Studies on the Ventilatory Functions of the Korean Children and Adolescents, with Special References to Prediction Formulas (한국 어린이 및 청소년의 폐환기능에 관한 연구 - 특히 표준치 예측 수식에 관하여 -)

  • The maximum breathing capacity (MBC) and the maximum mid-expiratory flow rate (MMF) are widely used in evaluation of the ventilatory function, among various parameters of pulmonary function. The MBC volume is the amount of gas which can be exchanged per unit time during maximal voluntary hyperventilation. Performance of this test, unlike that of single breath maneuvers, is affected by the integrity of the respiratory bellows as a whole including such factors are respiratory muscle blood supply, fatigue, and progressive trapping of air. Because of this, the MBC and its relation to ventilatory requirement correlates more closely with subjective dyspnea than does any other test. The MMF is the average flow rate during expiration of the middle 50% of the vital capacity. The MMF is a measurement of a fast vital capacity related to the time required for the maneuver and the MMF relates much better to other dynamic tests of ventilatory function and to dyspnea than total vital capacity, because the MMF reflects the effective volume, or gas per unit of time. Therefore, it is important to have a prediction formula with one can compute the normal value for the subject and the compare with the measured value. However, the formulas for prediction of both MBC and MMF of the Korean children and adolescents are not yet available in the present. Hence, present investigation was attempt to derive the formulas for prediction of both MBC and MMF of the Korean children and adolescents. MBC and MMF were measured in 1,037 healthy Korean children and adolescents (1,035 male and 1,002 female) whose ages ranged from 8 to 18 years. A spirometer (9L, Collins) was used for the measurement of MBC and MMF. Both MBC and MMF were measured 3times in a standing position and the highest values were used. For measurement, the $CO_2$ absorber and sadd valve were removed from the spirometer in order to reduce the resistance in the breathing circuit and the subject was asked to breathe as fast and deeply as possible for 12 seconds in MBC and to exhale completely as fast as possible after maximum inspiration for MMF. During the measurement, investigator stood by the subject to give a constant encouragement. All the measured values were subsequently converted to values at BTPS. The formulas for MBC and MMF were derived by a manner similar to those for Baldwin et al (1949) and Im (1965) as function of age and BSA or age and height. Ethanol Extract of Oldenlandia diffusa - an Effective Chemotherapeutic for the Treatment of Colorectal Cancer in Humans -Anti-Cancer Effects of Oldenlandia diffusa-

  • Objectives: Oldenlandia diffusa is traditionally used to relieve the symptoms of and to treat various diseases, but its anti-cancer activity has not been well studied. In the present study, the authors investigated the anti-cancer effects of an ethanol extract of Oldenlandia diffusa (EOD) on HT-29 human adenocarcinoma cells. Methods: Cells were treated with different concentrations of an EOD, and cell death was assessed by using a 3-[4,5-dimethylthiazol-2-yl]-2,5-diphenyltetrazolium bromide (MTT) assay. Analyses of the sub G1 peak, the caspase-3 and -9 activities, and the mitochondrial membrane depolarizations were conducted to confirm cell death by apoptosis. Also, intracellular reactive oxygen species (ROS) generation was determined using carboxy-H2DCFDA (5-(and-6)-carboxy-20,70-dichlorodihydrofluorescein diacetate). Results: EOD inhibited the proliferation of HT-29 cells for 24 hours by $78.6%{\pm}8.1%$ at $50{\mu}g/mL$, $74.4%{\pm}4.6%$ at $100{\mu}g/mL$, $65.9%{\pm}5.2%$ at $200{\mu}g/mL$, $51.4%{\pm}6.2%$ at $300{\mu}g/mL$, and by $41.7%{\pm}8.9%$ at $400{\mu}g/mL$, and treatment for 72 hours reduced the proliferation at the corresponding concentrations by $43.3%{\pm}8.8%$, $24.3{\pm}5.1mV$, $13.5{\pm}3.2mV$, $6.5{\pm}2.3mV$, and by $2.6{\pm}2.3mV$. EOD increased the number of cells in the sub-G1 peak in a dose-dependent manner. The mitochondrial membrane depolarization was elevated by EOD. Also, caspase activities were dose-dependently elevated in the presence of EOD, and these activities were repressed by a pan-caspase inhibitor (zVAD-fmk). The ROS generation was significantly increased by EOD and N-acetyl-L-cysteine (NAC; a ROS scavenger) remarkably abolished EOD-induced cell death. In addition, a combination of sub-optimal doses of EOD and chemotherapeutic agents noticeably suppressed the growth of HT-29 cancer cells. Conclusion: These results indicate that EOD might be an effective chemotherapeutic for the treatment of human colorectal cancer.

