Improvement Method of Sand Ground Using an Environmental Friendly Bio-grouting Material (친환경 바이오그라우팅을 이용한 모래지반 개량 공법)

  • The purpose of this research is to evaluate the improvements in the strength and injection range of loose ground after injecting $CaCO_3$(created by microorganism reaction). For this purpose, three cases of single-layer (Sand, SP, SW) specimens were made in a 150mm D ${\times}$ 200mm H space and two cases of multi-layer specimens (SW/SP, SP/SW) were made in a 150mm D ${\times}$ 300mm H space. The specimens were made with a relative density of 30% of soft ground and an injection was given over a time of one day. The uniaxial compression strength was measured with a cone penetrometer and the injection range was observed by checking the bulb formation around the injection nozzle. Also, the compositions of the specimens were assessed through XRD analyses. Based on the test results, a compressive strength of 500kPa and 15cm thick cementation were noted due to the cementation of the soil. This implies that there are significant effects of the pore condition and size on bio-grouting technology.

A Study on the Inhibition of Skin Pigmentation by Lobaric Acid as Protease Activated Receptor-2 Antagonist (Protease Activated Receptor-2의 길항제로서 Lobaric Acid의 피부 색소침착 억제 효능 연구)

  • Melanosome, the pigment granule in melanocyte, determines the color of skin when it moves into the keratinocyte. Inhibition of melanosome transfer from melanocyte to keratinocyte results in skin depigmentation. Protease activated receptor-2 (PAR-2) is involved in signal transduction systems via cell membrane and increases the melasome transfer when it is activated by cleavage of their extracellular amino acid sequence by trypsin or by a peptide such as SLIGKV. Here, we showed that lobaric acid inhibited PAR-2 activation and affected the mobilization of $Ca2^+$. The uptake of fluorescent microspheres and isolated melanosomes from melan-a melanocytes to keratinocytes induced by SLIGKV were inhibited by lobaric acid. Also, confocal microscopy studies illustrated a decreased melanosome transfer to keratinocytes in melanocyte-keratinocyte co-culture system by lobaric acid. In addition, lobaric acid induced visible skin lightening effect in human skin tissue culture model, melanoderm$^{(R)}$. Our data suggest that lobaric acid could be an effective skin lightening agent that works via regulation of phagocytic activity of keratinocytes.

Suitability evaluation of magnesium substituted biphasic calcium phosphates prepared by coprecipitation method (공침법을 이용한 마그네슘이 첨가된 biphasic calcium phosphate의 적합성 평가)

  • Magnesium-substituted BCP (biphasic calcium phosphate) powders were prepared by incorporating small amounts of magnesium into the structure of different hydroxyapatite (HAp)/${\beta}$-tricalcium phosphate (${\beta}$-TCP) ratios through coprecipitation method. A series of magnesium substitutions ranging from 0, 0.5, and 1.0 wt%, which are comparable to the measured magnesium contents, were performed. The obtained powders were characterized by the following analytical techniques: X-ray diffraction analysis (XRD), Thermo Gravimetric Analyzer (TGA) and Fourier transform infrared spectroscopy (FT-IR). The results have shown that substitution of magnesium in the calcium-deficient apatites resulted in the formation of biphasic mixtures of different HAP/${\beta}$-TCP ratios after heating above $1000^{\circ}C$. The 1.0 wt% magnesiumsubstituted-BCP were soaked in Hank's solutions after 2 weeks to observe the morphology of the biocement, especially needle-like hydroxyapatite crystals and to estimate the length and diameter of nanoneedle crystals.

Comparison of Biochemical Characterization of Korean and Chinese Mung Bean Lectin (한국산 녹두와 중국산 녹두에 있어서 Lectin의 생화학적 특성 비교)

  • The lectins were separated from Korean and Chinese mung bean seeds finally via chromatography using Sephadex G-100 and their biochemical features were studied and compared. They showed no hemagglutination with human red blood cells regardless of trypsin treatment and showed hemagglutination with only trypsin treated rabbit red blood cells. The molecular weights of two lectins were identified as 54 kDa and 28 kDa by SDS-PAGE. It was found that while the optimal reaction temperature of the lectin from Korean mung bean was $60^{\circ}C$, that of the lectin from Chinese mung bean seeds was $50^{\circ}C$. It was found also that the most thermal stable temperature of the seed lectin from Korean mung bean seeds was $50^{\circ}C$ and the lectin from Chinese mung bean was $40-50^{\circ}C$. The lectin from Korean mung bean seeds showed the highest activity at pH 3.2 and the lectin from Chinese mung bean showed the highest activity at pH 6.2. It was identified that when treating a denaturant, thiourea and guanidine-HCl resulted in no hemagglutination, so they induced denaturalization. It was identified also that there was no hemagglutination with urea, so it did not induced denaturalization. They showed no septicity to 6 types of carbohydrates including D-glucose. In addition, the lectins from the two mung bean seed had specificity to metal ions.

