Effects of Lead Acetate on the Uptake of [$^3$H]-serotonin by the Synaptosomes Separated from the Cerebrum and Brain Stem of the Rat (초산납이 흰쥐 synaptosome의 [$^3$H]-serotonin의 흡수에 미치는 영향)

  • This study was carried out to investigate the physiological changes induced acutely with the low doses of lead acetate in the synaptosomes from the cerebrum and brain stem of the rat. The general uptake patterns of [$^3$H]-serotonin were observed in synaptosomes, as a model of presynaptic nerve terminal, from the cerebrum and brain stem. And the effects of the low doses of lead acetate on the uptake process were investigated id vitro and in vivo. The Km value of the uptake of the [$^3$H]-serotonin by the synaptosomes was 0.5 $\mu$M in the cerebrum and 0.1 $\mu$M in the brain stem. These low values reveal that the synaptosomes from the cerebrum and the brain stem have a high affinity to [$^3$H]-serotonin, especially in brain stem. The uptake of $\mu$M-serotonin was dependant on the sodium and potassium ions. When being treated with ouabain, the $Na^+$ $-K^+$ ATPase inhibitor, the uptake of [$^3$H]-serotonin was reduced. This supports strongly that the uptake of [$^3$H]-serotonin was sensitive to the changes of the concentrations of the sodium and potassium ions. When the calcium channel blocker, verapamil, was treated, the uptake of [$^3$H]-serotonin was changed only in synaptosomes from the brain stem.   • A one-dimensional heat conduction problem in cylindrical coordinate system was solved using Crank-Nicolson finite difference method to predicting the temperature distribution in rice storage bin with wall insulator. The model can simulate the grain temperatures in insulated round bins using the input data of initial grain temperature. ambient air temperature, wind velocity, solar radiation on a horizontal surface, and thermal properties of grain, bin wall, wall insulator, insulator cover, and air. Temperatures were collected at the bin center, 0.65m in radial direction from the center, and near the bin wall in 2.7m diameter bin filled with rough rice to depth of 3.0m were used to validate the simulation model. Grain temperatures predicted by the model were in very good agreement with the measured temperatures. The residual mean square error between measured and predicted grain temperatures at the bin center was $1.38^{\circ}C$.

  • This study intended to measure the desorption and adsorption EMC of four years old Peeled ginseng, Unpeeled ginseng and Taegeuk ginseng under various conditions$20^{\circ}C$, $30^{\circ}C$, $40^{\circ}C$, $50^{\circ}C$) and five levels of relative humidity from 31% to 88%) by the static method. Four widely used EMC models were selected and evaluated. Also the empirical model was evaluated. The results are summarized as follows ; 1) EMC difference between ginseng size was not found but found between ginseng species. EMC difference between Peeled ginseng and Unpeeled ginseng was not found. EMC of Peeled ginseng and Unpeeled ginseng was higher than that of Taegeuk ginseng. 2) The hysteresis, which is difference between desorption and adsorption EMC, was found. Desorption EMC was higher than adsorption EMC. The hysteresis at the same temperature decreased as relative humidity increase. The difference of hysteresis between Peeled ginseng and Unpeeled ginseng was not large and the hysteresis of Taegeuk ginseng was smaller than those of other species. 3) Among the selected models, Henderson model was the best to predict the adsorption EMC of White ginseng(Peeled and Unpeeled ginseng), and Oswin model was the best to predict the desorption EMC of White ginseng and the desorption and adsorption EMC of Taegeuk ginseng. Statistical Optimization of Chitinase Production by Pantoea dispersa to Enhance Degradation of Crustacean Chitin Waste

  • A novel chitinase-producing bacterial strain of Pantoea dispersa was isolated from the sea near Bhavnagar, India for efficient disposal of chitinous waste from the seafood processing industry. The medium components were optimized by using a cubic model in the central composite design for increasing chitinase production. The optimal concentrations for higher production of chitinase were (g l-1) chitin, 10.0; urea, 0.35; MgSO4 7H2O, 0.08, and CaCl2, 0.15. Here, peptone (0.05 g l-1) was used as a constant variant in all trials. Using a statistical optimization method, the chitinase production was found to increase from 108 to 486.4 units ml-1. Chitin was prepared from the crustacean waste, and Fourier Transform Infrared (FTIR) Spectroscopy was used to characterize the isolated chitin. Chitinous waste degradation was studied in terms of chitinase production.

