방어 보통육과 혈합육의 단백질 및 아미노산조성의 사후변화 (Changes Occurred in Protein and Amino Acid Compositions during Postmortem Aging of White and Dark Muscle of Yellowtail at $2^{\circ}C$)

  • 방어의 배육 보통육과 측육 혈합육을 $2^{\circ}C$에 보장하여 두고 선도변화 총계별로 단백질의 조성과 각축의 구성 및 유리아미노산의 조성을 분석하였으며, 건향질단백질과 근원섬유단백질에 대하여는 구성단백질의 분포변화를 확인하기 위하여 $NaDodSO_4$변한 다음, polyacrylamide겔 전기영동상을 비교 해석함으로써 적색육어류의 선도변화와 보통육 및 혈합육의 단백질조성 변화와의 관련성을 검토하였다. K-값과 휘발성염기질소 및 pH값으로 판정했을 때, 혈합육은 보통육에 비하여 선도변화가 빨랐다. 사후 선도변화와 더불어 근원질단백질과 노원섬유단백질은 감소하는 반면, 세포내잔사단백질은 상대적으로 증가하였으며, 이러한 변화는 보통육에 비하여 혈합육이 훨씬 심한 편이었다. Sodium dodecyl sulfate 화한 단백질을 polyacrylamide겔 전기영동함으로 분석한 결과, 보통육의 근원질단백질은 16개 성분, 그리고 혈합육의 근원질단백질은 12개 성분으로 구성되어 있었으며, 보장 10일째부터 보통육의 근원질단백질은에는 41,000dalton과 18,000dalton의 양분이 조금씩 증가하였고, 26,000 dalton과 23,500 dalton 및 23,000 dalton되 각 성분은 점차 감소하였다. 혈합육의 근원질단백질에 있어서는 49,000 dalton의 성분은 감소하였고 47.000 dalton의 성분은 새로이 형성되었으며, 26,000 dalton의 성분은 소감하였다. 근원섬유단백질의 전기영동분석결과에 의하면, 보통육의 근원섬유단백질은 17개 성분, 그리고 혈합육의 근원조직단백질은 16개의 성분으로 구성되어 있었다. 보장 10일에부터 보통육의 근원섬유단백질중에는 40,000 dalton의 성분이 증가하였고, 37,500 dalton의 성분은 점차 감소하였으며, 32,000 dalton의 성분은 새로히 출현하였다. 혈합육의 근원섬유단백질은 보장 9일째부터 58,000 dalton과 64,000 dalton의 성분은 그 농도가 단가하였으며, 17,500의 light chain-2 단백질은 소감하였고, 32,000 dalton의 성분은 새로이 출현하였다. 이 같은 전기영동상의 변화는 근원질단백질이 근원섬유단백질에 비하여 빨랐으며, 근원섬유단백질의 경화에 있어서는 혈합육쪽이 보통육에 비하여 빨랐다. 양 육의 단백질을 구성하는 아미노산을 분석한 결과, 즉살한 방어의 보통육 단백질은 글루탐산, 아스팔트산, 류신. 알기닌, 알라닌 등은 그 양이 어느 정도 많았고, 프롤린과 트?토판은 그 양이 적었으며, 혈합육에 있어서도 비슷한 경형이었다. 그리고 10일이 경과한 보통육에 있어서는 글루탐산과 아스팔트산이 현저히 감소하였으며, 9일이 경과한 혈합육에 있어서는 알라닌, 글리신, 그리고 알기닌이 현저히 감소하였다. 유리아미노산의 조성을 분석 결과, 즉살한 방어의 보통육중에는 히스티딘이 총유리아미노산의 약 $63\%$를, 그리고 혈합육중에는 타우린이 총유리아미노산의 약 $67\%$를 차지하는 양을 보였다. 그리고 $2^{\circ}C$에서 10일이 경과했을 때, 보통육에서는 히스티딘, 발린, 타우린이 증가하였으며, 알라닌, 류신 및 글리신 등은 조금 감소하였다. 같은 온도에서 9일이 경과한 혈합육에서는 타우린, 페닐알라닌, 글리신은 증하였으며, 히스티딘, 알라닌, 세린 등은 감소하였다.

