Effects of Protein Kinases on Phospholipase C Activation and Intracellular $Ca^{2+}$ Mobilization Induced by Endothelin-1 (Endothelin-1에 의한 phospholipase C 활성화와 세포내 $Ca^{2+}$ 이동에 미치는 protein kinase들의 효과)

  • To investigate the effects of protein kinases on endothelin-1-induced phospholipase C activation and $Ca^{2+}$ mobilization in Rat-2 fibroblast, we measured the formation of inositol phosphates and intracellular $Ca^{2+}$ concentration with [$^3$H]inositol and Fura-2/AM, respectively. Endothelin-1 dose-dependently activated phospholipase C and increased intracellular $Ca^{2+}$ concentration. Protein kinase C activator PMA, significantly inhibited both phospholipase C activity and $Ca^{2+}$ mobilization induced by endothelin-1. Tyrosine kinase inhibitor, genistein, inhibited both. On the other hand, cyclic nucleotide (cAMP and cGMP) did not have any influence on the signaling pathway of phospholipase C-Ca$^{2+}$ mobilization induced by endothelin-1. These results suggest that protein kinase C and tyrosine kinase counteract on the signaling pathway of phospholipase C-Ca$^{2+}$ mobilization induced by endothelin-1 in Rat-2 fibroblast. fibroblast.

An Effect of $Al_{2}O_{3}$ on the Reaction between Molten Converter Slag and CaO pellet (용융전로(熔融轉爐)슬래그와 CaO펠렛의 상호반응(相互反應)에 미치는 $Al_{2}O_{3}$의 영향(影響))

  • As a basic study on the conversion of molten converter slag to the ordinary portland cement, the effects of $Al_{2}O_{3}$ addition on the interface reaction between solid CaO and molten converter slag has been studied. Alumina added converter slag whose basicity was controlled to 1 and 2 was melted and hold for 30 minutes in MgO crucible at $1500^{\circ}C$. Then sintered CaO pellet heated at the same temperature was dipped into the molten slag and held for 30minutes. After the reaction, the crucible was cooled in air and the specimen was cut off to the horizontal direction of the crucible. The dissolution rate of CaO pellet with the addition of $Al_{2}O_{3}$ was measured by the change of the radius or sintered CaO pellet and the interface layer was observed by SEM/EDX. As a result. At the basicity 2 slag, thickness of created $C_{3}S$ layer increased 3.5 times and quantity of $C_{6}AF_{2}\;or\;C_{4}AF$ phase increase 2 times than baisicy 1 slag.

  • This study was carried out to determine how the water soluble, the available $P_2O_5$ and urea-N would change in the course of time, when the mixtures of calcium superphosphate and urea with lime for fertilizer which occurred in Korea and largely contained calcium carbonate were made. Three kinds of materials, i. e., calcium superphosphate, urea and lime for fertilizer were used in this study. Three kinds of mixed fertilizer, i. e., A, B and C were made up by mixing these materials to satisfy the following formula. $$1)\;Ca(H_2PO_4)_2+CaCO_3+CO(NH_2)_2{\rightarrow}$$$$Ca_2H_2(PO_4)+H_2CO_3+NH_3$$ $$2)\;Ca(H_2PO_4)_2+CaCO_3+CO(NH_2)_2{\rightarrow}$$$$Ca_3(PO_4)_2+H_2CO_3+NH_3$$ $$3)\;Ca(H_2PO_4)_2+CaCO_3+CO(NH_2)_2{\rightarrow}$$$$Ca_3(PO_4)_2+H_2CO_3+CaCO_3+NH_3$$ A,B and C were placed in desiccators respectively a six month period. During the time of storage, the water soluble, the available phosphoric acid and urea-N were measured once a month, seven times with the control measurement. The results may be summarized as follows. 1. None of A, B and C showed any change in the urea-N with the lapse of time. This fact indicated that the combination of calcium superphosphate and urea with lime for fertilizer was not unfavourable. 2. A, B and C decreased in the amount of water soluble $P_2O_5$ with the passage of time. This fact indicated that the mixing of calcium superphosphate and urea with lime for fertilizer was unfeasible. 3. The available $P_2O_5$ in any of A,B and C did not undergo a change as time went by. This fact suggested that the combination of calcium superphate and urea with lime for fertilizer was favourable.

