영지 유래 가노더마논디올의 RAW 264.7 세포주에 대한 항염 효과 (Anti-inflammatory effect of ganodermanondiol from Ganoderma lucidumon RAW 264.7 cells)

  • Owing to its diverse range of bioactive compounds, Ganoderma lucidumhas garnered significant research attention for health promotion and disease prevention. Ganodermanondiol, which has a triterpenoid structure, is one of the major active compounds of G. lucidum. In the present study, the anti-inflammatory effects of ganodermanondiol were investigated to evaluate its usefulness as a functional ingredient. Ganodermanondiol (0.5-2 ㎍/mL) significantly inhibited the production of nitric oxide (NO), the expression of the cytokines tumor necrosis factor (TNF)??and interleukin 6 (IL-6), and the expression of cyclooxygenase (COX)-2 and inducible nitric oxide synthase (iNOS) in lipopolysaccharide-induced RAW 264.7 (murine macrophage) cells. Ganodermanondiol (0.5-2 ㎍/mL) also inhibited the phosphorylation of mitogen-activated protein kinase (MAPK) signal molecules, including p38 and c-Jun N-terminal protein kinase (JNK) in RAW 264.7 cells. Ganodermanondiol significantly inhibited the essential factors involved in the inflammatory responses of RAW 264.7 cells and would, therefore, serve as a potential prophylactic and therapeutic agent for immune-related diseases.

Inhibition of Cytopathic Effect of Human Immunodeficiency Virus-1 by Water-soluble Extract of Ganoderma lucidum

  • To examine components of Ganoderma lucidum for anti-human immunodeficiency virus (HIV) activity, the aqueous extracts of its basidiocarps were separated into high-molecular-weight (HMF) and low-molecular-weight (LMF) fractions. These fractions were used in XTT [2,3-bis (2-methoxy-4-nitro-5-sulfophenyl)-5-[(phenylamino) carbonyl]-2H-tetrazolium hydroxide] antiviral assay which can quantitatively measure cytopathic effects of HIV-1 on CEM, human T lymphoblastoid cell line. The CEM cell line added with serial diluted HMF or LMF was cultured in the absence or presence of HIV-1. The results showed that the LMF of the aqueous extract strongly inhibited cytopathic effect of the target cell induced by HIV-1. When two-fold serially diluted LMF ranging from $40.97{\mu}g/ml$4 to 125.00 .mu.g/ml was added to the virus-free culture system, no toxicity on the target cells was detected in all the concentrations tested. However, when it was added to the HIV-infected culture system, the viabilities of the target cell reached a plateau recovering its viabilities to 71.7% and 82.5% in experiment-1 and -2 at 15.60 .mu.g/ml, respectively. The cell viabilities were then gradually decreased but maintained at more than 50% above 31.20 .mu.g/ml concentration. On the contrary, HMF did not prevent any HIV-induced cytopathic effect at any concentrations tested on this cell line. From these results, negligible toxicities were observed by both HMF and LMF of G. luciolum, and recovery of cell viability in HIV infected target cell was induced only by LMF of the carpophores.

Ganoderma Lucidum Polysaccharides Target a Fas/Caspase Dependent Pathway to Induce Apoptosis in Human Colon Cancer Cells

  • Ganoderma lucidum polysaccharides (GLP) extracted from Ganoderma lucidum have been shown to induce cell death in some kinds of cancer cells. This study investigated the cytotoxic and apoptotic effect of GLP on HCT-116 human colon cancer cells and the molecular mechanisms involved. Cell proliferation, cell migration, lactate dehydrogenase (LDH) levels and intracellular free calcium levels ($[Ca^{2+}]i$) were determined by MTT, wound-healing, LDH release and fluorescence assays, respectively. Cell apoptosis was observed by scanning and transmission electron microscopy. For the mechanism studies, caspase-8 activation, and Fas and caspase-3 expression were evaluated. Treatment of HCT-116 cells with various concentrations of GLP (0.625-5 mg/mL) resulted in a significant decrease in cell viability (P< 0.01). This study showed that the antitumor activity of GLP was related to cell migration inhibition, cell morphology changes, intracellular $Ca^{2+}$ elevation and LDH release. Also, increase in the levels of caspase-8 activity was involved in GLP-induced apoptosis. Western blotting indicated that Fas and caspase-3 protein expression was up-regulated after exposure to GLP. This investigation demonstrated for the first time that GLP shows prominent anticancer activities against the HCT-116 human colon cancer cell line through triggering intracellular calcium release and the death receptor pathway.

