Nitrogen Removal in Column Wetlands Packed with Synthetic Fiber Treating Piggery Stormwater (축산단지 강우 유출수 처리를 위한 합성섬유충진 습지의 질소제거에 관한 연구)

  • A set of lab-scale polymer synthetic fiber packed column wetlands composing three columns (CW1, CW2 and CW3) with different hydraulic regimes, recirculation frequencies and pollutant loading rates, were operated in 2012. Synthetic fiber tested as an alternative wetland medium for soil mixture or gravel which has been widely used, has very high pore size and volume, so that clogging opportunity can be greatly avoided. The inflow to the wetland was artificial stormwater. All the wetlands achieved effective removal of TSS (94%~96%), TCOD (68%~73%), TN (35%~58%), TKN (62%~73%) and NH4-N (85%~ 99%). Particularly, it was observed that COD was released from the fiber during one distinct period in all wetlands. This was probably due to the degradation of polymer fiber, and the released organic matters were found to serve as carbon source for denitrification. In addition, with longer retention time and frequent recirculation, lower effluent concentration was observed. With higher pollutant loading rate, higher nitrification and denitrification rates were achieved. However, although organic matters were released from the fiber, the lack of carbon source was still the limiting factor for the system since the release persisted only for 40 days.

Growth Performance and Caecal Fermentation in Growing Rabbits Fed on Diets Containing Graded Levels of Mulberry (Morus alba) Leaves

  • Growth performance, nutrient digestibility and changes in caecal fermentation pattern was studied on four groups of 8 grower rabbits (soviet chinchilla) each, fed on diets containing 17 to 18% CP and $10.4-11.00MJ\;DE\;kg^{-1}$ feed. The complete diets contained mulberry leaves along with other feed ingredients at 0 (LH), 15 (ML15), 30 (ML30) and 45% (ML45) levels. In LH diet ground lucerne hay was added at 15% of the diet as roughage source. The diets were uniform in other nutritional parameters except the higher amount of hemicellulose (18.7 and 16.5%) in LH and ML15 diets. The performance of grower rabbits was better (p<0.05) on LH and ML15 diets in terms of twelve weeks weight (kg), total gain (g), average daily gain (ADG) and feed efficiency (%). Different levels of mulberry leaves in the diet did not influence the dry matter intake. Digestibility of nutrients for DM, OM, CP, NDF and energy was uniform in all the diets but the digestibility of CF, ADF and cellulose was higher (p<0.01) on ML15, ML30 and ML 45 diets. The hemicellulose digestibility was significantly higher (p<0.01) on LH (44.52%) and ML15 (48.00%) compared to ML30 (33.54%) and ML 45 (39.17%) diets. The nitrogen retention (% of intake) was higher (p<0.05) in LH and ML15 diets than ML30 and ML45 diets. The caecum weight as percent of intestine weight consistently increased (p<0.05) with increasing content of mulberry leaves in diets. Total nitrogen and $NH_3$-N was higher on LH and ML15 diets. It is concluded that incorporation of mulberry leaves can replace Lucerne hay in complete rabbit feed (15% in diet). Even at higher level i.e. 30 and 45% of the diet mulberry leaves based complete feed gave promising results.

Effect of increased dietary crude protein levels on production performance, nitrogen utilisation, blood metabolites and ruminal fermentation of Holstein bulls

