Simultaneous Detection of 10 Foodborne Pathogens using Capillary Electrophoresis-Based Single Strand Conformation Polymorphism

  • This report outlines the development of a rapid, simple, and sensitive detection system for pathogenic bacteria using a capillary electrophoresis-based, single strand conformation polymorphism (CE-SSCP) combined with PCR. We demonstrate that this method, used with primers targeting the V4 region of the16S rRNA gene, is capable of the simultaneous detection of 10 microbes that could be associated with foodborne illness, caused by animal-derived foods: Salmonella enterica, Listeria monocytogenes, Escherichia coli O157:H7, Campylobacter jejuni, Staphylococcus aureus, Bacillus cereus, Clostridium perfringens, Yersinia enterocolitica, Vibrio parahaemolyticus, and Enterobacter sakazakii. The traditional detection techniques are time-consuming and labor-intensive, due to the necessary task of separate cultivation of each target species. As such, the CE-SSCP-PCR method, that we have developed, has the potential to diagnose pathogens rapidly, unlike the traditional technique, in order to prevent foodborne illness in a much more efficient manner.

  • The effects of partial pressures of various gases on the qualities of coated conductors are studied, where the long samples were fabricated in a vacuum of a large physical vapour deposition system. The various gases such as $0_25,$H_2$,H_20$,$C_n H_m$, and CO come out from the large heating system and gives somewhat complicated effects on the interfacial chemistry at CeO/Ni which Is Important for the epitaxial growth of the film. The results of XRD measurements on the films fabricated in the controlled partial pressures of thoes gases will be presented.

열간금형용강의 희토류붕화처리에 관한 연구 (A Study on the Rare-earth Boronizing Treatment of STD 61 Steel)

  • The boronizing effects of STD 61 steel have been studied on the micro structure and hardness. The STD 61 Steel was soaked in molten salt, consisted of KCl, $BaCl_2$, NaF, $B_2O_3$, FeB, and Ce, at various temperatures and times. The boronizing conditions for the peak hardness were the temperature range of $900^{\circ}C$ to $950^{\circ}C$ for 5 hr and that of $1000^{\circ}C$ for 3 hr, respectively. Four boride layers such as FeB, $Fe_2B$, ${\alpha}$ and matrix layer surface were observed from the microscopic surface examination. The thickness of boride layer was increased by increasing the boronizing time and the temperature. The structure of boride layer was tooth shape.

Solubility of Mixed Lanthanide Hydroxide and Oxide Solid Solutions

  • The solubilities of different multicomponent lanthanide oxide (Ln2O3) solid solutions including binary (Ln1 and Ln2 = La, Nd, Eu, or Tm), ternary (Ln1, Ln2, and Ln3 = La, Nd, Eu, or Tm), and higher systems (Ln = La, Ce, Pr, Nd, Sm, Eu, Gd, Tb, Dy, Ho, Er, Tm, Yb, and Lu) were studied after aging for four weeks at 60℃. Our recent study revealed that the phase transformations in binary ((La, Nd) and (La, Eu)) and ternary (La, Nd, Eu) systems are responsible for the formation of (La, Nd)(OH)3, (La, Eu)(OH)3, and (La, Nd, Eu)(OH)3 solid solutions, respectively. The variations in the mole fractions of La3+, Nd3+, and Eu3+ in the sample solutions of these hydroxide solid solutions indicated that a thermodynamic equilibrium might account for the apparent La, Nd, and Eu solubilities. Conversely, the binary and ternary systems containing Tm2O3 as the heavy lanthanide oxide retained the oxide-based solid solutions, and their solubility behaviors were dominated by their congruent dissolutions. In the higher multicomponent system, the X-ray diffraction patterns of the solid phases, before and after contact with the aqueous phase indicated the formation of a stable oxide solid solution and their solubility behavior was explained by its congruent dissolution.