영지버섯 추출물을 이용한 제빵의 특성 (Bread Properties Utilizing Extracts of Ganoderma lucidum (GL))

  • 각종 성병 및 항암작용에 탁월한 효과가 입증된 영지버섯을 제빵시 첨가하여 기능성이 부여된 빵을 개발하고자 본 연구를 수행하였다. 영지추출액을 8%까지 첨가함에 따라 쓴맛이 증가하였지만, 1차 발효기간 동안 반죽의 부피는 증가하였고 굽기 손실률이 감소함을 관찰할 수 있었다. 영지추출물을 첨가한 빵의 관능검사결과, 2%첨가시 유의적인 차이가 나타나지 않았으나,4,6,8%를 첨가했을 때 4%는 보통으로 6,8%는 대조구에 비해 나쁘게 나타났다. 사진촬영 결과 역시 0, 2% 영지추출액 첨가식빵의 내부조직은 우수하였으나, 4%는 보통으로 6,8%는 나쁘게 평가되었다. 이러한 사실들로 보아, 일반식 빵과 차이가 없는 2% 영지추출액 첨가시 기능성이 부여된 식빵의 제조가 가능하리라 생각한다.

Analysis of Influence of Environmental Conditions on Ganoderic Acid Content: in Ganoderma lucidum Using Orthogonal Design

  • The influence of environmental conditions on the ganoderic acid (GA) content in the fungus Ganoderma lucidum was investigated using a one-factor-at-a-time design and orthogonal design. Among the various medium components examined, sucrose, soybean powder or peptone, ferrous sulfate, and pH 6.0 were the most suitable carbon source (factor A), nitrogen source (factor B), mineral source (factor C), and initial pH (factor D), respectively, for the GA content in the one-factor-at-a-time design. According to the orthogonal design, the order of effect for the four factors on the GA content was A>C>D>B. The best level of factor A was $A_2$ (sucrose) with a value of +0.34 mg/100 mg DW. The optimal treatment combination was $A_2B_1C_3D_1$ with which the GA content reached up to 2.63$\pm$0.011 mg/100 mg DW. The interactions between the mineral ion and the nitrogen source, and the mineral ion and the pH were both highly significant (P<0.01). The highest interaction effect was ($B_2{\times}D_2$) with a value of +0.19 mg/100 mg DW, which was higher than the level effect value for $B_2$ (peptone) and D$_2$ (pH 5.0). Therefore, the results proved that interactions between factors cannot be ignored. The results also indicated the importance of the interactions between the factors, which may help to understand the metabolic pathway leading to triterpene biosynthesis and the expression and regulation of the key enzymes involved.

LDL Receptor Knockout Mouse에서 영지추출물의 죽상경화증 개선 효과 (Improving Effect of Extract of Ganoderma lucidum in Atherosclerosis from LDL Receptor Knockout Mouse)