<사례보고> 농작업 환경개선을 위한 한국형 참여형 개선활동 교육(PAOT)의 개발과 실제 적용 사례 (<Field action report> Development and Application of Participatory Action Oriented Training(PAOT) for Improvement of Agricultural Working Environment in Korea)

  • 본 연구는 한국형 농업인 참여형 개선활동 기법(Participatory Action Oriented Training, PAOT) 교육 프로그램을 개발하고 적용함으로써 더 많은 지역으로 이 프로그램이 확산되는 데 기여하고자 시도하였다. 연구진은 농작업 환경 체크리스트를 노지, 과수, 시설, 축산, 복합농 등의 5개 범주로 구분하여 실제 우리나라 농작업 현장에 적합한 것으로 개발하였다. 체크리스트는 농작물 운반과 보관, 작업대와 연장 도구, 안전한 농기계 사용, 농작업 환경관리, 휴식과 일의 분담, 기초안전관리 등 6개의 대분류에 38개의 문항으로 구성하였다. 각 대분류별로 4-7개의 문항으로 구성이 된다. 또한 체크리스트와 교육에 포함할 모범사례들은 연구진이 직접 촬영한 사진과 외국의 사례를 참고로 하여 삽화 전문가에게 의뢰하여 제작한 삽화로 구성하였다. 이렇게 개발한 교재를 바탕으로 한국형 PAOT 프로그램을 개발하여, 경상북도의 4개 농촌 마을의 농업인 94명을 대상으로 2007년 7월에서 2008년 10월까지 4회의 참여형 농작업환경 개선교육을 수행하였다. 교육이 종료 3개월 후 연구진들이 직접 참여 농업인들의 농가를 방문하여 농업인들의 개선계획 실천여부를 확인하였고, 1시간 30분 정도 소요되는 "개선성과 중간 평가회"를 개최하여 개별 마을민들의 개선 성과를 전체 참가자들이 공유하는 시간을 가졌다. 전체 대상자 307명(남자 142명, 여자 165명)중 남자 59(41.5%)명, 여자 35(21.2%)명 등 총 94(30.6%)명이 PAOT 교육에 참가하였다. 전체 교육의 진행시간에 대한 적절성에서는 94.8%가 '만족한다' 이상으로 응답하였고, 교육의 전반적인 내용에는 98.9%가 '만족한다' 이상으로 응답하였고, 농작업 체크리스트 실습에는 100.0%가 '만족한다'고 응답하는 등 만족도 조사 문항 모두 90% 이상에서 만족한다고 응답하였다. 본 연구에서 개발된 농작업 환경 개선을 위한 PAOT 기법은 자발적이면서 지속적으로 농업인의 안전과 건강을 유지할 수 있는 하나의 대안이 될 수 있을 것으로 기대된다. 이를 위해서는 향후 보다 객관적인 연구가 수행되어야 할 것이며 더 많은 실제적인 적용 경험을 축적하는 것이 필요할 것이다.

한련초의 종자생존력(種子生存力) 및 생장특성(生長特性) (Seed Viability and Growth Characteristics of Eclipta prostrata (L.) L.)

  • 한련초의 종자(種子) 생존력(生存力), 심수관리(深水管理)에 의(依)한 초기생장반응(初期生長反應) 및 생장특성(生長特性)을 구명(究明)하기 위(爲)하여 수개(數個)의 시험(試驗)이 수행(遂行)되었다. 한련초 종자(種子)는 휴면성(休眠性)이 없었으며 실온(室溫)에 저장(貯藏)된 종자(種子)는 5개월(個月)까지 높은 발아율(發芽率)을 보였다. 수분흡수(水分吸收와 건조(乾燥)의 반복(反復)에 따른 종자(種子) 생존력(生存力)의 상실(喪失)은 건조방법(乾燥方法)에 따라 크게 달랐다. 흡수상태(吸水狀態)에서 건조상태(乾操狀態)로 빠른 전환(轉換)은 비고��(比較的) 높은 생존력(生存力)을 유지(維持)했으나 서서히 건조(乾操)된 종자(種子)는 발아중(發芽中)인 배(胚)의 손상(損傷)으로 극심(極甚)한 생존력(生存力) 상실(喪失)을 가져왔다. 토양(土壤) 5~10 cm 깊이에 있는 종자(種子)는 밭보다 논조건(條件)에서 더 오래 생존력(生存力)을 유지(維持)하였다 4 엽기(葉期) 이전(以前)의 심수관리(深水管理)(10 cm)는 신장(伸長)을 크게 억제(抑制)하였으나 그 이후(以後)부터의 심수관리(深水管理)는 오히려 신장伸長)을 자극(刺戟)하여 무처리(無處理)보다 초장(草長)이 유의(有意) 증가(增加)되었다. 분지(分枝)는 출현후(出現後) 2주(週)째부터 시작(始作)하였고 보통(普通) 10주(週)째까지 계속(繼續)되었다. 잎 크기는 동화산물(同化産物)의 공급(供給)과 관련(關聯)되어 분지시기(分枝時期)와 잎의 발생위치(發生位置)에 따라 결정(決定)되었다. 출현후(出現後) 5주(週)째부터 개화(開花)가 시작(始作)되었고 1개(個) 화서(花序)가 등숙(登熟)하는 데에는 10~14일(日)이 소요(所要)되었다. 개체당(個體當) 종자(種子)는 약(約) 14,000개(個)였다. 1일(日) 종자생산량(種子生産量)은 출현후(出現後) 10주(週)째까지 증가(增加)하다가 그 후(後) 감소(減少)하였으며 1 화서당(花序當) 종자수(種子數)는 늦게 발새(發生)된 화서(花序)일수록 감소(減少)하였다.

