Purpose: Inadequate hospital stocking and unavailability of essential antidotes is a worldwide problem. The purpose of this study was to determine the adequacy of antidote stocks in Korean hospitals based on a modified version of US antidote stocking guidelines. Methods: Both written and telephone surveys to collect information on hospital demographics and antidote stocking were given to the emergency departments or hospital pharmacies of 117 hospitals with emergency medical centers. The twenty antidotes included in the survey were taken from year 2000 US antidote stocking guidelines, except for activated charcoal and black widow spider antivenin. Antidote stocks were compared by hospital size, geographic location, and type of emergency medical center. Results: Complete responses were received from all hospitals. A mean of $12.4{\pm}2.9$ antidotes were adequately stocked per hospital. All hospitals stocked atropine, $CaCl_2$ naloxone, and sodium bicarbonate. However, digoxin Fab fragment (16%), cyanide kits (15%), EDTA (10%), BAL (9%), and fomepizole (1%) were not uniformly stocked. Large and teaching hospitals were significantly more likely to stock greater numbers of antidotes. Conclusions: Korean hospitals as a group do not have adequate antidote stocks. Korean stocking guidelines and an antidote management system are recommended in order to correct these deficiencies.
A strain producing a potent protease was isolated from turban shell. The strain was identified as Bacillus sp. S17110 based on phylogenetic analysis. The enzyme was purified from culture supernatant of Bacillus sp. S17110 to homogeneity by ammonium sulfate precipitation, SP-Sepharose, and DEAE-Sepharose anion exchange chromatography. Protease activity of the purified protein against casein was found to be stable at pH 7 to pH 10 and around $50^{\circ}C$. Approximately 70% of proteolytic activity of the enzyme was detected either in the presence of 100 mM SDS or Tween 20. The enzyme activity was enhanced in the presence of $Ca^{2+},\;Zn^{2+},\;Mg^{2+}$, but was inhibited by EDTA, indicating that it requires metal for its activity. The purified enzyme was found to be a monomeric protein with a molecular mass of 75 kDa, as estimated by sodium dodecyl sulfate polyacrylamide gel electrophoresis (SDS-PAGE) and gel filtration chromatography. The purified enzyme was analyzed through peptide fingerprint mass spectra generated from matrix-assisted laser desorption ionization-time-of-flight-mass spectrometry (MALDI-TOF-MS) and a BLAST search, and identified as immune inhibitor A (inhA) deduced from nucleotide sequence of B. cereus G9241. Since InhA was identified as protease that cleave antibacterial proteins found in insect, inhA-like protease purified from Bacillus sp. S17110 might be pathogenic to sea invertebrates.

A bacterial strain NS70 producing an alkaline protease was isolated from soil samples taken near a hot spring and identified as Bacillus licheniformis by its morphological and physiological properties and cellular fatty acid analysis. The isolated alkaline protease was purified by ammonium sulfate fractionation, DEAE-, CM-, and Phenyl-Sepharose column chromatography. The molecular weight of the purified enzyme was estimated to be 32, 000 Da by sodium dodecylsulfate polyacrylamide gel electrophoresis. Its optimal pH and temperature for proteolytic activity against Hammarsten casein were 12 and $65^{\circ}C$, respectively. The enzyme was stable at alkaline pH range from 6.0 to 12.0, and fairly stable up to $65^{\circ}C$. The enzyme was inhibited by phenylmethylsulfonyl fluoride but not by EDTA and N-ethylmaleimide indicating that the enzyme is serine protease. Enzyme activity was markedly inhibited by $Hg^{2+}$ and $Cu^{2+}$. Autolytic phenomena were observed on purified protease NS70 but autolysis was reduced by the addtion of $Ca^{2+}$ ion or bovine serum albumin.

The main objectives of root canal therapy are cleaning and shaping and then obturating the root canal system in 3 dimensions to prevent reinfection. Many instrumentation techniques and devices, supported by an irrigation system capable of removing pulp tissue remnants and dentin debris, have been proposed to shape root canals. But current regimens in chemomechanical debridement using instrumentation and irrigation with NaOCl are not predictably effective in root canal disinfection. These findings are not surprising because the root canal system is complex and contains numerous ramifications and anatomical irregularities. The microorganisms in root canals not only invade the anatomic irregularities of the root canal system but also are present in the dentinal tubules. Therefore further disinfection with an effective antimicrobial agent may be necessary and it well1mown that use of intracanal medication will lower bacterial count in infected root canals. Calcium hydroxide has a long history of use in endodontics, and more attention has been given to the use of calcium hydroxide as intracanal dressing for the treatment of infected pulp. However, when treatment is completed in one visit, no intracanal medications other than intracanal irrigants are used. Recently, a mixture of a tetracycline isomer, an acid, and a detergent(MTAD), has been introduced as a final rinse for disinfuction of the root canal system. It has been shown that MTAD is able to remove the smear layer with minimal erosive changes on the surface of dentin, and is effective against Enterococcus faecalis, a microorganism resistant to the action of other antimicrobial medications. In another study, the ability of MTAD was investigated to disinfect contaminated root canals with whole saliva and compared its efficacy to that of NaOCl Based on the results, it seems that MTAD is significantly more effective than 5.25% NaOCl in eradicating bacteria from infected root canals. In the cytotoxicity evaluation, MTAD is less cytotoxic than engenol, 3% $H20_2,\;Ca(OH)_2$ paste, 5.25% NaGCl, Peridex, and EDTA and more cytotoxic than 2.63%,1.31% and 0.66% NaOCl. Is it promising or transient?
