Fermentation Characteristics and Increased Functionality of Doenjang Prepared with Bamboo Salt (죽염 된장의 발효 특성 및 기능성 증진 효과)

  • The aim of this study is carried out to evaluate the fermentation characteristics and increased functionality when doenjang was prepared with bamboo salts. Grain type mejus were fermented with mixed starter cultures of Aspergillus oryzae, Bacillus subtilis, and Lactococcus lactis. These mejus were mixed with different kinds of salts-purified salt (PD), sea salt (SD), one-time baked bamboo salt ($1{\times}BD$), three-time baked bamboo salt ($3{\times}BD$), and nine-time baked bamboo salt ($9{\times}BD$)- when doenjangs were prepared. For doenjang fermentation period of 8 weeks at $37^{\circ}C$, the fermentation characteristics of all the groups were compared. The amino type nitrogen content and enzyme activities (protease and ${\alpha}$-amylase) in the samples were significantly increased. In DPPH radical scavenging activities and hydroxyl radical scavenging activities, $9{\times}BD$ (47% and 69%) showed the highest scavenging activities compared to PD (40% and 49%), SD (42% and 57%), $1{\times}BD$ (42% and 64%) and $3{\times}BD$ (45% and 65%) (P<0.05). The anticancer effects of doenjang in HT-29 cancer cells indicated all the groups, especially doenjang prepared with bamboo salts were higher than the others (P<0.05). Apoptosis related genes of Bax and Bcl-2, as well as inflammation related genes of iNOS and COX-2 were regulated by the treatment of doenjangs in HT-29 cancer cells. SD, $1{\times}BD$, $3{\times}BD$, and $9{\times}BD$ increased the expression level of Bax and decreased the expression level of Bcl-2, iNOS, and COX-2. These results suggest that sea salt and bamboo salt especially bamboo salt could improve fermentation characteristics and functionality of doenjang and play an important role in regulating apoptosis and inflammation related genes in cancer cells.

Study on the Anti-cancer, Anti-metastasis and Immune Response Improvement of Aqua-acupuncture with Amomum Amarum Lourerio Infusion Solution (익지인약침(益智仁藥鍼)이 항암(抗癌) 및 면역기능(免疫機能)에 미치는 실험적(實驗的) 연구(硏究))

  • Objective : This study was purposed to investigate the anti-cancer and anti-metastasis and immune response of Aqua-acupuncture with Amomum amarum Lourerio infusion solution. Methods : The Amomum amarum Lourerio infusion solution put into Chung-wan(CV12) of BALB/c or C57BL6 mice were rised to cancer by B15-F10 and HT1080, S-180 cancer cell line. Results : The following result have been obtained 1. The effect on expression of MMP-9 gene about the HT1080 cancer cell line was increased in 100, $50{\mu}g/m{\ell}$ diluent groups, compared with control group. 2. The effect on expression of MMP-9 gene about the B15-F10 cancer cell line was increased in all the sample groups, compared with control group. 3. S-180 cancer cell line transplants in BALB/c mice were inhibited significantly in weight inctease in all the sample groups, compared with control group. 4. The effect on spleen cell proliferation was decreased in all the sample groups, compared with control group. 5. The $IFN-{\gamma}$ in all the sample groups and the $TNF-{\alpha}$ in 25 and $12.5{\mu}g/m{\ell}$ diluent groups was increased. 6. In flow cytometry, the number of CD4+, CD19+ cell in all the sample group was increased and the number of CD8+ cell in $100{\mu}g/m{\ell}$ diluent group, compared with control group. Conclusion : According to the result, Aqua-acupuncture with Amomum amarum Lourerio infusion solution has significant anti-cancer, anti-metastasis and Immune response improvement.

  • The pharmacotherapy of schizophrenia exhibits wide inter-individual variabilities in clinical efficacy and adverse effects. Recently, human genetic diversity has been known as one of the essential factors to the variation in human drug response. This suggests that drug therapy should be tailored to the genetic characteristics of the individual. Pharmacogenetics is the field of investigation that attempts to elucidate genetic basis of an individual's responses to pharmacotherapy, considering drug effects divided into two categories as pharmacokinetics and pharmacodynamics. The emerging field of pharmacogenomics, which focuses on genetic determinants of drug response at the level of the entire human genome, is important for development and prescription of safer and more effective individually tailored drugs and will aid in understanding how genetics influence drug response. In schizophrenia, pharmacogenetic studies have shown the role of genetic variants of the cytochrome P450 enzymes such as CYP2D6, CYP2C19, and CYP2A1 in the metabolism of antipsychotic drugs. At the level of drug targets, variants of the dopamine $D_2$, $D_3$ and $D_4$, and 5-$HT_{2A}$ and 5-$HT_{2C}$ receptors have been examined. The pharmacogenetic studies in schizophrenia presently shows controversial findings which may be related to the multiple involvement of genes with relatively small effects and to the lack of standardized phenotypes. For further development in the pharmacogenomics of schizophrenia, there would be required the extensive outcome measures and definitions, and the powerful new tools of genomics, proteomics and so on.

