상황버섯균사체 쌀의 HT22 신경세포 보호 효과 (Neuroprotective Effect of Rice with Phellinus linteus Mycelium in HT22 Cells)

  • 상황버섯균사체 쌀의 뇌세포 보호 효과를 분석하여 상황버섯균사체 쌀에 대한 기초자료를 제공하고, 이를 이용하여 뇌 질환의 예방과 관련된 기능성 식품으로의 개발가능성을 찾고자 하였다. 상황버섯균사체 쌀 추출물을 처리하여 마우스 해마세포주 HT22의 세포보호 효과를 알아보기 위해 MTS assay를 이용한 세포생존율과 western blot을 이용한 세포사멸 단백질의 발현을 관찰하였다. 상황버섯균사체 쌀 추출물의 단일 독성을 측정한 결과 상황버섯균사체 쌀 추출물 처리군은 102.68% 이상의 생존율을 보여 신경세포의 사멸에 영향을 주지 않았다. 또한, 상황버섯균사체 쌀 추출물과 $H_2O_2$를 함께 처리하였을 때 $H_2O_2$로 자극된 세포는 63.80%의 세포생존율을 나타내었고, 상황버섯균사체 쌀 추출물과 $H_2O_2$가 함께 처리된 세포는 76.85%, 92.46%, 95.57%로 농도 의존적으로 세포생존율이 증가하였다. HT22 cell에 $H_2O_2$와 상황버섯균사체 쌀 추출물을 처리하여 apoptosis 유도 단백질을 측정한 결과, pro-apoptotic protein인 Bax는 발현이 억제되었고, anti-apoptotic protein인 Bcl-2는 발현이 증가함을 확인하였다. 또한, caspase-3와 PARP의 발현을 억제하여 상황버섯균사체 쌀 추출물이 $H_2O_2$로 유도되는 apoptosis를 억제하는 것을 확인할 수 있었다.

Protective effects of Tat-NQO1 against oxidative stress-induced HT-22 cell damage, and ischemic injury in animals

  • Oxidative stress is closely associated with various diseases and is considered to be a major factor in ischemia. NAD(P)H: quinone oxidoreductase 1 (NQO1) protein is a known antioxidant protein that plays a protective role in various cells against oxidative stress. We therefore investigated the effects of cell permeable Tat-NQO1 protein on hippocampal HT-22 cells, and in an animal ischemia model. The Tat-NQO1 protein transduced into HT-22 cells, and significantly inhibited against hydrogen peroxide ($H_2O_2$)-induced cell death and cellular toxicities. Tat-NQO1 protein inhibited the Akt and mitogen activated protein kinases (MAPK) activation as well as caspase-3 expression levels, in $H_2O_2$ exposed HT-22 cells. Moreover, Tat-NQO1 protein transduced into the CA1 region of the hippocampus of the animal brain and drastically protected against ischemic injury. Our results indicate that Tat-NQO1 protein exerts protection against neuronal cell death induced by oxidative stress, suggesting that Tat-NQO1 protein may potentially provide a therapeutic agent for neuronal diseases.

Ginsenoside Rg3 from Red Ginseng Prevents Damage of Neuronal Cells through the Phosphorylation of the Cell Survival Protein Akt

  • Neuronal cell death significantly contributes to neuronal loss in neurological injury and disease. Typically, neuronal loss or destruction upon exposure to neurotoxins, oxidative stress, or DNA damage causes neurodegenerative diseases such as Alzheimer's disease. In this study, we attempted to determine whether ginsenoside Rg3 from red ginseng has a neuroprotective effect via an anti-apoptotic role induced by S-nitroso-N-acetylpenicillamine (SNAP) at the molecular level. We also investigated the antioxidant effect of Rg3 using a metal-catalyzed reaction with $Cu^{2+}/H_2O_2$. Our results showed that Rg3 ($40-100\;{\mu}g/mL$) protected SK-N-MC neuroblastoma cells under cytotoxic conditions and effectively protected DNA from fragmentation. In the signal pathway, caspase-3, and poly (ADP-ribose) polymerase (PARP) were kept at an inactivated status when pretreated with Rg3 in all ranges. In particular, the important upstream p-Akt signal pathway was increased in a dose-dependent manner, which indicates that Rg3 may contribute to cell survival. We also found that oxidative stress can be mitigated by Rg3. Therefore, we have concluded that Rg3 plays a certain role in neurodegenerative pathogenesis via an anti apoptotic, antioxidative effect.