  • Objective: This study investigated the effect of dietary crude protein (CP) supplementation on nutrient intake, nitrogen (N) utilisation, blood metabolites, ruminal fermentation and growth performance of young Holstein bulls. Methods: Twenty-one young bulls weighing $277{\pm}11.2kg$ were equally divided into three groups and were offered diets formulated with low CP (LCP; 10.21% CP and 4.22% rumen degradable protein [RDP]), medium CP (MCP; 12.35% CP and 5.17% RDP) and high CP (HCP; 14.24% CP and 6.03% RDP). Yellow corn silage was used as a unique forage source and was mixed with concentrate. This mixed feed was given ad libitum to the young bulls included in the study. Results: Results showed that CP intake, blood urea nitrogen, N intake, total N excretion and N balance increased linearly with an increase in dietary CP level (p<0.05). However, no significant difference was observed in nutrient digestibility among the bulls receiving the different diets. Ruminal pH (p<0.05) and ammonia nitrogen ($NH_3-N$) concentration (p<0.01) were significantly higher in the bulls receiving the MCP and HCP diets than in those receiving the LCP diet. The bulls receiving the HCP diet showed significantly higher ruminal bacterial protein level, propionate, acetate and total volatile fatty acid (TVFA) concentrations than bulls receiving the LCP diet (p<0.05). Moreover, dietary CP level exerted a significant positive effect on the final body weight, average daily gain and gain-to-feed ratio of the bulls (p<0.05). Conclusion: High dietary CP level is optimal for achieving maximum growth and high profitability without exerting a negative effect on the physiology of growing Holstein bulls.

Influence of Transgenic Corn on the In vitro Rumen Microbial Fermentation

  • In this study, the comparative effects of transgenic corn (Mon 810 and Event 176) and isogenic corn (DK729) were investigated for their influence on in vitro rumen fermentation. This study consisted of three treatments with 0.25 g rice straw, 0.25 g of corn (Mon810/Event176/DK 729) mixed with 30 ml rumen fluid-basal medium in a serum bottle. They were prepared in oxygen free conditions and incubated at $39^{\circ}C$ in a shaking incubator. The influence of transgenic corn on the number of bacterial population, F. succinogenes (cellulolytic) and S. bovis (amylolytic), was quantified using RT-PCR. Fermentative parameters were measured at 0, 2, 4, 8, 12 and 24 h and substrate digestibility was measured at 12 and 24 h. No significant differences were observed in digestibility of dry matter, NDF, ADF at 12 and 24 h for both transgenic and isogenic form of corns (p>0.05) as well as in fermentative parameters. Fluid pH remained unaffected by hybrid trait and decreased with VFA accumulation as incubation time progressed. No influence of corn trait itself was seen on concentration of total VFA, acetic, propionic, butyric and valeric acids. There were no significant differences (p<0.05) in total gas production, composition of gas (methane and hydrogen) at all times of sampling, as well as in NH3-N production. Bacterial quantification using RT-PCR showed that the population number was not affected by transgenic corn. From this study it is concluded that transgenic corn (Mon810 and Event 176) had no adverse effects on rumen fermentation and digestibility compared to isogenic corn. However, regular monitoring of these transgenic feeds is needed by present day researchers to enable consumers with the option to select their preferred food source for animal or human consumption.

Tritium Distribution in Leachates from Domestic Solid Waste Landfills (생활폐기물 매립장 침출수의 삼중수소 분포)

  • It is for the purpose of investigating the tritium distribution in the leachates, the raw and treated leachates and the condensates of the methane gas, which have occurred from domestic solid waste landfills. Also it aims to measure the tritium distribution level on the colloid size of the leachates, the raw and treated leachates. It was found that the major inorganic contaminants of the leachates were Na, K, Ca, Mg, $NH{_4}^+$-N and $Cl^-$. The mean tritium level of the raw leachates of the investigated 13 landfill sites for 6 months was 17 ~ 1196 TU. It corresponded to a several scores or hundreds of magnitude higher value than that of the normal environmental sample level except for two landfill sites. Also such a high concentration of the tritium was found in the treated leachates and methane gas condensates as well. Nevertheless it is important to emphasize that the tritium level which was found in this research is about 100 times lower than the tritium limit for the drinking water quality. And most of the tritium existed in the dissolved colloid of the leachate of which the colloid size is below $0.45{\mu}m$. Also, according to the tritium analysis results of the leachates after filtration with $0.45{\mu}m$ membrane filter for some landfills, it is likely that some tritium of the leachate would be distributed in a colloid size over $0.45{\mu}m$. In general the relationship between the tritium and other contaminants in the raw leachate was low, but it was relatively high between the tritium and TOC. However, the tritium content in the leachate had no meaningful relationship with the scale, hydrological characteristics and age of the landfill.