Characteristics of Alcohol Metabolism of Hahyangju in Rats (하향주의 랫드에 대한 알코올 대사 특성)

  • This study was conducted to examine the volatile flavor compounds of Hahyangju, a traditional Korean liquor, and to evaluate the effect of the beverage on alcohol metabolism in rats. By GC/MS analysis, 17 volatile flavor compounds including iso-butyl alcohol and iso-amyl alcohol were detected in Hahyangju. The concentrations of acetaldehyde and ethylacetate in Hahyangju were decreased by filtration. Alcohol (0.035 mg/dL) and acetaldehyde (0.29 mg/dL) levels in the blood of rats given Hahyangju (HT animals) were lower than in rats given 17% (v/v) alcohol (AT rats). Also, alcohol dehydrogenase (ADH) and acetaldehyde dehydrogenase (ALDH) activities in HT rats were $24.63{\pm}1.8{\mu}$moles/mg protein and $9.8{\pm}1.3{\mu}$moles/mg protein, respectively, and were higher than in AT animals. The increases in ADH and ALDH activity in HT animals resulted in decreases in alcohol and acetaldehyde concentrations in blood, compared to the levels seen in rats given 17% (v/v) alcohol. These results suggest that Hahyangju may increase alcohol metabolizing activity, and consumption of Hahyangju may result in less of a hangover than follows ingestion of beverages (such as wine) containing about 17% (v/v) alcohol.

Effects of Neonatal Footshock Stress on Glucocorticoid and $5-HT_{2A/2C}$ Receptor Bindings and Exploratory Behavior

  • To investigate the effects of neonatal stress on behavior and neurochemistry, rats were exposed to the footshock stress on postnatal day (PND) 14 or PNDs 14 and 21. Rats were exposed to uncontrollable electric shocks delivered to the floor with a constant current (0.8 mA) for 5 sec period. Daily sessions consisted of 60 trials on a random time schedule with an average of 55 sec. The first exposure to footshocks on PND 14 decreased body weight gain for 1 day. However, the second exposure to footshocks on PND 21 did not affect body weight gain. Exploratory activity was measured by exposing a rat to a novel environment 24 h after experience of footshocks. Similar to the body weight changes, a decreased activity was noted after the first exposure to footshocks, while no changed activity was noted after the second exposure to footshocks. However, the Bmax value of $5-HT_{2A/2C}$ receptors in the cortex decreased by the second exposure to footshocks, but not by the first exposure to footshocks. Moreover, an autoradiographic study revealed that the density of $[^3H]dexamethasone$ binding in hippocampus decreased in rats exposed to footshocks 4 times during PND $14{\sim}20.$ These results suggest that the uncontrollable footshock stress changes 5-hydroxytryptamine and glucocorticoid receptor systems acutely and that the repeated exposure to the same stress may not elicit behavioral alterations by the compensatory activity of young brain although changes in some neurochemistry exist.

Cytotoxic Phenylpropanoids from the Rhizomes of Alpinia galanga

  • A bioassay-guided fractionation of the n-hexane and chloroform extracts of the rhizomes of Alpinia galanga led to the isolation of two active compounds, 1'S-1'-acetoxychavicol acetate (1) and p-coumaryl alcohol $\gamma$-O-methyl ether (2). 1'S-1'-acetoxychavicol acetate (1) exhibited significant cytotoxicity against all human cancer cell lines tested (A549; $IC_{50}$ 8.14, SNU 638; 1.27, HCTl16; 1.77, HT1080; 1.2, HL60; $IC_{50}$ 2.39 ${\mu}g/ml$), whereas p-coumaryl alcohol $\gamma$-O-methyl ether (2) showed selective cytotoxicity against the SNU638 cell ($IC_{50}$ = 1.62${\mu}g/ml$).

