MICROTENSILE BONDING OF RESIN FIBER REINFORCED POST TO RADICULAR DENTIN USING RESIN CEMENT (레진 시멘트를 이용한 레진 파이버 강화 레진포스트의 치근 상아질에 대한 미세인장결합강도)

  • Object The purpose of this study were to evaluate the microtensile bond strength of resin fiber reinforced post to radicular dentin using resin cement according to various dentin surface treatment and to observe the inter face between post and root dentin under SEM Material and Method A total 16 extracted human single rooted teeth were used. A lingual access was made using a #245 carbide bur in a high-speed handpiece with copious air water spray. The post space was mechanically enlarged using H-file(up to #60) and Gates Glidden bures(#3). This was followed by refining of the canal space using the calbrating drill set provided in ER Dentinpost(GEBR, BRASSELER GmbH&Co. KG). The 16 teeth were randomly distributed into 4 group of 4 teeth. Group 1 teeth had their post space prepared using 10% phosphoric acid as root canal surface treatment agent during 20s. The canal was then rinsed with saline and dried with paper point. Group 2 teeth had their post space prepared using 3% NaOCl as root canal surface treatment agent during 30min. The canal was then rinsed with saline and dried with paper point. Group 3 teeth had their post space prepared using 17% EDTA as root canal surface treatment agent during 1min. The canal was then rinsed with saline and dried with paper point. Group 4 teeth had their post space prepared using 17% EDTA as root canal surface treatment agent during 1min. After rinsing with saline, the canal was rinced 10m1 of 3% NaOCl for 30min. After drying with paper point, the post(ER Dentinpost, GEBR, BRASSELER GmbH&Co. KG) was placed in the treated canals using resin cement. Once the canal was filled with resin cement(Super bond C&B sunmedical co. Ltd.), a lentulo was inserted to the depth of the canal to ensure proper coating of the root canal wall. After 24 hours, acrylic resin blocks($10{\cdot}10{\cdot}50mm$) were made. The resin block was serially sectioned vertically into stick of $1{\cdot}1mm$. Twenty sticks were prepared from each group. After that, tensile bond strengths for each stick was measured with Microtensile Tester. Failure pattern of the specimen at the interface between post and dentin were observed under SEM. Results 1. Tensile bond strengths(meen{\pm}SD$) ) were expressed with ascending order as follows group 4, $12.52{\pm}6.60$ ; group 1, $7.63{\pm}5.83$ ; group 2, $4.13{\pm}2.31$ ; group 3, $3.31{\pm}1.44$. 2. Tensile bond strengths of Group 4 treated with 17% EDTA +3%NaOCl were significant higher than those of group 1, 2 and 3 (p<0.05). 3. Tensile bond strengths of Group 1 treated with 10% phosphoric acid were significant higher than those of group 2 (p<0.05). Tensile bond strengths of Group 4 treated with 17% EDTA +3% NaOCl was significant higher than those of other groups.

Anti-obesogenic Effect of Brassica juncea Extract on Bisphenol-A Induced Adipogenesis of 3T3-L1 Cells (비스페놀 A (Bisphenol-A)로 유도된 지방세포 분화에 미치는 갓 추출물의 항오비소겐 효과)

  • The purpose of the study was to investigate the content of sinigrin, an index component, in Brassica juncea extract and to evaluate the differentiation of lipocytes, inhibition of production of reactive oxygen species (ROS) and reduction of protein production by lipogenic factors (PPARγ, C/EBPα, aP2) in the processing of Brassica juncea extract and sinigrin in 3T3-L1 preadipocytes which induces Bisphenol A (BPA), an endocrine disrupting environmental hormone. From the investigation, the content of sinigrin in Brassica juncea extract, measured by HPLC, is found to be 21.27±0.2 mg/g. The XTT assay result on BPA-derived 3T3-L1 adipocytes shows there is no cytotoxicity found from 180 µM of sinigrin and 300 ㎍/mL of Brassica juncea extract. Moreover, both intracellular lipid accumulation and ROS production during differentiation of lipocyte are significantly reduced in cells processed with Brassica juncea extract and sinigrin. Lastly, it was also found that the production of transcription factors of lipocyte differentiation, PPARγ, C/EBPα and aP2, were found to be suppressed by the application of Brassica juncea extract and sinigrin. Such results reveals that Brassica juncea is effective in not only suppressing lipid accumulation in the environmental hormone bisphenol A-derived lipocyte, but also in reducing the ROS. The sinigrin-containing Brassica juncea is highly expected to be used in natural functional supplements that prevents the lipid metabolism disorders caused by BPA. There are necessities for additional clinical research and follow-up studies on the in vivo model to verify the relevant mechanisms.