  • The ferrimagnetic glass-ceramics in the system Fe2O3-CaO-SiO2 for hyperthermia were investigated. Glasses could be prepared up to the content of 40 wt% of Fe2O3 and below the weight ratio of 1.0 of CaO/SiO2. The maximum saturation magnetization and the maximum coercive force were 29.85 emu/g and 340.1 Oe respectively, for a glass 40Fe2O3.20CaO.40SiO2 composition heat-treated at 95$0^{\circ}C$ for 8 hours. And for a glass 40Fe2O3.30CaO.30SiO2 composition the maximum saturation magnetization and the maximum coercive force were 18.47 emu/g and 374.4 Oe heat-treated at 1,00$0^{\circ}C$ and 90$0^{\circ}C$ for 8 hours respectively. The maximum hysteresis loss was 1,726.3 cal/g for a glass 40Fe2O3.20CaO.40SiO2 composition heat-treated at 95$0^{\circ}C$ for 8 hours. It was found that the ferrimagnetic Fe2O3.CaO.SiO2 glass-ceramics was little injurious to human body as results of biocompatibility test and biotoxicity test.

Characteristics of Red $CaTiO_3$:Pr Phosphors Prepared by Sol-Gel Method (졸-겔법으로 제조된 $CaTiO_3$:Pr 적색형광체의 특성)

  • 적색 CaTiO$_3$:Pr 형광체 막을 졸-겔법으로 제조하고 형광체의 결정상 및 발광특성에 미치는 열처리 조건 및 Pr 첨가량의 영향에 대하여 연구하였다. CaTiO$_3$:Pr sol로부터 제조된 CaTiO$_3$:Pr 분말은 30~60nm 크기의 입자들이 0.3~0.5$mu extrm{m}$ 크기로 응집된 상태를 나타내었으며, 한편 sol을 borosilicate 기판 위에 4000rpm의 속도로 3회 스핀 코팅하고 열처리하였을 경우 두께 1.2$\mu\textrm{m}$의 CaTiO$_3$:Pr 박막이 형성되었다. CaTiO$_3$:Pr gel 분말을 여러 온도에서 열처리한 경우, $600^{\circ}C$ 이하에서는 비정질이지만, 온도가 증가하면서 erovskite 결정상이 생성되기 시작하여 90$0^{\circ}C$에서는 우수한 perovskite 결정상이 발달되었다. 반면, 박막의 경우에는 80$0^{\circ}C$의 열처리 온도에서는 결정성이 우수한 perovskite 상이 관찰되었으며 이러한 온도는 기존의 분말법의 소결온도에 비하여 300~40$0^{\circ}C$나 낮은 것이다. 0.2 mol% Pr이 첨가되고 90$0^{\circ}C$로 열처리된 분말시편은 최적의 Photoluminescence (PL) 특성을 나타낸 반면, 박막의 경우에는 80$0^{\circ}C$에서 열처리된 시편이 최적의 PL 특성을 나타내었다. 제조된 CaTiO$_3$:Pr 박막에 대하여 Cathodoluminescence (CL) 분석을 행한 결과, 6100$\AA$에서 폭이 좁고 강도가 큰 스펙트럼을 나타냄으로서, 본 실험에서 제조된 박막이 FED(field emission display)등의 적색형광체로 사용될 수 있음을 확인하였다.

Tributyltin Induces Apoptosis in R2C via Oxidative Stress and Caspase-3 Activation by Disturbance of $Ca^{2+}$

  • Tributyltin (TBT) used world-wide in antifouling paints toy ships is a wide-spread environmental pollutant. At low doses, antiproliferative modes of action have been shown to be involved, whereas at higher doses apoptosis seems to be the mechanism of toxicity in reproductive organs by TBT. In this study, we investigated that the mechanisms underlying apoptosis induced by TBT in R2C cell. Effects of TBT on intracellular $Ca^{2+}$ level and reactive oxygen species (ROS) were investigated in R2C cells by fluorescence detector. TBT significantly induced intracellular $Ca^{2+}$ level in a time-dependent manner. The rise in intracellular $Ca^{2+}$ level was followed by a time-dependent generation of reactive oxygen species (ROS) at the cytosol level. Simultaneously, TBT induced the release of cytochrome c from the mitochondrial membrane into the cytosol. Furthermore, ROS production and the release of cytochrome c were reduced by BAPTA, an intracellular $Ca^{2+}$ chelator, indicating the important role of $Ca^{2+}$ in R2C during these early intracellular events. In addition, Z-DEVD FMB, a caspase -3 inhibitor, decreased apoptosis by TBT. Taken together, the present results indicated that the apoptotic pathway by TBT might start with an increase in intracellular $Ca^{2+}$ level, continues with release of ROS and cytochrome c from mitochondria, activation of caspases, and finally results in DNA fragmentation.

