Effects of Acanthopanax senticosus Polysaccharide Supplementation on Growth Performance, Immunity, Blood Parameters and Expression of Pro-inflammatory Cytokines Genes in Challenged Weaned Piglets

  • To investigate the effect of dietary Acanthopanax senticosus polysaccharide (ASPS) on growth performance, immunity, blood parameters and mRNA expression of pro-inflammatory cytokines in immunologically challenged piglets, an experiment employing $2{\times}2$ factorial arrangement concerning dietary ASPS treatment (0 or 800 mg/kg) and immunological challenge (lipopolysaccharide [LPS] or saline injection) was conducted with 64 crossbred piglets (weaned at 28 d of age, average initial body weight of $7.25{\pm}0.21kg$) assigned to two dietary ASPS treatments with 8 replicates of 4 pigs each. Half of the piglets of per dietary treatment were injected with LPS or saline on d 14. Blood samples were obtained at 3 h after immunological injection on d 14 and piglets were slaughtered to obtain spleen samples on d 21. Dietary ASPS did not affect average daily gain (ADG) (p = 0.634), average daily feed intake (ADFI) (p = 0.655), and gain:feed (p = 0.814) prior to LPS challenge. After LPS challenge, for LPS-challenged pigs those fed ASPS had higher ADG and ADFI than the non-supplemented group (p<0.05), and an interaction between $LPS{\times}ASPS$ was observed on the two indices (p<0.05). Dietary ASPS improved lymphocyte proliferation among saline-injected and LPS-injected pigs (p<0.05). Interaction between $LPS{\times}ASPS$ was also revealed on lymphocyte proliferation (p<0.05). Circulatory concentration of IgG was influenced neither by ASPS (p = 0.803) or LPS (p = 0.692), nor their interaction (p = 0.289). Plasma concentration and spleen mRNA expression of interleukin-1beta (IL-$1{\beta}$), interleukin-6 (IL-6), and tumor necrosis factor (TNF)-${\alpha}$ were induced to increase (p<0.05) by LPS challenge, in contrast, these indices were decreased by dietary ASPS (p<0.05), and interactions were found on these cytokines (p<0.05). For LPS-challenged pigs, dietary ASPS also reduced the circulating concentration and spleen mRNA expression of IL-$1{\beta}$, IL-6 as well as TNF-${\alpha}$ (p<0.05). The interaction between $LPS{\times}ASPS$ was also observed on the circulating concentration of insulin-like growth factor-I, ${\alpha}$-acid glycoprotein (${\alpha}$-AGP), nonesterified fatty acid, and glucose (p<0.05). The results of this study demonstrate that dietary ASPS can modulate the release of pro-inflammatory cytokines during immunological challenge, which might enable piglets to achieve better growth performance.

National Genetic Evaluation (System) of Hanwoo (Korean Native Cattle)

  • Hanwoo (Also known as Korean native cattle; Bos taurus coreanae) have been used for transportation and farming for a long time in South Korea. It has been about 30 yrs since Hanwoo improvement began in earnest as beef cattle for meat yield. The purpose of this study was to determine the trend of improvement as well as to estimate genetic parameters of the traits being used for seedstock selection based on the data collected from the past. Hanwoo proven bulls in South Korea are currently selected through performance and progeny tests. National Hanwoo genetic evaluations are implemented with yearling weight (YW), carcass weight (CW), eye muscle area (EMA), backfat thickness (BF) and marbling score (MS). Yearling weights and MS are used for selecting young bulls, and EMA, BF, and MS are used for selecting proven bulls. One individual per testing room was used for performance tests, and five individuals per room for progeny tests. Individuals tested were not allowed to graze pasture, but there was enough space for them to move around in the testing room. Feeds including roughages and minerals were fed ad libitum, and concentrates were provided at the rate of about 1.8% of individual weight. Overall means of the traits were $352.8{\pm}38.56$ kg, $335.09{\pm}44.61$ kg, $77.85{\pm}8.838\;cm^2$, $8.6{\pm}3.7$ mm and $3.293{\pm}1.648$ for YW, CW, EMA, BF and MS. Heritabilities estimated in this study were 0.30, 0.30, 0.42, 0.50 and 0.63 in YW, CW, EMA, BF and MS, respectively, which are similar to results from previous research. Yearling weight was 315.54 kg in 1998, and had increased to 355.06 kg in 2011, resulting in about 40 kg of improvement over 13 yrs. YW and CW have improved remarkably over the past 15 yrs. Breeding values between 1996 and 2000 decreased or did not change much, but have moved in a desirable direction since 2001. These improvements correspond with the substantial increase in use of animal models since the late 1990s in Korea. Hanwoo testing programs have practically contributed to the improvement in aspects of quality and quantity. In sum, the current selection system is good enough to accommodate circumstances where fewer sires are used on many more cows. Although progeny tests take longer and cost more, they seem to be appropriate under the circumstances of the domestic market with its higher requirement for better meat quality. Consequently, accumulative data collection, genetic evaluation model development, revision of selection indices, as well as cooperation among farms, associations, National Agricultural Cooperative Federation, universities, research institutes, and government agencies must be applied to the Hanwoo selection program. All these efforts will assist the domestic market to secure a competitive position against imported beef under Free Trade Agreement trade system and will provide farmers with higher profits as well as the public with a higher quality of beef.