Heat Treatment Effects on the Phase Evolutions of Partially Stabilized Grade Zirconia Plasma Sprayed Coatings

  • Partially stabilized zirconia (PSZ) is an attractive material for thermal barrier coating. Zirconia exists in three crystallographic phases: cubic, tetragonal and monoclinic. Especially, the phase transformation of tetragonal phase to monoclinic phase accompanies significant volume expansion, so this transition generally results in cracking and contributes to the failure of the TBC system. Both the plasma sprayed ZrO$_2$-8Y$_2$O$_3$ (YSZ) coat and the ZrO$_2$,-25CeO$_2$,-2.5Y$_2$O$_3$ (CYSZ) coat are isothermally heat -treated at 130$0^{\circ}C$ and 150$0^{\circ}C$ for 100hr and cooled at different cooling rates. The monoclinic phase is not discovered in all the CYSZ annealed at 130$0^{\circ}C$ and 150$0^{\circ}C$. In the 150$0^{\circ}C$ heat-treated specimens, the YSZ contains some monoclinic phase while none exists in the 130$0^{\circ}C$ heat-treated YSZ coat. For the YSZ, the different phase transformation behaviors at the two temperatures are due to the stabilizer concentration of high temperature phases and grain growth. For the YSZ with 150$0^{\circ}C$-100hr annealing, the amount of monoclinic phase increased with the slower cooling rate. The extra oxygen vacancy, thermal stress, and c to t'phase transformation might suppress the t to m martensitic phase transformation.

  • $AB'_{0.5}B"_{0.5}O_3$ type complex perovskite structures which have been reported as proton conductors over $600^{\circ}C$ were studied. The $AB'_{0.5}B"_{0.5}O_3$ type complex perovskite structure is known to be more easily synthesized and has better stability than normal $ABO_3$ perovskite structure. And it is stable at about $800^{\circ}C$ in the $CO_2$ atmosphere, whereas the $BaCeO_3$ perovskite is easily decomposed into carbonate. In addition, this $AB'_{0.5}B"_{0.5}O_3$ type complex perovskite structure could simply produce oxygen vacancies within their structure not by introducing additional doping oxides but by just controling the molar ratio of $B'^{+3}$ and $B"^{+5}$ metal ions in the B site. Hence it is easy to design the structure which shows highly sensitive electrical conductivity to humidity. In this study, the single phase boundary of $BaR_{0.5+x}Ta_{0.5-x}O_{3-{\delta}}$(R = rare earth) complex perovskite structures and it's phase stability were investigated with changes in composition, x. And the humidity dependance of electrical conductivity at different $P_{H2O}$ conditions was investigated.

  • This research project focused on the production of hydrogen through ammonia decomposition reactions while investigating how the reactivity of this process varies when employing different catalysts. Several metal oxide supports (Al2O3, La2O3, CeO2) were utilized as catalysts, with active metals from both the transition metal group (Co, Ni, Fe, Cr, Cu) and the noble metal group (Ru, Rh, Pd, Pt) impregnated onto these supports. Furthermore, the study examined how the reactivity evolves with changes in reaction temperature when employing the prepared catalysts. Additionally, the research delved into the distinctive activation energies associated with each of the catalysts. In this research, In the noble metal catalyst system, the order of high activity for ammonia decomposition reaction to produce hydrogen is Ru > Rh > Pt ≈ Pd. In the transition metal catalyst system, the order of high activity is Co > Ni > Fe > Cr > Cu.

The developments of heavy hydrocarbon reformer for SOFC

  • Heavy hydrocarbon reforming is a core technology for "Dirty energy smart". Heavy hydrocarbons are components of fossil fuels, biomass, coke oven gas and etc. Heavy hydrocarbon reforming converts the fuels into $H_2$-rich syngas. And then $H_2$-rich syngas is used for the production of electricity, synthetic fuels and petrochemicals. Energy can be used efficiently and obtained from various sources by using $H_2$-rich syngas from heavy hydrocarbon reforming. Especially, the key point of "Dirty energy smart" is using "dirty fuel" which is wasted in an inefficient way. New energy conversion laboratory of KAIST has been researched diesel reforming for solid oxide fuel cell (SOFC) as a part of "Dirty energy smart". Diesel is heavy hydrocarbon fuels which has higher carbon number than natural gas, kerosene and gasoline. Diesel reforming has difficulties due to the evaporation of fuels and coke formation. Nevertheless, diesel reforming technology is directly applied to "Dirty fuel" because diesel has the similar chemical properties with "Dirty fuel". On the other hand, SOFC has advantages on high efficiency and wasted heat recovery. Nippon oil Co. of Japan recently commercializes 700We class SOFC system using city gas. Considering the market situation, the development of diesel reformer has a great ripple effect. SOFC system can be applied to auxiliary power unit and distributed power generation. In addition, "Dirty energy smart" can be realized by applying diesel reforming technology to "Dirty fuel". As well as material developments, multidirectional approaches are required to reform heavy hydrocarbon fuels and use $H_2$-rich gas in SOFC. Gd doped ceria (CGO, $Ce_{1-x}Gd_xO_{2-y}$) has been researched for not only electrolyte materials but also catalysts supports. In addition, catalysts infiltrated electrode over porous $La_{0.8}Sr_{0.2}Ga_{0.8}Mg_{0.2}O_3-{\delta}$ and catalyst deposition at three phase boundary are being investigated to improve the performance of SOFC. On the other hand, nozzle for diesel atomization and post-reforming for light-hydrocarbons removal are examples of solving material problems in multidirectional approaches. Likewise, multidirectional approaches are necessary to realize "Dirty energy smart" like reforming "Dirty fuel" for SOFC.