Effects of osteotropic hormones on the nitric oxide production in culture of ROS17/12.8 cells (뼈흡수유도호르몬이 ROS17/2.8세포로부터 Nitric Oxide 형성에 미치는 영향)

  • Purpose : We performed the present study to investigate whether osteotropic hormomes play roles on the nitric oxide (NO) production in culture of ROS 17/12.8 osteoblastic cells. Materials and Methods : The osteoblastic cell line ROS17/2.8 cells were cultured In F12 medium supplemented with $5\%$ fetal bovine serum (FBS) at $37^{\circ}C$ in a humidified atmosphere of $5\%\;CO_2$ in air. ROS17/2.8 cells were plated in 96-well plates at a density of $2-3\times10^3cells/well$ and grown to confluence. Then the cells were pretreated with osteotropic hormones (parathyroid hormone (PTH) 20-500 ng/mL, 1, 25-dihydroxycholecalciferol $(1,\;25[OH]_2D_3)$ 1-100 nM; prostaglandin $E_2 (PGE_2)$ 20-500 ng/mL in the medium supplemented with $0.4\%$ FBS for 72 hours and the cells were treated with cytokines $(TNF{\alpha}\;and\;IFN{\gamma})$ in phenol red-free F12 medium for an additional 48 hours. NO synthesis was assessed by measuring the nitrite anion concentration, the reaction product of NO, in the cell culture medium using Griess reagent. Results : PTH and $1,\;25[OH]_2D_3$ pretreatment induced a significant increase in NO production in the presence of $TNF{\alpha}\;and\;IFN{\gamma}.\;PGE_2$ slightly induced NO production compared to the control group. But, $PGE_2$ pretreatment did not affect in NO production in the presence of $TNF{\alpha}\;and\;IFN{\gamma}$. Conclusions : These results suggest that the actions of osteotropic hormones In bone metabolism may be partially mediated by NO in the presence of cytokines.

Combined Treatment of Activin A and Heparin Binding-EGF (HB-EGF) Enhances In Vitro Production of Bovine Embryos

  • This study was carried out to investigate the effects of tissue inhibitor of matalloproteinase-1 (TIMP-1), Activin A and Heparin binding epidermal growth factor (HB-EGF) on in vitro production of bovine embryos. In experiment 1, presumptive zygotes were cultured in the medium supplemented with TIMP-1 ($0.5{\mu}g/ml$), Activin A (100 ng/ml), or HB-EGF (100 ng/ml) at $39^{\circ}C$ in a humidified atmosphere of 5% (v/v) $CO_2$, 5% (v/v) $O_2$ and 90% (v/v) $N_2$. In experiment 2, TIMP-1 + HB-EGF or Activin A + HB-EGF combinations were supplemented in the culture medium. The developmental rate to blastocysts, hatching rate and total cell numbers of the blastocysts were evaluated in both experiments. The embryos cultured in medium without growth factor supplementation was used as control group. In experiment 1, the embryos cultured in medium supplemented with TIMP-1 and Activin A showed significantly higher developmental rate to blastocysts than those cultured with HB-EGF and control (36.9%, 34.1%, 21.2% and 23.1%, respectively) (P<0.0001). However, the hatching rate of blastocyst was significantly higher in embryos with HB-EGF than those with TIMP-1, Actvin A and Control groups (84.4%, 58.8%, 51.4% and 49.3%, respectively) (P<0.001). Total cell number per blastocyst was also significantly higher in embryos with HB-EGF group ($174.3{\pm}2.5$) than those with TIMP-1, Activin A (149.7 and 150.0, respectively) (P<0.05) and Control (119.0) (P<0.001). In experiment 2, embryos cultured with combined treatment of Activin A and HB-EGF resulted in significantly higher rates of blastocysts formation (48.0%), hatching rate (89.7%) and total cell number in blastocyst ($182.3{\pm}2.1$) than those with TIMP-1 and HB-EGF combination group (32.0%, P<0.001; 76.6%, P<0.05; $165.7{\pm}4.2$, P<0.001, respectively). Our data demonstrate that in vitro production of bovine embryos could be improved by combined supplementation of Activin A and HB-EGF in culture medium.

