Phenolic Compounds from Cercis chinensis Leaves (박태기나무엽의 페놀성분)

  • Studies on the pharmaco-constituents from the leaves of Cercts chinensis which have been used for the treatment of inflammation, contusion, dilated blood, pain of heart and stomach, edema, etc. in Korean folk remedies were carried out. Dried leaves of the plant were extracted with MeOH. The MeOH extract was suspended in distilled water and subsequently fractionated with $Et_{2}O$ and n-BuOH. From the $Et_{2}O$ and n-BuOH fractions, six phenolic compounds were isolated and identified as myricitrin($C_{21}H_{20}O_{12}, {\;}m.p.{\;}199~200^{\circ}$. $4myricetin-3-O-{\alpha}-L-rhamnopyranoside$), kaempferol($C_{15}H_{10}O_{6}, {\;}m.p. 276^{\circ}$), quercetin($C_{15}Ha_{10}O_{7}, {\;}m.p.{\;}313~314^{\circ}$), quercitrin ($C_{21}H_{20}O_{12}, {\;}m.p.{\;}176~178^{\circ}, {\;}quercetin-3-O-{\alpha}-L-rhamnopyranoside$), gallicin ($C_{8}H_{8}O_{5}, {\;}m.p.{\;}202~203^{\circ}$. methyl gallate), gallic acid ($C_{7}H_{6}O_{5}, {\;}m.p.{\;}260~265^{\circ}) through their physico-chemical data and UV, IR, EI-MS, $^{13}C-NMR$, and $^{1}H-NMR$ analysis with authentics.

Improved High-Performance Liquid Chromatographic Method for the Determination of N-vanillyl-n-nonivamide(nonivamide) in Korean Hot Peppers (HPLC 법에 의한 국산 고추에 함유되어 있는 Nonivamide의 신규 분석법 개발)

  • We report a novel HPLC method for analyzing the capsaicinoids and nonivamide. The following conditions were used: a total column and column temperature of $3^{\circ}C$, a detection wavelength of 280 nm, an eluent containing acetonitrile : 0.5% formic acid(31:69, v/v). The experimental data was compared with standard nonivamide and capsaicin. The results showed that nonivamide was eluted in 96.7 minutes with a recovery ratio of 96.5% after adding known nonivamide. In addition, the detection limit for nonivamide was 18.6 ng. The capsaicinoids from Korean pepper were extracted (after blooming 60th) from 3 breeds cultivated in Korea, Buchon, Chungyang and Hanbando. and crisis it analyzed sympathizes the nonivamide in the result nonivamide from the Buchon and the Chungyang which are not detected from the Hanbando $0.16{\sim}0.24{\mu}g/g$ it detects with very small amount it contains it confirmed the each capsaicinoid with the HPLC and the LC-MS. These results are expected contribute to the quality control of various product processes, which includes product delivery inspection as well as examining the capsaicinoids present in the product for possible medicine manufacture and various foodstuff, and the nonivamide content, which may be a feed for various manufacturing processes involving chemical synthesis.

Tyrosinase Inhibition Activity of Some Herbal Drugs (수종 생약의 티로시나제 억제효과)

  • To isolate biologically active compounds which exhibit tyrosinase inhibition activity and ultimately express skin whitening effect, 14 oriental herbal drugs were screened in ter ms of tyrosinase inhibition. For this purpose, in vitro enzyme assay system for tyrosinase, so called Pomerantz method with some modifications has been established. Crude methanolic extracts from 14 herbal drugs were made and examined for their inhibitory activity against tyrosinase. Those extracts from Cnidii Rhizoma, Arecae Semen, Caryophylli Flos, and Ephedrae Herba showed strong inhibitory activities on mushroom tyrosinase. Therefore, crude methanolic extracts from those 4 herbal drugs were further fractionated using ether, butanol and water. respectively. The ether and n-butanol extracts from Arecae Semen and the n-butanol and water extracts from Caryophylli Flos, respectively, showed relatively strong tyrosinase inhibitory activity compared to arbutin.