갈대(Reed, Phragmites communis Trin.)의 방제를 위한 제초제 스크리닝방법의 확립 - I. 갈대의 육묘 (Establishment of Herbicide Screening Methods for Reed (Phragmites communis Trin.) Control - I. Propagation of Reed)

  • 갈대방제용 제초제의 스크리닝재료를 효과적으로 확보할 수 있는 방법을 찾기 위해 갈대의 종자, 지하경, 달뿌리풀의 포복경 및 갈대의 마디절편을 이용하여 온실에서 육묘한 결과를 정리하면 다음과 같다. 1. 갈대의 종자, 지하경, 달뿌리풀의 포복경 및 갈대의 마디절편의 발아율은 각각 5, 46, 65, 75%로 나타났다. 2. 갈대의 종자를 이용하는 경우에는 보관이 용이하지만 발아율이 매우 낮고 육묘기간이 긴 문제점이 있었다. 3. 갈대의 지하경은 채취가 어렵고 채취시기도 11~3월로 제한되어 있었다. 또한 낮은 발아율, 지하경의 크기와 형태의 불균일, 초기 생육시 개체간의 차이, 보관상의 어려움 등의 문제점이 있었다. 한편 지하경을 이용하여 갈대를 육묘할 때는 원예용 부농상토를 사용하는 것이 바람직하였다. 4. 달뿌리풀의 포복경은 발아율이 비교적 높고 채취가 간편하지만 채취시기가 8~9월로 한정되어 있었으며 보관에 어려움이 있었다. 5. 갈대의 마디절편을 이용하는 경우에는 재료의 확보가 용이 하고 발아율이 비교적 양호할 뿐만 아니라 균일한 실험재료를 육성할 수 있었다. 마디절편은 채취 후 곧바로 사용해야 하며 중위절의 마디가 가장 높은 발아율을 보였으나 8월 이후에 채취한 마디에서의 발아율은 감소되는 경향이었다. 그러나 온실에서 모주를 재배하면서 필요시 절단하여 사용한다면 제초제 스크리닝용 갈대의 계속적인 육묘가 연중 가능할 것이다.

