황화수소(H2S) 감지를 위한 아세트산 납이 침염된 폴리에스터(PET) 섬유 기반의 변색성 센서 (Polyester (PET) Fabric dyed with Lead (II) acetate-based Colorimetric Sensor for Detecting Hydrogen Sulfide (H2S))

  • In this study, the colorimetric sensor, polyester (PET) fabric dyed with lead (II) acetate (Pb(C2H3O2)2), was fabricated and characterized for the detection of the hydrogen sulfide (H2S). The surface morphology of the fabric was determined using scanning electron microscope and energy-dispersive X-ray spectroscopy. The optical properties of the fabric were evaluated by measuring the variation in the blue value of an RGB sensor. The fabric showed a significant color change, high linearity (R2 : 0.98256), and fast response time (< 1.0 s) when exposed to H2S. This is because the sensor is highly porous and permeable to the gas. The fabric can not only be used as a hydrogen sulfide sensor but also be used to detect and prevent H2S influx using sticky tape on pipelines.

Effects of pH, $PCO_2$, and Adenosine on the Contractility of Pig Coronary Artery

  • Effects of pH, $PCO_2$, and adenosine on the vascular contractility were investigated in the pig coronary arteries. The helical strips of isolated coronary arteries were immersed in the HEPES or $HCO_3^-/CO_2$-buffered Tyrode's solution equilibrated with 100% $O_2\;or\;95%\;O_2-5%\;CO_2\;at\;35^{\circ}C$. The contraction was recorded isometrically using a force transducer. The amplitudes of contraction induced by ACh, high $K^+$, and electrical Held stimulation (EFS) were decreased by elevating extracellular pH (pHo) and were increased by lowering pHo. A shift from $0%\;CO_2\;to\;5%\;CO_2$ at constant pHo (pH 7.4) reduced the contractions induced by ACh, high $K^+$, EFS. However the contraction induced by 100mM $K^+$ was less influenced by the change of pHo or $CO_2$. The contraction induced by ACh in $Ca^{2+}$free Tyrode's solution as well as the contraction developed by the addition of extracellular of $Ca^{2+}$ were decreased by lowering pHo and were increased by elevating pHo. High $K^+$ (25mM) induced contraction at pH 6.8 was not returned to the level of the contraction at pH 7.4 by the elevation of extracellular. calcium $[Ca^{2+}]_o$. Adenosine-induced relaxation was more significant with 5% $CO_2$ than 0% $CO_2$ in the high $K^+$-induced contraction and was more significant with low pHo than high pHo in the contraction induced by EFS. From the above results, it is suggested that $H^+$ and $CO_2$ inhibit $Ca^{2+}$ influx as well as $Ca^{2+}$ release from intracellular $Ca^{2+}$ storage sites and enhance the relaxing effect of adenosine in the pig coronary artery.

낙동강 중·하류 지역의 남조류 개체수와 수질인자간의 상관관계 분석 (Analysis of correlation between cyanobacterial population and water quality factors in the middle and down stream region of nakdong river)

  • In this study, we investigated the correlation between the number of cyanobacterial species and the water quality factors after installation of Nakdong river weirs. Factor analyses of water quality factors were also performed. The results of correlations showed that there was no significant water quality with the cyanobacterial concentration but the correlation coefficient of nutrients increased from the upstream to the downstream. There was a significant correlation between W T and pH as important water quality factors for the growth of cyanobacteria. In the downstream region of Nakdong river, algae were more frequently detected than in the upstream region, and the blooms of algae species seem to be influenced by the higher pH and the influx of the phosphorus from the upstream tributaries. Nutrient concentration was higher in the downstream region than in the upstream region due to the effects of tributaries Geumho river and Nam river in Nakdong river. As a result of the factor analyses, nitrogen species were the principal factors in the upper region, and phorsphorus species were the principal factors in the downstream region.

Acidity in Precipitation and Solar North-South Asymmetry

  • We are motivated by both the accumulating evidence for the connection of solar variability to the chemistry of nitrogen oxide in the atmosphere and recent finding that the Galactic cosmic-ray (GCR) influx is associated with the solar north-south asymmetry. We have analyzed the measured pH in precipitation over the 109 stations distributed in the United States. We have found that data of pH in precipitation as a whole appear to be marginally anti-correlated with the solar asymmetry. That is, rain seems to become less acidic when the southern hemisphere of the Sun is more active. The acidity of rain is also found to be correlated with the atmospheric temperature, while not to be correlated with solar activity itself. We have carried on the analysis with two subsamples in which stations located in the east and in the west. We find that the pH data derived from the eastern stations which are possibly polluted by sulfur oxides and nitrogen oxides are not correlated with the solar asymmetry, but with the temperature. On the contrary, the pH data obtained from the western stations are found to be marginally anti-correlated with the solar asymmetry. In addition, the pH data obtained from the western stations are found to be correlated with the solar UV radiation. We conclude by briefly pointing out that a role of the solar asymmetry in the process of acidification of rain is to be further examined particularly when the level of pollution by sulfur oxides and nitrogen oxides is low.