Effects of L-Carnitine with Different Lysine Levels on Growth and Nutrient Digestibility in Pigs Weaned at 21 Days of Age

  • This study was conducted to investigate the effects of L-carnitine with different levels of lysine on performance of pigs weaned at 21 days of age. A total of 120 pigs were allotted into a $3{\times}2$ factorial design with three different levels of lysine (1.40%, 1,60% and 1.80%) and two levels of L-carnitine (0 and 1,000 ppm). Each treatment had 4 replications with 5 pigs per replicate. Pigs of $22{\pm}1$ days (5.9 kg of body weight) were grouped into a completely randomized block design. Treatments were 1) 1.4-Crt; 1.40% of lysine with 1,000 ppm of L-carnitine, 2) 1.4-N; 1.40% of lysine without L-carnitine, 3) 1.6-Crt; 1.60% of lysine with 1,000 ppm of L-carnitine, 4) 1.6-N; 1.60% of lysine without L-carnitine, 5) 1.8-Crt; 1.80% of lysine with 1,000 ppm of L-carnitine and 6) 1.8-N; 1.80% of lysine without L-carnitine. Growth performance was optimized in pigs fed 1.6% lysine regardless of carnitine addition. For the first 7 days of the experimental period, the best ADG and F/G were found in pigs within the 1.6-Crt group. Carnitine significantly improved (p<0.05) ADG of pigs when the lysine level in the diet was 1.6%. Only in the third week carnitine had a significant influence on growth performance of pigs. A lysine-sparing effect of L-carnitine was not detected in this study. The 1.6-Crt group showed the best proximate nutrient digestibility, and the crude fat and gross energy digestibility were higher when the L-carnitine was added in the diet. Lysine level significantly affected the digestibilities of DM (p<0.001), GE (p<0.001), CP (p<0.01) and C.fat (p<0.05). Carnitine also significantly improved digestibility of nutrients. Lysine level as well as carnitine level affected the amino acids digestibility, however, in 1.8% lysine diet carnitine did not influence on amino acids digestibility. Plasma carnitine content was significant higher (p<0.05) in pigs fed L-carnitine. This indicates the increased biological availability of carnitine within the body. L-carnitine supplementation tended to improve feed utilization during the third week (p<0.10) and during the entire period (p=0.10). Lysine level significantly affected feed utilization of pigs during the third week and entire period (p<0.05). As pigs grew, the lysine requirement was reduced.

Effects of Dietary Supplementation of Magnesium Hydrogen Phosphate (MgHPO4) as an Alternative Phosphorus Source on Growth and Feed Utilization of Juvenile Far Eastern Catfish (Silurus asotus)

  • The present study was conducted to investigate a supplemental effect of magnesium hydrogen phosphate (MHP, $MgHPO_4$) as an alternative phosphorus (P) source on growth and feed utilization of juvenile far eastern catfish (Silurus asotus) in comparison with three conventional P additives (monocalcium phosphate (MCP), dicalcium phosphate (DCP) and tricalcium phosphate [TCP]) as positive controls. A basal diet as a negative control was prepared without P supplementation and four supplemental P sources were added at the level of 2%. Five groups of 450 fish having mean body weight of 11.3 g following 24 h fasting after three week adaptation period were randomly distributed into each of 15 tanks (30 fish/tank). Fish were hand-fed to apparent satiety twice a day for 8 weeks. Fish fed MHP had weight gain (WG), protein efficiency ratio and specific growth rate comparable to those fed MCP. Fish fed MHP and MCP had feed efficiency (FE) significantly higher (p<0.05) than those fed DCP. Fish groups fed control and TCP showed the lower FE than the other groups which was significantly different (p<0.05) from those of fish fed the other diets. Survival rate was not significantly different (p>0.05) among treatments. Fish fed control had the lowest hematocrit, which was significantly different (p<0.05) from that of fish fed MHP. Fish fed MCP and MHP had plasma P higher (p<0.05) than fish fed the other diets. Relative efficiencies of MCP, DCP and TCP to MHP were found to be 100.5 and 101.3%, 92.0 and 91.6%, and 79.1 and 80.9% for WG and FE, respectively. P availability was determined to be 88.1%, 75.2%, 8.7%, and 90.9% for MCP, DCP, TCP, and MHP, respectively. Consequently, MHP recovered from wastewater stream showed that as an alternative P source its performance was comparative with MCP on growth and feed utilization of juvenile far eastern catfish.