  • Objectives : This study was designed to protect effect on atherosclerosis through regulation of low density lipoprotein(LDL) by 70 % ethanol extract Ganoderma lucidum (GL) in LDL receptor knockout mouse (LDLr ko mice) fed Western diet.Methods : The LDLr ko mice were divided into 3 groups ; Control, GL100, and GL300. After grouping, LDLr ko mice were fed Western diet. The GL (100 or 300 mg/kg body weight/day, p.o.) was administered every day for 8 weeks. The body weight and food intake were measured every day. The changes in the levels of alanine aminotransferase (ALT), aspartate aminotransferase (AST), creatinine, triglyceride (TG), total cholesterol (TC), and high-density lipoprotein (HDL) in serum were analyzed after experiment.Results : The LDLr ko mice fed Western diet were increased body weight gain and blood biochemistry parameters such as ALT, AST, TG, TC, and LDL. However GL300 group significantly reduced the body weight. Also TG, TC, and LDL level did not increase. The levels of ALT, AST, HDL were not changed. Also, LDLr ko mice model liver were observed lipid drop, but GL groups did not appear. Futhermore, histological analysis of GL groups aorta tissue were similar to NOR groups.Conclusions : We confirmed that whether GL administration is protect atherosclerosis or not. As the results, blood biochemistry and histological analysis did not changed much in GL administration groups. This study provides scientific evidence that GL protect the atherosclerosis through the reduction of LDL cholesterol. Therefore GL has potential medicine inhibition of atherosclerosis.

HPLC를 이용한 발효 감초의 지표 성분 분석 (Quantitative Analysis of Glycyrrhizic Acid in Fermented Glycyrrhizae Radix by HPLC)

  • The aim of this study was to study the quantitative analysis of glycyrrhizic acid in Glycyrrhizae Radix extract fermented with Paecilomyces japonica, Ganoderma lucidum, honey or Nuruk. The amounts of dry on loss were measured and the quantitative analysis of glycyrrhizic acid was performed by high performance liquid chromatographic (HPLC). HPLC method was performed on C18 column ($250\;mm\;{\times}\;4.6\;mm$, $5\;{\mu}m$, RS tech) using gradient solvent mixtures of water-acetonitrile with photodiode array detector (254 nm). The flow rate was $1.0\;m{\ell}/min$. Retention time of glycyrrhizic acid was about 23.96 min and linearity of calibration was $R^2$=0.9998. Contents of glycyrrhizic acid in Glycyrrhizae Radix extract (control) was $5.048\;{\pm}\;0.14$; Contents of glycyrrhizic acid in Glycyrrhizae Radix extract fermented with Paecilomyces japonica (SDT) was $1.975\;{\pm}\;0.07$; Contents of glycyrrhizic acid in Glycyrrhizae Radix extract fermented with Ganoderma lucidum (SYT) was $2.676 \;{\pm}\;0.07$; Contents of glycyrrhizic acid in Glycyrrhizae Radix extract fermented with honey (SST) was $5.191\;{\pm}\;0.06$; Contents of glycyrrhizic acid in Glycyrrhizae Radix extract fermented with Nuruk (SNT) was $5.305\;{\pm}\;0.34$, respectively. Contents of glycyrrhizic acid in SDT and SYT were decreased but that in SST and SNT was increased when compared to control.

영지버섯에서 추출한 포자오일의 항노화 및 보습 효능 (Anti-aging & Skin Hydration Effects of Spore oil Extracted from Ganoderma lucidum)

  • 본 연구에서는 영지버섯 포자오일(GLS)의 항노화, 항산화, 항염 그리고 보습에 대한 활성 평가를 진행하였다. 항산화 활성 실험에서 GLS은 DPPH 라디칼 소거 활성이 농도 의존적으로 증가하였다. 항염 평가는 LPS를 자극시킨 RAW264.7 세포에서 GLS에 대한 NO, TNF-α 그리고 IL-6 생성물의 억제 효능을 측정한 결과, GLS는 NO 그리고 전염증 사이토카인인 TNF-α, IL-6 생성물을 억제하였다. 또한, procollagen 생성물과 COL1A1 mRNA 발현 분석을 위해 인간 섬유아세포를, 그리고 AQP-3 mRMA 발현 분석을 위하여 인간 각질형성세포를 사용하였다. 그 결과, GLS는 procollagen 생성물과 COL1A1, AQP-3 mRNA 발현을 증가시켰다. 이러한 연구결과는 GLS가 항염, 주름 그리고 보습에 대한 잠재적인 효능을 가지고 있음을 시사한다.
