Studies on the Water Quality of the Han River Water, and Water Quality Standards (한강의 수질과 수질규준에 관한 고찰)

  • Monthly mean values for EDTA hardness in the Han River water were determined for March 1966 through May 1967. The hardness was retained a range of 22-59 mg/L the maximum value of 59 mg/L was approached in the months of mid-winter, indicating a seasonal variation. The annual ranges of Ca and Mg were 3.7-9.1 mg/L, 1.5-10.4 mg/L, respectively. The annual range of Mg:Ca ratio was 0.18-2.60. And the hardness correlated closely with Mg volume. In Korea, fortunately, most river waters are not yet suffering from damaging pollution. We must try to establish our water quality criteria based on scientific data, and make research recommendations for the following water uses; public water supplies, aquatic life and wildlife, industry, recreation and aesthetics.

Purification and Characterization of Carboxymethyl Cellulase from Stropharia rugosoannulata (독청버섯아재비 균주가 생산하는 Carboxymethyl Cellulase의 정제 및 효소학적 특성)

  • A Carboxymethyl Cellulase (CMCase) has been isolated and purified from the edible mushroom, Stropharia rugosoannulata. The molecular weight of CMCase was estimated to be 54 kDa by SDS polyacryl amide gel electrophoresis. The maximum activity of the purified CMCase was observed at pH 4.0 and $40^{\circ}C$, and stable for pH 3.0 to 11.0 to maintain 40% activity. The CMCase activity was activated by $AgNO_{3},\;MgSO_{4},\;and\;KCl$. However, its activity was inhibited by 1,10-phenanthroline, KCN and L-cysteine. Also, the enzyme activity was decreased by the addition of EDTA, suggesting that the purified CMCase is metalloenzyme.

Correlation of Arsenic and Heavy Metals in Paddy Soils and Rice Crops around the Munmyung Au-Ag Mines (문명 금은광산 주변 논토양에서 As 및 중금속의 토양과 벼작물의 상관성 평가)

  • This study has focused on investigation of correlation for As and heavy metals in paddy soil and rice crops sampled in the vicinity of the abandoned Munmyung Au-Ag mine. Soil samples extracted by various methods including aqua regia, 1 M $MgCl_2$, 0.01 M $CaCl_2$ and 0.05 M EDTA were analyzed for As and heavy metals (Cd, Cu, Pb and Zn). Rice grain samples grown on the soils were also analyzed for the same elements to evaluate the relationships between soils and rice crops. According to soil extraction methods, As and heavy metal contents in the soils were decreased in the order of aqua regia > 0.01 M $CaCl_2$ > 1 M $MgCl_2$ > 0.05 M EDTA. In addition to correlation analysis, statistically significant correlation with the four extraction methods (p<0.01) were found in the soil and rice samples. As calculation of biological accumulation coefficients (BACs) of the rice crops for As and heavy metals, the BACs for Cd, Zn and Cu were relatively higher than those for As and Pb. This study also carried out a stepwise multiple linear regression analysis to identify the dominant factors influencing metal extraction rates of the paddy soils. Furthermore, daily intakes of As and heavy metals from regularly consumed the rice grain (287 g/day) grown on the contaminated soils by the mining activities were estimated, and found that Cd and As intakes from the rice reached up to 73.7% and 51.8% for maximum allowance levels of trace elements suggested by WHO, respectively. Therefore, long-term consumption of the rice poses potential health problems to residents around the mine, although no adverse health effects have yet been observed.

  • Some biological activities showed as ATPase activity of myofibril, actomyosin and myosin extracted from chicken leg and pectoral skeletal muscle were investigated. The $Mg^{+2}$-ATPase activity at 0.05 M KCl were 0.82, 0.38 and 0.11 ${\mu}mole$ Pi/mg protein/min. in actomyosin, myofibril and myosin from pectoral muscle while 0.71, 0.32 and 0.08 ${\mu}mole$ Pi/mg protein/min. in actomyosin, myofibril and myosin from leg muscle. EDTA-ATPase activity at 0.6M KCl were 0.80, 0.42 and 0.40 ${\mu}mole$ Pi/mg protein/min. in actomyosin, myofibril and myosin from pectoral muscle. In case of leg muscle, that activity was noted as 0.69, 0.33 and 0.28 ${\mu}mole$e Pi/mg protein/min in proteins. ATPase activity of myosin from leg and pectoral muscle were inhibited in 10% at a higher concentration of $Mg^{+2}$ than molar concentration of EDTA, and the ATPase activity was increased to 400% compared with control at $10^{-3}M$ of $Ca^{+2}$. Actomyosin from pectoral muscle was solubilized at 0.1 M KCl above and that from leg muscle was solubilized at 0.15 M KCl above. In case of myosin, pectoral muscle was solubilized at 0.25M KCI above and leg muscle was solubilized at 0.30M KCl above.

