Effects of Gibberellin Application and Bagging on Ripening and Quality in 'Delaware' Grape Berries (Delaware 포도에서 Gibberellin 처리와 봉지씌우기가 과립의 성숙과 품질에 미치는 영향)

  • This experiment was carried out to clarify the effects of gibberellin(GA) aplication and bagging on repeening and quality in 'Delaware' grape berries. Treatments are 4 plots(2X2 factorial experiment); GA, GA+bagging, bagging and control. The clusters were dipped twice in 100 ppm GA with GA treatment : 10 days before and after the full bloom. The results obtained as follows: 1. GA treatment made the seedless grape berry reduced in the fresh weight but it hastened the ripening period about 2 weeks. 2. Total soluble solid(TSS), viscosity and pH value of berry juice increased with maturation. The concentration of TSS and viscosity were higher in GA treatment plot than GA non-treatment. 3. Berry-hardness, titratable acidity and alcohol inslouble solid(AIS) decreased with maturation. Expically berry-hardness and AIS decreased more greatly in GA non-treatment than GA treatment. 4. The concentration of anthocyanin increased with ripening but pectic substance didn't fluctuate nearly. These of anthocyanin and pectin were higher in GA non-treatment plot than GA treatment. 5. By analysis of factorial experiment GA treatment was highly significant with the $^{o}$Brix/Acidity ratio, juice viscosity and AIS, but high negatively, significant with berry-hardness and berry fresh weight. And it was significant with T S S and negatively, titratable acidity. Bagging was significant with $^{o}$Brix/Acidity ratio and AIS content, but negatively, titratable acidity. 6. Qualitative characters were high correlated with the $^{o}$Brix/Acidity ratio in simple correlation but direct effect by the path-coefficient analysis didn't coincide with simple correlation. The direct effect of pH was large and juice viscosity, the next. And that of berry-hardness was negligible but, AIS, small negatively.

Estimation of Volume Change and Fluid-Rock Ratio of Gouges in Quaternary Faults, the Eastern Blocks of the Ulsan Fault, Korea (울산단층 동부지역 제4기단층 비지대의 체적변화와 유체-암석비에 대한 고찰)

  • Many Quaternary faults are recognized as thin gouge and narrow cataclastic zone juxtaposing the Bulguksa granite and Quaternary deposit bed in the eastern block of the Using Fault, Korea: Gaegok 1, Caegok 2, Singye, Madong Wonwonsa and Jinhyeon faults. This study was performed to calculate chemical change, volume change, silica loss and fluid-rock ratio taken place in gouge zones of these Quaternary faults using XRF, XRD, EPMA. The chemical compositions of fault rocks reveal that the fault gouges are depleted in $SiO_2,\;Na_2\;O,and\;K_2O$ and enriched in $Al_2O_3,\;Fe_2O_3,\;P_2O_5,\;MgO,\;MnO,\;CaO,\;and\;LOI(H_2O+CO_2)$ relative to protoliths. The fact that there is enrichment of relatively immobile elements and depletion of the more soluble elements in the fault gouges relative to protoliths can be explained by fluid-assisted volume loss of $56\%$ for Caegok 1 fault, $22\%$ for Caegok 2 fault,$34\%$, for Singye fault, $8\%$ for Madong fault, $2\%$ for the Wonwonsa fault and $53\%$ for the linhyeon fault. Madong fault and Wonwonsa fault where ratios of the volume change, silica loss and fluid-rock are low might have acted as a closed system for fluid activity, whereas Caegok 1 fault and Jinhyeon fault with high ratios in those factors be an open system. The volumetric fluid-rock ratios range $10^2\sim10^4$ for all faults, being highest in Caegok 1 fault and Jinhyeon fault whose fluid activity was most significant.

