Development of Fibrinolytic Agents from Snake Venoms

  • Fibrinolytic proteases, piscivorase I (PI) and piscivorase II (PII), were isolated from Agkistrodon piscivorus piscivorus (eastern cotonmouth moccasin) venom using gel filtration on Bio-Gel P100 and ion-exchange chromatography on CM-Sepharose. The molecular welghts of two proteases were approximately 23400 and 29000. Their isoelectric points 6.6 and 8.5, respectively. The partial amino acid sequences of PI were characterized by tryptic digestion. PI readily cleaves the A${\alpha}$-and B${\beta}$-chaln of fibronogen, but PII rapidly cleaves A${\alpha}$-chain and more slowly the B${\beta}$-chain, They were activated by Ca$\^$2+/, Mg$\^$2+/ and Ba$\^$2+/, but inhibited by Zn$\^$2+/, Cu$\^$2+/ and Mn$\^$2+/. Two enzymes were also inhibited by cysten, ${\beta}$-mercapto -ethanol, and by metal chelators such as EDTA and EGTA, but not by benzamidine, PMSF, soybean trypsin inhibitor and aprotinin. They did not act like thrombin, plasmin and kallikrein, using specific chromogenllc substrates. Two protease did not induce platelet aggregation. PI showed low hemorrhagic activity at dosage of 50 $\mu\textrm{g}$.

Biochemical Characters of Polygalacturonase Produced by Botryosphaeria dothidea (사과 겹무늬썩음병균(Botryosphaeria dothidea)이 생산하는 Polygalacturonase의 생화학적 특성)

  • The polygalacturonase (PG) production in rotten apples by Botryosphaeria dothidea was purified by using gel filtration and ion exchange column chromatography, and the biochemical characters of PG were investigated. The purified PG appeared as a single band on sodium dodecyl sulfate-polyacrylamide gel electrophoresis (SDS-PAGE) with approximate molecular weight of 49 kilodalton (kDa). The molecular weight was equal to the native molecular weight estimated by gel filtration. The Km and Vmax values of PG were 0.51 mg/ml and 90.9 $\mu$M/min/ml, respectively. Optimum pH was 4.0~5.0, and the PG activity was stable from pH 5.0~10.0. Optimum temperature of the enzyme activity was 4$0^{\circ}C$. The PG activity was relatively stable at 2$0^{\circ}C$, but it was reduced 45% at 4$0^{\circ}C$ and completely inactivated at 8$0^{\circ}C$. The PG activity was considerably inhibited by Cu2+, Zn2+, SDS and EDTA, whereas it was not effected by Ca2+, K+, Mg2+ or Na+ ions.

Purification and Biochemical Characterization of a Serine Protease with Fibrinolytic Activity from Maggots of Mimela splendems

  • Maggot fibrolase (MsMg-1) was purified from the maggots of Mimela splendems using ammonium sulfate fractionation, DEAE Affi-gel affinity chromatography. This protease had a molecular weight of 85 kDa as determined by SDS-polyacrylarnide gel electrophoresis under reducing conditions. It showed strong proteolytic and fibrinolytic activities. The purified enzyme was strongly inhibited by phenylmethanesulfonyl fluoride, Mn/sup 2+/, and Zn/sup 2+/ but it was not by EDTA, EGT, Mg/sup 2+/, Ca/sup 2+/, and Li/sup 2+/ ions. In these experimental results, we have speculated that MsMg-1 is a serine protease with a strong fibrinolytic activity.

Cloning and Characterization of the Major Extracellular Neutral Protease (NprM) from Bacillus megaterium ATCC 14945

  • A gene, nprM, from Bacillus megaterium ATCC 14945 was obtained by PCR using primers synthesized based on two nprM sequences from two different strains, and cloned into Escherichia coli. The gene nprM encoded an extracellular neutral protease, and the molecular mass of the expressed enzyme was estimated to be approximately 36kDa on a denaturating gel. The enzyme was activated by $Ca^{2+}$, and the optimum concentration of $Ca^{2+}$ was 5 mM. The enzyme was inhibited by EDTA but not by PMSF. The optimal pH and temperature of the cloned enzyme were $50^{\circ}C and pH 7.5-8.0, respectively, and were similar to those of the enzyme from the gene gonor cell. The cloned NprM caused internal cleavage of the native endoglucanase of B. subtilis BSE616 as a model foregin protein, and resulted in a small truncated but still active endoglucanase.

