Effect of Italian Ryegrass(Lolium multiflorum) and Kentucky Bluegrass(Poa pratensis) Planted Singly or in Combination with Other Forage Species on Suppressing Weeds (단파 또는 혼파초지에서 Italian ryegrass와 Kentucky bluegrass의 잡초억제 효과)

  • A field plot experiment was carried out on mixed pasture from October 1995 to October 1997 to examine the effect of weed control as affected by the difference of mixed pasture species, planning with spilt plot design which main plots of 2 treatments (companion crop Italian ryegrass Lolium multiflorum and Kentucky bluegrass Poa pratensis) are divided into subplots of 4 treatments (T1: single companion crop, T2: Tall fescue Festuca arundinasea + Orchardgrass Dactylis glomerata + Ladino clover Trifolium repens, T3 : Perennial ryegrass Lolium perenne + orchardgrass + ladino clover, T4 : orchardgrass + ladino clover). Average 2 year-dry matter yields of companion crops as whole plots, Italian ryegrass and Kentucky bluegrass, were 15,437kg/ha and 14,629kg/ha, respectively and those of T1, T2, T3 and T4 as sub plots were 14,815, 15,407, 14,629 and 15,187kg/ha respectively without statistical1y significant difference(P>0.05). Though there was no significant difference between the dry matter yields of Italian ryegrass and Kentucky bluegrass, as whole plots, at 1st harvest of second year, dry matter yields of T3 and T4, as 4,591kg/ha and 4,816kg/ha respectively, were higher than those of T1, 3,376kg/ha(P<0.05), significantly. There was a significant difference among 2nd year-average DM yields of sub plots(P<0.05), showing highest with T4 15,131kg/ha and least with T1 13,085kg/ha and T3 13,269kg/ha. Italian ryegrass, as the companion crop, was better for the control of weeds in the early spring than Kentucky bluegrass, but reached to the similar botanical composition level with Kentucky bluegrass after summer with rapid decrease of Italian ryegrass. Percentage of Kentucky bluegrass in botanical composition increased slowly, but did'nt become an important factor for the control of weeds at the end of this experiment. Using orchardgrass in mixed pasture such as T2, T3 and T4 made weeds more decreased in mixed pasture than one in single conpanion crop. There was no difference among treatments of whole or sub plots in crude protein contents of pasture taken in this study. Contents of NDF and ADF of companion crop. Italian ryegrass plots harvested in October 1997 were hither than those of Kentucky bluegrass plots(P<0.05). ADF contents of Kentucky blue grass-plots taken in July 1997 has shown to be high significantly as compared to those of Italian ryegrass plots(P<0.01). The results of this study support that the companion crop Italian ryegrass was good for weed controls only in spring of 1st year after sowing and orchardgrass in mixed pastures played good roles for increase of dry matter yield and weed control.

Efficacy of Flaxseed Flour as Bind Enhancing Agent on the Quality of Extended Restructured Mutton Chops

  • Consumers have become very conscious about their nutrition and well being due to changes in their socio-economic lifestyle and rapid urbanization. Therefore, development of technology for production of low cost and functional meat products is urgently required. One such approach is innovative restructuring technology in which binding of meat pieces still remains the main challenge and extension of product is generally associated with poor binding and texture. Thus, the present study was envisaged as an attempt to solve this problem by the incorporation of flaxseed flour (FF) as bind enhancing agent. The FF was used at three different levels viz., 0.5%, 1%, and 1.5% to replace lean meat in pre-standardized restructured mutton chops formulation. The products were subjected to analysis for physico-chemical, sensory and textural properties. Cooking yield, moisture percentage and fat percentage increased with increase in the level of incorporation of FF, however, protein percent and pH decreased with increase in the level of incorporation. Shear force value of product incorporated with 1.5% FF was significantly higher (p<0.01) than control and product containing 0.5% FF level. Among the sensory attributes, product with 1% flaxseed flour showed significantly higher values (p<0.05) for general appearance, binding, texture and overall acceptability. Hardness showed significant increasing (p<0.01) values with increasing levels of incorporation of flaxseed flour, however all other parameters of texture profile analysis showed a decreasing trend. On the basis of sensory scores and physico-chemical properties, the optimum incorporation level of FF was adjudged as 1%. Products incorporated with optimum level of flaxseed flour (1%) were also assessed for water activity and microbiological quality during the storage period of 15 days. It was found that the extended restructured product could be safely stored under refrigeration ($4^{\circ}C{\pm}1^{\circ}C$) in low density polyethylene (LDPE) pouches for 15 days without marked deterioration in sensory and microbiological quality. Thus, it was concluded that flaxseed flour can be used as a good bind enhancing agent in extended restructured meat products at an economic cost.