Graphene Quantum Dot Interfacial Layer for Organic/Inorganic Hybrid Photovoltaics Prepared by a Facile Solution Process (용액 공정을 통한 그래핀 양자점 삽입형 유/무기 하이브리드 태양전지 제작)

  • This paper reports that the electronic properties at a $P3HT/TiO_2$ interface associated with exciton dissociation and transport can be tailored by the insertion of a graphene quantum dot (GQD) layer. For donor/acceptor interface modification in an $ITO/TiO_2/P3HT/Al$ photovoltaic (PV) device, a continuous GQD film was prepared by a sonication treatment in solution that simplifies the conventional processes, including laser fragmentation and hydrothermal treatment, which limits a variety of component layers and involves low cost processing. The high conductivity and favorable energy alignment for exciton dissociation of the GQD layer increased the fill factor and short circuit current. The origin of the improved parameters is discussed in terms of the broad light absorption and enhanced interfacial carrier transport.

A Study of Surface Electromyography Measurement of Wrist Exercise (Extension, Flexion, Grasping) in Healthy People (정상인의 완관절 신전, 굴곡, 악력(握力) 시 표면근전도 측정 연구)

  • Purpose : This study aimed to prove that surface electromyography(SEMG) can be used to identify the degree of symptoms of diseases in the upper extremities; it also aimed to confirm various potential therapeutic effects through an inquiry into the value measured by the SEMG on certain acupuncture points in the upper extremities. Methods : Fifty healthy people received instructions for the method of exercise: wrist flexion, extension and hand grasping. Disposable electrodes were attached to acupuncture points of Susamni($LI_{10}$), Naegwan($PC_6$), Oegwan($TE_5$) and below Sohae($HT_3$) two cun on both sides in flexion, extension and grasping to measure the SEMG values. The research results were analyzed using SPSS statistics Ver. 22.0(IBM, USA). Results : The average value was highest on Naegwan($PC_6$) in grasping, and the average SEMG value was higher in the order of grasping, extension and flexion. The asymmetry index of each point was Susamni($LI_{10}$) $16.26{\pm}13.59%$, Oegwan($TE_5$) $20.38{\pm}15.59%$, below Sohae($HT_3$) two cun $20.89{\pm}16.77%$, Naegwan($PC_6$) $22.49{\pm}14.91%$ in wrist extension, Susamni($LI_{10}$) $25.99{\pm}21.44%$, Oegwan($TE_5$) $21.15{\pm}15.94%$, below Sohae($HT_3$) two cun $19.62{\pm}15.46%$, Naegwan($PC_6$) $19.93{\pm}16.85%$ in wrist flexion, Susamni($LI_{10}$) $16.60{\pm}12.21%$, Oegwan($TE_5$) $10.94{\pm}8.29%$, below Sohae($HT_3$) two cun $15.20{\pm}12.60%$, Naegwan($PC_6$) $11.68{\pm}7.77%$ in grasping. Conclusions : In this study, to identify the degree of symptoms of diseases in the upper extremities and confirm therapeutic effects, it is necessary to analyze the calculated percentage and compare the SEMG measurement of special points with those of other points, and with the asymmetry index.

Inhibitory Effect of Zostera japonica on Growth of Human Cancer Cells (애기거머리말 추출물의 암세포 성장 억제효과)

  • In this study, crude extracts of the marine eelgrass Zostera japonica and their solvent-partitioned fractions were evaluated for their inhibitory effect against AGS, HT-1080 and MCF-7 human cancer cells using MTT assay. Each of the crude extracts (acetone/methylene, chloride, and methanol) of Z. japonica showed a significant inhibitory effect on the growth of human cancer cells. The combined crude extracts were partitioned between $CH_2Cl_2$ and water. The organic layer was further partitioned between 85% aq. MeOH and n-hexane, and the aqueous layer was then fractionated into n-BuOH and $H_2O$, successively. Growth inhibition effects of solvent-partitioned fractions from Z. japonica on human cancer cells increased in a dose-dependent manner. Among these tested samples, the 85% aq. MeOH fraction revealed good inhibitory effects on the growth of AGS and HT-1080 human cancer cells, while the n-hexane fraction exhibited good inhibitory effects on the growth of AGS and MCF-7 human cancer cells. In addition, 85% aq. MeOH and n-hexane fractions enhanced mRNA expression of p53 gene. These results suggest that there is further scope for the isolation of active compounds from Z. japonica, which should show much stronger anticancer activity.

The Association between the T102C Polymorphism of the HTR2A Serotonin Receptor Gene and HDL Cholesterol Level in Koreans

  • 5-HT2A is one of major serotonin receptor that is involved in the action of serotonin-targeting drugs. Previous clinical studies have shown an unexpected association between lower cholesterol level and psychiatric diseases, in which T102C polymorphism of HTR2A, gene of 5-HT2A serotonin receptor, might be involved. Therefore, we hypothesized a potential association between lower cholesterol level and T102C polymorphism. The effect of the T102C polymorphism on the serum lipid profiles of 646 subjects without specific psychiatric disease was investigated. Genotype was determined by polymerase chain reaction and restriction fragment length polymorphism analysis. There were significantly lower levels of total cholesterol ($193.6{\pm}35.0$ versus $202.1{\pm}45.5\;mg/dl$, p = 0.016) and HDL-cholesterol ($42.7{\pm}11.6$ versus $46.3{\pm}12.7\;mg/dl$, p = 0.004) in CC genotype than non-CC genotypes. Moreover, multivariate analysis showed that the CC genotype is a strong predictor of a lower HDL-cholesterol level (p < 0.001). In conclusion, this study shows that the CC genotype of the HTR2A gene is related to lower HDL-cholesterol level in Koreans. This is the first demonstration showing the potential genetic relationship between the serotonin receptor gene polymorphism and the HDL-cholesterol level.