오수유(吳茱萸) 물 추출물이 급성역류성 식도염에 미치는 효과 (Effect on Acute reflux Esophagitis by Evodiae Fructus Aquous Extract)

  • Objectives : This study was performed to investigate effect of evodiae fructus on acute reflux esophigitis rat induced by pylorus and forestomach ligation operation. Methods : Twenty-four laboratory rats were divided four groups and each group had six rats ; normal intact group, acute reflux esophagitis (RE) control group, two experiment RE group treated extract of evodiae fructus 600 mg/kg (EEF600) and 300 mg/kg (EEF300). All rats was fasted for 18 hr but free water, we induced RE by pylorus and forestomach ligation operation. Intact group and RE control group rats were orally administered a distilled water and two experiment groups were orally administed with EEF 600 mg/5ml/kg and 300 mg/5ml/kg. One hour after, rats were anesthetized, intact group was cut the abdomen open and sutured with 2.0 silk thread. RE control group and EEF group were cut the abdomen open, ligated pyloric canal and forestomach with 2.0 silk thread and sutured. Six hour after the operation, rats were sacrified, collected bloods in the abdominal vein, disectted a esophagus and stomach. The stomach was washed a 1 ml PBS and the esophagus was cut longitudinally and pictured a innter mucosa area to research damages in esophagus. Results : The esophagic tissue damage percentage of reflux esophagitis rat was increased compared to that of normal intact group. But esophagic damage percentage of EEF 600 were significantly decreased compared to that of RE control group. But there was no difference on gastric juice pH between control RE, alpha-tocopherol administration rat group and EEF administration rat group. In esophagus of RE control rat, gastric damage occurred severely and injury percentage of mucosa were increased, but EEF 600 mucous inflammatory damage percentage was significantly compared to that of RE control group. Proinflammatory cytokines such as TNF-alpha, IL-1beta and IL-6 in serum on RE control group were markedly grew than those of intact rat, those of vechicle group treated with EEF 600 and EEF 300 were remarkably decreased compared to production of proinflammatory cytokine of RE control group. In microscopic observation, intact group rat had no hyperemia, mucous injury and exclusion, ulcer and edema. But it could showed mucosa damages, submucosa edema and ulcer in RE control. However, administration of EEF 600 and EEF 300 made esophagus have less inflammation and injury by gastric acid. Conclusions : The results suggest that antiinflammatory Effect of EEF could attenuate the severity of reflux esophagitis and prevent the esophageal mucosal damage, and validate its therapeutic use in esophageal reflux disease.

AMINO ACID DIGESTIBILITY AS AFFECTED BY VARIOUS FIBER SOURCES AND LEVELS 2. THE RELATIONSHIP BETWEEN FIBER LEVELS AND AMINO ACIDS DIGESTIBILITY

