Rat에서 설파메타진의 대사 및 약물동태학 (Metabolic and pharmacokinetic profiles of sulfamethazine in the rat)

  • We used rats as the experimental animal for the elucidation of metabolic patterns and pharmacokinetic profiles of SMZ in the rat, by use of the urine and plasma from predetermined intervals, respectively. Information herefrom would give some insight into species differences and sex differences in the metabolism and pharamcokinetics of drugs, at least SMZ in particular. Results would be summarized as follows: 1. There were two hydorxy metabolites(5-hydroxysulfamethazine and 6-hydroxyethylsulfamethazine) and an acetyl derivative($N_4$-acetyl sulfamethazine) in the 24h-collected urine, on confirmation with each standard materials. There were also two unknown metabolites therein. 2. In the viewpoint of quantitative aspect, $N_4$-acetylsulfamethazine was the largest, hence it is assumed that the acetyl pathway is the major one in the metabolism of SMZ in the rat. 3. As regards sex difference in the rat, the male had more metabolic capacity than the female in metabolism of SMZ. 4. The concenteration-time curves of sulfamethazine(20mg/kg, po) in the plasma compartment were fitted to a one-compartment open model by use of a computer program(NONLIN). 5. There were significant differences(P<0.05) in the pharmacokinetics of sulfamethazine between two sexes in the rat, with higher disposition rate in the male. 6. The emergence of $N_4AcSMZ$ metabolized from SMZ was fast in the plasma of the rat. Half-life of $N_4AcSMZ$ was also. significantly different(P<0.05) between two sexes, suggesting differences in the eliminatory capacity of $N_4AcSMZ$.

고양이에서 상아질과민증 탈감작제에 의한 치수신경 활동성 변화 및 상아세관 폐쇄양상에 대한 연구 (CHANGES IN INTRADENTAL NERVE ACTIVITY AND OCCLUDING ASPECTS OF DENTINAL TUBULES BY CHEMICAL DESENSITIZING AGENTS APPLIED TO THE CAT DENTIN)

  • This experiment was performed to study mechanisms of desensitization by chemical desensitizing agents in hypersensitive dentin and compare effects of these agents by measuring the activity of intradental nerves and observing their occluding aspects on dentinal tubules with SEM over time after application of chemical desensitizing agents to the exposed dentinal surfaces. Canines of adult cats weighing 2-3 kg were cross-sectioned at 1.5 mm from incisal apex, and the smear layer of the exposed dentinal susface was removed by 32 % $H_3PO_4$ for 15 sec. Chemical desensitizing agents such as 10% $SrCl_2$, 5% $KNO_3$ and 30% $K_2C_2O_4$, were applied to the exposed dentin surfaces for 2 minutes. Intradental nerve activity was measured immediately after application of the agents, at 15 minutes and at 30 minutes by stimulating with 4M NaCl. To compare occluding ability of desensitizing agents on dentinal tubules in vivo and in vitro, the structures of the exposed dentinal surfaces of nonvital and vital teeth were morphologically observed by SEM. The results obtained were as follows : 1. Intradental nerve activity was decreased immediately after the application of 10 % $SrCl_2$, 5% $KNO_3$ and 30% $K_2C_2O_4$. (p<0.01), among which 30% $K_2C_2O_4$. showed the highest desensitizing effect(p<0.01). 2. The immediately decreased intradental nerve activity after application of 10 % $SrCl_2$ and 5% $KNO_3$ was increased over time. 10% $SrCl_2$ and 5% $KNO_3$ showed no desensitizing effect respectively at 30 minutes and at 15 minutes after application. 3. The immediately decreased intradental nerve activity after application of 30 % $K_2C_2O_4$ was persistently continued during the period of observation (p<0.01). 4. Precipitates of $SrCl_2$ and $KNO_3$ were not noted on the exposed dentinal surfaces and within dentinal tubules by SEM examination. On the other hand, 30 % $K_2C_2O_4$ produced precipitates on the exposed dentinal surfaces and openings of dentinal tubules without any formed preciptates within dentinal tubules. 5. Ten percent $SrCl_2$, 5 % $KNO_3$ and 30 % $K_2C_2O_4$ showed no differences in their occluding aspects on dentinal tubules either in vivo or in vitro studies and either immediately following application or at 30 minutes. These results suggest that the desensitizing effect of $SrCl_2$ and $KNO_3$ is resulted from their reducing effect on the intradental nerve activity rather than from their precipitates' occluding the dentinal tubules. However, desensitizing effect of 30 % $K_2C_2O_4$, is probably resulted from its precipitates' occluding the openings of the dentinal tubules as well as from it's reducing effect on the intradental nerve actibity.