Effects of O2 Plasma Pre-treatment and Post-annealing Conditions on the Interfacial Adhesion Between Ti Thin Film and WPR Dielectric (O2 플라즈마 전처리 및 후속 열처리 조건이 Ti 박막과 WPR 절연층 사이의 계면 접착력에 미치는 영향)

  • The effects of O2 plasma pre-treatment and post-annealing conditions on the interfacial adhesion of Ti thin film and WPR dielectric were investigated using 90° peel test for fan-out wafer level packaging (FOWLP) redistribution layer (RDL) applications. Peel strength between Ti film and WPR dielectric decreased from 8.9±1.3 g/mm to 2.7±0.9 g/mm for variation of O2 plasma pre-treatment time from 30s to 300s, which is closely related to C-O-C or C=O bonds breakage at the WPR dielectric surface due to excessive plasma pre-treatment conditions. During post-annealing at 150℃, the peel strength abruptly decreased from 0 h to 24 h, and then maintained constant until 100 h, which is also mainly due to the damage of WPR dielectric which is weak to high temperature. Therefore, the optimum plasma pre-treatment conditions on the surface of dielectric is essential to interfacial reliability of FOWLP RDL.

  • Electrum-sulfide mineralization of the Samgwang and Sobo mines of the Cheongyang Au-Ag area was deposited in two stages of quartz and calcite veins that fill fault zones in granite gneiss. Radiometric dating indicates that mineralization is Early Cretaceous age (127 Ma). Fluid inclusion and sulfur isotope data show that ore mineralization was deposited at temperatures between $340^{\circ}$ and $180^{\circ}C$ from fluids with salinities of 1 to 8 wt. % equiv. NaCl and a ${\delta}^{34}S_{{\sum}S}$ value of 2 to 5 per mil. Evidence of fluid boiling (and $CO_2$ effervescence) indicates a range of pressures from < 200 to $\approx$ 700 bars, corresponding to depths of ${\approx}1.5{\pm}0.3\;km$ in a hydrothermal system which alternated from lithostatic toward hydrostatic conditions. Au-Ag deposition was likely a result of boiling coupled with cooling. Meaured and calculated hydrogen and oxygen isotope values of ore-forming fluids indicate a significant meteoric water component, approaching unexchanged paleometeoric water values. Comparison of these values with those of other Korean Au-Ag deposits reveals a relationship among depth, Au/Ag ratio and degree of water-rock interaction. All investigated Korean Jurassic and Cretaceous gold-silver-bearing deposits have fluids which are dominantly evolved meteoric waters, but only deeper systems (${\geq}1.5\;km$) are exclusively gold-rich.

A Retrospective Study on the Effect of the Co-Administration of Ojeok-san and Hypoglycemic Agents on Blood Glucose Levels in Type 2 Diabetes Mellitus (오적산 병용투여가 혈당강하 치료를 받고 있는 제2형 당뇨병 환자의 혈당에 미치는 영향)

  • Objective: This study was conducted to investigate the hypoglycemic effect and safety of Ojeok-san in patients with type 2 Diabetes Mellitus. Methods: We investigated type 2 diabetes mellitus patients at Kyung-Hee University Korean Medical Hospital who were administered Ojeok-san for at least one day between January 2012 and September 2020, basal characteristics and laboratory tests were reviewed retrospectively. The hypoglycemic effect of Ojeok-san was assessed by comparing fasting blood sugar (FBS) and two hours post-prandial plasma glucose (PP2) levels from before and after taking Ojeok-san. Subgroup analyses were conducted according to baseline hypoglycemic treatments and glycated hemoglobin levels (< or ≥6.5%). The safety of Ojeok-san was assessed by comparing levels of aspartate aminotransferase, alanine aminotransferase, γ-glutamyl transferase, blood urea nitrogen, and creatinine. Results: After Ojeok-san administration, FBS and PP2 were significantly reduced to an average of 14.33 mg/dL and 27.67 mg/dL respectively. In the subgroup analysis, PP2 in patients receiving metformin mono therapy was significantly reduced to 28.67 mg/dL, and those receiving a dual therapy of metformin and DPP-4 inhibitor, it was significantly reduced to 32.33 mg/dL. In patients with glycated hemoglobin of lower than 6.5%, FBS was significantly reduced to 12.20 mg/dL, and both FBS and PP2 were significantly reduced, to 15.50 mg/dL and 40.00 mg/dL, respectively, in those with glycated hemoglobin levels of more than 6.5%. The safety profile showed no significant difference after Ojeok-san administration. Conclusions: Ojeok-san has significant hypoglycemic effects in patients with type 2 diabetes mellitus who are also taking hypoglycemic agents.