  • A number of correlation and regression analyses were performed on data from apparent and true digestibility of amino acids at ileal and fecal level with finishing pigs, in order to investigate whether these amino acid digestibilities could be predicted with certainly degree on their fiber fractions content (chemical analysis). The data comprised 16 diets varying in 4 levels of crude fiber and from 4 fiber sources. The relationships between fiber fractions including crude fiber (CF), NDF, ADF lignin and cellulose contents on apparent and true digestibility of almost all amino acids in both ileal and fecal level were negative, except glutamine at fecal level. In apparent digestibility at ileal level, the correlations of fiber fractions were moderate (r of NDF = 0.53 to 0.63; ADF, 0.50 to 0.77; cellulose, 0.50 to 0.75), with an exception of CF content was relatively high (r of 0.58 to 0.81). The correlations to true digestibility of amino acids were weaker. In case of at fecal level, the higher correlation (negative) was found with NDF than CF content. Estimations of amino acids digestibility were performed using regression equation. The data showed that apparent digestibility of amino acids could be estimated for almost amino acids except arginine, threonine, valine and tyrosine at fecal level and phenylalaine, valine and glycine at ileal level. The best prediction at ileal and fecal level ($r^2=0.55-0.77$ and 0.52-0.76), respectively was obtained with NDF content. Prediction for true digestibility of amino acids, none of fiber fractions could be estimated for arginine, leucine and valine at all collection levels. At ileal level, CF could be used for most of amino acids except phenylalanine, glycine and praline; cellulose, only for lysine and methionine and NDF, only for proline. At fecal level, glutamine digestibility could be estimated only from CF and ADF content. The best predictor at ileal level was ADF content whereas at fecal level was NDF content. These results indicate that lignin content could not be used as predictor for all amino acids at both levels neither in apparent nor true digestibility.

Effects of Conditioners (Standard, Long-Term and Expander) on Pellet Quality and Growth Performance in Nursery and Finishing Pigs

  • A series of experiments were conducted to determine the effects of standard (ST), long-term (LT), and expander (EX) conditioners on nutritional value of phase-three nursery and finishing swine diets. In Exp. 1, 180 pigs (average initial BW of 11.7 kg) were fed com-soybean meal based diets (1.3% lysine) during a 28 d growth assay. Gain/feed was improved (p<0.004) with pelleting and pellet durability index (PDI) increased with degree of conditioning (LT>ST). However, there was no advantage for LT vs ST conditioning in rate or efficiency of gain (p>0.5). In Exp. 2, 180 pigs (average initial BW of 10.4 kg) were fed con-soybean meal based diets (0.9% lysine) during a 28 d growth assay, Pelleted diets tended to support greater ADG (p<0.08) and gain/feed (p<0.002) with no marked advantage from EX vs ST conditioning. In Exp 3, a total of 70 barrows (average initial BW of 54 kg) was used in a growth assay to determine the effects of feeding a com-soybean meal based diet processed with a standard (ST) steam conditioner, a long-term (LT) steam conditioner, and an expander (EX) conditioner. The conditioned diets were fed as mash (M) or pellets (P) to give a $2{\times}3$ factorial plus a meal control. PDI increased with degree of conditioning (EX>LT>ST). There was a trend (p<0.07) for greater ADG in pigs fed diets that had been thermally conditioned. Also, there was a general advantage in gain/feed with pelleting (p<0.04), but this advantage was pronounced only with standard conditioning. Indeed, the greatest gain/feed was observed for pigs fed the expander treatments (p<0.03) and the expander mash was used as efficiently as the expander pellets. There was no difference in backfat thickness among pigs fed the treatments (p>0.3). but the more extreme the processing technique. the greater the incidence and severity of stomach lesions (p<0.04). These results suggest maximum rate and efficiency of growth with pelleting after standard steam conditioning or simply feeding an expanded mash.

Effects of Protein and Carbohydrate Supplementations on Fibre Digestion and Microbial Population of Sheep

  • The effects of two types of protein, soybean meal (SBM) and fish meal (FM); and two types of energy supplements, corn flour (CF) and paper pulp (PP), on intake of guinea grass (Panicum maximum), fibre digestion and microbial activities in four Merino rams with an average weight of $54.4{\pm}4.5kg$ were studied. Each animal was fitted with a ruminal cannula and a duodenal cannula at the proximal position. The animals were fed twice daily with chopped guinea grass (5 cm) ad libitum and one of the four dietary supplements: 170 g FM+268 g PP; 170 g FM+268 g CF; 200 g SBM+200 g PP or 200 g SBM+200 g CF. All the supplements were mixed with 100 g molasses. In sacco and in vivo digestibilities, digesta flow rates, fermentation and microbial population were studied in a $4{\times}4$ Latin square design with a $2{\times}2$ factorial arrangement of dietary treatments. The effects of energy or protein sources were not significant on grass intake of sheep. The potential degradabilities of NDF and ADF were not significantly affected by any of the supplements. However, the energy and protein sources had significant efects on disappearance rate of NDF and ADF. The disappearance rate of both NDF and ADF were significantly (p < 0.05) higher in animals fed PP when compared to animals fed CF. Animals fed FM also showed significantly (p < 0.03) higher disappearance rate of ADF than those fed SBM. Animals fed PP showed better digestion in the rumen and total tract. Total flow of NDF and ADF through the duodenum was not significantly affected by the various supplements. The mean rumen pH values (5.8-6.1) were not significantly different among the four different diets. The concentration of rumen ammonia was significantly (p < 0.0001) higher in animals fed SBM (235-266.4 mg N/L) supplement than in animals fed FM (174.9-179.7 mg N/L), while total VFA concentration was not significantly affected by both energy and protein supplements. Mean values of total VFA ranged from 72.5-82.3 mM. Molar proportions of acetate, propionate and butyrate were typical of a roughage type fermentation. Molar proportion of acetate was significantly (p < 0.0001) higher in sheep fed PP when compared to sheep fed CF. Animals fed FM had higher total viable bacterial counts, while animals fed CF showed higher protozoal numbers. Proportions of cellulolytic bacteria were only slightly higher in animals fed SBM or PP.