Neuroprotective effects of hesperetin on H2O2-induced damage in neuroblastoma SH-SY5Y cells

  • BACKGROUND/OBJECTIVES: Oxidative stress is a fundamental neurodegenerative disease trigger that damages and decimates nerve cells. Neurodegenerative diseases are chronic central nervous system disorders that progress and result from neuronal degradation and loss. Recent studies have extensively focused on neurodegenerative disease treatment and prevention using dietary compounds. Heseperetin is an aglycone hesperidin form with various physiological activities, such as anti-inflammation, antioxidant, and antitumor. However, few studies have considered hesperetin's neuroprotective effects and mechanisms; thus, our study investigated this in hydrogen peroxide (H2O2)-treated SH-SY5Y cells. MATERIALS/METHODS: SH-SY5Y cells were treated with H2O2 (400 µM) in hesperetin absence or presence (10-40 µM) for 24 h. Three-(4,5-Dimethyl-2-thiazolyl)-2,5-diphenyl-2H-tetrazolium bromide assays detected cell viability, and 4',6-diamidino-2-phenylindole staining allowed us to observe nuclear morphology changes such as chromatin condensation and apoptotic nuclei. Reactive oxygen species (ROS) detection assays measured intracellular ROS production; Griess reaction assays assessed nitric oxide (NO) production. Western blotting and quantitative polymerase chain reactions quantified corresponding mRNA and proteins. RESULTS: Subsequent experiments utilized various non-toxic hesperetin concentrations, establishing that hesperetin notably decreased intracellular ROS and NO production in H2O2-treated SH-SY5Y cells (P < 0.05). Furthermore, hesperetin inhibited H2O2-induced inflammation-related gene expression, including interluekin-6, tumor necrosis factor-α, and nuclear factor kappa B (NF-κB) p65 activation. In addition, hesperetin inhibited NF-κB translocation into H2O2-treated SH-SY5Y cell nuclei and suppressed mitogen-activated protein kinase protein expression, an essential apoptotic cell death regulator. Various apoptosis hallmarks, including shrinkage and nuclear condensation in H2O2-treated cells, were suppressed dose-dependently. Additionally, hesperetin treatment down-regulated Bax/Bcl-2 expression ratios and activated AMP-activated protein kinase-mammalian target of rapamycin autophagy pathways. CONCLUSION: These results substantiate that hesperetin activates autophagy and inhibits apoptosis and inflammation. Hesperetin is a potentially potent dietary agent that reduces neurodegenerative disease onset, progression, and prevention.

자완 수추출물(水抽出物)이 $H_2O_2$에 의해 유도(誘導)된 PC-12 세포주(細胞株)의 세포사(細胞死)에 미치는 영향(影響) (Study of Apoptotic Effect on Hydrogen Peroxide-induced Rat PC-12 Cells by Aster tataricus Water Extract)

  • Objective : Alzheimer's disease (AD) is a geriatric dementia that is widespread in old age. With an aging populace, AD is a looming problem in public health service. Alzheimer's disease is characterized by specific neuronal degeneration in certain areas of the brain. Mutations and abnormal expression of several genes are associated with ${\beta}-amyloid$ deposits and Alzheimer's disease; among them APP, PS1, and PS2, SOD, free radical, ROS. Methods:We studied herbal medicines that have a relationship to brain degeneration. From pre-modern times, although a variety of oriental prescriptions of Aster tataricus have been traditionally utilized for the treatment of AD, their pharmacological effects and action mechanisms have not yet been fully elucidated. Result : Based on morphological observations by phase-contrast microscope, TUNEL assay and MTT in the culture media, $H_20_2-induced$ cell death was significantly inhibited by Aticus. We examined by ROS formation, catalase activity and GSH activity. We studied the protective effect and inhibitory effects of neurotoxicity in $H_20_2-induced$ PC-12 cells by Aticus. Findings from our experiments show that Aticus inhibits apoptosis, which has neurotoxicities and cell damage in PC-12 cells. In addition, treatment with Aticus ($>25{\mu}g/ml$ for 6hrs) partially prevented $H_20_2-induced$ cytotoxicity in PC-12 cells, and induced a protective effect. Conclusion : As the result of this study, in the Aticus group, the apoptosis in the nervous system was inhibited, protected against the degeneration of PC-12 cells by $H_20_2$. Taken together, Aticus exhibited inhibition of $H_20_2-induced$ apoptotic cell death. Aticus was found to induce protective effect on GSH and catalase in PC-12 cells. Based on these findings, Aticus may be beneficial for the treatment of AD.