A New Receptor for site Clonidine in the Eel, Anguilla japonica Intestine (뱀장어(Anguilla japonica)장의 상피세포막에 존재하는 새로운 clonidine 결합 수용체에 관한 연구)

  • A novel clonidine binding sites were characterized in the intestinal membrane isolated from seawater eels, Anguilla japonica. The specific clonidine binding sites consisted of at least two classes, high affinity ($K_d=1.4{\pm}0.3$ nM n = 5) and low affinity ($K_d=175{\pm}34$ nM n = 5) sites. The specific binding of 2 nM [$^3H$]clonidine was most enhanced at $20^{\circ}C$ and pH 7.5, and reversed by unlabelled clonidine. Such binding was hardly inhibited by adrenaline, yohimbine or rauwolscine, indicating that most binding sites are distinct from $\alpha_2$-adrenoceptor. The specific clonidine binding sites was inhibited by various imidazoline/guanidinium drugs, indicating existence of imidazoline/guanidinium receptive sites (IGRS) or imidazoline receptors in the eel intestine. Competition experiments revealed that rank order to displace 2 nM [$^3H$]clonidine from their binding sites was as follows : guanabenz > cirazoline = naphazoline = UK14,304 = ST587 $\geq$ clonidine $\geq$ idazoxan = RX821002 = tolazoline > ST93 = oxymetazoline = amiloride = ST91 > yohimbine = efaroxan = rauwolscine $\geq$ adrenaline = ST567 = histamine = agmatine. Although physiological role of IGRS is not clear yet even in mammalian cell/tissues, eel intestine may be a good model to elucidate how the IGRS act in the cell and to decide what is the endogenous ligand for the IGRS.

Effects of neutron irradiation on superconducting critical temperatures of in situ processed MgB2 superconductors

  • Effects of neutron irradiation on the superconducting properties of the undoped $MgB_2$ and the carbon(C)-doped $MgB_2$ bulk superconductors, prepared by an in situ reaction process using Mg and B powder, were investigated. The prepared $MgB_2$ samples were neutron-irradiated at the neutron fluence of $10^{16}-10^{18}n/cm^2$ in a Hanaro nuclear reactor of KAERI involving both fast and thermal neutron. The magnetic moment-temperature (M-T) and magnetization-magnetic field (M-H) curves before/after irradiation were obtained using magnetic property measurement system (MPMS). The superconducting critical temperature ($T_c$) and transition width were estimated from the M-T curves and critical current density ($J_c$) was estimated from the M-H curves using a Bean's critical model. The $T_cs$ of the undoped $MgB_2$ and C-doped $MgB_2$ before irradiation were 36.9-37.0 K and 36.6-36.8 K, respectively. The $T_cs$ decreased to 33.2 K and 31.6 K, respectively after irradiation at neutron fluence of $7.16{\times}10^{17}n/cm^2$, and decreased to 22.6 K and 24.0 K, respectively, at $3.13{\times}10^{18}n/cm^2$. The $J_c$ cross-over was observed at the high magnetic field of 5.2 T for the undoped $MgB_2$ irradiated at $7.16{\times}10^{17}n/cm^2$. The $T_c$ and $J_c$ variation after the neutron irradiation at various neutron fluences were explained in terms of the defect formation in the superconducting matrix by neutron irradiation.

  • The Intelligent Power Unit (IPU) utilized in Honda's Civic Hybrid Integrated Motor Assist (IMA) system was developed with the aim of making every component lighter, more compact and more efficient than those in the former model. To reduce energy loss, inverter efficiency was increased by fine patterning of the Insulated Gate Bipolar Transistor (IGBT) chips, 12V DC-DC converter efficiency was increased by utilizing soft-switching, and the internal resistance of the IMA battery was lowered by modifying the electrodes and the current collecting structure. These improvements reduced the amount of heat generated by the unit components and made it possible to combine the previously separated Power Control Unit (PCU) and battery cooling systems into a single system. Consolidation of these two cooling circuits into one has reduced the volume of the newly developed IPU by 42% compared to the former model.