Characterization of an Aniline-degrading Bacterium, Delftia sp. JK-2 Isolated from Activated Sludge of Municipal Sewage Treatment Plant (도시폐수처리장의 활성슬러지에서 분리한 Aniline 분해세균 Delftia sp. JK-2의 특성연구)

  • Activated sludge samples were collected from a municipal sewage treatment plant and used for enrichment of microbial consortia with aniline as the sole carbon and nitrogen source. Threc aniline-degrading bacteria were obtained lrom microbial consortia and an isolate which has excellent aniline degradability was selected for this study. The isolate was Gram-negative, and identified and designated as Delfha sp. JK-2 on the basis of various physiological and biochemical tests. 10 mM aniline was completely degraded within 24 hours after inoculation of the culture. Ammonium ion was liberated in the medium transiently during the incubation and disappeared when aniline was completely degraded. Addition of glucose as a supplementary source to aniline minimal media showed significant decrease in aniline degradat~on rate for the strain Effective degradation of aniline was achieved by the addition of 0.5% nitrate as a nitrogen source, and resulted in approximately 80% higher aniline degradation compared to the absence of nitrate. Phylogenetic analysis based on 16s [DNA sequence revealed that the strain was closely related to De@ia acidovorans, with 96% overall similarity. The 16s [DNA sequence of JK-2 was also found to be closely related to those of six other clonal types, including Acidovoru, Aquaspirillum. Xylophilus, Variovorm, and Rhodofernr.

Assessment on Nitrous oxide (N2O) Emissions of Korea Agricultural Soils in 2009 (2009년 우리나라 농경지 토양에서의 N2O 배출량 평가)

  • This study was conducted to assess $N_2O$ emissions in agricultural soils of Korea. According to 1996 and 2006 IPCC (Intergovernmental Panel on Climate Change) methodology, $N_2O$ emission was calculated the sum of direct emission ($N_2O_{DIRECT}$) and indirect emission ($N_2O_{INDIRECT}$). To calculate $N_2O$ emissions, emission factor was used default of IPCC and activity data was used the food, agricultural, forestry and fisheries statistical yearbook of MIFAFF (Ministry for Food, Agriculture, Forestry and Fisheries). It was emitted 8,608 $N_2O$ Mg resulted from direct emission by application of chemical fertilizer and animal manure, input in n-fixation crops and input of crop residues and emissions converted $N_2O$ into $CO_2$ equivalent was 2,668 $CO_2$-eq Gg. Indirect emission as $N_2O_{(G)}$ (atmospheric deposition of $NH_3$ and $NO_X$) and $N_2O_{(L)}$ (leaching and runoffs) were 4,567 and 6,013 $N_2O$ Mg and emissions converted $N_2O$ into $CO_2$ equivalent were 1,416 and 1,864 $CO_2$-eq Gg, respectively. Total $N_2O$ emission in Korea agricultural soil in 2009 was 5,948 $CO_2$-eq Gg.

Changes in Heavy Metal Phytoavailability by Application of Immobilizing Agents and Soil Cover in the Upland Soil Nearby Abandoned Mining Area and Subsequent Metal Uptake by Red Pepper (광산 인근 밭토양에서 중금속 안정화제 처리 및 복토층 처리에 의한 토양 중 중금속 식물유효도 변화 및 고추의 중금속 흡수)

  • At the current situation of continuous utilization of heavy metal contaminated upland for agricultural purpose in Korea, minimizing transport of heavy metals from soil to crops is important for securing safety of human health. The present study (in field scale) examined the efficiency of several soil amendments (dolomite, steel slag, lime, zeolite, and compost) on reduction of phytoavailable heavy metals (Cd, Cu, Pb, and Zn) in soil through comparison with no amendment treatment and clean soil cover treatment. For determination of the phytoavailability, 1 M $NH_4NO_3$ extraction and red pepper cultivation were introduced. Among the amendments, in general, dolomite and steel slag were the most effective in reduction of metal (Cd, Pb, and Zn) phytoavailability resulting in less accumulation of these metals in shoot and fruit of red pepper. However, dolomite and steel slag treatment was not as effective as clean soil cover treatment which showed the least metal accumulation in red pepper fruit. Nevertheless, with taking into account the cost, treatment of dolomite or steel slag can be competitive method because the current study showed that dolomite or steel slag treatment reduced accumulated heavy metal concentration effectively in both shoot and fruit of red pepper compared to those from control soil and the concentration in fruit was within the standard value (<0.2 mg $kg^{-1}$ for both Cd and Pb).