  • Phytochemical study of Ganoderma lucidum (Polyporaceae) let to the isolation of five lanostane triterpenoid (1-5), along with two nitrogen derived phenolic compounds, N-phenylethylformamide (6) and N-acetylphenethylamine (7). The structures of the compounds were determined by 1D and 2D NMR, and MS experiments, as well as by comparison of their data with published values. Compounds 6 and 7 were isolated for the first time from the genus Ganoderma and this species. All the compounds were evaluated for cancer chemopreventive potential based on their ability to inhibit nitric oxide (NO) production induced by lipopolysaccharide (LPS) in mouse macrophage RAW 264.7 cells in vitro. Among the isolates, compounds 2 and 3 showed moderate inhibitory activity against NO production.

Synthesis, Characterization and in vitro Antibacterial Studies on Mixed Ligand Complexes of Iron(III) Based on 1,10-phenanthroline

  • As part of our attempt to discover novel active compounds against multi-drug resistant pathogens, we hereby report two new complexes of iron(III) with formulae: [Fe(L1)2(H2O)2]Cl3 and [Fe(L1)2(L2)(H2O)]Cl2 where L1 = 1,10-phenanthroline (C12H8N2) and L2 = guanide (C5H4N5O-). The synthesized complexes were characterized using spectroscopic analysis (ESI-MS, ICP-OES, FT-IR, and UV-Vis), cyclic voltammetry, CHN analysis, gravimetric chloride determination, melting point determination, and conductance measurement. Octahedral geometries are assigned to both complexes. In vitro antibacterial activity was tested on two Gram-positive (Staphylococcus aureus, Streptococcus epidermidis) and two Gram-negative (Escherichia coli and Klebsiella pneumoniae) bacteria using the disc diffusion method. The complexes demonstrated appreciable activity against these pathogens. Interestingly, the [Fe(L1)2(L2)(H2O)]Cl2 complex manifested a higher degree of inhibition against the drug-resistant Gram-negative bacteria than the commercially available drug, namely erythromycin.

Measurement and Estimation of VOC Composition from Gasoline Evaporation

  • Source profiles were developed for a total of 45 volatile organic compounds (VOC) that can be emitted from gasoline evaporation. The gasoline samples of five major brands (for each season) were blended on the basis of the market share in Seoul area and analyzed by a GC-MS/FID system. In addition, we calculated gasoline evaporative compositions using the Raoult's law from the liquid gasoline compositions. The measured and estimated gasoline vapor compositions agree well each other. As a group, alkanes are the most abundant in the gasoline vapors profiles (77.4% on average), followed by alkenes (19.1%), and aromatics (1.7%). As a specie in gasoline vapor, i-pentane is the most abundant, followed by n-butane, n-pentane, i-butane, trans-and cis-2-butenes, 2-methyl-2-butene, and trans-and cis-2-pentenes . It was also seen that aromatic content was much lower in the vapor phase compositions. From the comparison between experimental and calculated compositions, we identified the fact that once the gasoline vapor composition is reliably constructed entirely from the measured gasoline composition and the Raoult's law calculations, the need for doing separate chemical analyses of the gasoline vapor can be reduced.

A study on formation of hemoglobin adduct due to inhalation exposure with 1,3-butadiene in female mice