A Highly Selective and Sensitive Calcium(II)-Selective PVC Membrane Based on Dimethyl 1-(4-Nitrobenzoyl)-8-oxo-2,8-dihydro-1H-pyrazolo[5,1-a]isoindole-2,3-dicarboxylate as a Novel Ionophore

  • Dimethyl 1-(4-nitrobenzoyl)-8-oxo-2,8-dihydro-1H-pyrazolo[5,1-a]isoindole-2,3-dicarboxylate has been used as an ionophore and o-nitrophenyloctyl ether as a plasticizer in order to develop a poly(vinyl chloride)-based membrane electrode for calcium ion detection. The sensors exhibit significantly enhanced response towards calcium(II) ions over the concentration range $8.0{\times}10^{-7}\;1.0{\times}10^{-1}$ M at pH 3.0-11 with a lower detection limit of $5.0 {\times}10^{-7}$ M. The sensors display Nernstian slope of 29.5 ${\pm}$ 0.5 mV per decade for Ca(II) ions. Effects of plasticizers, lipophilic salts and various foreign common ions are tested. It has a fast response time within 10 s over the entire concentration range and can be used for at least 2 months without any divergence in potentials. The proposed electrode revealed good selectivity and response for $Ca^{2+}$ over a wide variety of other metal ions. The selectivity of the sensor is comparable with those reported for other such electrodes. The proposed sensor was successfully applied as an indicator electrode for the potentiometric titration of a Ca(II) solution, with EDTA.

Immobilization and Characterization of Inulinase from Bacillus sphaericus 188-1 (Bacillus sphaericus 188-1이 생성하는 Inulinase의 고정화의 특성)

  • 고정화 inulinase를 이용하여 inulin으로부터 fruc-tose 시럽을 연속적으로 생산하기 위하여 Bacillus sphaericus 188-1이 생성하는 inulinase를 부분 정제한후 5시간 NaIO$_4$로 산화시킨 cellulose에 고정화시킨 다음 이들의 성질을 조사하였다. 산화 cellulose에 고정화시킨 inulinase의 활성은 g당 47 Unit 이었고 고정화수율과 활성수율은 각각 41%와 39%이었다. 고정화시킨 inulinase의 작용 최적온도와 pH는 각각 5$0^{\circ}C$, pH 9.0이었고 5$0^{\circ}C$, pH 8.0~10.0에서 비교적 안정 하였다 고정화시킨 inulinas의 활성은 K+, Ca2+, Mn2+과 Hg2+에 의하여 활성화 되었으며 EDTA(10mM)에 의하여 심하게 저해되었다.

Studies on Degradation of Nucleic acid and Related Compounds by Microbial Enzymes (미생물 효소에 의한 핵산 및 그의 관련물질의 분해에 관한 연구)

  • As a series of studies on the nucleic acids and their related substances 210 samples were collected from 76 places such as farm soil, compost of heap, nuruk and meju to obtain microbial strains which produce 5'-phosphodiesterase. From these samples total of 758 strains were isolated by the use of dilution pour plate method. For all isolated strains primary screening of the productivity of RNA depolymerase was performed and useful strains with regard to 5'-phosphodiesterase productivities were identified. For these useful strains optimum condition, the effect of various compounds on the activity of 5'-phosphodiesterase, and the optimum condition for enzyme reaction were discussed. The quantitative of 5'-mononucleotides produced by the action of 5'-phosphodiesterase was performed using anion-exchange column chromatography and their identified was done by paper chromatography, thinlayer chromatography, ultra violet spectrophotometry, and characteristic color reaction using carbazole and schiff's reagent. (1) Penicillium citreo-viride PO 2-11 and Streptomyces aureus SOA 4-21 from soil were identified as a potent 5'-phosphodiesterase producing strains. (2) Optimum culture conditions for Penicillium citreo-viride PO 2-11 strain isolated were found to be pH 5.0 and $30^{\circ}C$, and the optimum conditions for enzyme action of 5'-phosphodiesterase were pH 4.2 and $60^{\circ}C$. Best carbon source for the production of 5'-phosphodiesterase was found to be sucrose and ammonium nitrate for nitrogen source. Addition of 0.01% corn steep liquor or yeast extract exhibited 20% increase in the amount of 5'-phosphodiesterase production compared to the control. 5'-phosphodiesterase produced by this strain was activated by $Mg^{++},\;Ca^{++},\;Zn^{++},\;Mn^{++}$ and was inhibited by EDTA, citrate, $Cu^{++},\;CO^{++}$. 5'-phosphodiesterase produced 5'-mononucleotide from RNA at a rate of 65.81%, and among the 5'-mononucleotides accumulated 5'-GMP only was found to have flavorous and the strain was also found lack of 5'-AMP deaminase. Productivity of flavorous 5'-GMP was found to be 186.7mg per gram of RNA. (3) Optimum culture canditions for the isolated Streptomyces aureus SOA 4-21 strain were pH 7.0 and $28^{\circ}C$, and the optimum conditions for the action of 5'-phosphodiesterase were pH 7.3 and $50^{\circ}C$. The best carbon source for 5'-phosphodiesterase production was found to be glucose and that of nitrogen was asparagine. Addition of 0.01% yeast extract exhibited increased productivity of 5'-phosphodiesterase by 40% compared to the non-added control. 5'-phosphodiesterase produced by this strain was activated by $Ca^{++},\;Zn^{++},\;Mn^{++}$ and was inhibited by citrate, EDTA, $Cu^{++}$. It was also found that the strain produce 5'-AMP deaminase in addition to 5'-phosphodiesterase. For this reason although decomposition rate was 63.58% the accumulation of 5'-AMP, 5'-CMP, 5'-GMP and 5'-UMP occurred by the breakdown of RNA. In the course of these reaction 5'-AMP deaminase converted 60% of 5'-AMP thus produced into 5'-IMP and flavorous 5'-mono nucleotide production was significantly increased by this strain over the above mentioned one. Production rates were found to be 171.8mg per grain of RNA for 5'-IMP and 148.2mg per gram of RNA for 5'-GMP, respectively.