마른 볏짚으로부터 fibrinolytic enzyme (BK)을 분비하는 균주를 분리하여 동정한 결과, Bacillus subtilis속으로 분류되었다. 분리된 균주(Bacillus subtilis BK-17를 배양하여 그 배양액으로부터 에탄올 침전, ion exchange, gel filtration 등의 과정을 거쳐 fibrinolytic enzyme (BK)를 분리 및 정제하였다. 이 정제된 효소를 SDS-PACE gel 전기영동한 결과 분자량은 약 31 kDa 이었다. BK의 활성 및 안정성에 대한 pH의 영향을 조사해본 결과, pH $6\∼8$범위에서 매우 높았고, 최적 pH는 7과 8이었다. 본 효소의 활성 및 안정성에 대한 최적 온도는 $50^{\circ}C$이었으며, $20^{\circ}C\∼50^{\circ}C$범위에서는 안정성이 그대로 유지되었지만 $60^{\circ}C\∼80^{\circ}C$범위에서는 현저히 감소하여 $50^{\circ}C$에 비해 $20\%\∼40\%$의 활성을 보였다. BK에 대한 활성 역시 안정성과 비슷한 양상을 보였고, 다만 $20^{\circ}C$에서 약 $62\%$의 활성을 보였고, 그 이후 $50^{\circ}C$까지 지속적으로 증가하여 $50^{\circ}C$에서 최대의 활성을 보였다. 실험한 금속이온들 중 1 mM의 $Zn^{2+}$와 $Ca^{2+}$에서 각각 $35\%$와 $23\%$의 저해활성을 보였지만 나머지 이온들에서는 의미 있는 영향이 없었다. 동일농도의 EDTA에 대해서는 $45\%$의 저해활성을 보였기 때문에 BK는 metallo enzyme으로 생각된다. Fibrinogen-rich fibrin plate 와 plasminogen-free fibrin plate에서의 분해활성을 검토해본 결과, 두 plate에서 비슷한 분해활성을 보였다. 따라서 본 효소는 plasminogen activator type 보다는 fibrin에 직접 활성을 가지는 것으로 생각된다.
사람의 혈액으로부터 Elastase를 분리하는 새로운 방법을 개발하여 순도 높은 효소를 얻고 이 효소에 의하여 발생되어지는 질병의 예방과 치료에 응용하기 위하여 이 효소의 근본적인 성질규명을 시도하였다. 정제 방법으로는 Sephodex G-75를 이용한 1회의 액체 크로마토그라피와 HPLC을 이용한 2회의 코마토그라피를 거치는 2단계 방법을 사용하였다. 이때 얻어진 효소는 분자량이 $26,000{\sim}29,700$ 사이에 있는 3개의 다른 분자량을 가진 물질로 확인되었으며, 항체 반응에서 사람의 결구내 Elastase의 특성을 나타내었다. 함유하고 있는 미량의 금속이온을 분석한 결과 정제하는 단계에 따라 Zn 이온의 효소분자에 대한비는 증가하였으며 가장 순수한 분자내의 효소분자와 Zn 이온과의 비는 1 : 2였다. 이 효소는 chelating agent에 의하여 활성도를 잃게 되었으며, 반대로 chelating agents에 의하여 활성도를 잃고 있는 반응 medium에 그 chelaing agents의 농도를 초과하는 2가 이온 즉 Zn 이온, Ca 이온, 그리고 Mn 이온을 넣으면 그 활성도는 원상복귀되나 Mg이온에 의하여는 그 활성도가 원상회복 되지 않았다. 이 모든 성질을 종합하면 사람의 Neutrophil granule에 있는 Elastase는 지금까지 알려진 바와 같이 Serine pretense임과 동시에 metalloenzyme으로 제고되어야 한다고 제안한다. 나아가서chelating agents에 의하여 이 효소의 활성도를 조정할 수 있다는 것은 이 효소에 의하여 일어나는 질병의 치료에 응용할 수 있는 가능성도 보여준다.