생약추출물의 acetylcholinesterase 저해, 항산화 및 신경세포보호 효과 in vitro 탐색 (In vitro screening of the acetylcholinesterase inhibition, antioxidant activity, and neuronal cell protective effect of medicinal plant extracts)

  • 본 연구에서는 20종 생약 열수추출물의 AChE 활성, 산화스트레스로 인한 지질 과산화물 생성 억제능 및 뇌신경세포 사멸에 대한 보호효과를 비교하였다. AChE의 억제활성은 산수유, 감초, 당귀 열수추출물에서 우수하였으며, 그 중 산수유가 가장 높은 억제율을 나타내었다. 또한 $H_2O_2/FeSO_4$로 산화 스트레스를 유발시켜 생약 열수추출물의 지질과산화물 생성 억제 활성을 조사한 결과 소엽, 하수오, 계피 및 감초 열수추출물에서 높은 항산화 활성을 보였다. L-Glutamate에 의해 유도된 신경세포 독성에 대한 보호효과는 감초, 계피, 길경, 박하 열수추출물의 $100{\mu}g/mL$ 농도에서 관찰되었다. 본 연구를 종합적으로 살펴보았을 때, 감초 열수추출물이 치매 예방 및 개선제로써의 활용가능성이 가장 뛰어난 것으로 판단된다. 향후 치매예방 효능을 가지는 새로운 화합물 발굴을 위한 기초자료로 활용될 것으로 사료된다.

산화적 스트레스에 대한 여주 (Momordica charantia) 추출물의 항산화 효과 및 세포사멸 억제 기전을 통한 신경세포보호효과 (Neuroprotective effects of Momordica charantia extract against hydrogen peroxide-induced cytotoxicity in human neuroblastoma SK-N-MC cells)

  • 건 여주로부터 얻은 70%에탄올 추출물의 항산화 효과를 측정하고, $H_2O_2$에 의해 유도된 산화적 스트레스에 대한 신경세포 보호효과를 알아보기 위해 human neuroblastoma cell인 SK-N-MC세포를 이용하여 실험을 수행하였다. 여주 추출물의 총 폴리페놀과 플라보노이드 함량은 각각 28.51 mg gallic acid/extract g과 3.95 mg catechin/extract g 이었고, 추출물의 DPPH 라디칼 소거능 ($IC_{50}$)은 $506.95{\mu}g/ml$ 이었다. 여주추출물을 신경세포에 전 처리한 후 $H_2O_2$을 처리하여 산화적 스트레스를 유도했을 때, 여주추출물에 의해 세포생존율은 증가되었고 세포내 ROS는 감소되는 것을 확인하였다. 그리고 세포내 항산화 방어시스템인 항산화효소 (SOD-1,2와 GPx-1)의 mRNA 발현이 여주추출물 처리에 의해 control 수준으로 회복되거나 control 보다 증가되는 결과를 보였으며, ROS 의존적 세포사멸과 연관 있는 것으로 알려진 MAPK pathway 중 p38과 JNK의 인산화를 여주추출물이 억제하였다. 또한 cleaved caspase-3와 cleaved PARP의 발현도 여주추출물의 처리에 의해 감소되었다. 본 연구 결과에서 70% 에탄올 여주추출물은 항산화효능이 우수하여 ROS를 직접적으로 제거할 뿐 아니라 세포내 ROS 축적을 억제시키는 효과를 보여주었다. 그리고 신경세포 내 항산화효소들의 발현 증가 기전과 p38, JNK의 인산화 억제 및 cleaved caspase-3, cleaved PARP의 발현 억제를 통한 세포사멸 억제 기전을 통해 산화적 스트레스로부터 신경세포를 보호하는 효과가 있음을 제시하고 있다. 따라서 여주추출물은 산화적 스트레스에 의한 알츠하이머병이나 파킨슨병 등과 같은 신경변성질환 (neurodegenerative disease)에 대한 예방 및 치료제의 소재로써 이용가치가 충분한 것으로 사료된다.