  • Objective: This study aimed to assess the safety and effect on glucose level of Bojungikgi-tang in patients with type 2 diabetes mellitus. Methods: To review patients' clinical characteristics and laboratory tests retrospectively, we investigated 15 hospitalized patients with type 2 diabetes mellitus who took Bojungikgi-tang at Kyung Hee University Korean Medical Hospital for at least one day between January 2012 and December 2022. The blood glucose levels, including fasting blood sugar (FBS), 2-hour postprandial glucose (PP2) levels, and glycated hemoglobin level, were collected to determine the effect of the Bojungikgi-tang on blood sugar changes. Furthermore, to evaluate the safety of Bojungikgi-tang, hepatic function and renal function tests were implemented, including aspartate aminotransferase, alanine aminotransferase, alkaline phosphatase, γ-glutamyltransferase, blood urea nitrogen, creatinine, and Modification of Diet in Renal Disease estimated glomerular filtration rate. Results: When Bojungikgi-tang and a standard treatment for diabetes were administered for patients with type 2 diabetes, it was confirmed that there were no statistically significant changes in FBS and PP2 levels in the analysis of each medication taken. There was no significant difference in the safety profile after taking Bojungikgi-tang. Conclusions: The combined administration of Bojungikgi-tang with standard hypoglycemic medication for patients with type 2 diabetes may not affect blood glucose levels and safety.

Expression of TRPV1 and iNOS in the Dorsal Root Ganglion Exposed by Autologous Nucleus Pulposus in the Rat

  • Purpose: To determine whether upregulation of inducible nitric oxide synthase (iNOS) transcription and translation is related to radicular pain in a model of lumbar disc herniation. Also, to investigate the temporal changes of mRNA expression of iNOS and the identity of iNOS and transient receptor potential vanilloid (TRPV) 1 channel expression cells in dorsal root ganglion (DRG) of a model of lumbar disc herniation. Methods: A lumbar disc herniated rat model was developed by implantation of the autologous nucleus pulposus, harvested from the coccygeal vertebra of each tail, on the left L5 nerve root just proximal to the DRG. Rats were tested for mechanical allodynia of the plantar surface of both hind paws 2 days before surgery and 1, 5, 10, 20 and 30 days postoperatively. Reverse transcription polymerase chain reaction (RT-PCR) was used to follow iNOS mRNA expression. To stain iNOS and TRPV1 in DRG, an immunohistochemical study was done 10 days after surgery. Results: A significant drop in mechanical withdrawal threshold on the ipsilateral and contralateral hind paws was observed 1 day after surgery and was prolonged to 30 days in rats with lumbar disc herniation. The expression of mRNA for iNOS peaked at postoperative day 10 on both sides of the DRG. iNOS-positive sensory neurons in the DRG varied in size from large to small diameter cells. A majority of small and intermediate sensory neurons were TRPV1-positive cells. Double immunofluorescence staining for TRPV1 and iNOS revealed that most intermediate TRPV1-positive sensory neurons co-localized with iNOS-positive neurons. Conclusion: Nucleus pulposus-induced mechanical allodynia can be generated without mechanical compression. This pain is related to temporal changes in expression of iNOS mRNA in the DRG. Co-localization of TRPV1 and iNOS in intermediate neurons of the DRG is correlated with pain modality and intensity.

A Study on the Development of Floor-Fixed Standpipe Sway Brace for Narrow Space (협소공간전용 바닥고정형 입상관 흔들림방지버팀대 개발에 관한 연구)

  • This paper proposes a solution to the problems of constructing and installing sway braces for existing standpipes in narrow spaces and pits. The study develops a floor-fixed sway brace for a narrow space that can support the ground area under horizontal seismic loads (X-axis, Y-axis) as well as vertical seismic loads (Z-axis). The results of structural analysis using SolidWorks simulation showed that the eccentric load was generated in the first design according to the anchored position along the vertical direction, and the problem of exceeding the allowable stress of the material along the horizontal and vertical directions. In the second design model, deformation caused by the eccentric load along the vertical direction, similar to the first design model, did not occur. The maximum strain rate was 0.17%, which is approximately 12.84% less than the first design model (Maximum strain rate of 13.01%). It was confirmed that the structural stability and durability improved. Compressive and tensile load testing of the prototypes showed that all of them meet the performance criteria of the standard.