Synthesis and Optical Property of (GaN)1-x(ZnO)x Nanoparticles Using an Ultrasonic Spray Pyrolysis Process and Subsequent Chemical Transformation (초음파 분무 열분해와 화학적 변환 공정을 이용한 (GaN)1-x(ZnO)x 나노입자의 합성과 광학적 성질)

  • In this study, (GaN)1-x(ZnO)x solid solution nanoparticles with a high zinc content are prepared by ultrasonic spray pyrolysis and subsequent nitridation. The structure and morphology of the samples are investigated by X-ray diffraction (XRD), field-emission scanning electron microscopy, and energy-dispersive X-ray spectroscopy. The characterization results show a phase transition from the Zn and Ga-based oxides (ZnO or ZnGa2O4) to a (GaN)1-x(ZnO)x solid solution under an NH3 atmosphere. The effect of the precursor solution concentration and nitridation temperature on the final products are systematically investigated to obtain (GaN)1-x(ZnO)x nanoparticles with a high Zn concentration. It is confirmed that the powder synthesized from the solution in which the ratio of Zn and Ga was set to 0.8:0.2, as the initial precursor composition was composed of about 0.8-mole fraction of Zn, similar to the initially set one, through nitriding treatment at 700℃. Besides, the synthesized nanoparticles exhibited the typical XRD pattern of (GaN)1-x(ZnO)x, and a strong absorption of visible light with a bandgap energy of approximately 2.78 eV, confirming their potential use as a hydrogen production photocatalyst.

Uptake of Butachlor by Rice Seedlings and Its Phytotoxic Action to the Physiological Activities (수도묘(水稻苗)의 Butachlor 흡수(吸收) 및 약해발생(藥害發生) 특성(特性)에 관한 생리적(生理的) 연구(硏究))

  • To clarify the mode of uptake of butachlor (2-chloro-2', 6'-diethyl-N-(butoxymethyl) acetanilide) by rice seedlings, its phytotoxic action to growth and physiological activities, studies were conducted with rice seedlings, at the 6th or 7th leaf-stage, which were treated with nutrient solution containing butachlor 0, 1.8, 3.6, 7.2, 10.8 or 14.4 ppm for 1, 2 or 4 days, in other case, the solutions were thereafter renewed with the untreated nutrient solution for further growth. Uptake of butachlor by rice seedlings increased linearly with increase of its concentration and duration of uptake. Butachlor inhibited root growth more than shoot growth, furthermore, the inhibitory effect on the shoot growth was greater in height than in weight or leafing rate. After 4 day-treatment, the rates of shoot growth in weight were delayed for 4 days. Butachlor inhibited water uptake rapidly and linearly with increase of its external concentration. The reduced uptake of water was followed by slow increase in the stomatal resistance of leaves. Upon completion of butachlor treatment, rate of water uptake was recovered rapidly, but the stomatal resistance with lag in time. Butachlor did not affect the uptake of cation such as ammonium, potassium and calcium, but inhibited substantially uptake of nitrate in proportion to its concentration. Especially, butachlor did not affect synthesis and degradation of nitrate reductase. In addition, butachlor has shown much greater binding to the lipidic substances from rice roots than the proteinous material. The primary mechanism of phytotoxic action of butachlor does not seem to be its effect on the protein synthesis, but great affinity to membranes. The inhibition of water uptake, and its subsequent closure of stomates is thought very important for reduced growth under mild phytotoxicity.