Effects of Expander Processing and Enzyme Supplementation of Wheat-based Diets for Finishing Pigs

  • Two experiments were conducted to determine the effects of expander processing and enzyme supplementation of wheat-based diets on growth performance and nutrient digestibility in finishing pigs. For Exp. 1, 60 finishing pigs (average initial BW of 49.5 kg) were fed meal, standard pellets and expanded pellets in a 70 d growth assay. From 49.5 to 79.0 kg, 79.0 to 111.8 kg, and overall (49.5 to 111.8 kg), ADG and ADFI were not affected by pelleting or standard vs expander conditioning (p>0.22). However, from 49.5 to 79.0 kg, pigs fed pellets have greater gain/feed than pigs fed mash (p<0.04), and pigs fed expanded pellets tended to have greater (p<0.10) gain/feed than pigs fed standard pellets. Overall (i.e. from 49.5 to 111.8 kg), gain/feed (p<0.02) and apparent fecal digestibilities of DM (p<0.001) and N (p<0.02) were improved by pelleting the diets. Also, expander processing further improved gain/feed (p<0.06) and digestibility of DM (p<0.04) compared to standard steam conditioning. Scores for keratinization (p<0.002) and ulceration (p<0.003) of the stomach were increased by pelleting, but the mean scores for the various treatments ranged only from 0.05 to 1.08 (i.e., low to mild keratosis and ulceration). For Exp. 2, 80 pigs (average initial BW of 54.1 kg) were fed mash and pellets (standard or expander) without and with xylanase. The enzyme was added to supply 4,000 units of xylanase activity/kg of diet. Adding xylanase to the mash diet improved gain/feed from 90.7 to 115.9 kg (p<0.04) of the growth assay and digestibility of DM (p<0.05) on d 39. However, in pelleted diets, adding the enzyme did not improve growth performance or digestibility of nutrients. Pelleting tended to increase scores for ulceration (p<0.06), and enzyme supplementation decreased stomach keratinization scores for pigs fed the standard pellets (p<0.01). However, as in Exp. 1, the mean scores for all treatment groups were quiet low (i.e., ranging from normal to mild). In conclusion, pelleting improved efficiency of growth, but additional benefits from expander conditioning were observed only in Exp. 1. Finally, xylanase tended to improve growth performance and nutrient digestibility, only in pigs fed mash diets but not in pigs fed pellets.

  • 본 연구에서는, 저온 및 약산성 조건에서 높은 활성을 보이는 티로시나아제인 tyrosinase-CNK의 반응 안정성을 조사하였다. Tyrosinase-CNK는 지금까지 알려진 중온성(mesophilic) 및 호열성(thermophilic) 환경 유래의 티로시나아제 보다는 열 안정성이 낮았으나 0 ℃에서 효소 안정성이 매우 뛰어났고, 반복적인 동결-해동(freeze-thaw) 과정에서도 효소 활성을 안정적으로 유지하였다. 또한, 물과 ethanol 및 acetonitrile이 혼합된 유기용매 환경에서 초기 상대 활성 값의 변화가 관찰되었으나 유기용매 농도의 증가로 인한 추가적인 활성 저하를 유발하지 않고 활성을 일정하게 유지하였다. 한편, 효소 반응의 염(salt)에 대한 저해는 chaotropic 특성이 많이 나타나는 염일수록 적게 나타났다. 결과적으로, tyrosinase-CNK는 통상의 티로시나아제를 이용하여 반응을 수행하기 어려운 환경에서도 원하지 않는 반응을 억제하고 효소 불활성화를 최소화면서 반응을 촉매할 것으로 기대된다.