  • In the oligomerization of p-aminophenol under the catalytic action of the metallic complexes, the effects of the ligands are studied. When the initial velocity of $O_2$ uptake at pH 8 using Fe(Ⅲ) as the central metal and N-hydroxylethylethylenediaminetriacetic acid (HEDTA), ethylenediaminetetraacetic acid (EDTA), diethylenetriaminepentaacetic acid (DTPA), 1,2-cyclohexanediaminetetraacetic acid(CyDTA) as the ligands respectively are compared, the velocities are as the following order: HEDTA > EDTA > DTPA > CyDTA. Further when the effect of the ligands, nitrilotriacetic acid (NTA), HEDTA, EDTA, and DTPA, on the yields of oligomers are compared, the result shows as the following order: NTA > HEDTA > EDTA > DTPA. These are nearly reverse order of the stability constants of the complexes. In order to determine the composition of the mixed complexes at the initial step, the method of continuous variation is used, and it is found that the composition ratio of Fe-EDTA complex to monomer in the mixed complexes is one at pH 5-8 range. It is also found that at pH 9 or in the more alkaline range, side reactions occur to form water soluble dimer of quinone type and the catalytic action of the metallic complex markedly decreases on account of the hydrolysis of the central metal by the $OH^-$ ion.

Changes in Hydration and Watertightness of Cement Containing Two-Component Fluosilicate Salt Based Chemical Admixture (2성분 규불화염계 혼화제가 첨가된 시멘트의 수화반응 및 수밀성 변화)

  • Fluosilicic acid ($H_2SiF_6$) is recovered as aqueous solution which absorbs $SiF_4$ produced from the manufacturing of industrial-graded $H_3PO_4$ or HF. Generally, fluosilicate salts prepared by the reaction between $H_2SiF_6$ and metal salts. Addition of fluosilicate salts to cement endows odd properties through unique chemical reaction with the fresh and hardened cement. In this study, two-component fluosilicate salt based chemical admixtures (MZ) of $4\%,\;6\%$, and $8\%$ concentration were prepared by the reaction of $H_2SiF_6$ ($25\pm2\%$) and metal salts. The effect of concentration of MZ at a constant adding ratio on the hydration and watertightness of cement were investigated respectively. In a cement containing MZ, metal fluorides such as $CaF_2$ and soluble silica by hydrolysis were newly formed during hydration. The total porosity of the hardened cement was lower in the presence of U because of packing role of metal fluoride and pozzolanic reaction of soluble $SiO_2$. Consequently, the watertightness of the hardened paste containing MZ was more improved than non-added (plain) due to an odd hydration between cement and MZ.

  • The Aster tataricus is a chinese traditional medicine called “Ziwan” which has an expectorative and remediable cough action. The anti-oxidant activities of A. tataricus were investigated. The MeOH extract of A. tataricus showed strong anti-oxidant activity in the NBT(nitroblue tetrazolium) method system, and thus fractionated with several solvents in to the EtOAc, n-BuOH, CH$_2$Cl$_2$, H$_2$O fraction. The EtOAc soluble fraction exhibiting strong anti-oxidant activity was further purified by repeated silica gel and sephadex LH-20 column chomatography. (omitted)

  • We examined the effects of four different types of cutting on the quality characteristics of sweet pumpkin. Two hundred grams of each of four samples were packed individually in polypropylene mm and stored at $10^{\circ}C$ for 9 days. Samples were evaluated for weight loss, change in hardness, color change, pH change, water-soluble materials, gas changes, and sensory evaluation. $CO_2$concentration increased during storage, whereas $O_2$ concentration rapidly decreased and then stabilized after 3 days. $C_2H_4$ was detected only after 3 days of storage, and steadily increased thereafter. The rate of weight loss steadily increased Analysis of Lab color space indicated no significant change in the L and b values, but an increase in the a value at the end of storage. Waster-soluble solids increased, but hardness showed no change. All the samples underwent a steady increase in pH. Samples cut into 8 pieces had the highest sensory scores.

