Screeing and Isolation of Antioxidant from Medicinal Plants

  • On the purpose of develcpement of antioxidative compound from natural sources, medicinal plants known to have antioxidant actuvity have been examined concening DPPH radical scavenging activity and SOD-liked activities. Among 8 plants exhibiting the activity, Houttuynia cordata THUNB was selected as resources to search for active compounds due to rareness of study. The antioxidative compounds from Houttuynia cordata THUNB,quercitrin was assayed using a DPPH free radical. The DPPH radical scavenging activity of quercitrin was similar to that of BHA and Ascorbic acid

Assessment of Radical Scavenging Activity and Phenolic Compounds of Xanthium occidentale

  • Common thistle contains water-soluble substances that are antioxidative to foods. Antioxidant activities measured by DPPH method for the ground samples were the greatest in leaves, although was less than that of commonly used antioxidants, BHT and ascorbic acid. Methanol extracts and fractions from Xanthium occidentale plants dose-dependently increased DPPH free radical scavenging activity, in vitro test. The extracts from leaves showed the strongest antioxidant activity. DPPH scavenging activity of the individual fraction was in order of n-butanol>water>ethyl acetate>n-hexane fraction. By means of HPLC analysis, leaf samples of Xanthium occidentale had the highest amount of phenolic compounds, related with antioxidant activity, and followed by stems and roots. Total content of these antioxidant phenolic com­pounds for leaves extracts were detected in water fraction (36.7 mg 100 $g^{-1}$) as the greatest amount, especially chlorogenic acid (39.4 mg 100 $g^{-1}$) was the greatest component. These results suggest that Xanthium occidentale plants had potent antioxidant activity, and their activities were differently exhibited depending on plant part and fraction.

삼백초(Saururus chinensis) 지상부의 에탄올 추출조건과 특성

  • 삼백초 지상부의 에탄올 추출농도와 추출 시간에 따른 수율, total phenol과 total flavonoid함량, DPPH 라디칼 소거능을 조사하였다. 추출 온도 9$0^{\circ}C$에서 에탄올농도를 증가시키면서 8시간동안 환류추출하였다. 추출수율과 DPPH 라디칼 소거능은 에탄올농도 40%까지는 증가하다가 60%이상에서는 감소하였고 total phenolic compound와 total flavonoid의 함량도 비슷한 경향을 보였다. total phenolic compound의 함량은 40%와 60%에서 큰 차이가 없는 반면 total flavonoid의 함량은 40%에서 더 높았다. total phenolic compound와 total flavonoid 함량, DPPH 라디칼 소거능 및 추출수율은 40% 이상에서는 큰 변화가 없어 추출용매의 조건은 40% 에탄올로 선정하였다. 40% 에탄올로 9$0^{\circ}C$에서 24시간 동안 4시간 간격으로 환류추출하였을 때 total phenolic compound와 total flavonoid 함량, DPPH 라디칼 소거능 및 추출수율은 4시간 이후 큰 차이가 없었다. 삼백초의 용매추출조건으로는 40% 에탄올, 4시간 추출하는 것이 적합하다고 판정된다.

Comparison of Activity of Angelica Gigas and Angelica Acutiloba from Kangwon (강원도 재배 참당귀 및 일당귀의 활성에 대한 비교연구)

  • In genus Angelica, three species have been used and cultivated for medical material in orient, A. gigas in korea, acutiloba in Japan and sinensis in China. The plant material of Angelica spp. is used for the treatment of women's disease as a hematic. The extracts from A. gigas and acutiloba were fractionated aqueous partitions. And study was performed to examine DPPH scavenging activities, BSA degradadion, anti-apoptosis and NO scavenging. DPPH radical scavenger activity was measured by DPPH method, it was shown dose-dependently effect. and BSA degradadion was shown same result. Treatment of cells with hydrogen peroxide, a reactive oxygen species was to indiced cell death and pretreatment with Angelica gigas and angelica acutiloba extract attenuated the occurrence of $H_2O_2$-induced cell death. In vitro nitric oxide (NO) scavenging effect on Angelica gigas and angelica acutiloba extracts. All extracts effectively reduced the generation of NO radicals in a dose-dependant manner.

Antioxidant Properties of Erigeron annuus Extract and Its Three Phenolic Constituents

  • The antioxidant activity of the extract of Erigeron annuus was assessed by means of two different in vitro tests: bleaching of the stable 1,1-diphenyl-2-picrylhydrazyl radical (DPPH test) and the scavenging of authentic peroxynitrite in company with peroxynitrite generation from 3-morpholinosydnonimine (SIN-1). In both tests, the 85% aq. MeOH and n-BuOH soluble fractions of the crude extract showed a significant scavenging effect on peroxynitrite and DPPH radical in comparison to L-ascorbic acid. And bioassay-guided fractionation of the n-BuOH soluble fraction led to the isolation of three compounds: Apigenin (1), quercetin-3-O-glucoside (2), and caffeic acid (3). The structures of the isolated compounds were elucidated on the basis of their spectroscopic data and their antioxidant activities were measured by determining their capacity to scavenge peroxynitrite and the DPPH radical.