  • The purpose of this study is the identification of (N-2-hydroxy-3-butenyl) valine(HBVal adduct) and (N-2,3,4-trihydroxy-butyl)valine(THBVal adduct)with mice inhalation exposure with 1,3-butadiene for 3 weeks($6\;hr/day\;{\times}\;5\;days/week$). Body weights were significantly lower from 4 or 9 exposure post-day in 1000 or 500ppm inhalation group than in control. The levels of HBVal adducts are 1.8, 3.7 and 6.2 pmol/mg globin in $1^{st}$, $2^{nd}$ , and $3^{rd}$ week for 500 ppm 1,3-butadiene(BD), and 5.7, 7.4 and 16.0 pmol/mg globin in $1^{st}$, $2^{nd}$ , and $3^{rd}$ week for 1000 ppm BD inhalation exposure. The levels of THBVal adducts are 32.0, 42.0 and 55.0 pmol/mg globin in $1^{st}$, $2^{nd}$ , and $3^{rd}$ week for 500 ppm BD, and 67.8, 72.7 and 83.5 pmol/mg globin in $1^{st}$, $2^{nd}$ , and $3^{rd}$ week for 1000 ppm BD inhalation exposure. Their ratios of THBVal and HBVal adducts are higher at earlier exposure and lower concentration. They are17.8, 11.4 and 8.87 in $1^{st}$, $2^{nd}$ , and $3^{rd}$ week for 500 ppm BD, and 11.9, 9.8 and 5.2 in $1^{st}$, $2^{nd}$ , and $3^{rd}$ week for 1000 ppm BD inhalation exposure. In conclusion, THBVal and HBVal adducts are a important hemoglobin adduct for monitoring of BD exposure, and the latter is more biomarker than the other.

  • The roots of Brassica campestris ssp rapa were extracted with 80% aqueous MeOH, and the concentrated extract was partitioned with EtOAc, n-BuOH and $H_{2}O$. From the EtOAc and n-BuOH fractions, five compounds were isolated through the repeated silica gel column chromatographies. From the result of spectroscopic data including NMR and MS, the chemical structures of the compounds were determined as palmitic acid methyl ester (1), linolenic acid methyl ester (2), linoleic acid methyl ester (3), ${\beta}-sitosterol$ (4) and daucosterol (5).

  • The stem barks of Fraxinus rhynchophylla was extracted with 95% EtOH, and the concentrated extract was successively partitioned with $CH_2Cl_2$, n-BuOH, and $H_2O$ in order to investigate the major phytochemicals. From the $CH_2Cl_2$ soluble fraction, a sterol (1) was isolated through the repeated silica gel column chromatographies. Three additional compounds (2-4) were isolated from the n-BuOH soluble fraction through silica gel, RP-18, and Sephadex LH-20 column chromatographies. Their chemical structures were elucidated as daucosterol $(1;{\beta}-sitosterol-3-O-{\beta}-D-glucopyranoside)$, caffeic acid (2), 6,8-dihydroxy-7-methoxycoumarin (3), and coniferaldehyde glucoside (4) by comparing their spectral data with those in the literatures. All isolates (1-4) were the first to be isolated from F. rhynchophylla.

Isolation of Candida albicans Chitin Synthase 1 Inhibitor from Streptomyces sp. A6705 and Its Characterization

  • In the course of searching for potent chitin synthase 1 inhibitors from natural resources, Streptomyces sp. A6705 was found to exhibit potent inhibitory activity against the chitin synthase 1 from C. albicans (CaCHS1p). As a result, the inhibitor was isolated and identified using a series of chromatographies. Through chemical analyses with UV spectrophotometry, MS spectrometry, and various NMR techniques, the inhibitor was identified as N,N-bis(2-phenylethyl)urea. The compound exhibited strong inhibitory activity against the chitin synthase 1 from C. albicans with an $IC_{50}$ of 14 ${\mu}g$/ml, representing a similar inhibitory activity to that of the well-known chitin synthase inhibitor, polyoxin D ($IC_{50}$ 15 ${\mu}g$/ml). However, the compound showed no inhibitory activity against the chitin synthase 2 of Saccharomyces cerevisiae up to 280 ${\mu}g$/ml, which is structurally and functionally analogous to CaCHS 1 p. In addition, the compound exhibited weak antifungal activities against Cryptococcus neoformans and Rhizoctonis solani.
