Antioxidant and Antiproliferative Activities of Methanolic Extract from Celandine

  • Celandine (Chelidonium majus, family Papaveraceae) is an herb used extensively in traditional Korean medicine. To investigate its antioxidant and antiproliferative activities, the methanolic extract of celandine was introduced. The antioxidant properties of the extract were tested using various in vitro systems, including hydroxyl radical scavenging assay, DNA damage protection assay, 1,1-diphenyll-2-2-pricylhydrazyl (DPPH) free radical scavenging activity, metal chelating activity, and reducing power assay. The extract exhibited stronger antioxidant activity ($IC_{50}=7.92{\mu}g/mL$) against hydroxyl radicals in the Fenton system than butylated hydroxyanisole ($IC_{50}=51.46{\mu}g/mL$) and $\alpha$-tocopherol ($IC_{50}=67.48{\mu}g/mL$). Likewise, damage to the plasmid pBR 322 induced by hydroxyl radicals was found to be protected by the extract at a concentration of $400{\mu}g/mL$. Cellular proliferation and the induction of apoptosis were also examined by a cellular proliferation assay, flow cytometry, and mRNA expression analysis. Taken together, the extract significantly inhibited the growth of HT-29 cells in a concentration- and time-dependent manner, and gradually increased both the proportion of apoptotic cells and the expression of caspase-3. Overall, our research suggests that celandine possesses antioxidant and antiproliferative properties.

Human ${\beta}$-Globin Second Intron Highly Enhances Expression of Foreign Genes from Murine Cytomegalovirus Immediate-Early Promoter

  • To develop a highly efficient mammalian expression vector, a series of vectors were constructed based on the murine cytomegalovirus (MCMV) immediate-early (IE) promoter and human ${\beta}$-globin second intron. The resulting MCMV promoter was several-fold stronger than the HCMV promoter in various mammalian cell lines, such as the NIH3T3, Neuro-2a, 293T, and HT1080 cell lines, and was only slightly weaker than the HCMV promoter in HeLa and CHO cells. The inclusion of the human ${\beta}$-globin second intron behind the MCMV promoter or HCMV promoter markedly enhanced the promoter activity in various mammalian cell lines, and the resultant MCMV/Glo-I expression system was stronger than the HCMV promoter from 4.7- to 11.2-fold in every cell line tested. Also, the MCMV/Glo-I promoter induced a higher level of the VSV-G protein in a transiently transfected 293T cell line, which is useful for the production of recombinant retrovirus and lentivirus vectors.

Effect of Acupuncture Stimulation ($HT_7$), on the Change of Standard Leads in ECG (신문혈(神門穴) 침자극(鍼刺戟)이 심전도(心電圖) 변화(變化)에 미치는 영향(影響))

  • Objectives : The aim of this study is to investigate the effect of Shinmun-acupuncture stimulation on the relationship of change in electrocardiopgraphy (ECG). Methods : For this purpose, 11 healthy volunteers were acupunctured at Shinmun acupoint using the reinforcing or reducing by inserting the needle in the same direction as the channel runs or in the opposite direction (迎隨補瀉). Then, we measured and observed the change of standard leads I, II and III in ECG. Results : In lead I , acupuncture treated groups were increased the activity of PR interval and PR segment compared to the control group. In lead II, during the acupuncture treated group by inserting the needle in the opposite direction as the channel runs was increased the activity of T wave duration, and after the acupuncture group by inserting the needle in the same direction as the channel runs was increased the activity of PR interval. In lead III, acupuncture treated groups were increased the activity of ST duration, PR interval and QT interval. Conclusions : These results suggested that Shinmun acupuncture stimulation plays an important role on the activities of ECG.