Effect of Fly Ash on the Yield of Chinese Cabbage and Chemical Properties of Soil (석탄회(石炭灰)(Fly ash) 시용(施用)이 배추의 수량(收量) 및 토양특성(土壤特性)에 미치는 영향(影響))

  • The effect of Bituminous and Anthracite coal ash(fly ash) on the performance of Chinese cabbage on an acid soil was studied through a pot experiment. The levels of application of the materials tested were five, 10 and 15% of dry soil weight. Regardless of the kind of fly ash, the application of it, tended to increase the yield of Chinese cabbage by 13 to 24% in fresh weight. Difference in application levels did not result in the difference in increasing the yield of Chinese cabbage. The application of fly ash tended to lower the all of the mineral nutrient contents in the Chinese cabbage, excepting boron. Boron content tended to increase along with the application of fly ash. Bituminous ash raised the pH of soil and increased available P, exchangeable Ca and soluble boron in the soil remarkably. Anthracite ash, on the other hand, did not increase the contents of other components in the soil, than soluble born.

Quality Changes in Delaware Grapes Treated with Chitosan during Storage (키토산 처리된 델라웨어 포도의 저장 중 품질 변화)

  • To maintain quality of Delaware grapes during storage, grape samples were treated with 0.1% chitosan dissolved in 0.5% acetic acid, packaged with low density polyethylene film, and stored at 4 or $20^{\circ}C$ for 12 days. Chitosan treatment reduced initial populations of yeast and molds in grapes by 1.86 log CFU/g compared to that of the control. During storage, oxygen contents in packages of samples decreased, whereas carbon dioxide contents increased. In addition, regardless of storage temperature, changes in oxygen and carbon dioxide concentrations of grapes treated with chitosan were lower than those of the control. Hardness of samples decreased, and Hunter L, a, and b values were not significantly different among treatments. Regarding pH and total soluble content, grapes stored at $4^{\circ}C$ maintained pH and had greater total soluble content than those stored at $20^{\circ}C$. These results suggest that chitosan treatment and low temperature storage can be useful for maintaining microbiological safety and quality of Delaware grapes during storage.

  • Histamine-releasing factors (HRFs) are soluble mediators that can release histamine and other mediators from basophils and mast cells and their activity can vary, depending on the type of IgE. The activity of HRFs is affected by the presence of IgE, although HRF is thought to bind to a specific receptor other than IgE. Until now, HRF signaling pathway including its receptor remains unclear in spite of numerous studies. Since there had been many reports about reactive oxygen species (ROS) as a signaling molecule rather than as a by-product of metabolism, we investigated the possibility of ROS as an intracellular messenger involved in HRF-mediated histamine degranulation. In RBL-2H3 cells, ROS was generated by HRF using $H_2O_2$-sensitive fluorescence of fluorescent 2', 7'-dichlorofluorescein ($H_2DCFDA$). These effects were blocked by anti-oxidant N-acetylcysteine (NAC). These results suggest that ROS generation could play a role as an intracellular messenger in histamine release by HRF.

Fabrication of Lipid Sensor Utilizing Photosensitive Water Soluble Polymer (감광성 수용성 고분자를 이용한 Lipid 센서의 제조)

  • A FET(field effect transistor) type lipid sensor was fabricated uy immobilizing lipase enzyme on the gate of pH-ISFET($SiO_{2}/Si_{3}N_{4}$). A water soluble polymer, polyvinyl alcohol(PVA) was modified with 1-methyl-4-(formyl-styryl) pyridinium methosulfate(SbQ) to give a photosensitive membrane(PVA-SbQ) in which lipase was immobilized. The optimum photolithographic conditions were ; spin coating speed $5,000{\sim}6,000$ rpm. UV exposure time $20{\sim}30$ seconds, developing time in water $30{\sim}40$ seconds, and vacuum drying time 45 min. at room temperature with the suspension containing PVA-SbQ aqueous solution(SbQ 1mol%, 10 wt %) $200{\mu}L$, bovine serum albumin (BSA) 7.5 mg, and lipase 10 mg. The lipid sensor showed good linear calibration curve in the range of $10{\sim}100$ mM triacetin as a lipid sample.
