Electron microscopy of Cytolysomes in plant cells(Glycine max Merr. and Zea mays L.) (식물세포(植物細胞)의 Cytolysomes에 관(關)한 전자현미경적(電子顯微鏡的) 연구(硏究))

  • The origin and the function of cytolysomes were studied in the mesophyll cells and the root-tip cells of Glycine max Merr. and Zea mays L. fixed by paraformaldehyde-glutaraldehyde-$OsO_4$. The cytolysome-like structures were found of three main types of configurations: multivesicular, myelin like (multilamllar) and multitubular. More complex and mixed ones were also observed. The origin of these structures seems to be initiated by invaginations or in holdings of the plasmalemma into the cell interior, and that by aggregation and convolution of endoplasmic reticulum in the cytoplasm. Invagination of the plasmalemma were found of two main types of configurations: concentric whorls of lamellar and multivesicular. The structures were also observed within vacuoles and cytoplasm. Since the structurers are widely distributed in the cells and are greatly varied in sizes and shapes. These structures originate from the plasmalemma and the cytoplasm subsequently protrudes into the vacuole, and that seem to play an important role on the formation of the autophagic vacuoles. The possible function and fate of these structures are discussed.

Effect of Airfoil Thickness on the Optimum Gurney Flap Height (최적 Gurney 플랩크기에 대한 익형두께의 영향)

  • A numerical investigation was performed to determine the effect of airfoil thickness on the optimum Gurney flap height using NACA 00XX series airfoils. Seven airfoils which have 3% chord thickness difference were used. These were NACA 0006, 0009, 0012, 0015, 0018, 0021, and 0024. A Navier-Stokes code, FLUENT, was used to calculate the flow field about airfoil. The fully turbulent results were obtained using the standard $k-{\varepsilon}$ two-equation turbulence model. To provide a check case fur our computational method, numerical studies for NACA 4412 airfoil were made and compared with already existing experimental data for this airfoil by Wadcock. For every NACA 00XX airfoil, Gurney flap heights ranging from 0.5% to 2.0% chord were changed by 0.5% chord interval and their effects were studied. With the numerical solutions, the relationship between $(L/D)_{max}$ and airfoil thickness as a function of flap height and the relationship between $(L/D)_{max}$ and flap height as a function of airfoil thickness were investigated. The same relationship for $(C_l)_{max}$ also were shown. From these results, the optimum flap size for each airfoil thickness can be determined and vice versa.

  • Recently new soybean saponins with D DMP (2,5-dihydroxy-6-methyl-2,3,- dihydro-4H-pyran-4-one) moiety have been isolated from legumes. The purpose of this study is to characterize ROS scavenging activities of DDMP saponins ($\alpha$g, $\beta$g saponin) isolated from Glycine max (L.) Merrill. The scavenging activity on OH was examined in terms of lipid peroxidation in the rat liver homogenates and the same activity on $O_2$ was also determined in the xanthine-xanthine oxidase system, respectively. Up to 0.25 mg DDMP saponins ($\alpha$g and $\beta$g saponins) did not cause any significant effects on the prevention of lipid peroxidation as compared with the control group. In terms of superoxide scavenging activities, 0.25 and 0.5 mg $\alpha$g saponin inhibits only 2.6% and 5.5% (p<0.05) of the control group, respectively. However, $\alpha$g saponin dose-dependently (p<0.01, r=0.955) inhibits the formation of superoxide radical unto 21.3% of the control group with a maximal dose of 0.5 mg (p<0.01), equivalent to 0.17 units of superoxide dismutase activity.

A Study on the Evaluation Metric of a Civil Aircraft Noise (민간항공기 소음평가 단위에 관한 연구)

  • The duration time of aircraft noise event is also an important factor for the evaluation of civil aircraft noise, which is considered as a notable characteristic of military aircraft noise. SEL is proved as a suitable noise metric for the measuring military aircraft noise of various flight pattern considering the duration time of noise event. This study reviews whether SEL is a suitable for measuring civil aircraft noise and study shows SEL is fairly compensating the duration time of civil aircraft noise event for the evaluation of aircraft noise. Noise metric for the evaluation aircraft noise based on SEL is more appropriate than based on $L_{MAX}$ for compensating duration time of noise event either military aircraft or civil aircraft. In this reason, current formula of WECPNL based on energy average of measured $L_{MAX}$ for the evaluation of aircraft noise impact in 'Test Method of Noise and Vibration of Korea' is recommended to be amended to formula of WECPNL based on energy average of measured SEL considering compensation of noise event duration time, if WECPNL is not based on measured EPNL, a metric compensating duration time.