시비관리 및 생 볏짚 처리가 수박연작 시설재배지 토양에 미치는 영향 (Fertilizer Management Practices with Rice Straw Application for Improving Soil Quality in Watermelon Monoculture Greenhouse Plots)

  • 수박재배에 적합한 양분함량보다 약 3배 이상 높고, 7년간 수박을 연작해온 시설 재배지에서 시비관리 및 생볏짚을 연용하면서 토양 중 염류경감 효과와 수박시들음증 발생률을 조사하였다. 시험은 관행구, 시비관리구, 시비관리 및 5, 10, 15 ton $ha^{-1}$ 생볏짚 처리구로 나누어 3년간 실시하였다. 유기물함량은 예상할 수 있는 바와 같이 대부분의 처리구에서 지속적으로 증가하였으며, 단지 시비관리구에서만 감소하였다. 토양 pH는 관행구 및 시비관리구에서는 변화가 거의 없었고, 시비관리-볏짚처리구에서 감소하는 경향을 보였다. 그러나 시험 3년차부터 10, 15 ton $ha^{-1}$ 시비관리-볏짚처리구에서 다시 증가하는 경향을 보였다. 치환성 양이온 즉 $Ca^{2+}$, $Mg^{2+}$, $K^+$, 중 $Ca^{2+}$$Mg^{2+}$의 함량과 수용성 음이온, ${NO_3}^-$, $Cl^-$, ${SO_4}^{2-}$, ${PO_4}^{3-}$의 함량은 시비관리구 및 시비관리-볏짚처리구에서 상당히 감소하는 경향을 보였다. 그러나 시비관리-볏짚처리구에서 생볏짚의 처리량과는 관계없이 시험 1년차에 많은 양이 감소한 후, 2~3년차에서는 더 이상 감소하지 않는 것으로 나타났다. 또한 치환성 $K^+$ 함량은 시비관리구에서 다고 감소하는 경향을 보였으나 다른 처리구에서는 시험기간 동한 거의 변화하지 않았다. 그리고 이러한 양이온과 음이온의 함량변화와 관계가 깊은 전기전도도(EC)를 조사한 결과 수용성 음이온 함량변화와 같은 경향을 보이며 감소하였다. 또한 수박생육과 관련하여 수박시들음증 발생률은 관행구에서 최고 약 20%까지 발생하였으나, 시비관리와 함께 생볏짚을 10 ton $ha^{-1}$ 이상 사용하면 최고3.5%까지 급격히 감소하였다.

An Automated Water Nitrate Monitoring System based on Ion-Selective Electrodes

  • Purpose: In-situ water quality monitoring based on ion-selective electrodes (ISEs) is a promising technique because ISEs can be used directly in the medium to be tested, have a compact size, and are inexpensive. However, signal drift can be a major concern with on-line management systems because continuous immersion of the ISEs in water causes electrode degradation, affecting the stability, repeatability, and selectivity over time. In this study, a computer-based nitrate monitoring system including automatic electrode rinsing and calibration was developed to measure the nitrate concentration in water samples in real-time. Methods: The capabilities of two different types of poly(vinyl chloride) membrane-based ISEs, an electrode with a liquid filling and a carbon paste-based solid state electrode, were used in the monitoring system and evaluated on their sensitivities, selectivities, and durabilities. A feasibility test for the continuous detection of nitrate ions in water using the developed system was conducted using water samples obtained from various water sources. Results: Both prepared ISEs were capable of detecting low concentrations of nitrate in solution, i.e., 0.7 mg/L $NO_3-N$. Furthermore, the electrodes have the same order of selectivity for nitrate: $NO_3{^-}{\gg}HCO_3{^-}$ > $Cl^-$ > $H_2PO_4{^-}$ > $SO{_4}^{2-}$, and maintain their sensitivity by > 40 mV/decade over a period of 90 days. Conclusions: The use of an automated ISE-based nitrate measurement system that includes automatic electrode rinsing and two-point normalization proved to be feasible in measuring $NO_3-N$ in water samples obtained from different water sources. A one-to-one relationship between the levels of $NO_3-N$ measured with the ISEs and standard analytical instruments was obtained.