Biological Activities of Water and Ethanolic Extracts from Allium victorialis L. Mature Leaves

  • Allium victorialis L. (A. victorialis) is a very popular vegetable in Korea. The most commonly used parts of this vegetable are the bulbs and young leaves. To determine if the mature leaves have any beneficial properties, we investigated antioxidant, anti-${\alpha}$-glucosidase, anti-inflammatory, and anticancer activities of water and ethanol extracts from A. victorialis. Antioxidant activity was evaluated by measuring total phenolic content, DPPH and superoxide radicals scavenging activities. The water extract from A. victorialis ($W{\cdot}A$. victorialis) exhibited higher antioxidant ability than the ethanol extract ($E{\cdot}A$. victorialis). Moreover, the water extract showed strong inhibitory effect on ${\alpha}$-glucosidase. On the other hand, the ethanol extract had greater anti-inflammatory activity on murine macrophage cells (RAW 264.7) and greater anticancer activities against human colon cancer cells (HT-29). These results suggest that mature leaves from E·A. victorialis may have health-enhancing effects.

Effects of Acanthopanacis cortex Extracts on the Cytokine-inducing and Immune response in Mice (생쥐에서 오가피에 의한 싸이토카인 유도와 면역반응에 관한 효과)

  • This experimental study was carried out to evaluate the effects of Acanthopanacis cortex on Cytokine-inducing and and immune response in Mice. In order to investigate the effect of Acanthopanacis cortex, the following was performed; Cytotoxicity, in vitro, the fraction of $CD4^+$, $CD8^+$, $B220^+$ in splenic cell, gene expression of IL-12(p35), IL-12(p40), IFN-${\gamma}$, and splenic cell proliferation by Acanthopanacis cortex. Analysis of cytokine gene expression was carried out by RT-PCR amplification. Amplified PCR products were electrophoresed on 1.2% agarose gel, and the analysis (Ht) was used to 1D-density program. The results were obtained as follows. Acanthpanacis cortex showed didn't have cell toxicity under $12{\mu}g/m{\ell}$ group on mouse lung fibroblast cells. In an in vitro model using mouse peripheral blood mononuclear cells (PBMCs), extract of Acanthpanacis cortex induced multiple cytokine, including interleukin-12 (p35), interleukin-12 (p40), interferon-gamma (IFN-${\gamma}$). The extract also enhanced the percentages of the $CD4^+$, and $CD8^+$ in the untreated control were $22.1{\pm}3.3$ to $38.4{\pm}2.1$, and $5.0{\pm}0.4$ to $10.7{\pm}0.3%$, respectively. From above findings, it is suggested that Acanthopanacis cortex is able to anti-cancer and activate immune response system.

  • To evaluate the antitumor activity and antimetastatic effects of Gobongeerlhobang(GG1B), studies were done experimentally. The results were obtained as follows: 1. GG1B extracts exhibited a significant cytotoxicity against HT1080, A549, SK-MEL-2 and SK-OV-3 cell lines. 2. The T/C% was 120.7% in GG1B treated group in S-180 bearing ICR mice. 3. GG1B extracts exhibited inefficient adhesive effect of A549, B16-BL6 cell to complex extracellular matrix. 4. GG1B extracts showed a significant inhibition of lung metastasis of B16-BL6 cells in C57BL/6. 5. In CAM assays, angiogenesis was insignificantly inhibited in GG1B treated group than control group. These results suggested that GG1B extracts might be usefully applied for prevention and treatment of cancer.

Evaluation on Dynamic Behavior of Friction Welded Joints in Alloy718 to SCM440 using Acoustic Emission Technique (Alloy718/SCM440 마찰용접재의 AE에 의한 동적 거동평가)

  • Dissimilar friction welding were produced using 15 mm diameter solid bar in superalloy(Alloy718) to chrome molybdenum steel(SCM440) to investigate their mechanical properties. Consequently, optimal welding conditions were n=2000 rpm, HP=60 MPa, UP=120 MPa, HT=10 sec and UT=10 sec when the metal loss(Mo) is 3.5 mm. Acoustic Emission(AE) technique was applied to analyze the dissimilar friction welding of Alloy718 and SCM440. The relationship between the AE parameters and dissimilar friction welding of both material was discussed. In the case of heating time of 6 sec, 10 sec, 14 sec and 20 sec, 5 AE events per 0.5 seconds and energy about $2.7{\times}10^{10}$ were exhibited in heating time. In upsetting time, resulting in various numbers of events per second and very low energy. The frequency range of the signal generated during the heating time was about 200 kHz. However, the upsetting time resulted in a wide range of signals from very low frequency to high frequency of 500 kHz due to rapid plasticity of the material.