Preparation of High Quality Grape Seed Oil by Solvent Extraction and Chemical Refining Process (용매추출 및 화학적 정제법에 의한 고품질의 포도씨유의 제조)

  • This study was conducted to prepare high quality grape seed oils by solvent extraction and chemical refining process. Additionally, quantitative analysis of several functional components in grope seed was carried out to compare quality characteristics of grape seeds from grapes grown by conventional and organic agricultural practices. There are no significant differences in several functional constituents of grape seeds between conventionally cultivated- and organically cultivated-grapes, although some functional compositions of grape seeds are different between two cultivation methods. The dried grape seed was pretreated with roasting heating for 5 min, milled and then extracted twice with n-hexane under reflux at $50^{\circ}C$ for overnight, followed by filtration and evaporation. The crude grape seed oil was successively purified by degumming with $0.1\%\;H_3PO_4$, deaciding with $20\%\;NaOH$, and then decoloring and deodorization by a steam distillation, and thereby producing purified grape seed oil(yield: $5.0\%/dried$ grape seed). Physicochemical characteristics of the purified grape seed oil were comparable to those of the imported grape seed oils.

Effects of Black Garlic on the Rheological and Functional Properties of Garlic Fermented by Leuconostoc mesenteroides (흑 마늘 첨가에 따른 마늘 젖산 발효물의 물성 및 기능성평가)

  • White and black garlic pastes were fermented by Leuconostoc mesenteroides and evaluated for its rheological and functional properties such as consistency, antioxidant and angiotensin converting enzyme inhibitory activity. The pH, acidity and solid content of black garlic paste were 4.60, 1.23%, 22.63%, respectively. The viable cell counts and consistency of fermented garlic was decreased by adding higher amounts of black garlic paste. Fermentation of white garlic (40%)/black garlic (10%) showed viable cell counts of $1.6\times10^{11}$, fluid consistency of 9.31 $Pa{\cdot}s^n$. Water and 70% ethanol extract from fermented garlic showed the polyphenol content of 6.29mg/mL and 5.99 mg/mL, respectively. Also, water extract indicated the DPPH radical scavenging effects and angiotensin converting enzyme (ACE) inhibitory activity with $IC_{50}$ 1.03 mg/mL and $IC_{50}$ 68.54 mg/mL, respectively. ACE inhibitory activity was increased with adding black garlic paste. Conversion of sucrose into dextran polymer in fermented garlic was drastically decreased by the addition of black garlic paste, indicating from 85% (0% black garlic) to 20% (20% black garlic) conversion yield. Garlic paste fermented with 10% black/40% white garlic showed the decrease in consistency and viable cell counts during both cold and freezing storages. In particular, consistency of fermented garlic was lower during freezing storage than cold storage, and the viable cell counts was drastically decreased after storage for 2 weeks.

Study on Control and Tuberization Inhibition of Eleocharis kuroguwai Ohwi (올방개의 괴경형성(塊莖形成) 억제(抑制) 및 방제(防除)에 관(關)한 연구(硏究))

  • The experiment was conducted to evaluate weeding effect and to inhibit tuberization of Eleocharis kuroguwai authority by single or systematic applications of several herbicides. E. kuroguwai was controlled above 90 percentage by pretilachlor and butachlor+bensulfuron application at 5-10 DAT(Days after transplanting) followed by bentazon at 50 DAT and weeded 88 percentage by mefenacet+bensulfuron application at 15 DAT. Tuberization inhibition rate of E. kuroguwai was higher at the application of butachlor+bensulfuron at 5 DAT followed by bentazon at 50 DAT than any other applications. The weight variation of E. kuroguwai tubers formed at 15-20cm soil stratum was larger than that at 10-15cm. Tuber of E. kuroguwai was hardly formed at top soil stratum (0-5cm) and deep soil (15-20cm) by the systematic application of bentazon at 50 DAT. Grain yield of rice from systematic application with bentazon at 50 DAT and mefenacet+bensulfuron at 10-15 DAT was similar to that of hand weeded plot.