Statistical Observation on Surgical Stomach Disturbance Patient (외과적 위질환환자의 통계적고찰)

  • 1972년 1월부터 1972년 12월말까지 충남대학교 의과대학 부속병원에서 위 수술을 받은 환자 66명을 대상으로 여러 가지 통계 결과 1. 성별 분포에선 남자가 51명 여자가 15명으로 약 3.4:1의 비율을 나타냈다. 2. 연령별 분포에선 $69.7\%$가 40세 이상에서 나타났다. 3. Chief complain은 Indigestion이었다. 4. 증상의 평균 연한은 1년 전후의 Chronic symptom을 경험했다. 5. Vital sign의 변화는 대부분이 별로 나타나지 않았다. 6. 수술 전 수혈은 1000cc 이내로 환자의 $78\%$에 필요했었다. 7. 수술 전의 Hb은 정상보다 낮은 율은 $45.4\%$였고 정상은 $54.6\%$였다. 8. 수술 전의 W.B.C는 정상보다 낮은 율은 $10.6\%$, 정상이 $42.4\%$, 정상보다 높은 율이 $47\%$였다. 9. 수술 전의 Ht치는 정상보다 낮은 율은 $60.6\%$, 정상은 $36.4\%$ 정상보다 높은 율은 $3\%$였다. 10. 수술 전 위액검사 결과는 Cancer인 경우는 Free acid와 Total acid가 모두 거의 정상보다 낮았고 Ulcer인 경우 대부분 높았다. 11. Liver function test에서는 표에서 나타난 대로 많은 변화는 없었다. 12. 병소 부위는 Cancer의 경우 $51.8\%$에서 Pylorus부위에 왔었고 Ulcer의 경우 $62.4\%$에서 Lesser curvature (ciratricial region포함)에 왔었다. 13. 수술방법은 $77.7\%$가 Subtotal gastrectomy였었다. 14. 위 수술을 위한 입원 일자는 $60\%$ 이상에서 $11\~15$일간의 평균을 보였었다.

Effects of Acanthopanacis cortex Extracts on the Cytokine-inducing and Immune response in Mice (생쥐에서 오가피에 의한 싸이토카인 유도와 면역반응에 관한 효과)

  • This experimental study was carried out to evaluate the effects of Acanthopanacis cortex on Cytokine-inducing and and immune response in Mice. In order to investigate the effect of Acanthopanacis cortex, the following was performed; Cytotoxicity, in vitro, the fraction of $CD4^+$, $CD8^+$, $B220^+$ in splenic cell, gene expression of IL-12(p35), IL-12(p40), IFN-${\gamma}$, and splenic cell proliferation by Acanthopanacis cortex. Analysis of cytokine gene expression was carried out by RT-PCR amplification. Amplified PCR products were electrophoresed on 1.2% agarose gel, and the analysis (Ht) was used to 1D-density program. The results were obtained as follows. Acanthpanacis cortex showed didn't have cell toxicity under $12{\mu}g/m{\ell}$ group on mouse lung fibroblast cells. In an in vitro model using mouse peripheral blood mononuclear cells (PBMCs), extract of Acanthpanacis cortex induced multiple cytokine, including interleukin-12 (p35), interleukin-12 (p40), interferon-gamma (IFN-${\gamma}$). The extract also enhanced the percentages of the $CD4^+$, and $CD8^+$ in the untreated control were $22.1{\pm}3.3$ to $38.4{\pm}2.1$, and $5.0{\pm}0.4$ to $10.7{\pm}0.3%$, respectively. From above findings, it is suggested that Acanthopanacis cortex is able to anti-cancer and activate immune response system.

  • The aim of this studies was to elucidate the meaning, theoretical propriety and use of the Sea points in PYOYOUBU. The results obtained were as follows : We reached a conclusion that Sea points, that is to say Xuehai($SP_{30}$), Xiaohai($SI_8$), Shaohai($HT_3$), Zhaohai($KI_6$), Qihai ($CV_6$) were gathering places of meridian energy. And the remedial function of Sea points against visera and bowel disease was closely connected with it's meridian pointological naming. Besides the remedial function of Sea points like this was closely connected with the cases in literature cited.

Potential Biological Activities of Magnoflorine: A Compound from Aristolochia debilis Sieb. et Zucc

  • Magnoflorine, an important compound in Aristolochia, was usually used as an anxiolytic chemical. In this study, the magnoflorine was isolated from Aristolochia and the biological activities such as antioxidant, ${\alpha}$-tyrosinase inhibitory, anti-inflammatory, and anticancer activities were investigated. The magnoflorine showed significant antioxidant activity as a 2,2-diphenyl-2-picryl-hydrazyl (DPPH) free radical scavenger, $50{\mu}g/mL$ of the magnoflorine scavenged about 70.8% of all the free radicals. And it was good at ${\alpha}$-tyrosinase inhibiting, $100{\mu}g/mL$ of the magnoflorine inhibited 36.5% of the tyrosinase. High dosage of magnoflorine inhibited the inflammation production nitric oxide (NO), and the magnoflorine protected the murine macrophage cells (RAW 264.7) from LPS-induced apoptosis. The cell viability of human colon cancer calls (HT-29) was around 100% when treated with different dose of magnoflorine, it's suggesting that magnoflorine had no anticancer effect.