  • On this paper we induce calcite($CaCO_3$) precipitation using microbial biomineralization of the Sporosarcina pasteurii and evaluate required performance evaluation by adjusting it to mortar. As a result carbonation normal mortar test piece(C3S-W) and mortar test piece(C3S-S.p) mixed with Sporosarcina pasteurii, reaction of C3S-S.p was late than C3S-W. Also, in the case of carbonation experiment of C3S-S.p curing in the Urea-CaCl2 aqueous solution(Medium) during 28days and durability of the C3S-W, durability of the mortar test piece(C3S-S.p) mixed with Sporosarcina pasteurii become higher than normal mortar test piece(C3S-W).

  • Microwave properties of $CaTiO_{3}$, MgO and CaO added $MgTiO_{3}$ ceramics are investigated. In the composition of $(Mg_{0.915}Ca_{0.085})TiO_3$, dielectric constant($varepsilon _T$) was 22, Qxf value was 50000, and temperature coefficient of resonant frequency($gamma_T$) approached to near zero $ppm/^{\circ}C$.$MgTiO_{3}$ ceramics with 0.1 mol of MgO showed the highest Qxf calue of 131000, dielectric constant of 17, and r, of -$50ppm/^{\circ}C$. Dielectric constant of 8 mol% CaO added to $MgTiO_{3}$-MgO was 20. Qxf valus was 52000, and sf was zero $ppm/^{\circ}C$. $MgTiO_{3}$-Mg0-Ca0 system showed higher Qxf value than ($Mg_{1-x}, Ca_x$) Ti03 system.

A Study on the Electromagnetic Property of NiCuZn Ferrite by Additive SnO2, CaO. (SnO2, CaO가 NiCuZn Ferrite의 전자기적 특성에 미치는 영향 연구)

  • The electromagnetic properties and microstructures of the basic composition of (N $i_{0.2}$C $u_{0.2}$Z $n_{0.6}$)$_{1.085}$(F $e_2$ $O_3$)$_{0.915}$ were invested by changing of the additive Sn $O_2$, CaO amounts and ferrite processes. There is no variation of grain size by changing additive amount. It can reduce the total loss when (N $i_{0.2}$C $u_{0.2}$Z $n_{0.6}$)$_{1.085}$(F $e_2$ $O_3$)$_{0.915}$ composition sintered at 1150 $^{\circ}C$ better than 130$0^{\circ}C$. Additive CaO confirmed of useful addition for the reduce total loss, because it increasing sintering density. Decreasing total loss were observed by adding both Sn $O_2$ 0.06 wt% and CaO 0.4 wt%.

Studies on the Effects of cAMP on the ATPase Activity and on the Calcium Uptake of the Sarcoplasmic Reticulum (근 소포체의 ATPase 활성과 Ca 능동수송에 미치는 cAMP의 영향)

  • The effect of adenosine 3', 5'-cyclic monophosphate on the ATPase activity and on the active transport of Ca of the sarcoplasmic reticulum fragments of the rabbit skeletal muscle was studied. Cyclic AMP (cAMP) had no effect on the ATPase activity of the fragments (8,000 ~ 20,000 $\times$ G and 20,000 ~ 36,000 $\times$ G fractions). $N^6$, O^{2'} -Dibutyryl cAMP (DBcAMP) had either no effect on the activity. On the other hand, theophylline (1 mM) increased the activity by about 20%. The active uptake of Ca by the sarcoplasmic reticulum fragments was inhibited by the presence of 1$\times$$10^{-6}$ ~ 1 $\times$ $10^{-3}$M of cAMP. The presence of DBcAMP or theophylline also inhibited the uptake. It is, therefore, concluded that the Ca uptake of the sarcoplasmic reticulum seems to be controlled by cAMP.
