was higher than that of the sulfate type electrolyte due to the higher generation of OH radical and oxidants.

울릉도 북면 천부리 추산 용출소의 수질화학적 특성 (The Hydrochemistry of ChusanYongchulso Spring, Cheonbu-ri, Buk-myeon, Northern Ulleung Island)

  • 울릉도 북면 천부리에 소재하는 자분샘인 추산용출소 지하수의 수질특성을 규명하고, 이를 통하여 대수층의 발달특징을 고찰하였다. 상류구배인 나리분지 일대에 광범위하게 분포하는 부석층과 화산쇄설층, 칼데라 함몰과 관련된 단층과 절리와 같은 파쇄대는 투수계수가 높아 용출소의 지하수함양에 유리한 조건을 제공한다. 특히 다공성이며 표면적이 넓은 알칼리성 화산쇄설물로 구성된 화산암류로 인해 상류하천수와 용출소는 독특한 수질특성을 보인다. 용출소의 수질유형은 $Na-HCO_3$형이며, 상류의 샘과 하천수는 $Na-HCO_3$형과 Na-Cl형의 경계에 놓인다. EC과 상관성이 높은 성분으로는 $HCO_3$와 Na를 비롯하여 F, Ca, Mg, Cl, $SiO_2$, $SO_4$ 등이 있다. 높은 Na, Cl 함량은 울릉도 지질 전반에 알칼리계열의 화산암이 분포하며, 고기 화산활동에 영향을 받은 것으로 판단된다. 물-암석반응을 잘 반영하는 요소인 Eh와 pH는 어떤 수질성분과도 상관성이 거의 없는 것으로 나타났다. 요인분석결과에 의하면, 요인 1의 영향력은 54%로 나타났으며, 요인 2의 경우 25.8%이다. 요인 1에 높은 적재량을 가지는 성분은 F, Na, EC, Cl, $HCO_3$, $SO_4$, $SiO_2$, Ca, $NO_3$, Mg 등이다. 요인 2에 대해 적재량이 높은 성분은 Mg, Ca이며, 음의 적재값이 높은 성분은 K, $NO_3$, DO 등이다. 이 지역의 독특한 수질특성 즉 높은 Na, Cl, F, $SO_4$ 등은 알칼리계열의 화산쇄설암류의 세립질 입자, 높은 공극률 등이 물-암석반응을 가속화시킨 결과로 해석된다. 그러나 울릉도의 수질특성과 대수층의 발달, 수리순환을 규명하기 위하여서는 광범위한 지구물리탐사와 더불어, 동위원소, 미량원소, 추적자 등을 통한 추가연구가 필요하다.

Streptomyces sp. SL-387에 의한 Aminopeptidase M 저해제 MR-387A 및 B의 생산 배지 최적화 (Media Optimization for the Production of Aminopeptidase M Inhibitors MR-387A and B by Streptomyces sp. SL-387)

  • 토양으로부터 분리한 Streptomyces sp. SL-387 균주에 의하여 신규 aminopeptidase M 저해제 MR-387A 및 B를 생산하기 위한 최적 배지 조건을 검토하였다. 최적 배지 조건으로 glucose 1%, soybean meal 3%, yeast extract 0.2%, beef extract 0.1%, NaCl 0.3%, $K_2HPO_4$ 0.01%, $CaCO_3$ 0.03%, $MnCl_2{\cdot}4H_2O$ 0.001%, $ZnCl_2{\cdot}7H_2O$ 0.001%, $MgSO_4{\cdot}7H_2O$ 0.0005%, pH 7.0가 적당한 것으로 나타났다. 이 배지를 발효용 배지로 사용하였을때 저해제의 생산은 배양 120시간에 최대에 도달하였으며, 그때의 최대 생산성(total productivity)은 909.1 U/ml이었다.