  • The wall shear stress in the vicinity of end-to end anastomoses under steady flow conditions was measured using a flush-mounted hot-film anemometer(FMHFA) probe. The experimental measurements were in good agreement with numerical results except in flow with low Reynolds numbers. The wall shear stress increased proximal to the anastomosis in flow from the Penrose tubing (simulating an artery) to the PTFE: graft. In flow from the PTFE graft to the Penrose tubing, low wall shear stress was observed distal to the anastomosis. Abnormal distributions of wall shear stress in the vicinity of the anastomosis, resulting from the compliance mismatch between the graft and the host artery, might be an important factor of ANFH formation and the graft failure. The present study suggests a correlation between regions of the low wall shear stress and the development of anastomotic neointimal fibrous hyperplasia(ANPH) in end-to-end anastomoses. 30523 T00401030523 ^x Air pressure decay(APD) rate and ultrafiltration rate(UFR) tests were performed on new and saline rinsed dialyzers as well as those roused in patients several times. C-DAK 4000 (Cordis Dow) and CF IS-11 (Baxter Travenol) reused dialyzers obtained from the dialysis clinic were used in the present study. The new dialyzers exhibited a relatively flat APD, whereas saline rinsed and reused dialyzers showed considerable amount of decay. C-DAH dialyzers had a larger APD(11.70$\pm$1.32mmHg/min)compared to CF dialyzers(4.32$\pm$0.55mmHg/min)(p<0.05). However, there was no observable difference in the UFR between the two dialyzers. Neither APD nor UFR showed any significant increase with an increasing number of reuses for up to more than 20reuses. A substantial number of failures observed in APD(larger than 20mmHe/min)on the reused dialyzers(2 out of 40 CP and S out 26 C-DAK) were attributed to the Possible damage on the fibers. The CF 15-11 HFDs which failed APD test did not show changes in the UFR compared to normal dialyzers indicating that APD is a more sensitive test than UFR test to evaluate the integrity of the fibers. 30527 T00401030527 ^x For quantitative measurement of reflected light from a clinical diagnostic strip, a prototype old reflectance photometer was designed. The strip loader and cassette were made to obtain more accurate reflectance parameters. The strip was illuminated at 45˚c through optical fiber and the intensity of reflected light was determined at rectanguLat angle using a photodiode. The kubelka-munk coefficient and reflection optical density were determined ar four different wavelengths(500, 550, 570 and 610nm) for blood glucose strip. For higher concentration than 300mg/41 about glucose, a saturation state of abforbance was observed at 500, 550 and 570nm. The mice were divided into two groups. One group was injected with Photofrin II, intravenously (2.5mg/kg body weight). The other group received no Photofrin II. Mice from both groups were irradialed for approximately 15 minutes at 100, 300, or 500 mW/cm2 with the argon (488nm/514.5 nm), dye(628nm) and gold vapor (pulsed 628 nm) laser light. A photosensitizer behaved as an added absorber. Under our experimental conditions, the presence of Photolfrin II increased surface temperature by at least 40% and the temperature rise due to 300 mW/cm2 irradiation exceeded values for hyperthermia. Light and temperature distributions with depth were estimated by a computer model. The model demonstrated the influence of wavelength on the thermal process and proved to be a valuable tool to investigate internal temperature rise. 30536 T00401030536 ^x We investigated the structural geometry of thirty-eight Korean femurs. The purpose of this study is to identify major geometrical differences between Korean femurs 3nd others that we believe belong to Caucasians so that we would be able to get insights into the femoral component design that fits Asians including Koreans. We utilized computerized tomography (CT) images of femurs extracted from cadavers. The CT images were transformed into bitmap data by using a film scanner, and then analyzed by using a commercially available software called Image v.1.0 and a Macintosh IIci computer.The resulting data were compared with already published data. The major results show that the geometry of the Korean femurs is significantly different from that of Caucasians: (1) the anteversion angle and the canal flare index are greater by the amount of approximately 8˚ and 0.5, respectively, (2) the shape of the isthmus cross section is more round, and (3) the distance between the teaser trochanter and the proximal border of the isthmus is shelter by about 15 mm. The results suggested that the femoral component suitable for Asians should be different from the currently-used components designed and manufactured mostly by European or American companies. 