Effect of Light Quality on Growth and Fatty Acid Production in Chlorella vugaris Using Light Emitting Diodes (발광다이오드를 이용한 광파장에 따른 Chlorella vulgaris의 생장과 지방산 생산에 미치는 효과)

  • Microalgae are considered as superior biodiesel producers, because they could effectively produce high amount of lipid with fast growth rate. In this study, Chlorella vulgaris was exposed to various light wavelengths (${\lambda}_{max}$ 470 nm, ${\lambda}_{max}$ 525 nm, and ${\lambda}_{max}$ 660 nm) using light emitting diodes (LEDs) to examine effect of light quality on their growth and fatty acid production in 0.4-L bubble column photobioreactors. Fluorescent lamps were also used as polychromatic light sources (control). From the results, biomass productivity was varied by light wavelength from 0.05 g/L/day to 0.30 g/L/day. Maximum biomass productivity was obtained from red LED among tested ones. We also observed that contents of oleic acid and linolenic acid, which affect biodiesel properties, were significantly changed depending on supplied wavelength. These results indicated that production of algal biomass, and fatty acid content and productivity could be improved or controlled by supplying specific light wavelength.

  • Experiments of alcohol fermentation of the yeast,K. fragi1is CBS 1555 were performed to obtain the following results. In these experiments, the initial concentrations of sugar which was composed of inulin and fructose as weight ratio of one to one were 30, 50, 75, 100 and 150g/l and the initial densities of the microorganism were less than 0.5g/l, 10g/1 to 15g/1, and 50g/l. The functional relationship among specific growthrates, sugar concentrations, and alcohol concentrations could be expressed by Aiba-shoda equation and the specific growth rate represented the trend that decreased with increase in the initial concentration of the microorganism. Also, $\mu$max and Ks of Monod's equation could be expressed as the function of initial cell concentration like the following equations. $\mu$max=0.8-0.008X Ks=0.54X+8 In the region that sugar, alcohol and cell concentrations were 10g/1 to 120g/l, 0g/l to 60g/l and 0.5g/l to 50g/l respectively, the differences between the experimental values and the calculated ones for specific growth rate approached to 40% with respect to experimental values at the worst cases, but in most cases, those were distributed in the range of less than 20%.

Kinetic Analysis for Paper-mill Wastewater Treatment Using Pure Oxygen Activated Sludge Process (순산소 활성오니공정을 이용한 제지폐수처리의 동력학적 해석)

  • An experimental study was conducted to evaluate the treatment efficiency of paper-mill wastewater using pure oxygen activated sludge process. Effects of hydraulic retention time (HRT) and organic loading on process performance and kinetics were investigated. The raw paper-mill wastewater(BOD concentration ${\leq}500mg/L$) and the effluent from dissolved air flotation(DAF) treatment(BOD concentration ${\geq}500mg/L$) were used as influent for pure oxygen activated sludge process. Average BOD removal efficiencies were above 89.3% under 6hours or longer of HRT, while under 3hours of HRT they decreased to about 82%. With the effluent from DAF process, the half saturation constants($K_S$) and the maximum specific substrate removal rate($K_{max}$) were 85 mg/L and 2.25 L/day, respectively. However, with the raw paper-mill wastewater, both $K_S$ and $K_{max}$ increased to 156 mg/L and 3.84 L/day, respectively. The microbial yield coefficient(Y) and the decay coefficient($K_d$) were 0.46 gVSS/gBOD and 0.03 L/day, respectively, with effluent from DAF process. While, Y and $K_d$ were 0.24 gVSS/gBOD and 0.035 L/day, respectively, with the raw paper-mill wastewater.

The Evaluation of Difference according to Image Scan Duration in PET Scan using Short Half-Lived Radionuclide (단 반감기 핵종을 이용한 PET 검사 시 영상 획득 시간에 따른 정량성 평가)