Identification of Differentially Expressed Genes in Murine Hippocampus by Modulation of Nitric Oxide in Kainic Acid-induced Neurotoxic Animal Model

  • Kainic acid (KA) causes neurodegeneration, but no consensus has been reached concerning its mechanism. Nitric oxide may be a regulator of the mechanism. We identified differentially expressed genes in the hippocampus of mice treated with kainic acid, together with or without L-NAME, a nonselective nitric oxide synthase inhibitor, using a new differential display PCR method based on annealing control primers. Eight genes were identified, including clathrin light polypeptide, TATA element modulatory factor 1, neurexin III, ND4, ATPase, $H^+$ transporting, V1 subunit E isoform 1, and N-myc downstream regulated gene 2. Although the functions of these genes and their products remain to be determined, their identification provides insight into the molecular mechanism(s) involved in KA-induced neuronal cell death in the hippocampal CA3 area.

Tat-indoleamine 2,3-dioxygenase 1 elicits neuroprotective effects on ischemic injury

  • It is well known that oxidative stress participates in neuronal cell death caused production of reactive oxygen species (ROS). The increased ROS is a major contributor to the development of ischemic injury. Indoleamine 2,3-dioxygenase 1 (IDO-1) is involved in the kynurenine pathway in tryptophan metabolism and plays a role as an anti-oxidant. However, whether IDO-1 would inhibit hippocampal cell death is poorly known. Therefore, we explored the effects of cell permeable Tat-IDO-1 protein against oxidative stress-induced HT-22 cells and in a cerebral ischemia/reperfusion injury model. Transduced Tat-IDO-1 reduced cell death, ROS production, and DNA fragmentation and inhibited mitogen-activated protein kinases (MAPKs) activation in H2O2 exposed HT-22 cells. In the cerebral ischemia/reperfusion injury model, Tat-IDO-1 transduced into the brain and passing by means of the blood-brain barrier (BBB) significantly prevented hippocampal neuronal cell death. These results suggest that Tat-IDO-1 may present an alternative strategy to improve from the ischemic injury.

SK-N-SH 신경세포내 항산화 효과와 p38 인산화 억제에 의한 곤드레, 누룩치 그리고 산마늘의 신경 보호 효과 (Neuroprotective Effects of Cirsium setidens, Pleurospermum kamtschaticumin, and Allium victorials Based on Antioxidant and p38 Phosphorylation Inhibitory Activities in SK-N-SH Neuronal Cells)

  • 신경계 질환은 산화적 스트레스에 의한 신경세포 손상에 의해 발생하는 것이 하나의 기전으로 알려져 있다. 본 연구는 $H_2O_2$에 의해 유도된 산화적 스트레스에 대항하여 곤드레(Cirsium setidens, CS), 누룩치(Pleurospermum kamtschaticumin, PK) 그리고 산마늘(Allium victorials, AV)의 뇌신경 보호 효과 및 그 기전에 대한 것이다. CS와 AV 처리는 대조군과 비교하여 $400{\mu}g/mL$까지 인간의 신경세포주인 SK-N-SH 세포에 대해 세포독성이 없었다. 산화적 유도자인 $H_2O_2$를 SK-N-SH 세포에 처리하였을 때 세포사멸 및 활성산소종(ROS) 생산이 현저하게 증가하였으나 CS 또는 AV 처리에 의해 산화적 스트레스에 의해 증가된 세포사멸과 ROS 생산이 현저하게 감소하였다. 실험한 산채 중에 CS와 PK가 AV보다 더 강한 DPPH 라디칼 소거 작용이 있었으나 PK는 대조군과 비교하여 SK-N-SH 세포를 사멸시키는 강한 세포독성을 가지고 있었다. CS는 AV보다 산화적 스트레스에 대항하여 세포사멸 및 ROS 생성에 더 높은 저해적 영향력을 보여주었다. 따라서 계속되는 실험에는 CS를 사용하였다. CS의 순차적 용매 분획물들인 헥산, 클로로포름, 에틸아세테이트, 부탄올 및 물 분획물들(CS-HE, CS-CH, CS-EA, CS-BU, CS-AQ)은 산화적 스트레스에 대항하여 SK-N-SH 세포사멸 및 세포내 ROS 생성을 억제하였다. CS-EA는 5개의 분획물들 중 가장 강한 DPPH 라디칼 소거작용 및 세포내 ROS 소거 활성을 가지고 있었고, 가장 강한 뇌신경세포 보호 효과를 가지고 있었다. CS-EA는 산화적 스트레스에 의해 증가된 세포자멸사(apoptosis)의 신호전달 경로에 관여하는 p38의 인산화를 저해함으로써 활성화되는 것을 약화시켰다. 이 결과들은 CS-EA가 뇌신경세포에서 항산화 효과 및 p38 인산화 억제에 의한 뇌신경 보호 효과를 나타낼 것이라 제안하였다.