Effects of Histamine on Cultured Interstitial Cells of Cajal in Murine Small Intestine

  • Interstitial cells of Cajal (ICCs) are the pacemaker cells in the gastrointestinal tract, and histamine is known to regulate neuronal activity, control vascular tone, alter endothelial permeability, and modulate gastric acid secretion. However, the action mechanisms of histamine in mouse small intestinal ICCs have not been previously investigated, and thus, in the present study, we investigated the effects of histamine on mouse small intestinal ICCs, and sought to identify the receptors involved. Enzymatic digestions were used to dissociate ICCs from small intestines, and the whole-cell patch-clamp configuration was used to record potentials (in current clamp mode) from cultured ICCs. Histamine was found to depolarize resting membrane potentials concentration dependently, and whereas 2-PEA (a selective H1 receptor agonist) induced membrane depolarizations, Dimaprit (a selective H2-agonist), R-alpha-methylhistamine (R-alpha-MeHa; a selective H3-agonist), and 4-methylhistamine (4-MH; a selective H4-agonist) did not. Pretreatment with $Ca^{2+}$-free solution or thapsigargin (a $Ca^{2+}$-ATPase inhibitor in endoplasmic reticulum) abolished the generation of pacemaker potentials and suppressed histamine-induced membrane depolarization. Furthermore, treatments with U-73122 (a phospholipase C inhibitor) or 5-fluoro-2-indolyl des-chlorohalopemide (FIPI; a phospholipase D inhibitor) blocked histamine-induced membrane depolarizations in ICCs. On the other hand, KT5720 (a protein kinase A inhibitor) did not block histamine-induced membrane depolarization. These results suggest that histamine modulates pacemaker potentials through H1 receptor-mediated pathways via external $Ca^{2+}$ influx and $Ca^{2+}$ release from internal stores in a PLC and PLD dependent manner.

Prediction of potential habitats and distribution of the marine invasive sea squirt, Herdmania momus

  • The influx of marine exotic and alien species is disrupting marine ecosystems and aquaculture. Herdmania momus, reported as an invasive species, is distributed all along the coast of Jeju Island and has been confirmed to be distributed and spread to Busan. The potential habitats and distribution of H. momus were estimated using the maximum entropy (MaxEnt) model, quantum geographic information system (QGIS), and Bio-ocean rasters for analysis of climate and environment(Bio-ORACLE), which can predict the distribution and spread based only on species occurrence data using species distribution model (SDM). Temperature and salinity were selected as environmental variables based on previous literature. Additionally, two different representative concentration pathway (RCP) scenarios (RCP 4.5 and RCP 8.5) were set up to estimate future and potential habitats owing to climate change. The prediction of potential habitats and distribution for H. momus using MaxEnt confirmed maximum temperature as the highest contributor(77.1%), and mean salinity, the lowest (0%). And the potential habitats and distribution of H. momus were the highest on Jeju Island, and no potential habitat or distribution was seen in the Yellow Sea. Different RCP scenarios showed that at RCP 4.5, H. momus would be distributed along the coast of Jeju Island in the year 2050 and that the distribution would expand to parts of the Korea Strait by the year 2100. RCP 8.5, the distribution in 2050 is predicted to be similar to that at RCP 4.5; however, by 2100, the distribution is predicted to expand to parts of the Korea Strait and the East Sea. This study can be utilized as basic data to effectively control the ecological injuries by H. momus by predicting its spread and distribution both at present and in the future.

Onion peel extract and its constituent, quercetin inhibits human Slo3 in a pH and calcium dependent manner

  • Sperm function and male fertility are closely related to pH dependent $K^+$ current (KSper) in human sperm, which is most likely composed of Slo3 and its auxiliary subunit leucine-rich repeat-containing protein 52 (LRRC52). Onion peel extract (OPE) and its major active ingredient quercetin are widely used as fertility enhancers; however, the effect of OPE and quercetin on Slo3 has not been elucidated. The purpose of this study is to investigate the effect of quercetin on human Slo3 channels. Human Slo3 and LRRC52 were co-transfected into HEK293 cells and pharmacological properties were studied with the whole cell patch clamp technique. We successfully expressed and measured pH sensitive and calcium insensitive Slo3 currents in HEK293 cells. We found that OPE and its key ingredient quercetin inhibit Slo3 currents. Inhibition by quercetin is dose dependent and this degree of inhibition decreases with elevating internal alkalization and internal free calcium concentrations. Functional moieties in the quercetin polyphenolic ring govern the degree of inhibition of Slo3 by quercetin, and the composition of such functional moieties are sensitive to the pH of the medium. These results suggest that quercetin inhibits Slo3 in a pH and calcium dependent manner. Therefore, we surmise that quercetin induced depolarization in spermatozoa may enhance the voltage gated proton channel (Hv1), and activate non-selective cation channels of sperm (CatSper) dependent calcium influx to trigger sperm capacitation and acrosome reaction.