Investigation on the Microbiological and Biochemical Properties of Kimchi in the Solid-state Model System Designed for Fermented Sausages

  • The objective of this study was to investigate the potential of the application of kimchi LAB as starter culture in the production of fermented sausages. For this, the solid-state model media composed to simulate the substantial conditions of meat mixtures were fermented for 120 h after the treatment with different concentrations of kimchi (0.5, 1.0, 1.5, 3.0, and 5.0%) and lyophilized kimchi-powder (0.2 % and 0.5%). During the fermentation period, the growth of total viable cells and LAB, and the changes of pH and titratable acidity were investigated. The initial LAB counts ranged from 7.18 to 8.34 Log CFU/ mL for kimchi media and from 6.93 to 6.94 Log CFU/mL for kimchi-powder media depending on the added concentrations. The kimchi LAB in this study were not influenced by the immobilized condition for their adaptation and growth by showing no lag phase and thus acted similar as in the submerged medium. The initially increased counts reached around 9 Log CFU/ mL in 12 h independent of the concentrations of a ded kimchi. However, the growth and metabolic activity of kimchi-powder LAB were influenced by the immobilized condition. Supposedly, as the nutrient supply in solid-state depended solely on diffusion, these differences in the souring properties were caused by the LAB topography in the medium matrix. Nevertheless, the differences in the numbers of LAB between two media were less than 0.5 Log units and the pH drop in the solidstate batches was quite rapid and reached low values. Therefore, it can be assumed that kimchi and kimchi-powder LAB showed the utility as the substitute of commercial starter culture even without a rehydrating pretreatment.

Effect of Ethanol on the Protolytic Properties of the Vitamins B Group

  • A multiwavelength spectrophotometric titration method was applied to study protolytic constants of four water-soluble vitamins, folic acid(vitamin B9 or B0), thiamine(vitamin B1), riboflavin(vitamin B2) and pyridoxal (vitamin B6) in binary ethanol-water mixtures at 25oC and an ionic strength of 0.1M NaNO3. The protolytic equilibrium constants, spectral profiles, concentration diagrams and also the number of components has been calculated from the curve fitting of the pH-absorbance data with appropriate mass balance equations by an established factor analysis model. DATAN program was used for determination of acidity constant and SPECFIT program was used for calculation of standard deviations and partial correlation coefficients. A glass electrode calibration procedure based on the four parameter equation pH=α+SpcH+JH+[H+]+ JOH-Kw/[H+] based on the Gran,s plots was used to obtain pH-readings in the concentration scale (pcH). The effect of the solvent on the protolytic constants was discussed.

Effective Dose Equivalent due to Inhalation of Indoor Radon-222 Daughters in Korea (한국인의 라돈-222 자핵종 호흡 실효선량당량 평가)

  • Effective dose equivalents resulting from inhalation of indoor radon-222 daughters at 12 residential areas in Korea were assessed by a simple mathematical lung dosimetry model based on the measurements of long-term averaged radon concentrations at 340 dwellings. The long-term averaged indoor radon-222 concentrations and corresponding eqilibrium equivalent radon $concentration(EEC_{Rn})$ measured by passive time-integrating CR-39 radon cups are in the range of $33.82{\sim}61.42Bq/m^3(median\;:\;48.90Bq/m^3)$ and of $13.53{\sim}24.57Bq/m^3(median\;:\;19.55Bq/m^3)$, respectively. The effective dose equvalent conversion factor for the exposure to unit $EEC_{Rn}$ derived in this study was estimated $1.07{\times}10^{-5}mSv/Bq\;h\;m^{-3}$ for a reference adult and agreed well with those recommended by the ICRP and UNSCEAR. The annual average dose equivalent to the lung $(H_{LUNG})$ from inhalation exposure to measured $EEC_{Rn}$ was estimated to be 20.90 mSv and resulting effective dose $equivalent(H_E)$ was to be 1.25 mSv, which is about 50% of the natural radiation exposure of 2.40 mSv/y to the public reported by the UNSCEAR.