메탄 부분산화반응 촉매에 La 첨가 및 제조방법에 따른 촉매활성에 미치는 영향 (Effects of La Addition and Preparation Methods on Catalytic Activities for Methane Partial Oxidation Catalysts)

  • 메탄의 부분산화에 의하여 합성가스를 제조하였다. 저렴하며 본 반응에 활성이 좋은 것으로 알려진 Ni을 활성 물질로, 우수한 산소 저장능력과 높은 산화 환원 특성을 지닌 $CeO_2$를 담체로 하여, 함침법과 우레아법으로 촉매를 제조하였다. 반응은 고정층 반응기를 이용하여 1 atm, $650{\sim}800^{\circ}C$에서 실시하였다. 표면적 측정 장치를 이용하여 촉매 제조법에 따른 표면적 차이를 비교해 본 결과 우레아법으로 제조한 촉매가 함침법으로 제조한 촉매보다 표면적이 약 11배 이상 큼을 알 수 있었고, SEM으로 표면 구조를 조사해본 결과 우레아법으로 제조한 촉매가 훨씬 더 미세하고 균일함을 알 수 있었다. 우레아법으로 제조한 촉매가 함침법으로 제조한 촉매보다 더 높은 메탄 전환율 및 합성가스 선택도를 나타내었다. 본 반응의 문제점인 탄소 침적을 줄이기 위하여 La을 첨가하여 탄소 침적에 미치는 La의 영향을 알아보았다. TGA(열중량 분석기) 분석 결과 La이 첨가되지 않은 촉매에는 약 16%의 탄소 침적이, La을 첨가한 촉매에는 약 2%의 탄소 침적이 형성되었다. 따라서 La 첨가는 탄소 침적을 줄여서 촉매 비활성화를 막은 것으로 추정된다.

아말감과 복합레진의 수복 과정과 수복 후 발생하는 상아세관액 흐름의 실시간 측정 (REAL-TIME MEASUREMENT OF DENTINAL TUBULAR FLUID FLOW DURING AND AFTER AMALGAM AND COMPOSITE RESTORATIONS)

  • 본 연구의 목적은 아말감과 복합레진의 수복 과정과 수복 후 상아세관액의 흐름(dentinal fluid flow, DFF)을 측정하기 위함이다. 치근을 절제한 제 3 대구치를 자체 제작한 미세 유체 흐름 측정장치에 연결한 후 1급 와동을 형성하여 아말감과 복합레진 수복을 시행하였다. DFF의 측정은 와동 형성부터 수복 후 30분까지 연속적으로 이루어졌고, 수복 후 3, 7일에 재 측정하였다. 주수 하에 와동 형성 시 DFF는 inward로, 끝난 후에는 outward로 바뀌었다. 아말감 충전 시 DFF는 inward로 바뀌었고 충전 완료 후 미약한 outward DFF를 보였다. 복합레진 수복의 경우, 산 처리 후 수세와 건조 시 각각 inward와 outward DFF를 보였고 primer도포 후 공기 분사는 급격한 outward, 소수성 본딩제를 적용하는 단계에서는 outward 였던 DFF가 감소하여 0에 가까워지거나 약간 inward 흐름을 보였다. 접착제와 복합레진의 광중합은 급격한 inward의 DFF를 일으켰다. 수복 후 30분, 3일, 7일째 수복 재료에 따른 DFF의 감소에 통계적으로 유의한 차이는 없었다 (p>0.05).