A Study on Characterization of Expansion Agent in Mortar with Light Burned Dolomite By-Product (경소백운석(輕燒白雲石) 부산물(副産物)을 활용(活用)한 바닥 몰탈용 팽창재(膨脹材)의 특성검토(特性檢討))

  • This study investigated a utilization technology of light burned dolomite. light burned dolomite ($CaMg(CO_3)_2$) generated in furnace (steel manufacturer) is an alternative to quick lime. Using light burned dolomite has an effect on reducing the consumption of fluorite slag MgO concentration in supersaturated solution by prolonging the life of softening effect. Armophous MgO, not containing periclase is formed by firing dolomite under $800^{\circ}C$. It has larger surface area and higher reactivity than periclase, and also shows better expansion effect than quicklime. Due to those effects, therefore, armophous MgO produced from light burned dolomite is used as an alternative expansion agent in mortar. In the experiment, characteristics of light burned dolomite were compared to those of existing expansion agents such as anhydrite and quicklime. Then, each expansion and shrinkage rates were measured over a period of about 3 months in both of 1m Jis mold at labscale and apartment mortar flooring at field scale. In the result, it was observed that light burned dolomite in mortar flooring more compensates for the expansion and shrinkage rates than the existing expansion agents, showing low expansion rate of below 0.05% and also decreases the crack.

Synthesis of Sulfonated PET-g-GMA Fine Ion-exchange Fibers for Water Treatment by Photopolymerization and Their Adsorption Properties for Metal Ions (광중합법을 이용한 수처리용 설폰산형 PET-g-GMA 극세 이온 교환 섬유의 합성 및 금속 이온 흡착 특성)

  • The sulfonated PET-g-GMA ion-exchange fine fibers were synthesized by UV radiation-induced graft copolymerization using a photoinitiator, and their chemical structure and adsorption properties were investigated. The optimum values for synthetic conditions - UV intensity, reaction time, and reaction temperature were 450 W, 60 min, and $40^{\circ}C$, respectively. Maximum values of the degree of sulfonation and ion exchange capacity were 8.12 mmol/g and 3.25 meq/g, respectively. Tensile strength of sulfonated PET-g-GMA fine ion exchange fibers was lower than that of PET trunk polymer as the grafting reaction rates increased. It was shown that as for the adsorption rate of $Ca^{2+}$ and $Mg^{2+}$ by the sulfonated PET-g-GMA fine ion exchange fibers, magnesium ion is slower than calcium ion in the solution. However, in the mixture of the calcium and magnesium ions, the adsorption rate of calcium ion was much slower than that of magnesium ion.

Behavior of AKD in AKD-sized Paper by Heating Treatment During Ageing (AKD 사이즈 처리한 종이의 저장중 가열처리에 의한 AKD의 거동)

  • This study was performed to investigate the reaction between alkylketene dimer(AKD) and cellulose molecules in AKD-sized paper sheet. AKD was added to highly beaten($80{\pm}3^{\circ}SR$) SwBKP(ca. 0.8% on pulp) in order to have much AKD retention in the paper sheet. This AKD-sized paper sheet was aged at different temperatures, $60^{\circ}C,\;80^{\circ}C,\;105^{\circ}C\;and\;125^{\circ}C$. Changes in FT-IR spectra of AKD in paper sheet during the ageing were measured. In addition, sizing degrees of the AKD-sized paper sheet pretreated for 30 sec. at $105^{\circ}C$ were measured by HST size tester during the storage at different temperature. IR spectra of AKD-sized paper sheet preheated at $105^{\circ}C$ for 30 sec. showed unchanged spectra two absorption bands at $1849cm^{-1}\;and\;1722cm^{-1}$ which refer to the typical AKD IR bands. However, these typical AKD bands were gradually reduced with increasing ageing, completely disappeared after 6 hrs. and formed new absorption band at $1706cm^{-1}$, which refers to carbonyl stretching vibration of dialkylktone. Eventually the AKD molecule was hydrolyzed to diakylketone without formation of ${\beta}$-ketoester with cellulose in paper sheet. After 6 days ageing, a little amount of ${\beta}$-ketoester bands was identified in 6 or 7 days ageing, because of the absence of water due to long-term heating. The same tendency was observed at different ageing conditions. At the practical papermaking process, AKD reacts prevailing with water, and mostly seems to be hydrolyzed to dialkylketene. Concerned to the sizing development, AKD-sized paper sheet was shown no sizing development at the initial stage of ageing at $60^{\circ}C$ after heating treatment at $105^{\circ}C$ for 30 sec., and gradually increased the sizing degree with increasing ageing, such as Hercules Sizing Tester (HST) 130 see for 12 hr, HST 300 sec. for 3 days and HST 400 sec. for 5 days. It was concluded that hydrolyzed AKD could contributed to the sizing development of the paper sheet.