A Study on the Remediation of Lead Contaminated Soil in a Clay Shooting Range with Soil Washing (토양세척법에 의한 클레이사격장 납 오염토양의 정화에 관한 연구)

  • For an efficient remediation of Pb-contaminated soil (S-1) in a clay shooting range, a soil washing test was performed with mineral acid, organic acid, chelating agent, and chloride. The Pb extraction efficiency of extractant (0.1 M) used in the washing test showed the order of HCl > $Na_2$-EDTA > NTA > DTPA > citric acid > malic acid > succinic acid > acetic acid > $CaCl_2$ > $MgCl_2$, for S-1 soil. As compared to initial Pb concentration, extraction efficiency by the concentration of extractant was 93.35%, 80.80%, 73.92%, and 24.57% in S-1 soil for HCl (0.5 M, pH 1.10), $Na_2$-EDTA (0.01 M, pH 3.99), citric acid (0.5 M, pH 1.27), and $MgCl_2$ (0.1 M, pH 8.82), respectively. S-1 soil had 56.83% of residue form and 43.17% of non-residue form (18.04% of exchangeable form), respectively. Although the concentrations of these fractions sharply decreased after HCl washing, since the exchangeable forms with relatively large mobility are still distributed as high as 18.78% (to Pb total content in residual soils) in S-1 soil, it is necessary to devise a proper management plan for residual soils after soil washing application.

Effects of Calcium Antagonists on Superoxide Generation, NADPH Oxidase Activity and Phagocytic Activity in Activated Neutrophils (칼슘 길항제가 활성화된 호중구에서의 $O_{\overline{2}}$의 생성, NADPH oxidase활성도 및 탐식작용에 미치는 영향)

  • NADPH oxidase dependent superoxide generation and phagocytosis in neutrophils stimulated with opsonized zymosan or heat aggregated IgG were coincided with the process of calcium uptake. The responses in activated neutrophils were enhanced with increasing concentrations of extracellular calcium and these effects were significantly inhibited by calcium chelators, EGTA and EDTA. The superoxide generation in activated neutrophils was reduced by dantrolene and chlorpromazine. Calcium antagonists, bepredil, diltiazem, verapamil, nifedipine and nimodipine effectively inhibited the calcium uptake, superoxide generation and phagocytosis in activated neutrophils, and NADPH oxidase activity was also inhibited. The results suggest that calcium antagonists may inhibit the superoxide generation and phagocytosis in activted neurtophils by the inhibition of calcium influx and by the action on intracellular redistribution of calcium and NADPH oxidase system.

Purification and Characterization of Inulinase from Penicillium sp. (Penicillium sp. 유래 Inulinase의 정제 및 특성)

  • An extracellular inulinase from Penicillium sp. which isolated from soil sample was purified to a single protein th-rough ammonium sulfate fractionation, DEAE-Sephacel ion exchange chromatography and Toyopearl HW 65 F gel filtration. The temperature and pH for the enzyme reaction were around 6$0^{\circ}C$ and 4.0, respectively. The enzyme was stable at 3$0^{\circ}C$-5$0^{\circ}C$ and in the pH range of 4 to 5. $CuCl_2$, $HgCl_2$ and EDTA inhibited the enzyme activity strongly. By contrast, $MnCl_2$ and $CaCl_2$ activated the enzyme activity. The molecular weight of the purified enzyme was esti-mated to be 77,000 dalton by SDS-polyacrylamide gel electrophoresis. The Km values of the enzyme for inulin were calculated to be $2.2\times10^{-3}$M. TLC analysis suggested that purified enzyme is exo-type inulinase. The NH2-terminal amino acid sequences of the purified enzyme was determined to be $NH_2$-X-Glu-Ser-Tyr-Thr-Glu-Lys/Leu-Tyr-Arg-Pro.