Determination of $^{226}Ra$ Isotope in the Leachate around Phosphogypsum Stack Using Ethylenediaminetetraacetic Acid (EDTA) (Ethylenediaminetetraacetic acid (EDTA)를 이용한 인산석고 야적장 침출수 중의 $^{226}Ra$ 분석법 개발)

  • Ba is the most useful element to get the $Ba(Ra)SO_4$ precipitate. However, when the high concentrations of ions such as sulfate, calcium are existed in the leachate of phosphogypsum stack, it is difficult to get the $Ba(Ra)SO_4$ precipitate. Since this reason, the developed method for the Ba coprecipitate using EDTA was performed to determine the $^{226}Ra$ concentration in the high sulfate sample. The average concentration of $^{226}Ra$ in a leachate of phosphogypsum using this method was 0.102 $Bq{\cdot}kg^{-1}$ and the minimal detectable activity is 3.4 $mBq{\cdot}kg^{-1}$. The $mBq{\cdot}kg^{-1}$ method was 0.102 $Bq{\cdot}kg^{-1}$ and the minimal detectable activity is 3.4 $mBq{\cdot}kg^{-1}$. The $^{226}Ra$ stock solution and the CRM (Certified Reference Material) were analyzed to verify this method. In analyzed $^{226}Ra$ stock solution, bias with added concentration was approximately 1% and the correlation curve between $^{226}Ra$ concentration in simulated standard sample and measured $^{226}Ra$ concentration showed good agreement with a correlation coefficient ($R^2$) of 0.99. In analyzed CRM, maximum bias with reference value was 5.8% (k=1) and the analytical results were in good agreement with the reference value.

  • Calcification in patients with endstage renal disease on renal replacement therapy can occur in extraskeletal area such as conjunctiva and cornea. Conjunctival and corneal calcification(CCC) has mostly has been reported in adults with endstage renal disease on hemodialysis. CCC seems to be associated with the duration of renal replacement therapy, and high Ca$\times$P value. We report a 10-year-old girl who was on peritoneal dialysis for 31 months and presented with CCC on both eyes. Her corneal calcification was resolved after the epithelial debridement and ethylenediaminetetraacetic acid(EDTA) soaking therapy.

  • This experiment was conducted to investigate the characteristics of alkaline protease from Aspergillus fumigatus which was isolated from soil as a superior strain for the production of the alkaline protease. The optimum temperature for enzyme activity was $50^{\circ}C$ and optimum pH was 9.0. The enzyme was stable at pH 8.0 to 10.0 and thermal inactivation was shown $30^{\circ}C$. The activity of the enzyme was increased by the addition of $Mn^{++},\;Cu^{++},\;Ba^{++},\;Mg^{++},\;$wheras it was inhibitied by $K^+,\;Fe^{+++},\;Ag^+,\;Pb^{++},\;Na^+,\;Ca^{++},\;Hg^+,\;Zn^{++}$. EDTA. 2, 4-DNP, ${\varepsilon}-amino$ caproic acid did not show inhibitory effect on the proteolytic activity of alkaline protease but P-chloromercuribenzoic acid inhibited the enzyme activity, indicating that reactive sulfhydryl group is required for the enzymatic activity. The reaction of this enzyme followed typical Michael-Menten Kinetics with the Km value of $8.33{\times}10^{-4}mole/{\ell}$ with the Vmax of $47.62{\mu}g/min$. This enzyme had stronger proteolytic activity than trypsin on substrate such as casin and hemoglibin.

