Methanolic Extract Isolated from Root of Lycoris aurea Inhibits Cancer Cell Growth and Endothelial Cell Tube Formation In Vitro

  • In this study, we investigated the effect of methanolic extract isolated from the root of Lycoris aurea (LA) on the growth of cancer cells and the tube formation activity of endothelial cells. Various cancer cells were treated with LA at doses of 0.3, 1, 3, 10 or 30 ${\mu}g/ml$ and LA significantly suppressed the growth of several cancer cell lines, including ACHN, HCT-15, K-562, MCF-7, PC-3 and SK-OV-3, in a dose-dependent manner. We also found that LA induced cell cycle arrest at G2/M phase in ACHN renal cell adenocarcinoma cells. Further study demonstrated that LA concentration-dependently inhibited the tube formation, which is a widely used in vitro model of reorganization stage of angiogenesis, in human umbilical vein endothelial cells. Collectively, these results show that LA inhibits the growth of cancer cells and tube formation of endothelial cells and the growth-inhibitory effect of LA might be mediated, at least in part, by blocking cell cycle progression.

Low temperature deposition of LaMnO3 on IBAD-MgO template assisted by plasma (IBAD-MgO 기판상에 플라즈마를 이용한 LaMnO3 저온 증착)

  • LMO($LaMnO_3$) buffer layer of superconducting coated conductor was deposited on IBAD-MgO template in the plasma atmosphere at $650^{\circ}C$ which is relatively low compared with conventional deposition temperature of more than $800^{\circ}C$. Deposition method of LMO was DC sputtering, and target and deposition chamber were connected to the cathode and anode respectively. When DC voltage was applied between target and chamber, plasma was formed on the surface of target. The tape substrate was located with the distance of 10 cm between target and tape substrate. When anode bias was connected to the tape substrate, electrons were attracted from plasma in target surface to the tape substrate, and only tape substrate was heated by electron bombardment without heating any other zone. The effect of electron bombardment on the surface of substrate was investigated by increasing bias voltage to the substrate. We found out that the sample of electron bombardment had the effect of surface heating and had good texturing at low controlling temperature.

Optimal Cardiac Magnetic Resonance Contrast-Enhanced Timing Robust Angiography (CMR-CENTRA) for the Three-Dimensional Reconstruction of the Bilateral Atria in the Electroanatomic Mapping (EAM) of Atrial Fibrillation

  • Purpose: To optimize the timing of scans using cardiac magnetic resonance contrast-enhanced timing robust angiography (CMR-CENTRA) for electroanatomic mapping (EAM) of the right atrium (RA) and left atrium (LA) in patients with atrial fibrillation (AF). Materials and Methods: Fifty patients with AF (38 men; mean age, $59.6{\pm}9.3years$) underwent CMR-CENTRA in preparation for EAM. The CMR-CENTRA data were acquired at five different scan times: 0 seconds, 5 seconds, 10 seconds, 15 seconds, and 20 seconds after an intravenous injection of contrast media. To evaluate the degree of contrast enhancement, right atrial relative contrast (RA-RC) and left atrial relative contrast (LA-RC) on the CMR-CENTRA scans were assessed at each time point. The three-dimensional (3D) reconstruction of the RA and LA for the EAM system was performed using the CMR-CENTRA data. Results: A CMR-CENTRA at a scan time of 10 seconds showed significantly greater LA-RC (P < 0.05) compared with all other scan times. A CMR-CENTRA at a scan time of 15 seconds showed significantly greater RA-RC (P < 0.05) compared with all other scan times. In the 3D reconstruction of the RA, the success rates of CMR-CENTRA at scan times of 10 seconds and 15 seconds were 18% and 100%, respectively. In the 3D reconstruction of the LA, the success rates of CMR-CENTRA at 10- and 15-second scan times were 100%. Conclusion: The CMR-CENTRA data acquired at 15 seconds after the injection of contrast media is appropriate for the preparation of an EAM system that is focused on the RA and LA in patients with AF.