  • The effect of ${\alpha}_1$-adrenoceptor(${\alpha}_1$-AR) stimulation on intracellular pH($pH_i$), $Na^+$ activity($a_{Na}{^i}$) and contractility were investigated in isolated papillary muscles of euthyroid or hyperthyroid guinea pig with conventional microelectrode, $Na^+$ or $H^+$-selective microelectrodes, and tension transducer. Stimulation of the ${\alpha}_1$-AR by phenylephrine produced a decrease in $a_{Na}{^i}$ in euthyroid preparations. This decrease in $a_{Na}{^i}$ was abolished in presence of PKC activator, phorbol dibutyrate, and increased contrary to decrease. Phenylephrine also increased $a_{Na}{^i}$ in hyperthyroid ones. However, phenylrephtine produced an increase in $pH_i$ in both euthyroid and hyperthyroid ones. These changes were blocked by prazosin, an antagonist of ${\alpha}_1$-AR. These findings suggest that the changes in $a_{Na}{^i}$ and $pH_i$ are mediated by a stimulation of $Na^+-H^+$ exchange via ${\alpha}_1$-AR stimulation. This study focused on the increase in $a_{Na}{^i}$, $pH_i$ and contractility. The increase in $pH_i$ was blocked by amiloride or EIPA, $Na^+-H^+$ exchange inhibitors. Therefore, the increase in $a_{Na}{^i}$ and $pH_i$ mediated by ${\alpha}_1$-AR appeared to be due to an influx of $Na^+$ and a reduction of $H^+$ through $Na^+-H^+$ exchange. This study also revealed that the increase in $pH_i$ and $a_{Na}{^i}$ might be related to the sustained positive inotropic response. The $a_{Na}{^i}$ increase may contribute to the intracellular $Ca^{2+}$ through the $Na^+-Ca^{2+}$ exchange, and the $pH_i$ increase could cause an increase in the $Ca^{2+}$ sensitivity of myofilaments and may augment the ${\alpha}_1$-AR-mediated positive inotropic response.

Ca-dependent Alteration in Basal Tone, Basal $^{45}Ca$ Uptake and $^3H-nitrendipine$ Binding in the Aorta of Spontaneously Hypertensive Rats

  • We investigated the alterations in basal tone of aortic strips by changing the Ca concentration, basal $^{45}Ca$ uptake and $^3H-nitrendipine$ binding of the single cells of aortic smooth muscles in the spontaneously hypertensive rats (SHR) and Wistar-Kyoto (WKY) rats. While the basal tone of the aortic strips in WKY rats was not affected by alteration of Ca concentration, that in SHR was decreased by the removal of Ca from the bath solution and was recovered by the restoration of Ca to normal levels. This contraction increased in a Ca concentration-dependent manner and reached a maximum at 2 mM Ca. The basal tone of aorta in SHR was suppressed by verapamil $(10^{-6}M)$. The basal tone of aorta in SHR increased about 50% in the strips of endothelial rubbing, compared with that of intact endothelium. Basal $^{45}Ca$ uptake in the aortic single smooth muscle cells of SHR was greater than that of WKY (p<0.01), Specific bindings of $[^3H]nitrendipine$ in the aortic single smooth muscles of SHR and WKY were saturable. The dissociation constant $(K_d)\;was\;0.71{\pm}0.15\;and\;1.18{\pm}0.08nM$ SHR, respectively, and the difference in $K_d$ between two strains was statistically significant (p<0.03). The maximal binding capacity $(B_{max})\;was\;34.6{\pm}3.2\;and\;47.4{\pm}4.3\;fmol/10^6$ SHR respectively, and the difference of $(B_{max})$ between two strains was statistically significant (p<0.05). from the above results, it is suggested that the increase of Ca influx via potential-operated Ca channels and the increase of the number of dihydropyridine-sensitive Ca channels contribute to high basal tone of the aortic strips in SHR.

Regulation of Choline Transport by Oxidative Stress at the Blood-Brain Barrier In Vitro Model

  • In the present study, we examined how the transport of choline is regulated at the blood-brain barrier (BBB) under the central nervous system (CNS) cellular damages by oxidative stress using a conditionally immortalized rat brain capillary endothelial cells (TR-BBB), in vitro the BBB model. It was also tested whether the choline uptake is influenced by membrane potential, extracellular pH, protonophore (FCCP) and amiloride in TR-BBB cells. In result, $[^3H]choline$ uptake was inhibited by FCCP and dependent on extracellular pH. The treatment of TR-BBB cells with 20 ng/mL tumor necrosis $factor-{\alpha}$ $(TNF-{\alpha})$, 10 ng/mL lipopolysaccharide (LPS), 100 ${\mu}M$ diethyl maleate (DEM) and 100 ${\mu}M$ glutamate resulted in 3.0-fold, 2.6-fold, 1.8-fold and 2.0-fold increases of $[^3H]choline$ uptake at the respective peak time, respectively. In contrast, hydrogen peroxide and raffinose did not show any significant effects on choline uptake. In addition, choline efflux was significantly inhibited by $TNF-{\alpha}$, LPS and DEM producing cell damage states. In conclusion, the influx and efflux transport system for choline existed in TR-BBB cell line and this process was affected by several oxidative stress inducing agents.