Characterization and Production of Thermostable and Acid-stable Extracellular Fibrinolytic Enzymes from Cordyceps militaris

  • Biochemical and enzymatic characterization for extracellular protease isolated from Cordyceps militaris cultivated on rice bran medium was investigated. C militaris produced proteolytic enzymes from 10 days after inoculation, maximum enzyme production was found at 25 days. The optimum temperature and pH of proteases production was at $25^{\circ}C$ and pH 7.0, respectively. The protease activity was observed in the four peaks (Pro-I, Pro-II, Pro-III, and Pro-IV) separated through Sephadex G-100 column chromatography. The separated protease was optimally active at $25^{\circ}C$. Optimum pH of the protease was between 7 and 8. Enzyme was also stable over at $30-80^{\circ}C$. The enzyme was highly stable in a pH range of 4-9. Protease activity was found to be slightly decreased by the addition of $Mg^{2+}$, $Mn^{2+}$, $Zn^{2+}$, $Fe^{2+}$ and $Cu^{2+}$, whereas inhibited by the addition of $Ca^{2+}$ and $Co^{2+}$ Protease activity was inhibited by protease inhibitor PMSF. On the other hand, the partially purified protease was investigated on proteolytic protease activity by zymogram gel electrophoresis using three substances (casein, gelatin and fibrin). Four active bands (F-I, FII, F-III, and F-IV) of fibrin degradation were revealed on fibrin zymogram gels. Both of F-II and FIII showed caseinolytic, fibrinolytic and gelatinolytic activities in three gels. Thermostability, pH stability, and pH-thermostability of the enzyme determined the residual fibrinolytic activity also displayed on fibrin zymogram gel. The only one enzyme (F-II) displayed over a broad range of temperature at $30-90^{\circ}C$. The FII displayed fibrinolytic activity in the pH range 3-5, but was inactivated in the range of pH 6-11. The F-I and F-III showed enzyme activity in the pH range of 6-11. In the pH-thermostability, the F-II only kept fibrinolytic activity after heating at $100^{\circ}C$ for 10, 20 and 30 min at pH 3 and pH 7, respectively. On the other hand, the F-II was retained activity until heating for 10 min under pH 11 condition. By using fibrin zymogram gel electrophoresis, extracellular fibrinolytic enzyme F-II from C. militaris showed unusual thermostable under acid and neutral conditions.

Durability Properties of Ultra Rapid Hardening Mortar Produced with Alumina-based Binder for Repairing Sewage Treatment Pipes (하수관거 보수용 알루미나계 결합재 초속경 모르타르의 내구 특성)

  • In this study, the durability of ultra rapid hardening mortar for sewage pipe was evaluated by type of mortar binder. As a result of analyzing the internal structure for each type of mortar, it was confirmed that Al2(OH)3 was generated in the internal structure of the CAC-based mortar, and its corrosion resistance was superior to that of other types of mortar. As a result of the compressive strength test, OPC had the tsmallest strength, followed by CAC100 > CAC100P > CAC80. This trend was similar to the previous study results. Chloride ion penetration resistance and freeze-thaw test showed similar trends. That is, CAC and C12A7 were better than OPC, and CSA was worse than OPC. This is mostly beacuse of cracks caused by expansion of CSA-based mortar. CAC100P mix showed the best chemical resistance. It is thought that this is because the alumina gel formed inside the mortar and the polymer combine to make the internal structure more dense.

Ziziphus spina christifor Sustainable Agroforestry Farming in Arid Land of Khartoum State of Sudan

  • Cow pea (Vigna unguiculata) was intercropped with Ziziphus spina-christi as summer forage in two consecutive seasons of 2017 and 2018. The aims to find out suitable agroforestry practice for saline soils of Khartoum State. And to investigate effect of tree spacing on forage biomass yield under semi -irrigated systems. Completely randomized block design with 3 replicates was conducted for this trial. Thus Z.spina-christi that fixed at 4×4 m was intercropped with cowpea at 1 m and 1.5 m spacing from trees trunk. Tree growth parameters were measured in terms of tree height, tree collar diameter, tree crown diameter and fruit yield per tree. While crop were parameters were determined in terms of plant height, number of plant, forage biomass yield per ha and land equivalent ratio. Soil profile of 1×1 m and 1.5 m depth was excavated and its features were described beside its chemical and physical properties were analyzed for 0-10 cm, 0-30 cm, and 30-60 cm and 60-100 cm layers. The results revealed that soil pH, CaCO3, SAR, ESP, and EC ds/m were increased by increasing soil depths. Meanwhile tree growth in terms of tree height was significant in the first season 2017 when compared with tree collar diameter and tree crown diameter. Also significant differences were recorded for tree growth when compared with sole trees in the second season in 2018. Tree fruit showed marked variations between the two seasons, but it was higher under intercropping particularly at ZS2. Crop plant height was highly significant under sole cropping than intercropping in first season in 2017. In contrast forage biomass yield was significant under intercropping in ZS1 and ZS2 treatments. Land equivalent ratio was advantageous under this agroforestry system particularly under ZS2. Thus it recorded 5 and 9 for ZS2 in the two consecutive seasons respectively. Therefore, it is feasible to introduce this agroforestry system under such arid lands to provide summer forage yield of highly nutritive value and low cost for animals feed as well as to increase farmers' income and to halt desertification and to sequester carbon.