Effects of Albizziae Cortex Extracts on the Elastase Activity and DPPH and NO Scavenging Activities

  • Elastic fibers are found in the skin, lungs, arteries, veins and other structures. Elastases destroy the elastic fibers and cause the emphysema and pulmonary hypertension. Oxidative stress is needed for these pathologic changes. Accordingly, present study was designed to investigate the effect of Albizziae Cortex extracts (ACE) on elastase activity and anti-oxidative effects of ACE. The in vitro inhibitory effects on elastase and di(phenyl)-(2,4,6-trinitrophenyl)iminoazanium (DPPH) and nitric oxide (NO) free radical scavenging activities of ACE were measured. The elastase activity was significantly inhibited by ACE. DPPH and NO free radicals were significantly scavenged as well. ACE showed the elastase-inhibiting effects and anti-oxidative activities in vitro. These results suggest that ACE may have potential roles in the treatment of pulmonary emphysema and pulmonary hypertension.

Convergence study on the antioxidant effect of crude extracts of Nelumbo nucifera Gaertner (연밥 조추출물의 항산화 효능에 관한 융합 연구)

  • The present study was performed to evaluate the antioxidant effect of crude extracts of Nelumbo nucifera Gaertner. Antioxidant effect was analysed by DPPH-radical scavenging activity, lipid peroxidation and superoxide dismutase (SOD)-like activity. DPPH-radical scavenging activity and SOD-like activity of linoleic acid, glutamic acid, ethyl acetate crude extract and ethyl alcohol crude extract of Nelumbo nucifera Gaertner were dose-dependently increased. However, lipid peroxidation of glutamic acid, linoleic acid, ethyl acetate crude extract and ethyl alcohol crude extract of Nelumbo nucifera Gaertner were time-dependently decreased. The data suggests that crude extracts of Nelumbo nucifera Gaertner may be a putative antioxidant substance and apply the development of medicine through convergence study.

Varietal Variation in Antioxidative Activity of Rice Grain by DPPH and TBA Methods

  • This study was to investigate antioxidative activity of rice grain using 1, 1-Diphenyl-2-picrylhydrazyl(DPPH) and thiobarbituric acid (TBA) method and germination ability for screening rice varieties with high antioxidative activities on korean native and foreign rice varieties harvested in 1998 and 1999. The average antioxidative activity of foreign rice varieties (DPPH 63.5% and TBA 55.2%) was significantly higher than that of native rice varieties (DPPH 47.2% and TBA 45.6%) on varieties harvested in 1999. The promptness index (PI) of native rice varieties was higher in stored rices for three months (mean PI=160.7) than that of stored rices for a year (mean PI=141.6). On the other hand, the PI of foreign rice varieties was higher in stored rices stored for a year (mean PI=176.7) than that of stored rices for three months (mean PI=157.5). Varieties with high redness of hulled rice (a-value) showed significant lipid peroxidation inhibitory activity to DPPH in a stored rices for a year (r=${0.5744}^**$) and stored rices for three months (r=${0.5630}^**$) . These results indicate that the pigments of hulled rice varieties may play important antioxidative roles and colored rice varieties with higher antioxidative potentials can be developed and also may provide information with rice breeder to breed rice variety with a high antioxidative activity for a rapid screening of a small amounts of a large number of samples using color value.

Screening for Antioxidative Activity of Oriental Medicinal Plant Materials (시판 한약재에 대한 항산화 활성 검정)

  • Antioxidative effect of 42 medicinal plant extracts was screened to search natural antioxidants. The extracts of Terminalia chebula, Caesalpinia sappan, Sanguisorba officinalis, Rubus coreanus, and Alpinia katsumadai showed strong inhibition effect on DPPH radical, and LDL oxidation. Those of Eugenia caryophyllata, Alpinia officinarum, Zingiber officinale, Xanthium strumarium, Sophora japonica, Aristolochia contorta, and Alpinia katsumadai exhibited potent antioxidative activity on linoleic acid peroxidation. Among 42 medicinal plant materials, the extract of Terminalia chebula showed the highest scavenging activity $(3.08\;{\mu}g/ml)$ on DPPH radical which was higher than that $(13.52\;{\mu}g/ml)$ of ${\alpha}-tocopherol$. The extract of Terminalia chebula showed also stronger inhibition activity on LDL oxidation than that of ${\alpha}-tocopherol$ did.