고성능 액체 크로마토그래피를 이용한 마황 및 월비가출탕 전탕액에서 에페드린류의 동시분석 (Stimultaneous Determination of Ephedrine Alkaloids in Ephedra sinica and Wolbigachul-tang by High Performance Liquid Chromatography)

  • Objectives: Ephedra sinica and Wolbigachul-tang which contains Ephedra sinica are used to treat obesity in Korean medicine. The aim of this study was to analyze the quantities of ephedrine alkaloids by high-performance liquid chromatography. Methods: The analysis was performed using a YMC-Triat C18 column with operating at 25℃, and UV detection at 210 nm. The mobile phase used a gradient flow with 0.1% H3PO4 in water and acetonitrile. Specificity, linearity, precision, accuracy, limit of detection, and limit of quantification were measured for validation anaylsis. This method was applied to analyze the quantities of ephedrine alkaloids in Ephedra sinica and Wolbigachul-tang. Results: The concentration per Ephedra sinica (gram) of ephedrine and pseudoephedrine in Ephedra sinica decoction are 4.74±0.22 mg and 2.19±0.10 mg, respectively and in Wolbigachul-tang decoction are 6.39±0.34 mg and 2.97±0.21 mg, respectively. The retention time of ephedrine was 23.6 min and that of pseudoephedrine was 25.8 min, and norephedrine and methylephedrine were not detected. Conclusions: In conclusion, analyzed the concentration of ephedrine alkaloids in Ephedra sinica and Wolbigachul-tang by the developed validation method.

광릉(光陵) 삼림(森林)의 식물군집구조(植物群集構造)(I) -Classification 및 Ordination 방법에 의한 소리봉(蘇利峯)지역의 식생분석(植生分析)- (The Structure of Plant Community in Kwangnung Forest(I) -Analysis on the Forest Community of Soribong Area by the Classification and Ordination Techniques-)

  • 광릉(光陵) 소리봉(蘇利峯)지역 삼림의 식물군집구조분석(植物群集構造分析)을 위하여 46개소에 조사지(1개조사지당 $10{\times}10m$ 방형구(方形區) 5개씩 설치)를 설정하고 식생조사를 실시하여 얻은 자료에 대하여 TWINSPAN에 의한 classification방법과 4종류의 ordination(PO, PCA, RA, DCA) 방법을 적용하였다. Classification에 의하여 갈참나무-서어나무군집, 서어나무-갈참나무-소나무군집, 갈참나무-서어나무-까치박달나무군집, 까치박달-갈참나무군집 등 4개의 group으로 조사지가 분리되었다. Ordination에 의한 조사지분석에서는 DCA가 가장 효과적이었으며 4개의 group으로 분리되었다. 종에 대한 두 기법분석에 의해 추정된 천이과정(遷移過程)은 교목상층(喬木上層)에서 소나무${\rightarrow}$신갈나무, 졸참나무, 갈참나무, 서어나무, 팥배나무${\rightarrow}$까치박달나무, 물푸레나무, 층층나무의 순이었고, 교목하층(喬木下層) 및 관목층(灌木層)은 진달래, 개옻나무, 참싸리, 병꽃나무${\rightarrow}$참개암나무, 생강나무${\rightarrow}$고추나무, 작살나무, 괴불나무의 순이었다. 환경인자(環境因子)의 ordinationqnstjr에 의하면 소나무군집에서 까치박달나무군집으로 갈수록 토양산도, 유기물함량, 전질소함량, 치환성양이온함량등의 토양성질들이 양호하여졌다. 서어나무, 갈참나무, 신갈나무등은 까치박달나무, 층층나무와 상이한 niche에 존재함이 나타났다.

남매지에서 Eubacteria 군집구조의 계절적 변화와 그에 영향을 미치는 환경요인 (Seasonal Variation of Eubacterial Community Structure and Their Structure Affecting Environmental Parameters in Reservoir)