Phase Evolution and Thermal Conductivities of (La1-xGdx)2Zr2O7 Oxides for Thermal Barrier Coatings (열차폐코팅용 (La1-xGdx)2Zr2O7 산화물의 상형성과 열전도도)

  • With increase in operating temperature of gas turbine for higher efficiency, it is necessary to find new materials of TBC for replacement of YSZ. Among candidate materials for future TBCs, zirconate-based oxides with pyrochlore and fluorite are prevailing ones. In this study, phase structure and thermal conductivities of $(La_{1-x}Gd_x)_2Zr_2O_7$ oxide system are investigated. $(La_{1-x}Gd_x)_2Zr_2O_7$ system are comprised by selecting $La^{3+}/Gd^{3+}$ as A-site ions and $Zr^{4+}$ as B-site ion in $A_2B_2O_7$ pyrochlore structures. With powder mixture from each oxide, $(La_{1-x}Gd_x)_2Zr_2O_7$ oxides are fabricated via solid-state reaction at $1600^{\circ}C$. Either pyrochlore or fluorite or mixture of both appears after heat treatment. For the developed phases along $(La_{1-x}Gd_x)_2Zr_2O_7$ compositions, thermal conductivities are examined, with which the potential of $(La_{1-x}Gd_x)_2Zr_2O_7$ compositions for TBC application is also discussed.

Effect of n-3 Fatty Acids on Estrogen Dependency and Protein Kinase C Activity of Human Breast Cancer Cell

  • To investigate the effect of long chain n-3 polyunsaturated fatty acids on breast cancer cell growth, estrogen-dependent MCF-7 human breast cancer cells were cultured serum-free DMEM media containing 0.5$\mu\textrm{g}$/ml of differnet kinds of fatty acids; linoleic acid(LA), arachidonic acid(AA), eicosapentaenoic acid(EPA) and docosahexaenoic acid acid(DHA) and 1, 0.1, 0.2, 0.5and 1.0ng/ml 17$\beta$-estradiol as well as 10$\mu\textrm{g}$/mi insulin and 1.25 mg/ml delipidized bovine serum albumin for 3 days. Cell growth monitored by MTT assay was lower in DHA and EPA treatments as compared to LA treatment, but not with AA treatment. Estrogen concentrations at which cell growth was initially stimulated were 0.1ng/ml for LA and DHA treatments and 0.2ng/ml for EPA and AA treatments, but the degree of stimulation was 25~30% lower in DHA and EPA treatments than in LA treatment. Fatty acid analysis showed that each fatty acid in culture medium was well incoporated into celluar lipid. Protein kinase C activity of cells was most elevated in LA treatment from 2 to 8 hours of culture followed by DHA, EPA, and AA treatments. It is concluded that inhibitions of n-3 DHA and EPA on breast cancer cell growth as compard with n-6 LA is mediated via changes in membrane fatty acid composition reducing estrogen sensitivity and increasing protein kinase C activity.

Ultraviolet Radiation-Induced Apoptosis is Inversely Correlated with the Expression Level of Poly(ADP-ribose) Polymerase