Protease생산(生産)을 위한 Asp. oryzae의 배양조건(培養條件), 생산효소(生産酵素)의 정제(精製) 및 정제효소(精製酵素)의 육연화(肉軟化)에 관한 효과(效果)에 대(對)하여 연구 하였다. Asp. oryzae가 생산(生産)하는 proteolytic enzyme이 육연화(肉軟化)에 미치는 영향(影響)을 조사한 결과(結果)는 다음과 같다. 1. Asp. oryzae를 밀기울에 고체배양(固體培養)한 결과(結果) 최적조건(最適條件)은 배양일수(培養日數) 3일, 산수량(散水量) 130%, pH 6.5와 탄소원(炭素原)으로는 glucose 2%, 질소원으로는 urea 0.03%, mineral salts로 $MgSO_4$ 0.1%가 가장 좋았다. 2. Asp. oryzae 고정(固定)배양액으로부터 효소(酵素)를 추출(抽出)하고 그 추출액으로부터 유안염석(硫安鹽析) 및 Sephadex G-75 columm chromatography에 의하여 정제(精製)하였다. 3. 정제(精製)된 enzyme은 산성(酸性)에서는 hemoglobin, 중성(中性), alkali성(性)에서는 casein를 기질(基質)로 사용한 결과 작용최적(作用最適) pH가 3.0, 6.6, $8.4{\sim}8.5$, $10{\sim}10.5$이었으며 pH의 안정성(安定性)범위는 $pH\;6{\sim}10$이었다. 4. 최적온도(最適溫度)는 $50^{\circ}C$ 이었으나 안정성(安定性)은 $40^{\circ}C$ 까지였다. 5. Metal ion 및 EDTA에 미치는 영향은 protease는 Ag ion에 저해 되었다. 또 ion 농도가 낮아짐에 따라 금속 ion에 의한 조해(阻害)는 감소되었다. EDTA에 의하여서도 조해(阻害)되었다. 6. Chicken과 bovine으로부터 myofibril과 actomyosin을 추출(抽出) 정제(精製)하여 attack시킨 결과(結果) 근원섬유단백질(筋原纖維蛋白質)의 MgATPase활성(活性) 및 Ca-ATPase활성(活性)은 현저하게 변화(變化)하였다. 따라서 본(本) 효소(酵素)는 연육소(軟肉素)로서 이용(利用)할 수 있음을 알았다.
고등어 surimi를 효율적으로 생산하기 위한 연구의 일환으로 감압수세장치를 설계 제작하고 이 장치를 이용하여 각각 상압과, 560 및 660 mmHg의 감압 하에서 각각 1, 3, 5 및 7회 알칼리 수세하고 압력 및 수세횟수에 따른 고등어 surimi의 품질 특성을 검토하였다. 설계 제작한 감압수세장치는 연속수세가 가능하였으며 양질의 surimi를 효율적으로 제조할 수 있었다. 전체의 수세조건을 통하여 surimi의 수분함량은 $72.0{\sim}72.9%$, 조지방 $4.8{\sim}5.7%$ (수세하지 않은 것, 7.0%), pH $6.9{\sim}7.0$ (수세하지 않은 것, 6.0), VBN $6.9{\sim}7.0\;mg/100\;g$ 및 가압드립 $6.7{\sim}8.3%$이었으며, 단백질 추출성은 560 mmHg, 5회 수세 시 염용성, 수용성 및 기질 단백질추출성이 각각 3,694, 6,036 및 1,424 mg/100 g으로써 각각 가장 높았다. $Mg^{2+}-$ 및 $Ca^{2+}-ATPase$ 활성은 560 mmHg, 5회 수세 시 각각 0.25 및 $0.17\;{\mu}mol\;Pi/min/mg$ actomyosin으로써 가장 높았다. Setting gel의 TGase 활성은 560 mmHg, 5회 수세 시 3.932 nmol/mg이었으며 gel 강도는 setting gel 및 cooked gel에서 각각 420 g cm (상압, 320 g cm) 및 485 g cm (상압, 412 g cm)로써 각각 가장 높았다. 전반적으로 보아 고등어 surimi 제조를 위한 가장 적합한 수세조건은 760 mmHg, 660 mmHg 및 560 mmHg 압력 중 560 mmHg의 감압 하에서 5회 반복 수세하는 것이었다.
Strontium doped lanthanum manganite (LSM) with perovskite structure for SOFC cathode material shows high electrical conductivity and good chemical stability, whereas the electrical conductivity at intermediate temperature below $800^{\circ}C$ is not sufficient due to low oxygen ion conductivity. The approach to improve electrical conductivity is to make more oxygen vacancies by substituting alkaline earths (such as Ca, Sr and Ba) for La and/or a transition metal (such as Fe, Co and Cu) for Mn. Among various cathode materials, $LaSrMnCuO_3$ has recently been suggested as the potential cathode materials for solid oxide fuel cells (SOFCs). As for the Cu doping at the B-site, it has been reported that the valence change of Mn ions is occurred by substituting Cu ions and it leads to formation of oxygen vacancies. The electrical conductivity is also affected by doping element at the A-site and the co-doping effect between A-site and B-site should be described. In this study, the $La_{1-x}Sr_xMn_{0.8}Cu_{0.2}O_{3{\pm}{\delta}}$ ($0{\leq}x{\leq}0.4$) systems were synthesized by a combined EDTA-citrate complexing process. The crystal structure, morphology, thermal expansion and electrical conductivity with different Sr contents were studied and their co-doping effects were also investigated.