  • Purpose : Because of the rapid physical decay of the short half-lived radionuclide, counting of event for image is very limited. In this reason, long scan duration is applied for more accurate quantitative analysis in the relatively low sensitive examination. The aim of this study was to evaluate the difference according to scan duration and investigate the resonable scan duration using the radionuclide of 11C and 18F in PET scan. Materials and Methods : 1994-NEMA Phantom was filled with 11C of $30.08{\pm}4.22MBq$ and 18F of $40.08{\pm}8.29MBq$ diluted with distilled water. Dynamic images were acquired 20frames/1minute and static image was acquired for 20minutes with 11C. And dynamic images were acquired 20frames/2.5minutes and static image was acquired for 50minutes with 18F. All of data were applied with same reconstruction method and time decay correction. Region of interest (ROI) was set on the image, maximum radioactivity concentration (maxRC, kBq/mL) was compared. We compared maxRC with acquired dynamic image which was summed one bye one to increase the total scan duration. Results : maxRC over time of 11C was $3.85{\pm}0.45{\sim}5.15{\pm}0.50kBq/mL$ in dynamic image, and static image was $2.15{\pm}0.26kBq/mL$. In case of 18F, the maxRC was $9.09{\pm}0.42{\sim}9.48{\pm}0.31kBq/mL$ in dynamic image and $7.24{\pm}0.14kBq/mL$ in static. In summed image of 11C, as total scan duration was increased to 5, 10, 15, 20minutes, the maxRC were $2.47{\pm}0.4$, $2.22{\pm}0.37$, $2.08{\pm}0.42$, $1.95{\pm}0.55kBq/mL$ respectively. In case of 18F, the total scan duration was increased to 12.5, 25, 37.5, and 50minutes, the maxRC were $7.89{\pm}0.27$, $7.61{\pm}0.23$, $7.36{\pm}0.21$, $7.31{\pm}0.23kBq/mL$. Conclusion : As elapsed time was increased after completion of injection, the maxRC was increased by 33% and 4% in dynamic study of 11C and 18F respectively. Also the total scan duration was increased, the maxRC was reduced by 50% and 20% in summed image of 11C and 18F respectively. The percentage difference of each result is more larger in study using relatively shorter half-lived radionuclide. It appears that the accuracy of decay correction declined not only increment of scan duration but also increment of elapsed time from a starting point of acquisition. In study using 18F, there was no big difference so it's not necessary to consider error of quantitative evaluation according to elapsed time. It's recommended to apply additional decay correction method considering decay correction the error concerning elapsed time or to set the scan duration of static image less than 5minutes corresponding 25% of half life in study using shorter half-lived radionuclide as 11C.

The effect of powder A2/powder A3 mixing ratio on color and translucency parameters of dental porcelain

  • PURPOSE. The purpose of this study is to mix dental ceramic powder in varying ratios and evaluate the effect of the mixing ratio on color and translucency. MATERIALS AND METHODS. The ceramic powder of shade A3 of the same product was mixed with the shade A2 of three products: IPS e.max Ceram (Ivoclar Vivadent, Schaan, Liechtenstein), Vintage Halo (SHOFU Inc., Kyoto, Japan), and Ceramco 3 (Ceramco-Dentsply, Burlington, NJ, USA) in the following fixed ratios (0 wt%, 25 wt%, 50 wt%, 75 wt%, and 100 wt%) and then fired. A total of 150 specimen of ceramic fired were manufactured in a regular size (W: 8.5 mm, L: 10.5 mm, and H: 1.5 mm). For color and translucency, $L^*$, $a^*$, and $b^*$ were measured and Two-way analysis of variance (ANOVA) and One-way analysis of variance (ANOVA) were used for data analysis (${\alpha}$=0.05). RESULTS. The higher the mixing ratio was, $L^*$, $a^*$, and $b^*$ of IPS e.max Ceram were all increased, and $L^*$ of Vintage Halo was reduced and $a^*$ and $b^*$ were increased. $L^*$ and $a^*$ of Ceramco3 were reduced and $b^*$ of Ceramco3 was increased. Color difference (${\Delta}E^*ab$) was increased in all three products as the mixing ratio got higher. Increased mixing ratios resulted in decreased translucency parameter (TP) values for IPS e.max Ceram but increased TP values for Vintage Halo and Ceramco3. CONCLUSION. In this limited study, CIE $L^*$, $a^*$, and $b^*$ were influenced by the mixing ratio of the A3 powders and porcelain powder mixtures represented a various color and translucency.

Growth Characteristics of Acidithiobacillus thiooxidans in Different Sulfur Concentrations (황 농도에 따른 Acidithiobacillus thiooxidans의 생장 특성)

  • The growth characteristics of sulfur-oxidizing bacteria, Acidithiobacillus thiooxidans AZ11, MET, and TAS were investigated in mineral salt media supplemented with elemental sulfur of 1$\sim$50 g $L^{-1}$. The sulfur oxidation rates of A. thiooxidans. MET and TAS increased highly with increasing sulfur concentration up to 10 g L$^{-1}$, but the rates increased slowly in sulfur concentration over 10 g L$^{-1}$. A. thiooxidans AZ11 showed the parallel increase of sulfur oxidation rate until sulfur concentration increased up to 40 g L$^{-1}$. The maximum sulfur oxidation rates (V$_{max}$) of AZl1, MET and TAS were 1.88, 1.38 and 0.43 g S L$^{-1}$ d$^{-1}$, respectively. The maximum specific growth rates (${\mu}_{max}$) of AZ11, MET, and TAS were 0.33 d$^{-1}$, 0.30 d$^{-1}$ and 0.45 d$^{-1}$, respectively. Although MET and TAS couldn't grow at sulfate concentration of 40 g L$^{-1}$, AZ11 could grow in the presence of 58 g L$^{-1}$ sulfate, the final oxidation product of elemental sulfur.