  • The present study was conducted to elucidate whether the expression level of poly(ADP-ribose) polymerase (PARP) is related to the ultraviolet radiation (UV)-induced apoptosis. After treatment of the mammalian cell lines HeLa S3 and Chinese hamster ovary (CHO) with 50 J/m2 UV, induction of apoptosis was determined by several means during 24 h post-incubation. Incidence of apoptosis was much lower in CHO than HeLa S3 cells based on the percentage of apoptotic cells in terms of morphological changes in nucleus or direct counting of viable cells and qualitative or quantitative DNA fragmentation. Interestingly, when the expression level of PARP was measured by western blotting, the amounts of PARP that was retained at each time point inversely correlated with the incidences of apoptosis in these cells. Concomitant with generation of the 85 kDa fragment, 116 kDa PARP disappeared in HeLa S3 within 6 h after UV treatment, whereas a fair amounts of 116 kDa band was still retained in CHO cells at 36 h post-incubation. This inverse relationship was also observed in the adaptive response system, in which cells weve treated with a high dose of UV after pretreatment with a low dose. As expected, typical adaptive responses appeared in CHO cells but not in HeLa cells, showing greater cell viability and lesser DNA fragmentation. During the adaptive response in CHO cells, PARP was expressed at much higher level compared to the single, high dose-treated cells. Interestingly, even though PARP was induced at 6 h post-incubation In both cell types, its expression was more prominent in CHO cells. Thus, our data indicate that the retained level of intact PARP against UV damage inversely correlates with incidence of apoptosis in mammalian cells, and also suggest that a machinery to protect the PARP degradation against UV damage exists in CHO but not in HeLa S3 cells.

  • Voltammetric behavior of some light lanthanide ions (La$^{3+}$, Pr$^{3+}$, Nd$^{3+}$, Sm$^{3+}$, and Eu$^{3+}$) in various supporting electrolytes has been investigated by several electrochemical techniques. The peak potentials and the peak currents, their dependency on the concentration, temperature and pH effects, the reversibility of the electrode reactions are described. The reduction of La$^{3+}$, Pr$^{3+}$ and Nd$^{3+}$ in 0.1 M lithium chloride proceeds by a three-electron change directly to the metallic state (Ln$^{3+}$ + 3e- → Ln$^0$) and charge transfer is totally irreversible. However, the reduction of Sm$^{3+}$ in 0.1 M tetramethylammonium iodide and Eu$^{3+}$ in 0.1 M lithium chloride proceeds in two stages (Ln$^{3+}$ + e- → Ln$^{2+}$ and Ln$^{2+}$ + 2e- → Ln$^0$). At pH values lower than ca.4 the hydrated lanthanide species (Ln(OH)$^{2+}$) reduced before the lanthanide ions (Ln$^{3+}$) due to the catalytic effect of hydrogen ions, and peak current increase with in the order Eu$^{3+}$ < Sm$^{3+}$ < Nd$^{3+}$ < Pr$^{3+}$ < La$^{3+}$ in differential pulse polarography. Some representative plots of $i_{pc}V^{-1/2} (proportional to current function) vs. V show considerable influence of hydrogen ion/lanthanide ion concentration in cyclic voltammetry. It is shown that a reaction of lanthanide ions with proton and/or water and catalytic reaction is enhanced at lower pH and at decreased lanthanide ion concentration.

Ginsenosides from the fruits of Panax ginseng and their cytotoxic effects on human cancer cell lines (인삼(Panax ginseng) 열매로부터 분리한 ginsenoside의 동정 및 암세포독성 효과)

  • The fruits of Panax ginseng were extracted with 80% aqueous MeOH and the concentrates were partitioned into EtOAc, n-BuOH, and $H_2O$ fractions. The repeated $SiO_2$ and octadecyl $SiO_2$ column chromatographies for the EtOAc fraction led to isolation of five ginsenosides. The chemical structures of these compounds were determined as ginsenoside F1 (1), ginsenoside F2 (2), ginsenoside F3 (3), ginsenoside Ia (4), notoginsenoside Fe (5) based on spectroscopic analyses including nuclear magnetic resonance, MS, and infrared. Compounds 2-5 were isolated for the first time from the fruits of P. ginseng in this study. All isolated compounds were evaluated for cytotoxic activities against human cancer cell lines such as HCT-116, SK-OV-3, human cervix adenocarcinoma (HeLa), HepG2, and SK-MEL-5. Among them compounds 2, 4, and 5 showed significant cytotoxicity on cancer cells. Compound 2 exhibited cytotoxicity on SK-MEL-5, HepG2, and HeLa cells with $IC_{50}$ values of 82.8, 86.8, and $78.3{\mu}M$, respectively. Compound 4 showed cytotoxicity on HCT-116, SK-MEL-5, SK-OV-3, HepG2, and HeLa cells with $IC_{50}$ values of 24.5, 25.4, 26.3, 22.0, and $24.9{\mu}M$, respectively. Compound 5 did on SK-MEL-5 cell with $IC_{50}$ value of $81.7{\mu}M$. The cytotoxicity of ginsenoside 2, 4, and 5 isolated from the fruits of Panax ginseng showed strong inhibition effect against on cancer cells, all of which have a glucopyranosyl moiety on C-3.