Growth, Behavior, and Carcass Traits of Fattening Hanwoo (Korean Native Cattle) Steers Managed in Different Group Sizes

  • This study was conducted to evaluate growth, behavior and carcass traits of fattening Hanwoo (Korean native cattle) steers managed in different group sizes. A total of 48 animals, 6 months of age, were allocated to one of three group sizes and the experiment was conducted from 12 to 30 months of age. Groups were balanced for weight and the group sizes consisted of 4, 8, or 12 steers which were named 4sG, 8sG and 12sG, respectively. When animals were 12 months of age, initial fasted body weight (BW, $304.51{\pm}12.40\;kg$) was measured. All animals were housed at a constant space allowance of $8.82\;m^2$ per animal, and a feeder and drinker were provided per 4 animals. The whole fattening stage was divided into three phases: phase I (from 12 to 18 month of age), phase II (from 19 to 24 month of age), and phase III (from 25 to 30 month of age). Steers managed in 12sG showed low (p<0.05) growth rate and feed conversion rate (FCR) in phase I and phase II when compared to other treatment groups. However, this difference was not observed for the whole fattening phase (p>0.05). Steers managed in 4sG had a thick (p<0.05) ultrasound back fat thickness at 15 and 18 months of age. However, group size had no effect on meat yield and quality traits of area and marbling score. Animals managed in 8sG yielded a better meat grade of "A" than the "B" grade in other treatment group sizes. Lean color, fat color, firmness and maturity scores did not differ among group sizes. Hanwoo steers housed under 12sG spent less time on eating concentrate, relevant higher eating rate, less frequency of allogrooming, and more time on walking (p<0.05). It could be concluded that a large group size retarded growth rate and back fat thickness in the fattening stage, which was mainly focused on 15 and 18 months of age.

Study of four week repeated dose toxic test of Sweet Bee Venom in Beagle Dogs (Sweet Bee Venom의 비글견을 이용한 4주 반복 근육시술 독성시험)

  • Objectives: This study was performed to analyse four week repeated dose toxicity of Sweet Bee Venom(Sweet BV) extracted from the bee venom in Beagle dogs. Methods: All experiments were conducted under the regulations of Good Laboratory Practice (GLP) at Biotoxtech Company, a non-clinical study authorized institution. Male and female Beagle dogs of 5-6 months old were chosen for the pilot study of four week repeated dose toxicity of Sweet BV which was administered at the level of 0.56mg/kg body weight which is eighty times higher than the clinical application dosage as the high dosage, followed by 0.28 and 0.14mg/kg as midium and low dosage, respectively. Equal amount of excipient(normal saline) to the Sweet BV experiment groups was administered as the control group every day for four weeks. Results: 1. No mortality was witnessed in all of the experiment groups. 2. All experiment groups were appealed pain sense in the treating time compared to the control group, and hyperemia and movement disorder were observed around the area of administration in all experiment groups, and higher occurrence in the higher dosage treatment. 3. For weight measurement, Neither male nor female groups showed significant changes. 4. In the urine analysis, CBC and biochemistry didn't show any significant changes in the experiment groups compared with control group. 5. For weight measurement of organs, experiment groups didn't show any significant changes compared with control group. 6. To verify abnormalities of organs and tissues, thigh muscle which treated with Sweet BV, cerebrum, liver, lung, kidney, and spinal cords were removed and conducted histologocal observation with H-E staining. In the histologocal observation of thigh muscle, cell infiltration, inflammatory, degeneration, necrosis of muscle fiber, and fibrosis were found in both thigh tissue. And the changes were depend on the dose of Sweet BV. But another organs were not detected in any abnormalities. 7. The proper high dosage of Sweet BV for the thirteen week repeated test in Beagle dogs may be 0.28mg/kg in one time. Conclusion: Above findings suggest that Sweet BV is relatively safe treatment medium. Further studies on the subject should be conducted to yield more concrete evidences.