Binding Mode Prediction of 5-Hydroxytryptamine 2C Receptor Ligands by Homology Modeling and Molecular Docking Analysis

  • Serotonin or 5-hydroxytryptamine subtype 2C ($5-HT_{2C}$) receptor belongs to class A amine subfamily of G-protein-coupled receptor (GPCR) super family and its ligands has therapeutic promise as anti-depressant and -obesity agents. So far, bovine rhodopsin from class A opsin subfamily was the mostly used X-ray crystal template to model this receptor. Here, we explained homology model using beta 2 adrenergic receptor (${\beta}$2AR), the model was energetically minimized and validated by flexible ligand docking with known agonists and antagonists. In the active site Asp134, Ser138 of transmembrane 3 (TM3), Arg195 of extracellular loop 2 (ECL2) and Tyr358 of TM7 were found as important residues to interact with agonists. In addition to these, V208 of ECL2 and N351 of TM7 was found to interact with antagonists. Several conserved residues including Trp324, Phe327 and Phe328 were also found to contribute hydrophobic interaction. The predicted ligand binding mode is in good agreement with published mutagenesis and homology model data. This new template derived homology model can be useful for further virtual screening based lead identification.

Effects of SSP Therapy on Plasma Catecholamine of the Body (SSP요법이 인체의 혈장 카테콜라민에 미치는 효과)

  • The goal of this article is to estimate the change in plasma catecholamine when ST-36 (Leg Three Li) and HT-7 (Spirit Gate) among the meridian points were stimulated for the group of the young and the old, using SSP therapy, thereby clarifying its effects on the responses of catecholamine that generates various physiological effects on the body, in particular, the effects on catecholamine, in the senile body, thereby providing its availability for physical therapy for the old, which is now the major concern of the society. As for research subjects, a total of 10 healthy male and female adults in their twenties were selected as the group of the young, and 10 old male and female, aged over 60, as the group of the old. The findings are as follows: When SSP stimuli were applied to ST-36 and HT-7, as for the change in the content of plasma catecholamine for the group of the young, norepinephrine significantly increased from $309.40{\pm}23.81pg/ml$ before stimulation to $374.90{\pm}35.31pg/ml$ 20 minutes later and to $406.70{\pm}42.43pg/ml$ 40 minutes later (P < 0.05); epinephrine significantly increased from $100.70{\pm}28.55pg/ml$ before stimulation to $95.70{\pm}24.16pg/ml$ 20 minutes later and to $128.80{\pm}25.02pg/ml$ 40 minutes later (P < 0.05); and dopamine showed no significant differences as its content was $245.20{\pm}40.01pg/ml$ before stimulation, $233.50{\pm}59.56pg/ml$ 20 minutes later and $250.90{\pm}56.13pg/ml$ 40 minutes later.

Anxiolytic effect of chronic ginseng treatment using elevated T-maze in mice

  • The roots of the plant Korean ginseng have been extensively used in the traditional Chinese herbal medicine. The effects of chronic administration of Korean ginseng extract (KGE) were investigated on two different anxiety models: the elevated T-maze (for inhibitory avoidance and escape measurements) and the open field test (OFT). Diazepam (1 mg/kg), KGE (10, 30 and 100 mg/kg) were administered orally for 15 days. On the 14th day, mice were previously exposed for 30 min to one of the open arms of the T-maze, 24 h before the test. On 15th day, mice had two exposures to the enclosed and open arm of the elevated T-maze followed by exposure to the open field apparatus. The number of line crossings in the apparatus was used to assess locomotor changes. Cumulative Concentration Response Curve of 5-HT was plotted using rat fundus which were pre-treated in a similar way. Treatment with Diazepam (1 mg/kg) and KGE (10, 30 and 100 mg/kg) significantly (P < 0.05) impaired inhibitory avoidance performance but did not impair escape latency. In OFT, diazepam facilitated locomotion as compared to vehicle and other treatment groups. KGE at any of the selected doses did not impair locomotion. Concentration response curve of 5-HT was shifted towards the right with suppression of maxima in rats treated with KGE. The results suggest that KGE exerts anxiolytic like behaviour in a specific subset of defensive behaviour, particularly those related to generalized anxiety disorder.

  • The purpose of this study is to investigate the home trading system (HTS) of the security companies in order to examine the critical factors of HTS service quality and the effect of these factors on HTS customer satisfaction and loyalty. The results show : (1) the quality factors of HTS are assurance, reliability, tangible, responsiveness, and empathy, (2) and these quality factors significantly affect customer satisfaction on HTS and customer satisfaction and loyalty to the security company. (3) Also, customer satisfaction on HTS plays an important role in improving customer satisfaction and loyalty to the security company.