Effect of Aeration and Agitation Conditions on the Production of Glucoamylase with Aspergillus niger No. PFST-38

  • Aspergillus niger No. PFST-38 was grown on complex media in 30L agitated fermentors at various aeration rates and stirrer speeds. We could correlate the mixing time as a function of the Reynolds number and the apparent viscosity, as follows. ${\theta}_M=2.95\;\NRe^{-0.52},\;{\theta}_M=1.88\;{\eta_a}^{0.57}$ Also, the effects of the apparent viscosity (${\theta}_a$), the impeller rotational speed (N), the air flow rate ($V_s$), and the mixing time (${\theta}_M$) on the oxygen transfer coefficient, $K_L a$ were determined experimentally, and equated as follows. $K_La=12.04N^{0.88}Vs^{0.71}{n_a}^{-0.83},\;K_La=30.2N^{0.88}Vs^{0.71}{\theta_M}^{-1.45}$ $K_La$ increased as the agitation speed and the air flow rate increased. The rate of $K_La$ increase was dependent more on the rotational speed of impeller than on the air flow rate. The glucoamylase production increased with the increase of the agitation speed upto at 500 rpm and increased with the increase of air flow rate upto at 1.0 vvm. The values calculated from the above equation confirmed that the experimental maximum production of glucoamylase was achieved when the $K_La$ and the apparent viscosity of the broth were $260\;hr^{-1}$ and 1800 cps, respectively.

Cell differentiation and Anti-oxydative effect of Dioscoreae Rhizoma on HeLa Cell (산약(山藥)의 HeLa cell 분화에 미치는 영향과 항산화효과에 대한 연구)

  • Purpose: This study examined the Cell differentiation and the anti-oxidative effect of Dioscoreae Rhizoma on HeLa cells. We are interested in whether Dioscoreae Rhizoma is capable of causing apoptosis processes on HeLa cell, and whether cotreatment of NCS with Dioscoreae Rhizoma reduces cell viability. Methods: We used aqueous extract to treat HeLa cell with different concentrations treated with a water or a MeOH extract of Dioscoreae Rhizoma (0, x10, x20, x40, x80). The MTT (3, (4, 5-dimethyl-thiazol) 2, 5-diphenyl-tetraxolium bromide) reduction assay was employed to quantify the differences in cell activity and viability. Cells were stained with DAPI and visualized by fluorescent Microscope. The caspase-3, Bcl-2, PARP, p53 expression level was monitored using western-blotting techniques. The patterns of the changes in expression were scanned and analyzed. Results: The survival rate of cells treated with Dioscoreae Rhizoma extracts increased by 20% at specific concentration. The other side Dioscoreae Rhizoma extracts induced apoptotic features including chromatin condensation and fragmentation. And Dioscoreae Rhizoma extracts increased the expression of caspase-3, p53 and the cleavage of PARP protein. However, co-treatment with Dioscoreae Rhizoma with NCS attenuated the activations of p53 and PARP protein that were mediated by NCS treatment alone. This is indicated Dioscoreae batatas extracts attenuated cytotoxicity induced by oxidative agents including NCS. Conclusion: Our results suggest Dioscoreae Rhizoma extracts induce cell differentiation or apoptosis connected with concentration. Further elucidation of concentration of Dioscoreae Rhizoma awaits many other biochemical investigative studies.