Improvement of Transglycosylation Efficiency using a Glycosynthase Mutant derived from Thermoplasma acidophilum ${\alpha}$-Glucosidase (Thermoplasma acidophilum 유래 ${\alpha}$-glucosidase로 부터 생산된 glycosynthase 돌연변이 단백질의 개선된 당전이 효율)

  • Glycosynthase is an active site nucleophile mutant enzyme, prepared from glycosidase, which is capable of synthesizing oligosaccharide derivatives without the hydrolysis of the product. Thermoacidophilic ${\alpha}$-glucosidase of Thermoplasma acidophilum (AglA) exhibits a transglycosylating activity yielding various glycosides. AglA was converted to glycosynthase by the substitution of the catalytic nucleophile Asp-408 residue into non-nucleophile glycine in order to increase its ability to synthesize various glycosides by transglycosylation. The glycosynthase mutant was purified by Ni-NTA chromatography and its glycoside-synthesizing activity was measured by using an external nucleophile, sodium formate buffer, providing maltose as a donor and p-nitrophenyl-${\alpha}$-D-glucopyranoside ($pNP{\alpha}G$) as an acceptor, respectively. In addition, $pNP{\alpha}G$ was examined for its feasibility to act as both a donor and an acceptor, and products were compared with those of the wildtype enzyme. The mutant enzyme was found to catalyze the formation of a specific product from $pNP{\alpha}G$ with a yield of 42.5% without further hydrolysis, while the wild-type enzyme produced two $pNP{\alpha}G$ products at low yields. The results demonstrate the possibility of satisfactory yields for the reactions in the presence of small amounts of acceptor, and demonstrate that the high activity of the mutant, at pHs below neutrality, was applicable in the transfer of glucose from the natural donor.

Study on Nutrient Balance in Paddy Field of Fluvio-Marine Deposit (하해혼성(河海混成) 논토양(土壤)의 양분수지(養分收支)에 관한 연구)

  • To test for the effect of applied fertilizer and nutrients on uptake and transport for paddy rice, two paddy field trials were conducted with Dongjinbyeo in degenerated salt paddy field of Jeonbuk series from 1999 to 2000. After experiment, soil acidity, content of organic matter phosphate, silicate, potassium, calcium, and total nitrogen was increased by application of fresh cattle manure(FCM). Content of Nitrogen in soil layer leached inorganic nitrogen $NO_3$ was higher that that of $NH_4$ and was high in treatment of FCM. Content of $PO_4$ was higher in FCM than other treatments. But content of potassium was in high control. During the growth of rice plant, the amount of water consumption was 477mm. The amount of supplied nitrogen was high in treatment of no nitrogen(NN), 20% reduced application of LCU(LCU-20%), and no fertilizer. In case of phosphate, the supplied amount was more than the consumed amount with the exception of treatment "no phosphate(NP) and no fertilizer(NF)". In case of potassium, the consumed amount was more than the supplied amount in all treatments. The amount of applied nitrogen in the nutrient infiltrated water was high in treatment soil test(ST), C+FCM+Si(Silicate) and the ratio of recovered nitrogen was high in 20% reduced application of LCU. The amount of applied phosphate in the nutrient infiltrated water was high in FCM and that of applied potassium was high in 20% reduced application of LCU. Nitrogen use efficiency of paddy rice was high in 20% reduced application of LCU and use efficiency of phosphate and potassium was high in C+Si(Silicate). Grain yield of rice was high in order of 20% reduced application of LCU>C+Si=C+FCM+Si>C+FCM.

Establishment of 60 Mesh Nets to Reduce Crop Loss by Bemisia tabaci (Gennadius) (Homoptera: Aleyrodidae) in Tomato Greenhouse (토마토 온실에서 방충망 설치에 의한 담배가루이 피해 경감)

  • Investigations were conducted to prevent entrance of Bemisia tabaci (Gennadius) with very fine mesh nets. B. tabaci vectors tomato yellow leaf curl virus in tomatoes. B. tabaci could be completely prevented to penetrate into an 83 mesh net, though few of them passed the 60 mesh net. An independent 60 mesh net cage was built within one of the three interconnected common plastic greenhouse ($21.0{\times}18.3m$) in order to find its usefulness as a crop protection tool from insect pests. TYLCV infected tomato plants were not detected, though a few eggs of B. tabaci and mines of Liriomyza trifolii (Burgess) were detected locally at the entrance of the sliding door. There were practically no significant differences between treatments of nets and the conventional plant height, leaf length and width, number of floret clusters, sugar brix, and product yield. In viewpoints of commercialization, it could be considered highly prospectful to apply to the nets to glasshouses or other greenhouses in which the growing period of crops is long, if the nets were used in the openings of sides, ceilings, and entrances of the greenhouse. However, it might be necessary to take preventive measures for occurrences of insect pests living in soil, temporarily or permanently, as well as high humidity diseases.