  • A New lectin from baby clam, Tapes japonica, was isolated and purified through the following procedures; acetone powder, 0.15M NaCl extraction, ammonium sulfate fractionation, N-acetyl-D-galactosamine-agarose affinity column, and ion exchange Mono Q of FPLC. This lectin nonspecifically agglutinated human erythrocytes but didn't agglutinate mouse and rabbit erythrocytes. And the lectin neither stimulated human lymphocytes nor agglutinated Sarcoma 180 cells. On polyacrylamide gel electrophoresis, the lectin migrated as a major single band indicating homogeneous. A molecular weight was estimated to be about 131,000 daltons by Biogel P-300 and 125,000 daltons by SDS-PAGE without ${\beta}-mercaptoethanol$. This lectin is supposed to be a tetramer composed of heterogeneous subunits, about 30,000 and 33,000 daltons. Baby clam lectin was inhibited by EDTA and recovered agglutinating activity by $Ca^{++}\;and\;Mn^{++}$. This lectin is revealed as glycoprotein that contained about 4.2% neutral sugar.

The Effects of Laundering Conditions on Calcium Deposition on the Fabric (세척조건이 직물에의 Calcium 침착에 미치는 영향)

  • The influence of laundering conditions on calcium deposition on the fabric was studied by repeated laundering the cotton fabric with soap in the hard water of 200 P.P.M. $CaCO_3$. The experimental variables were: 1) soap concentrations ($0.06\%$, $0.13\%$, $0.25\%$), 2) water contents in the fabric after hydroextraction. ($65\%$, $150\%$, $315\%$), 3) builders (Na-EDTA, sodium carbonate, sodium metasilicate), 4) washing cycle (5, 10, 15, 20 cycles). The fabric was washed for 15 minutes at $23\pm1^{\circ}C$ in a washing machine (Model; Gold Star Wp-2005) under the similar conditions with those of home laundering, and rinsed 5 times for 5 minutes. The amount of calcium deposits on the fabric was determined by the EDTA-Back titration method described by Wasserman and Basch. Results of this study were follows: 1) The amount of calcium deposits on the fabric increased with increasing wash cycles. 2) During the rinsing process, residual calcium content on the fabric increased with water content in the fabric after hydroextraction. 3) The amount of calcium deposits on the fabric decreased with the increasing soap concentration above the equivalent amount of calcium ion content in the water. 4) Sequestering agents and alkaline builders influenced the amount of calcium deposits on the fabric. The amount of calcium deposits on the fabric was in the order of sodium metasilicate, sodium carbonate, nonbuilder, and EDTA.

Characterization of ${\alpha}$-amylase Producing Hybrid Constructed between Saccharomycopsis and Saccharomyces (Saccharomycopsis속과 Saccharomyces속의 잡종형성 균주에서 생산하는 ${\alpha}$-amylase의 특성)

  • This study has been performed to deveope a yeast strain having high ${\alpha}$-amylase production ability using nuclear transfer method. Hybrids formed between the strains of Saccharomyces fiburigera KCTC 7393 and Saccharomyces cerevisiae KCTC 7049 (tyr-, ura-)were obtained by nuclear transfer technique. Nuclei isolated from the wild type S. fiburigera strain were transfered into auxotrophic mutants S. cerevisiae and selected the hybrids showing an increased starch degrading capability were selected (MN-16). This transformant grew best and produced maximal ${\alpha}$-amylase activity on the medium containing 2% (V/V) soluble starch. ${\alpha}$-Amylase from MN-16 was purified electrophoretically homogenety and its properties were investigated. The enzyme was purified about 10.6 fold with an overall yield 9.7% from the culture medium by ammonium sulfate fractionation. DEAE-Sephacel column chromatography, and Sephacryl S-200 column chromatography. The purified enzyme showed a single band on SDS-polyacrylamide gel electrophoresis. The molecular weight of the ${\alpha}$-amylase was estimated to be 53,000 daltons by SDS-PAGE and by gel permeation chromatography on Sephacryl S-200. The purified enzyme showed the maximum activity at pH 5.5 and 40${\circ}C$. The km value for soluble starch was 2.5㎎/㎖. The enzyme activity increased in the presence of $Ca^{2+}, Co^{2+}, EDTA, Mg^{2+}, Mn^{2+}, Zn^{2+}$, but inhibited by $Cu^{2+}, Fe^{2+}$, and $Ni^{2+}$