Chemical Constituents of Fatsia japonica Stem (팔손이나무(Fatsia japonica) 줄기의 성분연구)

  • Fatsia japonica is grown wild to Eastern Asia, including Korea, Japan, and Taiwan and it is known as ornamental plant, and it is also known that pharmacological action. In this study, we have selected the stem of F. japonica with consideration about biological activities and amount of yield. In addition, four compounds (1-4) were isolated from the stem of F. japonica. Extensive spectroscopic and chemical studies established the structures of these compounds as maltose (1), begoniifolide A (2), leiyemudanoside B (3), leonticin F (4). All of the compounds were investigated for their anti-inflammatory, anti-neuroinflammatory, and neuro-protective effects on RAW264.7, BV2, and HT22 cells. However, among four compounds, there were no effects by maltose (1), begoniifolide A (2), leiyemudanoside B (3), leonticin F (4) on the anti-inflammatory, anti-neuroinflammatory, and neuro-protective action. This is the first report on the isolation of maltose (1), begoniifolide A (2), leiyemudanoside B (3), leonticin F (4) from the stem of F. japonica. Begoniifolide A (2), leiyemudanoside B (3), leonticin F (4) were isolated for the first time from this plant. It might be necessary to continue the further studies to find the biological active compounds isolated from the stem of F. japonica.

Change of growth performance, hematological parameters, and plasma component by hexavalent chromium exposure in starry flounder, Platichthys stellatus

  • The study investigated the changes in growth performance, hematological parameters, plasma components, and stress indicators of juvenile starry flounder, Platichthys stellatus, depending on varying exposure to hexavalent chromium. P. stellatus was exposed to waterborne chromium at 0, 50, 100, 200, and 400 ppb for 4 weeks. The result showed that Cr exposure resulted in decreased daily length gain (DLG), daily weight gain (DWG), condition factor (CF), and hepatosomatic index (HIS) in P. stellatus. In terms of hematological parameters, red blood cell (RBC) count, hematocrit (Ht), and hemoglobin (Hb) significantly decreased at 400 ppb after 2 weeks. In terms of plasma components, inorganic analysis was unchanged and cholesterol, an organic component, considerably increased at 400 ppb after 4 weeks. Plasma enzyme components including glutamic oxalate transaminase (GOT) and glutamic pyruvate transaminase (GPT) were significantly increased. Stress indicators such as cortisol and glucose were notably increased over 100 ppb after 4 weeks with increasing chromium concentration. The results indicate that exposure to waterborne Cr induced toxic effects on growth, hematological parameters, plasma components, and stress indicators.

Structural and Thermal Analysis and Membrane Characteristics of Phosphoric Acid-doped Polybenzimidazole/Strontium Titanate Composite Membranes for HT-PEMFC Applications

  • A series of novel PBI/SrTiO3 nanocomposite membranes composed of polybenzimidazole (PBI) and strontium titanate (SrTiO3) with a perovskite structure were fabricated with various concentrations of SrTiO3 through a solution casting method. Various characterization techniques such as proton nuclear magnetic resonance, thermogravimetric analysis, atomic force microscopy (AFM) and AC impedance spectroscopy were used to investigate the chemical structure, thermal, phosphate absorption and morphological properties, and proton conductivity of the fabricated nanocomposite membranes. The optimized PBI/SrTiO3-8 polymer nanocomposite membrane containing 8wt% of SrTiO3 showed a higher proton conductivity of 7.95 × 10-2 S/cm at 160℃ compared to other nanocomposite membranes. The PBI/SrTiO3-8 composite membrane also showed higher thermal stability compared to pristine PBI. In addition, the roughness change of the polymer composite membrane was also investigated by AFM. Based on these results, nanocomposite membranes based on perovskite structures are expected to be considered as potential candidates for high-temperature PEM fuel cell applications.