30537 T00401030537 ^x It is well known that nonlinear propagation characteristics of the wave in the tissue may give very useful information for the medical diagnoisis. In this paper, a new method to detect nonlinear propagation characteristics of the internal vibration in the tissue for the low frequency mechanical vibration by using bispectral analysis is proposed. In the method, low frequency vibration of f0( = 100Hz) is applied on the surface of the object, and the waveform of the internal vibration x (t) is measured from Doppler frequency modulation of silmultaneously transmitted probing ultrasonic waves. Then, the bispectra of the signal x (t) at the frequencies (f0, f0) and (f0, 2f0) are calculated to estimate the nonlinear propagation characteristics as their magnitude ratio, w here since bispectrum is free from the gaussian additive noise we can get the value with high S/N. Basic experimental system is constructed by using 3.0 MHz probing ultrasonic waves and the several experiments are carried out for some phantoms. Our department has developed an information system for routine hematology using down-sized computer system. We have used an IBM 486 compatible PC with 16MB main memory, 210 MB hard disk drive, 9 RS-232C port and 24 pin dot printer. The operating system and database management system were SCO UNIX and SCO foxbase, respectively. For program development, we used Xbase language provided by SCO foxbase. The C language was used for interface purpose. To make the system use friendly, pull-down menu was used. The system connected to our hospital information system via application program interface (API), so the information related to patient and request details is automatically transmitted to our computer. Our system interfaced with fwd complete blood count analyzers(Sysmex NE-8000 and Coulter STKS) for unidirectional data tansmission from analyzer to computer. The frame and leaflet of the polymer valve were made from polyurethane, To evaluate the hemodynamic performance of the polymer valve a comparative study of flow dynamics past a polymer valve and a St. Jude Medical prosthetic valve under physiological pulsatile flow conditions in vitro was made. Comparisons between the valves were made on the transvalvular pressure drop, regurgitation volume and maximum valve opening area. The polymer valve showed smaller regurgitation volume and transvalvular pressure drop compared to the mechanical valve at higher heart rate. Because the point of maximun temperature of laser-irradiated gelatin exists below the surface due to surface cooling, evaporation at the boiling temperature is made explosively from below the surface. The important parameters of this process are the conduction loss to laser power absorption (defined as the conduction-to-laser power parameter, Nk), the convection heat transfer at the surface to conduction loss (defined as Bi), dimensionless extinction coefficient (defined as Br.), and dimensionless irradiation time (defined as Fo).   • The wall shear stress in the vicinity of end-to end anastomoses under steady flow conditions was measured using a flush-mounted hot-film anemometer(FMHFA) probe. The experimental measurements were in good agreement with numerical results except in flow with low Reynolds numbers. The wall shear stress increased proximal to the anastomosis in flow from the Penrose tubing (simulating an artery) to the PTFE: graft. In flow from the PTFE graft to the Penrose tubing, low wall shear stress was observed distal to the anastomosis. Abnormal distributions of wall shear stress in the vicinity of the anastomosis, resulting from the compliance mismatch between the graft and the host artery, might be an important factor of ANFH formation and the graft failure. The present study suggests a correlation between regions of the low wall shear stress and the development of anastomotic neointimal fibrous hyperplasia(ANPH) in end-to-end anastomoses. 30523 T00401030523 ^x Air pressure decay(APD) rate and ultrafiltration rate(UFR) tests were performed on new and saline rinsed dialyzers as well as those roused in patients several times. C-DAK 4000 (Cordis Dow) and CF IS-11 (Baxter Travenol) reused dialyzers obtained from the dialysis clinic were used in the present study. The new dialyzers exhibited a relatively flat APD, whereas saline rinsed and reused dialyzers showed considerable amount of decay. C-DAH dialyzers had a larger APD(11.70${\pm}$1.32mmHg/min)compared to CF dialyzers(4.32${\pm}$0.55mmHg/min)(p<0.05). However, there was no observable difference in the UFR between the two dialyzers. Neither APD nor UFR showed any significant increase with an increasing number of reuses for up to more than 20reuses. A substantial number of failures observed in APD(larger than 20mmHe/min)on the reused dialyzers(2 out of 40 CP and S out 26 C-DAK) were attributed to the Possible damage on the fibers. The purpose of this study is to identify major geometrical differences between Korean femurs 3nd others that we believe belong to Caucasians so that we would be able to get insights into the femoral component design that fits Asians including Koreans. We utilized computerized tomography (CT) images of femurs extracted from cadavers. The CT images were transformed into bitmap data by using a film scanner, and then analyzed by using a commercially available software called Image v.1.0 and a Macintosh IIci computer.The resulting data were compared with already published data. The major results show that the geometry of the Korean femurs is significantly different from that of Caucasians: (1) the anteversion angle and the canal flare index are greater by the amount of approximately 8˚ and 0.5, respectively, (2) the shape of the isthmus cross section is more round, and (3) the distance between the teaser trochanter and the proximal border of the isthmus is shelter by about 15 mm. Then, the bispectra of the signal x (t) at the frequencies (f0, f0) and (f0, 2f0) are calculated to estimate the nonlinear propagation characteristics as their magnitude ratio, w here since bispectrum is free from the gaussian additive noise we can get the value with high S/N. We use combination of MLP and fuzzy filter for pattern recognition to estimate the direction of a muscle and Hogan's method for the force estimation. EMG signals are acquired by using a amputation simulator and 2 dimensional joystick motion.