  • 경북 경산시 에 위치한 남매지 에서 세균군집 의 구조를 fluorescent in situ hybridization(FISH) 방법을 이용하여 분석하였고 아울러 세균군집크기에 영향을 주는 환경요인을 측정하여 그 상관성을 파악하고자 하였다. 담수계에 우점하는 것으로 알려진 proteobacteria($\alpha$.$\beta$.$\gamma$-subclasses)와 글Cytophaga-Flavobacterium (CF) group에 속하는 진정세균군의 연중변화를 정점별로 측정하였다. 총세균수에 대한 각 세균군집 비율은 정점간의 큰 차이 없이 $\alpha$-subclass가 4.0~29.2%,$\beta$-subclass가 1.7~25.8%, $\gamma$-subclass가 1.8~12.8%를, 그리고 CF group은 4.9~36.3%의 군집 비율을 나타냈다. 온도,SS, pH, DOC,질산염, 조류 현존량 등 환경요인과 세균군집 간의 상관관계를 분석한 결과, 총세균수는 수온, DOC,55, eubacteria는 DOC, 녹조류 현존량, $\gamma$-subclass 세균군은 DOC, CF group세 균은 녹조류 현존량과 양의 상관성을 나타낸 반면 $\beta$-subclass는 남세균 . 총 조류 현존량과 음의 상관성을 나타냈다.

확인적 요인모형을 이용한 낙동강 유역의 오염특성 분석 (Analysis of Pollutant Characteristics in Nakdong River using Confirmatory Factor Modeling)

  • The study was conducted to analyze the spatio-temporal changes in water quality of the major 36 sampling stations of Nakdong River, depending on each station, season using the 17 water quality variables from 2000 to 2010. The result was verified to interpret the characteristics of water quality variables in a more accurate manners. According to the Principal component analysis (PCA) and Exploratory factor analysis (EFA) results; the results of these analyses were identified 4 factors, Factor 1 (nutrients) included the concentrations of T-N, T-P, $NO_{3}-N$, $PO_{4}-P$, DTN, DTP for sampling station and season, Factor 2 (organic pollutants) included the concentrations of BOD, COD, Chl-a, Factor 3 (microbes) included the concentrations of F.Coli, T.Coli, and Factor 4 (others) included the concentrations of pH, DO. The results of a Cluster analysis indicated that Geumhogang 6 was the most contaminated site, while tributaries and most of the down stream sites of Nakdong River were mainly affected by each nutrients (Factor 1) and organic pollutants (Factor 2). The verification consequence of Confirmatory factor analysis (CFA) from Exploratory factor analysis (EFA) result can be summarized as follows: we could find additional relations between variables besides the structure from EFA, which we obtained through the second-order final modeling adopted in CFA. Nutrients had the biggest impact on water pollution for each sampling station and season. In particular, It was analyzed that P-series pollutant should be controlled during spring and winter and N-series pollutant should be controlled during summer and fall.

  • 폴리비닐알콜, 규소텅스텐산, 인산 및 수용액으로 구성된 $80{\mu}m$의 두께의 고분자겔 전해질 필름을 제조하여 활성탄소계 전기이중층 케페시터를 제조하였다. 제조한 고분자겔 전해질 필름은 상온에서 $10^{-2}S\;cm^{-1}$의 높은 이온전도도를 나타내었으며, 본 전해질 필름을 적용한 활성탄소계 전기이중층 케패시터는 100 mV/s에서 $58F\;g^{-1}$의 높은 캐패시턴스 특성과 우수한 수명특성을 나타내었다.

우모 케라틴 분해세균의 분리, 특성 및 우모 분해산물의 식물 생육촉진 효과 (Isolation and Characterization of Feather Keratin-Degrading Bacteria and Plant Growth-Promoting Activity of Feather Hydrolysate)

  • This study was conducted to isolate and characterize a novel feather-degrading bacterium producing keratinase activity. A strain K9 was isolated from soil at poultry farm and identified as Xanthomonas sp. K9 by phenotypic characters and 16S rRNA gene analysis. The cultural conditions for the keratinase production were 0.3% fructose, 0.1% gelatin, 0.04% $K_2HPO_4$, 0.06% $KH_2PO_4$, 0.05% NaCl and 0.01% $FeSO_4$ with an initial pH 8.0 at $30^{\circ}C$ and 200 rpm. In an optimized medium containing 0.1% chicken feather, production yield of keratinase was approximately 8-fold higher than the yield in basal medium. The strain K9 effectively degraded chicken feather meal (67%) and duck feather (54%), whereas human nail and human hair showed relatively low degradation rates (13-22%). Total free amino acid concentration in the cell-free supernatant was about 25.799 mg/l. Feather hydrolysate produced by the strain K9 stimulated growth of red pepper, indicating Xanthomonas sp. K9 could be not only used to increase the nutritional value of chicken feather but also a potential candidate for the development of natural fertilizer applicable to crop plant soil.
